Hearing loss and deafness,as a worldwide disability disease,have been troubling human ***,the auditory organ of the inner ear is highly heterogeneous and has a very limited number of cells,which are largely uncharacterized in ***,with the development and utilization of single-cell RNA sequencing(scRNA-seq),researchers have been able to unveil the complex and sophisticated biological mechanisms of various types of cells in the auditory organ at the single-cell level and address the challenges of cellular heterogeneity that are not resolved through by conventional bulk RNA sequencing(bulk RNAseq).Herein,we reviewed the application of scRNA-seq technology in auditory research,with the aim of providing a reference for the development of auditory organs,the pathogenesis of hearing loss,and regenerative *** about spatial transcriptomic scRNA-seq,single-cell based genome,and Live-seq technology will also be discussed.

Preclinical evaluation of cyclophosphamide and fludarabine combined with CD19 CAR-T in the treatment of B-cell hematologic malignancies in vivo

Background:Chimeric antigen receptor T(CAR-T)cell therapy has achieved marked therapeutic success in ameliorating hematological ***,there is an extant void in the clinical guidelines concerning the most effective chemotherapy regimen prior to chimeric antigen receptor T(CAR-T)cell therapy,as well as the optimal timing for CAR-T cell infusion *** and Methods:We employed cell-derived tumor xenograft(CDX)murine models to delineate the optimal pre-conditioning chemotherapy regimen and timing for CAR-T cell ***,transcriptome sequencing was implemented to identify the therapeutic targets and elucidate the underlying mechanisms governing the treatment ***:Our preclinical in vivo evaluation determined that a combination of cyclophosphamide and fludarabine,followed by the infusion of CD19 CAR-T cells five days subsequent to the chemotherapy,exerts the most efficacious therapeutic effect in B-cell hematological ***,RNA-seq data indicated that the therapeutic efficacy predominantly perturbs tumor cell metabolism,primarily through the inhibition of key mitochondrial targets,such as C-Jun Kinase enzyme(C-JUN).Conclusion:In summary,the present study offers critical clinical guidance and serves as an authoritative reference for the deployment of CD19 CAR-T cell therapy in the treatment of B-cell hematological malignancies.

Molybdenum disulfide(MoS_(2))-based nanostructures are highly desirable for applications such as chemical and biological sensing,photo/electrochemical catalysis,and energy storage due to their unique physical and chemical *** this work,MoS_(2)core-shell nanoparticles were first prepared through the liquid-phase processing of bulk MoS2by a femtosecond *** core of prepared nanoparticles was incompletely and weakly crystalline MoS_(2);the shell of prepared nanoparticles was highly crystalline MoS_(2),which wrapped around the core layer by *** femtosecond laser simultaneously achieved liquid-phase ablation and light *** formation mechanism of the core-shell nanoparticles is to prepare the nanonuclei first by laser liquid-phase ablation and then the nanosheets by light exfoliation;the nanosheets will wrap the nanonuclei layer by layer through van der Waals forces to form core-shell *** MoS_(2)core-shell nanoparticles,because of Mo-S bond breakage and recombination,have high chemical activity for chemical ***,the nanoparticles were used as a reducing agent to directly prepare three-dimensional(3D)Au-MoS_(2)micro/nanostructures,which were applied as surface-enhanced Raman spectroscopy(SERS)substrates to explore chemical sensing *** ultrahigh enhancement factor(1.06×10^(11)),ultralow detection limit(10-13M),and good SERS adaptability demonstrate highly sensitive SERS activity,great ability of ultralow concentration detection,and ability to detect diverse analytes,*** work reveals the tremendous potential of 3D Au-MoS_(2)composite structures as excellent SERS substrates for chemical and biological sensing.

Sensory hair cells(HCs)in the cochlea cannot regenerate spontaneously in adult mammals after being damaged by external or genetic ***,several genes and signaling pathways are reported to induce cochlear HC regeneration at the early neonatal ***14 encodes a ribosomal protein that is involved in the regulation of cell differentiation and proliferation in ***,its roles in the cochlea have not been reported in ***,we specifically overexpressed Rps14 in Lgr5+progenitor cells in the newborn mouse cochlea and found that Rps14 conditional overexpression(cOE)mice had significantly increased the ectopic HCs,including inner and outer *** further explored the source of these ectopic HCs and found no EdU+supporting cells observed in the Rps14 cOE *** lineage tracing results,on the other hand,revealed that Rps14 cOE mice had significantly more tdTomato+HCs in their cochleae than control *** results indicated that regenerated HCs by cOE of Rps14 are most likely derived from inducing the direct trans-differentiation of Lgr5+progenitor cells into ***,real-time qPCR results suggested that the transcription factor genes Atoh1 and Gfi1,which are important in regulating HC differentiation,were upregulated in the cochlear basilar membrane of Rps14 cOE *** summary,this study provides in vivo evidence that,in the postnatal mouse cochlea,Rps14 is a potential gene that can promote the spontaneous trans-differentiation of Lgr5+progenitor cells into *** gene may one day be exploited as a therapeutic target for treating hearing loss.

