The role of emergency incident type in identifying first responders' health exposure risks

Fire-based emergency management service(EMS)personnel are dispatched to various incidents daily,many of which have unique occupational *** fully understand the variability of incident types and how to best pre-pare and respond,an exploration of the *** system of incident types is *** study uses potential exposure to SARS-CoV-2 as a case example to understand if and how coding categories for incident call types may be updated to improve data standardization and emergency response decision *** received emergency response incident data generated by three fire department computer-aided dispatch(CAD)systems between March and September *** incident was labeled EMS,Fire,or *** the 162,766 incidents,approximately 8.1%(n=13,144)noted potential SARS-CoV-2 exposure within their narrative descriptions of which 86.3%were coded as EMS,9.9%as Fire,and 3.9%as *** assess coding variability across incident types,researchers used the original 3-incident type variable and a new 5-incident type variable reassigned by researchers into EMS,Fire,Other,Hazmat,and Motor *** regressions compared differences in potential exposure using the 3-and 5-incident type *** evaluating the 3-incident type variable,those respond-ing to a Fire versus an EMS incident were 84%less likely to be associated with potential exposure to *** the 5-incident type variable,those responding to Fire incidents were 77%less likely to be associated with a potential exposure than those responding to EMS *** in potential exposure between the 3-and 5-incident type models show the need to understand how incident types are *** demonstrates the need for data standardization to accurately categorize incident types to improve emergency preparedness and *** have implications for incident type coding at fire department municipality and national levels.

Genome-wide analysis reveals signatures of complex introgressive gene flow in macaques(genus Macaca)

The genus Macaca serves as an ideal research model for speciation and introgressive gene flow due to its short period of diversification(about five million years ago)and rapid radiation of constituent *** understand evolutionary gene flow in macaques,we sequenced four whole genomes(two *** and two ***)and combined them with publicly available macaque genome data for genome-wide *** analyzed 14 individuals from nine Macaca species covering all Asian macaque species groups and detected extensive gene flow signals,with the strongest signals between the fascicularis and silenus species ***,we detected bidirectional gene flow between *** and *** estimated proportion of the genome inherited via gene flow between the two species was 6.19%.However,the introgression signals found among studied island species,such as Sulawesi macaques and ***,and other species were largely attributed to the genomic similarity of closely related species or ancestral ***,gene flow signals varied in individuals of the same species(***,***,***,*** and ***),suggesting very recent gene flow after the populations *** sequentially Markovian coalescence(PSMC)analysis showed all macaques experienced a bottleneck five million years ago,after which different species exhibited different fluctuations in demographic history trajectories,implying they have experienced complicated environmental variation and climate *** results should help improve our understanding of the complicated evolutionary history of macaques,particularly introgressive gene flow.

Elevated Stream Pathogenic Indicator Bacteria Concentrations in Livestock Grazing Areas across a Single National Forest

Livestock presence in proximity to forest streams has been shown to contribute pathogenic bacteria in excess of water quality standards established to protect human health. However, the degree to which livestock fecal contamination in streams on national forest lands is either a limited or a potentially widespread occurrence is still debated. This study provides additional insight into the matter. We analyzed water in small streams near known cattle grazing areas within the Stanislaus National Forest in the Sierra Nevada, California from 2012 to 2016. Fourteen stream sites were sampled before and after cattle were released onto the forest (four of these sites were sampled across multiple years) to compare indicator bacteria concentrations (fecal coliform—FC, Escherichia coli—EC) to standards established for recreational contact for surface waters. One control site was also sampled. There were 194 water quality violations of either state or federal regulatory standards for recreational contact, all of which occurred once cattle were on the forest. Mean (max) FC and EC concentrations were on orders of magnitude higher after cattle were released onto the forest [FC 1307 (30,000) and EC 1033 (17,000) MPN/100 mL] than during the time period before cattle were on the forest [FC 19 (220) and EC 17 (220) MPN/100 mL;FC, F1,210 = 105, p 1,210 = 85.5, p 0.001]. In addition, the presence of cattle, visual evidence of recent cattle-related disturbances, and sampling week were important predictor variables of FC and EC. These findings support the link between cattle presence and increased levels of stream pathogenic bacteria, and also demonstrate that stream pathogenic bacteria pollution occurs widely across the forest. This research indicates the need to consider alternative range management practices to better protect water quality and human health.

Ly49 receptors activate angiogenic mouse DBA＋ uterine natural killer cells

In humans, specific patterns of killer immunoglobulin-like receptors （KIRs） expressed by uterine natural killer （uNK） cells are linked through HLA-C with pregnancy complications （infertility, recurrent spontaneous abortion, intrauterine growth restriction and preeclampsia）. To identify mechanisms underpinning the associations between NK cell activation and pregnancy success, pregnancies were studied in mice with genetic knockdown （KD） of the MHC-activated Ly49 receptor gene family. ***49KD pregnancies were compared to normal control ***49z29 and C57BL/6 （B6） pregnancies. At mid-pregnancy （gestation day （gd9.5））, overall uNK cell （TCRI^-CD122＋DBA＋DX5- （DBA＋DX5-）） and TCRIβ-CD122＋DBA-DX5＋（DBA-DX5＋）） frequencies in pregnant uterus were similar between genotypes. Ly49KD lowered the normal frequencies of Ly49＋ uNK cells from 90.3% to 47.8% in DBA-DX5＋ and 78.8% to 6.3% in DBA＋DX5- uNK cell subtypes. ***49KD matings frequently resulted in expanded blastocysts that did not implant （subfertility）. ***49KD mice that established pregnancy had gestational lengths and litter sizes similar to controls. ***49KD neonates, however, were heavier than controls. ***49KD implantation sites lagged in early （gd6.5） decidual angiogenesis and were deficient in mid-pregnancy （gd 10.5） spiral arterial remodelling. Ultrastructural analyses revealed that ***49KD uNK cells had impaired granulogenesis, while immunocytochemistry revealed deficient vascular endothelial cell growth factor （VEGFA） production. Perforin and IFNG expression were normal in ***49KD uNK cells. Thus, in normal mouse pregnancies, Ly49 receptor signaling must promote implantation, early decidual angiogenesis and mid-pregnancy vascular remodelling. Disturbances in these functions may underlie the reported genetic associations between human pregnancy complications and the inability of specific conceptus MHCs to engage activating KIR on uNK cells.

Mucosal healing(MH)is vital in maintaining homeostasis within the gut and protecting against injury and *** factors and signaling pathways contribute in a dynamic and coordinated manner to maintain intestinal homeostasis and mucosal regeneration/***,when intestinal homeostasis becomes chronically disturbed and an inflammatory immune response is constitutively active due to impairment of the intestinal epithelial barrier autoimmune disease results,particularly inflammatory bowel disease(IBD).Many proteins and signaling pathways become dysregulated or impaired during these pathological conditions,with the mechanisms of regulation just beginning to be ***,there remains a relative lack of broadly effective therapeutics that can restore MH due to the complexity of both the disease and healing processes,so tissue damage in the gastrointestinal tract of patients,even those in clinical remission,*** increased understanding of the molecular mechanisms of IBD and MH,tissue damage from autoimmune disease may in the future be ameliorated by developing therapeutics that enhance the body’s own healing *** this review,we introduce the concept of mucosal healing and its relevance in IBD as well as discuss the mechanisms of IBD and potential strategies for altering these processes and inducing MH.
