Is there a correlation between the outcome of transurethral resection of prostate and preoperative degree of bladder outlet obstruction？

To compare the impact of transurethral resection of prostate （TURP） on symptom scores and maximal flow rates （Qmax） in patients with equivocal bladder outlet obstruction （BOO） and definite BOO and to assess the relationship between the surgical outcomes and degree of preoperative BOO, we prospectively evaluated men with lower urinary tract symptoms and bladder outlet obstruction index （BOO1） greater than 20, who were refractory to conventional medical treatment and underwent TURP. Urodynamic evaluation, International Prostate Symptom Score （IPSS）, uroflowmetry, post-void residual volume （PVR） check and transrectal ultrasound were performed. 20〈B001〈40 was defined as equivocal BOO and BOO1 〉~40 as definite BOO. Changes of IPSS, Qmax, PVR and correlation analysis was performed between the degree of improvement of Qmax, subdomains of I PSS and BOO1. Fifty-four patients showed equivocal BOO and 80 patients showed definite BOO. Preoperatively equivocal BOO group and definite BOO group showed significant differences in maximal bladder capacity and prevalence of detrusor overactivity, whereas no difference was noted in prostate volume. Postoperatively both groups showed improvements in Qmax, obstructive （IPSSO） and irritative （IPSSI） subdomain of IPSS, but the degree of improvement in Qmax and IPSSI subdomain was statistically significantly greater in definite BOO group. The degree of improvement of Qmax and IPSSI showed weak correlation with preoperative BOO1. As a weak correlation was identified between preoperative degree of BOO and outcome of TURP, other factors other than BOO1 such as severity of patients＇ symptoms should be considered in deciding treatment modality.

nfluence of paradoxical sleep deprivation and sleep recovery on testosterone level in rats of different ages

This study was performed to assess serum testosterone alterations induced by paradoxical sleep deprivation （PSD） and to verify their attenuation during sleep recovery （SR） based on different durations and ages. Wistar male rats aged 12 weeks for the younger group and 20 weeks for the elder group were randomly distributed into one of the following groups： a control group （cage and platform）, 3-day SD, 5-day SD, 7-day SD, 1-day SR, 3-day SR and 5-day SR groups. For PSD, the modified multiple platform method was used to specifically limit rapid eye movement （REM） sleep. Differences in the testosterone and luteinizing hormone levels between the younger group and the elder group according to duration of PSD and SR recovery were analysed. Testosterone continued to fall during the sleep deprivation period in a time-dependent manner in both the younger （P=-0.001, correlation coefficient r=-0.651） and elder groups （P=0.001, correlation coefficient r=-0.840）. The elder group showed a significantly lower level of testosterone compared with the younger group after PSD. Upon SR after 3 days of PSD, the testosterone level continued to rise for 5 days after sleep recovery in the younger group （P=0.013）, whereas testosterone concentrations failed to recover until day 5 in the elder group. PSD caused a more detrimental effect on serum testosterone in the elder group compared to the younger group with respect to decreases in luteinizing hormone （LH） levels. The replenishment of serum testosterone level was prohibited in the elder group suggesting that the effects of SD/SR may be age-dependent. The mechanism by which SD affects serum testosterone and how age may modify the process are still unclear.

The effect of diet-induced insulin resistance on DNA methylation of the androgen receptor promoter in the penile cavernosal smooth muscle of mice

Population studies have suggested an association between diabetes and the symptoms of testosterone deficiency. Recently, the expression of the androgen receptor （AR） has been shown to be decreased in diabetic patients. Furthermore, diabetes has been shown to induce global methylation. In this study, we used an animal model to investigate whether diabetes results in increased methylation of the AR promoter and whether these changes are associated with the decreased expression of AR in penile cavernosal smooth muscle tissue. Twenty C57BL/6J mice were divided into two groups, receiving either high- （mature diabetic） or low- （mature control） caloric meals for 14 weeks. Another 10 mice were killed at 1 week （young control）. Animals in the mature diabetic group showed decreased testosterone levels, although this was not statistically significant. In both control groups, no significant methylation was observed in the AR promoter region CpG island （-85 to ＋339）. In the mature diabetic group, significant methylation was observed at ＋ 185 and ＋200 of the AR promoter. These changes were associated with increased homeostatic model assessment for insulin resistance （HOMA-IR） and decreased corpus cavernosal tissue mass and expression of AR mRNA and protein. We conclude that in these animals, insulin resistance increased the methylation of the GC-rich regions of the AR promoter, leading to decreased AR expression.

