目的考察黄芪水提物效应成分对高糖(25 mmol/L)诱导的人脐静脉内皮细胞(HUVEC)损伤的保护作用,并探讨其作用机制。方法给雄性SD大鼠连续灌胃黄芪水提物[20 g/(kg·d)]7 d,分别于末次灌胃后1、2、4 h取血,合并上清,行超高效液相色谱质谱(UPLC⁃MS)分析,筛选黄芪水提物在大鼠体内代谢后的效应成分。将细胞分为正常组、模型组、不同浓度效应成分组及效应成分配伍组。用CCK-8试剂盒检测细胞活力,探讨黄芪水提物效应成分的最佳浓度。通过活性氧(ROS)实验检测细胞氧化损伤情况。酶联免疫吸附测定法检测各组细胞超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、丙二醛(MDA)含量;蛋白质印迹法检测核因子E2相关因子(Nrf2)、血红素氧合酶1(HO⁃1)蛋白表达水平。结果芒柄花素、大豆苷元、毛蕊异黄酮是黄芪水提物的效应成分。与正常组比较,模型组细胞活力增加(P<0.05);与模型组比较,芒柄花素各浓度组(26、13、6.5μmol/L)细胞活力降低(P<0.05),大豆苷元高浓度组(0.11μmol/L)细胞活力降低(P<0.05)。与正常组比较,模型组细胞ROS水平升高(P<0.05);与模型组比较,芒柄花素高、中浓度组(26、13μmol/L),大豆苷元不同浓度组(0.11、0.06、0.03μmol/L),毛蕊异黄酮高浓度组(7.00μmol/L)及各配伍组细胞ROS水平均降低(P<0.05)。效应成分配伍对细胞的抑制率高于单个效应成分,且高浓度配伍组抑制率最高。与正常组比较,模型组细胞Nrf2、HO⁃1蛋白表达量降低(P<0.05);与模型组比较,效应成分高浓度配伍组Nrf2、HO⁃1蛋白表达量升高(P<0.05)。与正常组相比,模型组细胞SOD、CAT含量降低(P<0.05)、MDA含量升高(P<0.05);与模型组相比,效应成分高浓度配伍组SOD、CAT含量均升高(P<0.05)、MDA含量降低(P<0.05)。结论黄芪水提物效应成分芒柄花素、大豆苷元及毛蕊异黄酮有保护高糖诱导的HUVEC损伤的作用,其机制可能与调节Nrf2、HO⁃1、SOD、CAT及MDA表达,发挥抗氧化作用有关。

Background Clostridium butyricum(CB)is a probiotic that can regulate intestinal microbial composition and improve meat *** protected fat(RPF)has been shown to increase the dietary energy density and provide essential fatty ***,it is still unknown whether dietary supplementation with CB and RPF exerts beneficial effects on growth performance and nutritional value of goat *** study aimed to investigate the effects of dietary CB and RPF supplementation on growth performance,meat quality,oxidative stability,and meat nutritional value of finishing ***-two goats(initial body weight,20.5±0.82 kg)were used in a completely randomized block design with a 2 RPF supplementation(0 vs.30 g/d)×2 CB supplementation(0 vs.1.0 g/d)factorial treatment *** experiment included a 14-d adaptation and 70-d data and sample collection *** goats were fed a diet consisted of 400 g/kg peanut seedling and 600 g/kg corn-based concentrate(dry matter basis).Result Interaction between CB and RPF was rarely observed on the variables measured,except that shear force was reduced(P<0.05)by adding CB or RPF alone or their combination;the increased intramuscular fat(IMF)content with adding RPF was more pronounced(P<0.05)with CB than without CB *** pH24h(P=0.009),a*values(P=0.007),total antioxidant capacity(P=0.050),glutathione peroxidase activities(P=0.006),concentrations of 18:3(P<0.001),20:5(P=0.003)and total polyunsaturated fatty acids(P=0.048)were increased,whereas the L*values(P<0.001),shear force(P=0.050)and malondialdehyde content(P=0.044)were decreased by adding ***,CB supplementation increased essential amino acid(P=0.027),flavor amino acid(P=0.010)and total amino acid contents(P=0.024)as well as upregulated the expression of lipoprotein lipase(P=0.034)and peroxisome proliferator-activated receptorγ(PPARγ)(P=0.012),and downregulated the expression of stearoyl-CoA desaturase(SCD)(P=0.034).The RPF supplementation increased dry

