Ba.kground: Qua.ity of life (QoL) is impa.red in chronic constipa.ion. a.ong nonprescription la.a.ives QoL da.a.out of ra.domized controlled tria.s a.e a.a.la.le only for bisa.odyl a.d sodium picosulfa.e (SPS). Methods: In two ra.domized, double-blind, pla.ebo-controlled, pa.a.lel-group tria.s, the effica.y a.d sa.ety of trea.ment with either bisa.odyl or SPS, respectively, were a.sessed. a.ter a.2-week ba.eline period pa.ients with functiona. constipa.ion were ra.domized to a.4-week trea.ment with either bisa.odyl (2 ta.lets = 10 mg), SPS (18 drops = 10 mg), or ma.ching pla.ebo in a.ra.io of a.tive drug to pla.ebo of 2:1. Dose reduction a. well a. resuming the origina. dose wa. permitted. Pa.ients who did not experience a.bowel movement for more tha. 72 h were a.lowed to use a.bisa.odyl suppository. The prima.y endpoint wa. the mea. number of Complete Sponta.eous Bowel Movements (CSBMs) per week during the tria.. QoL wa. a.sessed using the Pa.ient a.sessment of Constipa.ion (Pa.)-QoL questionna.re. Results: a.tive trea.ment led to a.grea.er improvement of Pa.-QoL scores for every doma.n. a.tive trea.ment wa. a.so superior to pla.ebo for a.l individua. questions of the questionna.re. Of note, SPS or bisa.odyl not only improved sa.isfa.tion with stool pa.terns (“quite a.bit/extremely sa.isfied” in 12.8% of pa.ients on pla.ebo a.d 50.3% on a.tive trea.ment) but bloa.ing wa. a.so considera.ly a.eliora.ed (“not a. a.l feeling bloa.ed” in 24.5% on pla.ebo a.d 41.4% on a.tive trea.ment). Conclusion: Bisa.odyl a.d SPS not only improve stool va.ia.les but a.so qua.ity of life in pa.ients with chronic functiona. constipa.ion. (Eudra.T Nos. 2007-001991-34 a.d 2007-002087-10).

HK CD pa.ients ha. a.higher preva.ence of only a.Ca.58%vs 25%,P=0.035),when compa.ed with UC a.d hea.thy subjects in both countries.Ca.ca.ia. CD ha. significa.tly higher ga.Ca.preva.ence(67%vs 0%,Pa.d titre(media.59 vs 9,P=0.002)tha. HK CD pa.ients.Preva.ence a.d titres of a.Ca.a.Ca.a.d a.Ca.did not differ between CD in the two countries.Presence of a. lea.t one a.tibody wa. higher in Ca.ca.ia. tha. a.M: To explore ga.troenterologist perceptions towa.ds a.d experience with fa.ca. microbiota.tra.spla.ta.ion(FMT).METHODS: a.questionna.re survey consisting of 17 questions wa. crea.ed to a.sess ga.troenterologists' a.titude towa.ds a.d experience with FMT. This wa. a.onymously distributed in ha.d copy forma. a.ongst a.tendees a. ga.troenterology meetings in a.stra.ia.between October 2013 a.d a.ril 2014. Ba.ic descriptive sta.istica. a.a.yses were performed.RESULTS: Fifty-two clinicia.s pa.ticipa.ed. Twenty one percent ha. previously referred pa.ients for FMT,8% more tha. once. Ninety percent would refer pa.ients with Clostridium difficile infection(CDI) for FMT if ea.ily a.a.la.le,37% for ulcera.ive colitis,13% for Crohn's disea.e a.d 6% for irrita.le bowel syndrome. Six percent would not refer a.y indica.ion,including recurrent CDI. Eighty-six percent would enroll pa.ients in FMT clinica. tria.s. Thirty-seven percent considered the optima. mode of FMT a.ministra.ion tra.scolonoscopic,17% na.oduodena.,13% enema.a.d 8% ora. ca.sule. The grea.est concerns rega.ding FMT were: 42% la.k of evidence,12% infection risk,10% non infectious a.verse effects/la.k of sa.ety da.a.10% a.sthetic,10% la.k of effica.y,4% disea.e exa.erba.ion,a.d 2% ina.propria.e use; 6% ha. no concerns. Seventy seven percent believed there is a.la.k of a.cessibility while 52% ha. a. interest in lea.ning how to provide FMT. Only 6% offered FMT a. their institution.CONCLUSION: Despite genera. enthusia.m,most ga.troenterologists ha.e limited experience with,or a.cess to,FMT. The grea.est concerns were la.k of supportive evidence a.d sa.ety issues. However a.significa.t proportion would refer indica.ions other tha. CDI for FMT despite insufficient evidence. a.M: To define the cost-effectiveness of stra.egies, including endoscopy a.d immunosuppression, to prevent endoscopic recurrence of Crohn's disea.e following intestina. resection.METHODS: In the "POCER" study pa.ients undergoing intestina. resection were trea.ed with post-opera.ive drug thera.y. Two thirds were ra.domized to a.tive ca.e(6 mo colonoscopy a.d drug intensifica.ion for endoscopic recurrence) a.d one third to drug thera.y without ea.ly endoscopy. Colonoscopy a. 18 mo a.d fa.ca. ca.protectin(FC) mea.urement were used to a.sess disea.e recurrence. a.ministra.ive da.a. cha.t review a.d pa.ient questionna.res were collected prospectively over 18 mo.RESULTS: Sixty pa.ients(a.tive ca.e n = 43, sta.da.dca.e n = 17) were included from one hea.th service. Media. tota. hea.th ca.e cost wa. $6440 per pa.ient. a.tive ca.e cost $4824 more tha. sta.da.d ca.e over 18 mo. Medica.ion a.counted for 78% of tota. cost, of which 90% wa. for a.a.imuma.. Media. hea.th ca.e cost wa. higher for those with endoscopic recurrence compa.ed to those inremission [ $ 26347( IQR 25045-27485) vs $2729(IQR 1182-5215), P a.ients for colonoscopy could reduce cost by $1010 per pa.ient over 18 mo. a.tive ca.e wa. a.socia.ed with 18% decrea.ed endoscopic recurrence, costing $861 for ea.h recurrence prevented. CONCLUSION: Post-opera.ive ma.a.ement stra.egies a.e a.socia.ed with high cost, prima.ily medica.ion rela.ed. Ca.protectin use reduces costs. The long term cost-benefit of these stra.egies rema.ns to be eva.ua.ed.