Extraction of rare earth elements from neodymium(NdFeB) magnet scrap using magnesium halides

The demand for neodymium（NdFeB） permanent magnets for electric vehicles and eco-friendly generators is ***,NdFeB magnets contain rare earth elements（REEs）,which are limited in *** this study,we performed an exchange reaction between magnesium halides（fluoride and chloride） and waste NdFeB scrap and then compared the characteristics of the extracted halides *** compositions of the ternary Mg fluoride（LiF:NaF:MgF2=50:40:10 in mole ratio） and chloride（LiCl:NaCl:MgCl2=10:50:40 in mole ratio） salts were thermodynamically determined for achieving low eutectic *** reactions between the NdFeB scrap powder（1-2 mm） and Mg halide salts were carried out at 1073 and 873 K for the fluoride and chloride systems,respectively,in an argon *** the reaction,we separated Nd halide from the residual salt and evaluated the Ndextraction *** phase formation of the salt was analyzed using X-ray diffraction（XRD）,and the extraction rate of Nd was analyzed using inductively coupled plasma optical emission spectroscopy（ICPOES）.Nd was extracted in the form of Nd halide（NdF3or NdCl3）,and the extraction rates in the fluoride and chloride systems are 98.64% and 84.59%,***,the fluoride system is more effective than the chloride system for Nd *** study provides a comprehensive comparative analysis of the effectiveness of fluo ride and chlo ride systems in extracting REEs from NdFeB magnet *** study findings can be used to develop an effective method for recycling magnet scraps.

Polystyrene nanoparticles pose significant toxicological risks to aquatic ecosystems, yet their impact on zebrafish(Danio rerio) embryonic development, particularly erythropoiesis, remains underexplored. This study used single-cell RNA sequencing to comprehensively evaluate the effects of polystyrene nanoparticle exposure on erythropoiesis in zebrafish embryos. In vivo validation experiments corroborated the transcriptomic findings,revealing that polystyrene nanoparticle exposure disrupted erythrocyte differentiation, as evidenced by the decrease in mature erythrocytes and concomitant increase in immature erythrocytes. Additionally, impaired heme synthesis further contributed to the diminished erythrocyte *** findings underscore the toxic effects of polystyrene nanoparticles on hematopoietic processes, highlighting their potential to compromise organismal health in aquatic environments.

干旱-半干旱地区地下水重金属污染受到广泛关注,但是地下水中重金属分布特征、污染来源及潜在人体健康风险尚未完全清楚.对采集的46组地下水进行测试分析,运用水文地球化学模拟和多元统计分析的方法,揭示大同盆地地下水中11种重金属(As、B、Pb、Sb、Tl、Mn、Ba、Cd、Co、Cr和Al)的污染特征和形态分布,结合绝对主成分-多元线性回归模型和健康风险评估模型,厘定地下水中重金属的来源和健康风险水平.结果表明:①地下水中As、B、Pb、Sb和Tl超过地下水质量标准Ⅲ类水标准限值,属于重度污染,Mn为轻度污染,重金属平均浓度总体呈现中部>南部>东部>西部的分布规律;②As、B、Pb、Sb和Tl主要存在形态分别为HAsO42(−91.71%)、H3BO(397.61%)、PbCO(388.08%)、Sb(oh)6(−99.81%)和Tl(+94.84%),pH是影响重金属形态分布的关键因素之一;③重金属污染来源主要为土壤迁移源(Co、Cd和Cr)、工业来源(Al、Pb、Sb和B)、矿区来源(Tl和Mn)及自然来源(Ba和As),其中工业来源占比最大(34%),其次为自然来源(29%)、矿区来源(20%)和土壤迁移来源(18%);④重金属特定来源健康风险评价结果表明,工业活动和自然来源分别是成人(54.68%)和儿童(67.51%)致癌风险的主要来源,As的致癌风险最高.矿区活动是非致癌风险的主要贡献者(成人贡献率56.69%,儿童贡献率56.97%),Tl和As的非致癌风险较高.研究成果对于指导干旱-半干旱地区水环境重金属污染防治具有积极的意义.