Antibacterial mechanism of kojic acid and tea polyphenols against Escherichia coli O157:H7 through transcriptomic analysis

Escherichia coli O157:H7 is one of the major foodborne pathogenic bacterial that cause infectious diseases in *** previous found that a combination of kojic acid and tea polyphenols exhibited better activity against *** O157:H7 than using either *** study aimed to explore responses underlying the antibacterial mechanisms of kojic acid and tea polyphenols from the gene *** functional enrichment analysis by comparing kojic acid and tea polyphenols individually or synergistically against *** O157:H7 found that acid resistance systems in kojic acid were activated,and the cell membrane and genomic DNA were destructed in the cells,resulting in“oxygen starvation”.The oxidative stress response triggered by tea polyphenols inhibited both sulfur uptake and the synthesis of ATP,which affected the bacteria's life metabolic ***,we found that kojic acid combined with tea polyphenols hindered the uptake of iron that played an essential role in the synthesis of DNA,respiration,tricarboxylic acid *** results suggested that the iron uptake pathways may represent a novel approach for kojic acid and tea polyphenols synergistically against *** O157:H7 and provided a theoretical basis for bacterial pathogen control in the food industry.

The circadian clock coordinates rhythms in numerous physiological processes to maintain organismal homeostasis. Since the suprachiasmatic nucleus(SCN) is widely accepted as the circadian pacemaker, it is critical to understand the neural mechanisms by which rhythmic information is transferred from the SCN to peripheral clocks. Here, we present the first comprehensive map of SCN efferent connections and suggest a molecular logic underlying these projections. The SCN projects broadly to most major regions of the brain, rather than solely to the hypothalamus and thalamus. The efferent projections from different subtypes of SCN neurons vary in distance and intensity, and blocking synaptic transmission of these circuits affects circadian rhythms in locomotion and feeding to different extents. We also developed a barcoding system to integrate retrograde tracing with in-situ sequencing, allowing us to link circuit anatomy and spatial patterns of gene expression. Analyses using this system revealed that brain regions functioning downstream of the SCN receive input from multiple neuropeptidergic cell types within the SCN, and that individual SCN neurons generally project to a single downstream brain *** map of SCN efferent connections provides a critical foundation for future investigations into the neural circuits underlying SCNmediated rhythms in physiology. Further, our new barcoded tracing method provides a tool for revealing the molecular logic of neuronal circuits within heterogeneous brain regions.

层次结构数据广泛存在于各类机器学习场景中,双曲空间能够以极低的失真编码层次结构数据,引入核方法后,可进一步提高双曲空间的表征能力.然而,现有的双曲核仍然存在自适应能力较低或数据失真的缺陷.为了解决这些问题,文中提出基于莫比乌斯陀螺矢量空间的双曲正定核方法.利用莫比乌斯陀螺矢量空间与庞加莱模型之间的关系,构造莫比乌斯径向基核.具体使用莫比乌斯陀螺距离代替欧几里得距离,构造莫比乌斯高斯核和莫比乌斯拉普拉斯核,并进一步证明核函数的正定性.另外,将该核函数从复空间转换到实空间上,更适用于大多数机器学习任务.在多组真实的社交网络数据集上的实验验证文中方法的有效性.

