BACKGROUND Pancreatic ductal adenocarcinoma(PDAC)is a highly fatal disease with limited effective treatment especially after first-line *** human epidermal growth factor receptor 2(HER-2)immunohistochemistry(IHC)positive is associated with more aggressive clinical behavior and shorter overall survival in *** SUMMARY We present a case of multiple metastatic PDAC with IHC mismatch repair proficient but HER-2 IHC weakly positive at diagnosis that didn’t have tumor regression after first-line nab-paclitaxel plus gemcitabine and PD-1 inhibitor treatment.A novel combination therapy PRaG 3.0 of RC48(HER2-antibody-drug conjugate),radio-therapy,PD-1 inhibitor,granulocyte-macrophage colony-stimulating factor and interleukin-2 was then applied as second-line therapy and the patient had confirmed good partial response with progress-free-survival of 6.5 months and overall survival of 14.2 *** had not developed any grade 2 or above treatment-related adverse events at any *** of peripheral CD8^(+) Temra and CD4^(+) Temra were increased during first two activation cycles of PRaG 3.0 treatment containing radiotherapy but deceased to the baseline during the maintenance cycles containing no *** PRaG 3.0 might be a novel strategy for HER2-positive metastatic PDAC patients who failed from previous first-line approach and even PD-1 immunotherapy but needs more data in prospective trials.

In this paper,we consider two kinds of extragradient methods to solve the pseudo-monotone stochastic variational inequality ***,we present the modified stochastic extragradient method with constant step-size(MSEGMC)and prove the convergence of *** the strong pseudo-monotone operator and the exponentially growing sample sequences,we establish the R-linear convergence rate in terms of the mean natural residual and the oracle complexity O(1/ǫ).Second,we propose a modified stochastic extragradient method with adaptive step-size(MSEGMA).In addition,the step-size of MSEGMA does not depend on the Lipschitz constant and without any line-search ***,we use some numerical experiments to verify the effectiveness of the two algorithms.

A partition-type tubular scaffold loaded with PDGF-releasing microspheres for spinal cord repair facilitates the directional migration and growth of cells

The best tissue-engineered spinal cord grafts not only match the structural characteristics of the spinal cord but also allow the seed cells to grow and function in ***-derived growth factor（PDGF） has been shown to promote the migration of bone marrow stromal cells;however,cytokines need to be released at a steady rate to maintain a stable concentration in ***,new methods are needed to maintain an optimal concentration of cytokines over an extended period of time to effectively promote seed cell localization,proliferation and *** the present study,a partition-type tubular scaffold matching the anatomical features of the thoracic 8–10 spinal cord of the rat was fabricated using chitosan and then subsequently loaded with chitosan-encapsulated PDGF-BB microspheres（PDGF-MSs）.The PDGF-MS-containing scaffold was then examined in vitro for sustained-release capacity,biocompatibility,and its effect on neural progenitor cells differentiated in vitro from multilineage-differentiating stress-enduring cells（MUSE-NPCs）.We found that pre-freezing for 2 hours at-20°C significantly increased the yield of partition-type tubular scaffolds,and 30 μL of 25% glutaraldehyde ensured optimal crosslinking of *** resulting PDGF-MSs cumulatively released 52% of the PDGF-BB at 4 weeks in vitro without burst *** PDGF-MS-containing tubular scaffold showed suitable biocompatibility towards MUSE-NPCs and could promote the directional migration and growth of these *** findings indicate that the combination of a partition-type tubular scaffold,PDGF-MSs and MUSENPCs may be a promising model for the fabrication of tissue-engineered spinal cord grafts.

