Ly49 receptors activate angiogenic mouse DBA＋ uterine natural killer cells

In humans, specific patterns of killer immunoglobulin-like receptors （KIRs） expressed by uterine natural killer （uNK） cells are linked through HLA-C with pregnancy complications （infertility, recurrent spontaneous abortion, intrauterine growth restriction and preeclampsia）. To identify mechanisms underpinning the associations between NK cell activation and pregnancy success, pregnancies were studied in mice with genetic knockdown （KD） of the mHC-activated Ly49 receptor gene family. ***49KD pregnancies were compared to normal control ***49z29 and C57BL/6 （B6） pregnancies. At mid-pregnancy （gestation day （gd9.5））, overall uNK cell （TCRI^-CD122＋DBA＋DX5- （DBA＋DX5-）） and TCRIβ-CD122＋DBA-DX5＋（DBA-DX5＋）） frequencies in pregnant uterus were similar between genotypes. Ly49KD lowered the normal frequencies of Ly49＋ uNK cells from 90.3% to 47.8% in DBA-DX5＋ and 78.8% to 6.3% in DBA＋DX5- uNK cell subtypes. ***49KD matings frequently resulted in expanded blastocysts that did not implant （subfertility）. ***49KD mice that established pregnancy had gestational lengths and litter sizes similar to controls. ***49KD neonates, however, were heavier than controls. ***49KD implantation sites lagged in early （gd6.5） decidual angiogenesis and were deficient in mid-pregnancy （gd 10.5） spiral arterial remodelling. Ultrastructural analyses revealed that ***49KD uNK cells had impaired granulogenesis, while immunocytochemistry revealed deficient vascular endothelial cell growth factor （VEGFA） production. Perforin and IFNG expression were normal in ***49KD uNK cells. Thus, in normal mouse pregnancies, Ly49 receptor signaling must promote implantation, early decidual angiogenesis and mid-pregnancy vascular remodelling. Disturbances in these functions may underlie the reported genetic associations between human pregnancy complications and the inability of specific conceptus mHCs to engage activating KIR on uNK cells.

Helper-type innate lymphoid cells(ILC)play an important role in intestinal *** of the NKR-P1 gene family are expressed in various innate immune cells,including natural killer(NK)cells,and their cognate Clr ligand family members are expressed in various specialized tissues,including the intestinal epithelium,where they may play an important role in mucosalassociated innate immune *** this study,we show that the inhibitory NKR-P1B receptor,but not the Ly49 receptor,is expressed in gut-resident NK cells,ILC,and a subset ofγδT cells in a tissue-specific ***3 cells constitute the predominant cell subset expressing NKR-P1B in the gut lamina *** known NKR-P1B ligand Clr-b is broadly expressed in gut-associated cells of hematopoietic *** genetic deletion of NKR-P1B results in a higher frequency and number of ILC3 andγδT cells in the gut lamina ***,the function of gut-resident ILC3,NK,andγδT cells in NKR-P1B-deficient mice is impaired during gastrointestinal tract infection by Citrobacter rodentium or Salmonella typhimurium,resulting in increased systemic bacterial dissemination in NKR-P1B-deficient *** findings highlight the role of the NKR-P1B:Clr-b recognition system in the modulation of intestinal innate immune cell functions.

The conventional perception of astrocytes as mere supportive cells within the brain has recently been called into question by empirical evidence, which has revealed their active involvement in regulating brain function and encoding behaviors associated with ***, astrocytes in the basolateral amygdala have been found to play a role in the modulation of anxiety-like behaviors triggered by chronic stress. Nevertheless, the precise molecular mechanisms by which basolateral amygdala astrocytes regulate chronic stress–induced anxiety-like behaviors remain to be fully elucidated. In this study, we found that in a mouse model of anxiety triggered by unpredictable chronic mild stress, the expression of excitatory amino acid transporter 2 was upregulated in the basolateral amygdala. Interestingly, our findings indicate that the targeted knockdown of excitatory amino acid transporter 2 specifically within the basolateral amygdala astrocytes was able to rescue the anxiety-like behavior in mice subjected to stress. Furthermore, we found that the overexpression of excitatory amino acid transporter 2 in the basolateral amygdala, whether achieved through intracranial administration of excitatory amino acid transporter 2agonists or through injection of excitatory amino acid transporter 2-overexpressing viruses with GfaABC1D promoters, evoked anxiety-like behavior in mice. Our single-nucleus RNA sequencing analysis further confirmed that chronic stress induced an upregulation of excitatory amino acid transporter 2 specifically in astrocytes in the basolateral amygdala. moreover, through in vivo calcium signal recordings, we found that the frequency of calcium activity in the basolateral amygdala of mice subjected to chronic stress was higher compared with normal *** knocking down the expression of excitatory amino acid transporter 2 in the basolateral amygdala, the frequency of calcium activity was not significantly increased, and anxiety-like behavior was ob

Deciphering the role of ultra-low-loaded rhodium in NiFe-mIL-53 for superior oxygen evolution reaction

