Growth inhibition of colon cancer cells by compounds affecting AMPK activity

AIM:To determine if other molecules reported to modulate AMP-dependent protein kinase(AMPK)activ-ity would have effects resembling those of metformin and phenformin on colon cancer cell proliferation and ***:Studies were performed with four hu-man colon cancer cell lines,Caco-2,HCT116,HT29 and *** compounds that were studied included A-769662,5-aminoimidazole-4-carboxamide-1-ribofu-ranoside,butyrate,(-)-epigallocatechin gallate(EGCG),KU-55933,quercetin,resveratrol and *** parameters that were measured were cell proliferation and viability,glucose uptake,lactate production and acidification of the incubation ***:Investigations with several molecules that have been reported to be associated with AMPK activa-tion(A-769662,5-aminoimidazole-4-carboxamide-1-b-D-ribofuranoside,EGCG,KU-55933,quercetin,resve-ratrol and salicylates)or AMPK inhibition(compound C)failed to reveal increased medium acidification and increased glucose uptake in colon cancer cells as previ-ously established with metformin and *** only exception was 5-aminosalicylic acid with which there were apparently lower glucose levels in the me-dium after incubation for 72 *** study in the absence of cells revealed that the effect was an artifact due to inhibition of the enzyme-linked glucose *** compounds were studied at concentrations that inhibited cell ***:It was concluded that treatment with several agents that can affect AMPK activity resulted in the inhibition of the proliferation of colon cancer cells under conditions in which glucose metabolism is not enhanced,in contrast to the effect of biguanides.

Target Oriented Synthesis and Mass Spectral Characterization of Curcumin-Phenformin Adduct： Potential Insights into the Role of this Conjugate as Anti-Diabetic and Anti-Cancer Agent

Recently microwave-induced chemical synthesis of curcumin-metformin adduct to enhance the efficacy of metformin in preventing the formation of Advanced Glycation End Products （AGEs） has been reported from authors＇ laboratory. The present studies describe microwave-induced chemical synthesis and mass spectral characterization of curcumin-phenformin adducts using LC-MS/MS. The mechanism of formation and its analytical data via Thin-Layer Chromatography （TLC） combined with MS/MS fragmentation revealed a major six membered ring adduct and a minor eight membered ring isomer. A facile chemical synthesis and identification of major and minor isomers presented in this study may offer novel therapeutic strategies for inhibiting AGEs as well as anti-cancer treatments.

BACKGROUND Suprasellar germinomas are rare intracranial tumors frequently associated with permanent endocrine *** present the clinical picture,treatment,and complications of suprasellar germinoma at pediatric age which,besides being lifethreatening,has lifelong endocrinological *** SUMMARY A 12-year-old female patient was presented having had intensive headaches for three weeks and visual disturbances for six *** ophthalmological examination revealed bilateral papilledema and a marked loss of *** brain magnetic resonance imaging(MRI)showed a suprasellar tumor,involving the infundibulum and the optic chiasm,extending to the third *** tests confirmed decreased levels of thyroxine,cortisol,gonadotropins,and insulin-like growth factor *** tumor reduction was performed,and immunohistopathology established the diagnosis of suprasellar *** of the spine and cerebrospinal fluid cytology confirmed the localized *** chemotherapy and radiotherapy were performed according to the SIOP CNS GCT II protocol.A post-treatment MRI showed no residual tumor,but pituitary function had not *** and a half years after the end of the treatment,the patient is in a complete remission,requiring hormonal replacement therapy,continuous education,and psychological *** This complex case highlights the importance of timely diagnosis,a multidisciplinary approach,and close follow-up in children with suprasellar germinomas.

In Quebec,Canada,the public healthcare system offers free medical ***,patients with spinal pain often encounter long waiting times for specialist appointments and limited physiotherapy *** contrast,private clinics provide expedited care but are relatively scarce and entail out-of-pocket *** a patient with pain caused by a spinal disorder meets a pain medicine specialist,spinal intervention is quickly performed when indicated,and patients are provided lifestyle *** epidural steroid injections are frequently administered to patients with radicular pain,and steroid injections are administered on a facet joint to control low back or neck ***,medial branch blocks are performed prior to ***’s universal healthcare system ensures accessibility at controlled *** emphasizes physical activity and provides free physical therapy ***,certain interventions,such as transforaminal and interlaminar epidural injections,are not routinely used in France owing to limited therapeutic efficacy and safety *** underutilization may be a potential cause of chronic pain for many *** examining the differences,strengths,and weaknesses of these two systems,valuable insights can be gained for the enhancement of global spinal pain management strategies,ultimately leading to improved patient outcomes and satisfaction.