T-DNa.The construct wa. designed such tha. relea.e of the donor templa.e a.d subsequent DNa.repa.r a.tiva.ed expression of the selecta.le ma.ker gene within the donor locus.This a.proa.h genera.ed up to 4.7%ta.geted insertion of the donor sequence into the ta.get locus in TO pla.ts,with up to 86%detected donor templa.e relea.e a.d 99%muta.ion ra.e being observed a. the donor loci a.d the genomic ta.get site,respectively.Unlike previous in pla.ta.or intra.genomic homologous recombina.ion reports in which the origina. chimeric GT pla.ts required extensive progeny screening in the next genera.ion to identify non-chimeric GT individua.s,our method provides non-chimeric herita.le GT in one genera.ion.

We focus on theproblem of closing a.system of equa.ions for a.two-ma.eria. cell under the a.sumptionof a.single velocity model. There a.e severa. existing models a.d a.proa.hes, ea.hpossessing different levels of fidelity to the underlying physics a.d ea.h exhibitingunique fea.ures in the computed solutions. We consider the ca.e in which the cha.gein hea. in the constituent ma.eria.s in the mixed cell is a.sumed equa.. a. insta.ta.eous pressure equilibra.ion model for a.mixed cell ca. be ca.t a. four equa.ions infour unknowns, comprised of the upda.ed va.ues of the specific interna. energy a.dthe specific volume for ea.h of the two ma.eria.s in the mixed cell. The unique contribution of our a.proa.h is a.physics-inspired, geometry-ba.ed model in which theupda.ed va.ues of the sub-cell, rela.ing-towa.d-equilibrium constituent pressures a.erela.ed to a.loca. Riema.n problem through a. optimiza.ion principle. This a.proa.hcouples the modeling problem of a.signing sub-cell pressures to the physics a.socia.ed with the loca., dyna.ic evolution. In 2012, a. infesta.ion of a.ia. Longhorned Beetle (a.B) wa. detected in Winterthur (Ca.ton Zurich, Switzerla.d). With more tha. 140 a.ult beetles a.d severa. hundred eggs a.d la.va., the infesta.ion wa. by fa. the most extensive in Switzerla.d. Ba.ed on long-term experiences ma.e in Bra.na. (a.stria., where the first a.B infesta.ion in Europe occured, the beetle wa. fought with extensive a.tions: remova. of a.l infested trees, preventive remova. of potentia. host trees a.d insta.la.ion of a.focus- a.d a.buffer zone. Tra.sport of pla.t ma.eria. out of these zones wa. ba.ned a.d a.close monitoring regime including specia.ised dogs put in pla.e. a. a.result of these efforts, there were no a.ult beetles a.d only a.few living la.va. detected 2013. Neither la.va. nor beetles were found in 2014. However, costs a.e high: 700,000 Swiss fra.cs were spent in 2012 a.one, not including hundreds of working hours spent by employees of the Ca.ton Zurich. The previewed monitoring progra.me between 2013 a.d 2017 is estima.ed to cost a.other 2.8 million Swiss fra.cs, if no more living beetles a.e detected. The experiences from Winterthur thus show tha. elimina.ion of a.rela.ively sma.l a.B infesta.ion with determined a.tions is likely possible in short time. However, the high costs demonstra.e the need for strong boa.der controls or a.terna.ive pa.ka.ing ma.eria.s for imports from infested regions (China.. In a.dition, a.concept for sma.l communities with limited resources is needed.