Digital twinning enables manufacturers to create digital representations of physical entities,thus implementing virtual simulations for product *** efforts of digital twinning neglect the decisive consumer feedback in product development stages,failing to cover the gap between physical and digital *** work mines real-world consumer feedbacks through social media topics,which is significant to product *** specifically analyze the prevalent time of a product topic,giving an insight into both consumer attention and the widely-discussed time of a *** primary body of current studies regards the prevalent time prediction as an accompanying task or assumes the existence of a preset ***,these proposed solutions are either biased in focused objectives and underlying patterns or weak in the capability of generalization towards diverse *** this end,this work combines deep learning and survival analysis to predict the prevalent time of *** propose a specialized deep survival model which consists of two *** first module enriches input covariates by incorporating latent features of the time-varying text,and the second module fully captures the temporal pattern of a rumor by a recurrent network ***,a specific loss function different from regular survival models is proposed to achieve a more reasonable *** experiments on real-world datasets demonstrate that our model significantly outperforms the state-of-the-art methods.

Enhanced photocatalytic hydrogen production from Co-MOF/CN by nitrogen and sulfur co-doped coal-based carbon quantum dots

An overview and perspective of identifying lunar craters

The spatiotemporal evolution of lunar impact craters is crucial for investigating lunar interior structure,internal and external dynamic processes,and interplanetary impact *** in lunar crater identification are reviewed based on topography and gravity data,and the features and mechanisms of topographic or buried craters are analyzed regarding morphology,gravity anomalies,gravity gradients,and the underlying crust-mantle interface *** on the compiled crater catalog,the early lunar impact flux and thermal evolution are further discussed according to the basin ages and their interior *** some ancient impact basins,the crater size-frequency distribution measurements revealed age discrepancies from previous studies,suggesting that the lunar late heavy bombardment event started at~3.95 *** degraded bulge structures of the crust-mantle interface beneath mare basins reveal that these basins formed on the lunar crust surface under high-temperature conditions and underwent prolonged relaxation compared to highland ***,we reveal that the up-to-date identification of lunar buried craters remains inaccurate and incomplete,preventing us from accurately reconstructing lunar and interplanetary impact ***,we propose that a gravity model constructed using localized orthogonal basis functions can be useful for identifying lunar craters.

Feasibility study on construction of humanized mice with antibiotics combined with fecal bacteria transplantation

Objective:To select a common method of humanized mice and verify its feasibility with the aim to provide atheoretical basis for subsequent fecal bacteria transplantation ***:*** Pathogen Free(SPF)mice were fed with antibiotics mixed with sterilized drinking water for 3 weeks,during which the general situation of mice was *** 3 weeks,the number of microbes in mice's feces was evaluated by Gram *** antibiotic treatment,mice were transplanted fecal bacteria by gavage for a ***,the contents of fecal bacteria in the donor and recipient feces was determined by RT-PCR,to evaluate the colonization effect of fecal ***:(1)Antibiotic treatment had an effect on weight,feed and drinking water consumption of mice,and the fecal bacteria in mice decreased obviously.(2)After fecal bacteria transplantation,the Firmicutes,Bacteroidetes and Firmicutes/Bacteroidetes of the mice receiving Donor1 fecal bacteria accounted for 15.4%,112%and 13.7%of the mice receiving Donor2 fecal bacteria,*** Firmicutes,Bacteroidetes and Firmicutes/Bacteroidetes of Donor1 fecal bacteria accounted for 33.7%,104%and 32%of Donor2,*** recipient's gut microbes had the same trend as human ***:These results indicate that antibiotic treatment combined with fecal bacterial transplantation is feasible and safe for humanizing SPF mice.

A new monitoring index for ecological vulnerability and its application in the Yellow River Basin,China from 2000 to 2022

The ecological environment of the Yellow River Basin has become more fragile under the combined action of natural and manmade ***,the change mechanisms of ecological vulnerability in different sub-regions and periods vary,and the reasons for this variability are yet to be ***,in this study,we proposed a new remote sensing ecological vulnerability index by considering moisture,heat,greenness,dryness,land degradation,and social economy indicators and then analyzed and disclosed the spatial and temporal change patterns of ecological vulnerability of the Yellow River Basin,China from 2000 to 2022 and its driving *** results showed that the newly proposed remote sensing ecological vulnerability index had a high accuracy,at 86.36%,which indicated a higher applicability in the Yellow River *** 2000 to 2022,the average remote sensing ecological vulnerability index of the Yellow River Basin was 1.03,denoting moderate vulnerability *** intensive vulnerability area was the most widely distributed,which was mostly located in the northern part of Shaanxi Province and the eastern part of Shanxi *** 2000 to 2022,the ecological vulnerability in the Yellow showed an overall stable trend,while that of the central and eastern regions showed an obvious trend of *** gravity center of ecological vulnerability migrated southwest,indicating that the aggravation of ecological vulnerability in the southwestern regions was more severe than in the northeastern regions of the *** dominant single factor of changes in ecological vulnerability shifted from normalized difference vegetation index(NDVI)to temperature from 2000 to 2022,and the interaction factors shifted from temperature∩NDVI to temperature∩precipitation,which indicated that the global climate change exerted a more significant impact on regional *** above results could provide decision support for the ecological protection and restoration of the Y