Designing highly active and stable electrocatalysts of oxygen evolution reaction （OER） is one of the crucial *** this study,a novel OER electrocatalyst,NiFe-mIL-53 modified with ultra-low rhodium（Rh@NiFe-mIL-53）,is successfully prepared via the hydrothermal ***-situ Raman spectroscopy and electrochemical impedance spectroscopy reveal that the doped Rh accelerates the phase transformation of NiFe-mIL-53 and the in-situ formed Rh@NiFeOOH is the actual active *** importantly,the enhanced reversibility of electrochemical reconstruction between NiFeOOH and NiFe（OH）2after doping Rh is beneficial for improving the electrochemical stability of the catalyst.X-ray photoelectron spectroscopy spectra show the strong electronic interaction between single-atom Rh and Ni/Fe in Rh@***,theoretical calculations confirm that the integration of single-atom Rh into the NiFeOOH successfully reduces the band gap,regulates the d-band center （εd）,accelerates the charge transfer,and optimizes the adsorption behavior of oxygen-containing intermediates,thereby lowering the energy barrier of rate-determining ***,the optimized Rh@NiFe-mIL-53 exhibits excellent OER activity （240 mV） with a small Tafel slope of 48.2 mV dec-1and long-term durability (over1270 h at 10 m A cm-2and 110 h at 200 mA cm-2).This work presents a new perspective on designing highly efficient OER electrocatalysts.

This paper presents an analytical solution for the free vibration behavior of functionally graded carbon nanotube-reinforced composite(FG-CNTRC) doubly curved shallow shells with integrated piezoelectric layers. Here, the linear distribution of electric potential across the thickness of the piezoelectric layer and five different types of carbon nanotube(CNT) distributions through the thickness direction are considered. Based on the four-variable shear deformation refined shell theory, governing equations are obtained by applying Hamilton's principle. Navier's solution for the shell panels with the simply supported boundary condition at all four edges is derived. Several numerical examples validate the accuracy of the presented solution. New parametric studies regarding the effects of different material properties, shell geometric parameters, and electrical boundary conditions on the free vibration responses of the hybrid panels are investigated and discussed in detail.

Theoretical analysis on the deformation characteristics of coal wall in a longwall top coal caving face

Against the background of analyzing coal wall stability in 14101 fully mechanized longwall top coal caving face in majialiang coal mine,based on the torque equilibrium of the coal wall,shield support and the roof strata,an elastic mechanics model was established to calculate the stress applied on the coal *** displacement method was used to obtain the stress and deformation distributions of the coal *** study also researched the influence of support resistance,protective pressure to the coal wall,fracture position of the main roof and mining height on the coal wall *** following conclusions are drawn:(1) The shorter the distance from the longwall face,the greater the vertical compressive stress and horizontal tensile stress borne by the coal *** coal wall is prone to failure in the form of compressive-shear and tension;(2) With increasing support resistance,the revolution angle of the main roof decreases *** the support resistance and protective force supplied by the face guard increases,the maximum deformation of the coal wall decreases linearly;(3) As the face approaches the fracture position of the main roof,coal wall horizontal deformation increases significantly,and the coal wall is prone to instability;and(4) The best mining height of 14101 longwall face is 3.0 m.

EFFECT OF CARBON mIGRATION ON CREEP PROPERTIES OF Cr5mo DISSImILAR WELDED JOINTS WITH Ni-BASED AND AUSTENITIC WELD mETAL

In this paper, the effect of carbon migration on creep properties of Cr5mo dissimilar welded joints with Ni-based (Inconel 182) and Cr23Nil3 (A302) austenitic weld metal was investigated. Carbon migration near the weld metal/ferritic steel interface of Cr5mo dissimilar welded joints was analyzed by aging method. Local creep deformations of the dissimilar welded joints were measured by a long-term local creep deformation measuring technique. The creep rupture testing was performed for Cr5mo dissimilar welded joints with Inconel 182 and A302 weld metal. The research results show that the maximum creep strain rate occurs in the decarburized zone located on heat affect zone (HAZ) of Cr5mo ferritic steel. The creep rupture life of Cr5mo dissimilar welded joints with A 302 weld metal decreases due to carbon migration and is about 50% of that welded with Inconel 182 weld metal.

Although logistic regression is the most popular for modelling regression relationships with binary responses,many find relative risk(RR),or risk ratio,easier to interpret and prefer to use this measure of risk in regression ***,since Zou published his modified Poisson regression approach for modelling RR for cross-sectional data,his paper has been cited over 7000 times,demonstrating the popularity of this alternative measure of risk in regression analysis involving binary *** longitudinal studies have become increasingly popular in clinical trials and observational studies,it is imperative to extend Zou’s approach for longitudinal *** two most popular approaches for longitudinal data analysis are the generalised linear mixed-effects model(GLmm)and generalised estimating equations(GEE).However,the parametric GLmm cannot be used for the extension within the current context,because Zou’s approach treats the binary response as a Poisson variable,which is at odds with the Bernoulli distribution for the binary *** the other hand,as it imposes no mathematical model on data distributions,the semiparametric GEE is coherent with Zou’s modified Poisson *** this paper,we develop a GEE-based longitudinal model for binary responses to provide inference about RR.

Towards Efficient and Secure Access Control System for mobile Cloud Computing