In order to improve the bioavailability of lutein(LUT),a novel lutein-stevio side nanoparticle(LUT-STE)were prepared previously,but the information about LUT-STE on protecting of eye health was *** study investigated the effect of LUT-STE on antioxidant activity of H_(2)O_(2)-induced human retinal pigment epithelial(ARPE)*** and LUT-STE(final concentration of 5μg/mL)significantly enhanced cell viability from(74.84±5.10)%to(81.92±10.01)%(LUT)and(89.33±4.34)%(LUT-STE),and inhibited the cell apoptosis(P<0.05).After pretreatment with LUT-STE in ARPE cells,the levels of superoxide dismutase(SOD),catalase(CAT)and glutathion peroxidase(GSH-Px)in ARPE cells were significantly increased(P<0.05),the contents of reactive oxygen species(ROS)and malondialdehyde(MDA)were *** addition,the vascular endothelial growth factor(VEGF)levels were inhibited by 13.61%and 17.39%,respectively,pretreatment with LUT and *** blotting results showed that the pretreatment with LUT-STE inhibited the expression of caspase-9 and caspase-3 and up-regulated Bcl-2/Bax pathway to inhibit H_(2)O_(2)-induced *** summary,the novel delivery LUT-STE had more pronounced inhibitory effect on H_(2)O_(2)-induced damage in human ARPE cells.

针对物流机器人在物料配送中对路径规划的安全性、实时性与高效性要求逐步提升的问题,本文提出了一种物流机器人调度系统及其双向同步跳点搜索算法。首先,分析物流机器人调度系统的功能需求并设计总体实现方案。其次,采用正六边形栅格地图取代传统栅格地图,并在此基础上提出改进的双向同步跳点搜索算法(JPS)。该算法调整了传统JPS算法的节点拓展规则以适应新地图,并引入灰狼优化算法指导扩展的相对方向,确保两侧搜索能够相遇。通过仿真测试对比所提算法与传统算法,所提算法比正四边形栅格图传统JPS算法快34%,比正六边形栅格图传统JPS算法快23%,结果表明所提算法的规划效率与安全性更高。此外,实物实验显示本文提出的双向同步JPS算法相较对比方法路径长度更优,在调度系统监控下物流机器人行走更加安全。

Isotropic volumetric MRI for displaying cranial perineural spread of cranial nerve in nasopharyngeal carcinoma

Objective To observe the value of isotropic volumetric MRI for displaying perineural spread(PNS)of cranial nerve(CN)in nasopharyngeal *** Eighty-seven patients with pathologically proven nasopharyngeal carcinoma were prospectively *** scanning,including three-dimensional liver acquisition with volume acceleration-flexible(3D LAVA_Flex)image,T2WI with fat suppression(T2WI-FS),T1WI,contrast enhancement(CE)T1WI-FS of nasopharynx and neck region were *** displaying rates of CN PNS were evaluated and compared between 3D LAVA_Flex and T2WI-FS,T1WI,CE-T1WI-FS at patient level,CN group level and neural level,*** The displaying rate of CN PNS in all 87 nasopharyngeal carcinoma patients by 3D LAVA_Flex sequence was 49.43%(43/87),higher than that of conventional MRI(30/87,34.48%,P=0.001).Among 59 patients with advanced nasopharyngeal carcinoma diagnosed with conventional sequences,the displaying rate of CN PNS was 71.19%(42/59)by 3D LAVA-Flex sequence,higher than that of conventional MRI(30/59,50.85%,P=0.001).At both patient level and posterior CN level,significant differences of the displaying rate of CN PNS were found between 3D LAVA-Flex sequence and T2WI-FS,T1WI,CE-T1WI-FS,while at CN level,the displaying rates of mandibular nerve PNS,CNⅨ—ⅪPNS in jugular foramen(P<0.05)and CNⅨ—ⅫPNS in carotid space of 3D LAVA_Flex sequence were all significantly higher than that of T2WI-FS,T1WI and CE-T1WI-FS(all P<0.05),of PNS of CNⅢ—Ⅴin cavernous sinus were higher than that of T2WI-FS(P<0.05),while of PNS of hypoglossal nerve were significantly higher than that of T2WI-FS and T1WI(both P<0.05).Conclusion 3D LAVA_Flex sequence could be used to effectively display CN PNS of nasopharyngeal carcinoma.