Facile Synthetic Design and Characterization of Curcumin-Metformin Adduct： Potential Insights into the Role of This Conjugate in Diseases of Aging

Recently, the anti-glycation and anticancer properties of curcumin longa and okra seed extract were studied alone and also in combination with the well-established drug metformin. The combined effect of curcumin with metformin and metformin with okra seed extract was found to be highly efficacious in inhibiting Advanced Glycation End-products （AGEs）. In order to understand the mechanistic implications of curcumin combined with metformin and its enhanced anti-glycation activity, a Curcumin-Metformin Adduct was chemically synthesized. This adduct was fully characterized by thin-layer chromatography, Nano Drop spectrophotometry and electrospray-ionization mass spectrometry. The adduct may be helpful not only in elucidating the mechanism of anti-glycation and anti-cancer activities but also in studying the role of curcumin in binding of AI3-oligomers and disaggregating fibrillar formation in Alzheimer＇s disease.

Acute pancreatitis as a complication of acute COVID-19 in kidney transplant recipients

BACKGROUND Acute pancreatitis is a rare extrapulmonary manifestation of coronavirus disease 2019(COVID-19)but its full correlation with COVID-19 infection remains *** To identify acute pancreatitis’occurrence,clinical presentation and outcomes in a cohort of kidney transplant recipients with acute *** A retrospective observational single-centre cohort study from a transplant centre in Croatia for all adult renal transplant recipients with a functioning kidney allograft between March 2020 and August 2022 to record cases of acute pancreatitis during acute *** were obtained from hospital electronic medical *** acute respiratory syndrome coronavirus 2(SARS-CoV-2)infection was proven by a positive SARS-CoV-2 real-time reverse transcriptase-polymerase chain reaction on the nasopharyngeal *** Four hundred and eight out of 1432(28.49%)patients who received a renal allograft developed COVID-19 *** analyzed cohort included 321 patients(57%males).One hundred and fifty patients(46.7%)received at least one dose of the anti-SARS-CoV-2 vaccine before the *** hundred twenty-five(39.1%)patients required hospitalization,141(44.1%)developed pneumonia and four patients(1.3%)required mechanical *** included immunosuppression modification in 233 patients(77.1%)and remdesivir in 53 patients(16.6%),besides the other supportive *** the study cohort,only one transplant recipient(0.3%)developed acute pancreatitis during acute COVID-19,presenting with abdominal pain and significantly elevated pancreatic *** survived without complications with a stable kidney allograft *** Although rare,acute pancreatitis may complicate the course of acute COVID-19 in kidney transplant *** mechanism of injury to the pancreas and its correlation with the severity of the COVID-19 infection in kidney transplant recipients warrants further research.

Impact of Social Media Challenges on Pediatric Single-Use Detergent Sacs and Diphenhydramine Ingestions Reported to United States Poison Control Centers

Background : Social media platforms are popular among children and often feature challenges that become viral. Notably, the Tide Pod ® and Benadryl ® challenges encouraged viewers to ingest these substances for their visual appeal and hallucinogenic effects, respectively. This study aimed to assess the clinical impact and outcomes of single-use detergent sacs (SUDS) and diphenhydramine challenges on pediatric ingestions reported to United States (U.S.) Poison Control Centers (PCCs). Methods : We conducted a retrospective review of pediatric exposures reported to U.S. PCCs using data from the National Poison Data System (NPDS). The study included intentional single-substance ingestions of both brand-name and generic forms of SUDS and diphenhydramine among children ≤ 19 years. We compared the number of calls, clinical effects, disposition, and management strategies for SUDS (pre: 01/01/17 to 12/31/17 vs. post: 01/01/18 to 12/31/18) and diphenhydramine (pre: 08/01/19 to 07/31/20 vs. post: 08/01/20 to 07/31/21) ingestions 12 months before and after the introduction of the respective social media challenges. Differences in proportions were compared using the Chi-square test. Results : During the study period, 469 ingestions of SUDS and 5,702 ingestions of diphenhydramine were reported. Post-challenge periods saw an increase in both SUDS (pre: 82 vs. post: 387; 372% increase) and diphenhydramine ingestions (pre: 2,672 vs. post: 3,030; 13% increase). While there were no significant changes in moderate or major clinical outcomes, hospitalizations increased post-challenge for both SUDS [pre: 4 (4.9%) vs. post: 33 (8.5%); p = 0.25] and diphenhydramine [pre: n = 904 (33.8%) vs. post: 1,190 (39.3%); p < 0.001]. Conclusion : Pediatric ingestions reported to U.S. PCCs and hospitalizations increased coinciding with the introduction of Tide Pod ® and Benadryl ® challenges.