Identification of differentially expressed miRNAs associated with chronic kidney disease-mineral bone disorder

The purpose of this study is to characterize a meta-signature of differentially expressed mRNA in chronic kidney disease （CKD） to predict putative microRNA （miRNA） in CKD-mineral bone disorder （CKD- MBD） and confirm the changes in these genes and miRNA expression under uremic conditions by using a cell culture system. PubMed searches using MeSH terms and keywords related to CKD, uremia, and mRNA arrays were conducted. Through a computational analysis, a meta-signature that characterizes the significant intersection of differentially expressed mRNA and expected miRNAs associated with CKD-MBD was determined. Additionally, changes in gene and miRNA expressions under uremic conditions were confirmed with human Saos-2 osteoblast-like cells. A statistically significant mRNA meta-signature of upregulated and downregulated mRNA levels was identified. Furthermore, miRNA expression profiles were inferred, and computational anaIyses were performed with the imputed mieroRNA regulation based on weighted ranked expression and putative microRNA targets （IMRE） method to identify miRNAs associated with CKD occurrence. TLR4 and miR-146b levels were significantly associated with CKD-MBD. TLR4 levels were significantly downregulated, whereas pri- miR-146b and miR-146b were upregulated in the presence of uremic toxins in human Saos-2 osteoblast-like cells. Differentially expressed miRNAs associated with CKD-MBD were identified through a computational analysis, and changes in gene and miRNA expressions were confirmed with an in vitro cell culture system.

Oxidative hair dyes containingρ-phenylenediamine(PPD)are reported to induce an allergic reaction by promoting oxidative stress when absorbed through the *** the associated risk,these hair dyes remain popular owing to their convenience and sharpness of *** makes it important to minimize the cytotoxicity and oxidative stress induced by PPD-containing hair *** cava extract has been evaluated in different studies for its protective effects against external stress in fibroblasts and *** study was aimed at using in-vitro and in-vivo models to investigate the extract’s effects on cytotoxicity of and oxidative stress induced by PPD-containing hair *** of CIEL*a*b*Color space was first used to determine the range of *** extract that would not interfere with the coloring ability of the dye upon ***,the set ranges of *** extract(5% and 7%)were added to the hair dye and their toxicity assessed by evaluating the viability of fibroblasts and *** effects on developmental phenotypes and induction of oxidative stress by hair dye were evaluated and compared with those of hair dyes containing different contents of *** extract using an in-vivo zebrafish *** results showed that *** extract in hair dye could significantly decrease the cytotoxicity and levels of oxidative stress caused by hair dyes containing PPD in both in-vitro and in-vivo *** results suggest that the addition of 7% *** extract to 250μg/mL hair dye does not interfere with the coloring ability of the dye while showing significant protective eff ects against the hair *** study proposes that the use of *** extract as an adduct to hair dyes containing PPD reduces the cytotoxicity and oxidative stress induced by these hair dyes.

Application of cationic liposomes for delivery of nucleic acids

Cationic liposomes have been studied as a means to enhance thestability of nucleic acid therapeutics in the bloodstream and improve their cellular *** nucleic acid therapeutics, siRNA and plasmid DNA have been extensively tested fordelivery using cationic liposomes. This review discusses recent progress in the applicationof cationic liposomes for the delivery of nucleic acid therapeutics.