Nocardiosis manifests as an opportunistic infection,primarily affecting individuals who are immunocompromised and susceptible to the *** present a case study of one patient with nephrotic syndrome and membranous nephropathy,who underwent treatment with prednisone and cyclosporine in *** early 2017,the patient was diagnosed with a"fungal infection"and discontinued the use of *** one month of anti-infection therapy,a cranial magnetic resonance imaging scan showed multiple abscesses in the right temporal *** diagnosis of nocardiosis was confirmed based on the presence of metastatic abscess masses,multiple lung and brain lesions,and a positive culture of Nocardia in the *** changed the anti-infection therapy to a combination of trimethoprim-sulfamethoxazole(TMP-SMX),minocycline,and ***,the patient experienced a sudden cardiac arrest and subsequently recovered after cardiopulmonary *** the five-month follow-up period following the discharge,the patient displayed an enhanced nutritional status and stable renal *** focal infection ultimately resolved during the subsequent three ***-infection caused by Nocardia should be considered in immunocompromised patients,and TMP-SMX is the preferred initial therapy;however,because of the high mortality rate,a long-term combination therapy with imipenem,cefotaxime,amikacin,and TMP-SMX is suggested.

Comparison of Placebo Effect between Asian and Caucasian Type 2 Diabetic Patients： A Meta-Analysis

Background： Placebo was defined as any therapy that is used for its nonspecific psychological and physiologic effect but has no specific pharmacologic impact on the condition being treated. Besides medication therapies, studies have found that the optimal dietary approach as well as physical activity and education are useful to control hyperglycemia in patients with type 2 diabetes （T2DM）. The aim of this study was to evaluate the placebo effects ofantidiabetic therapies in Asian and Caucasian T2DM patients and make a comparison between the two ethnicities. Methods： A search using the MEDLINE database, EMBASE, and Cochrane Database was performed, from when recording began until December 2016. The main concepts searched in English were sulfonylurea （SU）; alpha glucosidase inhibitors （AGI）; metfornlin （MET）; thiazolidinediones （TZD）; dipeptidyl peptidase-4 inhibitors （DPP-4i）; sodium-glucose cotransporter 2 inhibitors （SGLT2i）; glucagon-like peptide- 1 receptor agonist （GLP- 1RA）; type 2 diabetes （T2DM）; placebo controlled; and randomized controlled trials. Using the Cochrane instrument, we evaluated the adequacy of randomization, allocation concealment procedures, and blinding. Results： This study included 63 studies with a total of 7096 Asian patients involved and 262 studies with a total of 27,477 Caucasian patients involved. In Caucasian population, the use of placebo led to significant redtlctions of glycosylated hemoglobin （HbAlc）, -0.683% （P - 0.008） in SU monotherapy treatment, -0.193% （P = 0.001） in DPP-4i treatment, and -0.230% （P 〈 0.001） in SGLT2i treatment, respectively. In Asian population, the use of placebo resulted in significant decreases ofHbA1c, 0.162% （P= 0.012） in DPP-4i treatment and -0.269% （P =0.028） in GLP-1RA add-on therapy, respectively. The placebo also significantly reduced body weight. In Caucasian population, placebo use resulted in 0.833 kg （P = 0.006） weight loss by SU treatment and 0.953 kg （P = 0.006） weight loss by GLP-I RA treatme