近年来,线上授课的形式被广大高校师生所接受,但线上授课的不足也日益显现,医学教育强调理论与实践并行,因此结合线上和线下教学优势,探索新型教学模式对于培养高素质的创新型医学人才具有重要的意义。研究将BOPPPS模型的引入、目标、预测、参与式学习、后测和小结6个过程与小规模限制性在线课堂(small private online course,SPOC)教学结合,形成了一种以学生为中心的新型教学模式。为了评估学生在新教学模式下的表现,研究还构建了创新型医学人才培养评价体系,并通过发放调查问卷初步评估教学效果。结果显示83.2%学生认为该教学模式能提高学习兴趣,79.3%学生认为可提高学习自觉性,85.9%学生认为可提高其知识掌握程度。研究工作表明,积极开展BOPPPS模型结合SPOC的教学模式和构建创新型人才培养评价体系,对于临床医学本科学教学的改革和发展将起到积极的促进作用。

Propolis as a promising functional ingredient: A comprehensive review on extraction， bioactive properties， bioavailability， and industrial applications

Propolis is a resinous complex mixture made from plant resins collected by worker bees and mixed with their own secretions. It is rich in polyphenols and flavonoids and thus has a wide range of biological activities and is considered a functional source for promoting human health. However， propolis and its bioactive compounds have poor water solubility， rapid and intense metabolism， and low oral bioavailability， which limits their wide application. In this paper， the main bioactive substances in propolis were summarized， and the biological characteristics and therapeutic potential of propolis and its bioactive substances were discussed. In addition， this paper discussed the factors affecting the bioavailability of propolis and its functional ingredients， focusing on the research progress in improving the bioavailability and bioactivity of propolis and its functional ingredients using nanoencapsulation technology. Finally， the current situation of the global propolis market and the applications of propolis products in the pharmaceutical， food， cosmetic and other industrial fields are discussed， providing useful references for promoting the development of the propolis industry.

综合考虑充电需求和配电网接纳能力的电动汽车充电站规划对电动汽车和配电网协同发展具有重要意义。对电动汽车出行大数据进行数理统计,获取出行和充电行为特征指标;以此为基础用马尔科夫原理和轮盘赌法模拟目标数量下电动汽车的出行链,预测充电需求时空分布,获得配电网各节点充电需求不可达率和不满足率;进而筛选候选站址和接入方案,采用熵权修正的层次分析法评估配电网对候选接入方案的接纳能力;以建设运营总成本最小为目标建立充电设施规划模型,采用遗传算法求解,通过配电网潮流计算检验规划结果的合理性。IEEE33节点系统案例验证了所提方法的有效性。

A Blockchain-Based Access Control Scheme for Reputation Value Attributes of the Internet of Things

The Internet of Things(IoT)access controlmechanism may encounter security issues such as single point of failure and data *** address these issues,a blockchain-based IoT reputation value attribute access control scheme is ***,writing the reputation value as an attribute into the access control policy,and then deploying the access control policy in the smart contract of the blockchain system can enable the system to provide more fine-grained access control;Secondly,storing a large amount of resources fromthe Internet of Things in Inter Planetary File System(IPFS)to improve system throughput;Finally,map resource access operations to qualification tokens to improve the performance of the access control *** simulation experiments based on the Hyperledger Fabric *** simulation experimental results,it can be seen that the access control system can achieve more fine-grained and dynamic access control while maintaining high throughput and low time delay,providing sufficient reliability and security for access control of IoT devices.

Transformer and GAN-Based Super-Resolution Reconstruction Network for Medical Images

Super-resolution reconstruction in medical imaging has become more demanding due to the necessity of obtaining high-quality images with minimal radiation dose,such as in low-field magnetic resonance imaging(MRI).However,image super-resolution reconstruction remains a difficult task because of the complexity and high textual requirements for diagnosis *** this paper,we offer a deep learning based strategy for reconstructing medical images from low resolutions utilizing Transformer and generative adversarial networks(T-GANs).The integrated system can extract more precise texture information and focus more on important locations through global image matching after successfully inserting Transformer into the generative adversarial network for picture ***,we weighted the combination of content loss,adversarial loss,and adversarial feature loss as the final multi-task loss function during the training of our proposed model *** comparison to established measures like peak signal-to-noise ratio(PSNR)and structural similarity index measure(SSIM),our suggested T-GAN achieves optimal performance and recovers more texture features in super-resolution reconstruction of MRI scanned images of the knees and belly.

Robust adaptive radar beamforming based on iterative training sample selection using kurtosis of generalized inner product statistics

In engineering application,there is only one adaptive weights estimated by most of traditional early warning radars for adaptive interference suppression in a pulse reputation interval(PRI).Therefore,if the training samples used to calculate the weight vector does not contain the jamming,then the jamming cannot be removed by adaptive spatial *** the weight vector is constantly updated in the range dimension,the training data may contain target echo signals,resulting in signal cancellation *** cope with the situation that the training samples are contaminated by target signal,an iterative training sample selection method based on non-homogeneous detector(NHD)is proposed in this paper for updating the weight vector in entire range *** principle is presented,and the validity is proven by simulation results.
