Phytochemical analysis and antioxidant profile of methanolic extract of seed, pulp and peel of Baccaurea ramiflora Lour.

Objective: To analyze the phytochemical constituents responsible for the plausible antioxidant effect of methanolic extract of the seed, pulp and peel of Baccaurea ramiflora Lour. Methods: Fresh seed, pulp, and peel of Baccaurea ramiflora fruits were extracted with methanol(MEBRse, MEBRpu, MEBRpe) and evaluated by phytochemical analysis for their content of innumerable metabolites(primary and secondary) viz. carbohydrates, alkaloids, glycosides, tannins, phenols, terpenoids, flavonoids, proteins, and fixed oils. The antioxidant efficacy was assessed through different assay methods viz. 2,2-diphenyl-1-picrylhydrazyl(DPPH) free radical scavenging activity, total antioxidant capacity(TAC) and reducing power capacity(RPC). Estimation of total phenolic content(TPC), and total flavonoid content(TFC) was also done to confirm the presence of these phytochemicals. Results: It was revealed from the phytochemical analysis of MEBRse that alkaloids, glycosides, carbohydrates, phenols, and flavonoids were present, while that of MEBRpu showed the existence of carbohydrates, proteins, alkaloids, glycosides, phenols, saponins, flavonoids, and fixed oils. Presence of carbohydrates, alkaloids, phenols, tannins, flavonoids, and terpenoids were found in the MEBRpe. A significant antioxidant activity was revealed by the MEBRpu [EC_(50):(27.612 ± 1.375) μg/mL], compared to MEBRpe, and MEBRse in DPPH assay. The ranking order for RPC was MEBRpu > MEBRpe > MEBRse respectively. The EC_(50) value of TAC of the MEBRpu, MEBRpe, and MEBRse were(25.107 ±0.744) μg/mL,(241.127 ± 7.463) μg/mL and(372.364 ± 11.030) μg/mL, respectively. Quantity of TPC and TFC were the highest in the MEBRpu(124.360 ± 2.078 mg GAE/g and 107.527 ±1.900 mg QRE/g extract) rather than MEBRpe and MEBRse extracts. Conclusions: This study suggests that MEBRpu has a significantly higher antioxidant property than MEBRpe and MEBRse. In recent years, the popularity of medicinal plants as a remedy has been increased manifold due to having minimal adverse effects. The current study aimed to compare the hypoglycemic, hypolipidemic and hepatoprotective effects of the ethanolic extract of Asparagus racemosus (EEAR) Linn. alone and combinedly with conventional antidiabetic agents (gliclazide and pioglitazone) in alloxan-induced diabetic rats. Diabetes was induced in male Wister albino rats by the administration of single intra-peritoneal injection of alloxan monohydrate (120 mg/kg b.w.). Effect of oral administration of two different doses of EEAR (200 and 400 mg/kg b.w.), gliclazide (10 mg/kg b.w.) and pioglitazone (10 mg/70kg/b.w.) alone for 2 weeks and a combination of EEAR (200 mg/kg b.w.) with either gliclazide (10 mg/kg b.w.) or pioglitazone (10 mg/70kg/b.w.) for 2 weeks were examined on hypoglycemic activity on 0th, 5th, 10th and 14th day of treatment. After 2 weeks of treatment, hypolipidemic and hepatoprotective effects were estimated by serum biochemical markers such as total cholesterol (TC), triglyceride (TG), low density lipoprotein (LDL), very low density lipoprotein (VLDL), high density lipoprotein (HDL), serum glutamate oxaloacetate transaminases (SGOT), serum glutamate pyruvate trans-aminases (SGPT) and total protein (TP) with the help of commercially available kits. The survival rate, body weight and organ weight were also measured. Alloxan treatment resulted in persistent hyperglycemia, hyperlipidemia and liver dysfunction in rats. Treatment with EEAR at different doses improved hyperglycemia significantly (p th and 14th day of treatment in a dose-dependent mood when compared to the disease control rats, gliclazide treated rats and pioglitazone treated rats. Grass pea(Lathyrus sativus L.)is an imperative food crop cultured in dryland agricultural *** is a vital source of dietary protein to millions of populaces living in low-income countries in South-East Asia and *** study highlights the improvement of genomic properties and their application in marker-trait relationships for 17 yield-related characters in 400 grass pea genotypes from China and *** characters were assessed via 56 polymorphic markers using general linear model(GLM)(P+G+Q)and mixed linear model(MLM)(P+G+Q+K)in the tassel software based on the linkage disequilibrium and population structure *** structure analysis showed two major groups and one admixed group in the *** significant loci pairs of linkage disequilibrium(LD)mean value(D′)was 0.479.A total of 99 and 61 marker-trait associations in GLM and MLM models allied to the 17 traits were accepted at a 5%level of *** these markers,21 markers were associated with more than one trait;12 marker-trait associations passed the Bonferroni correction *** models found six markers C41936,C39067,C34100,C47146,C47638,and C43047 significantly associated with days to maturity,flower color,plant height,and seed per pod were detected in the Hebei and Liaoyang location(p≤0.01),and the interpretation rate(R^(2)value)11.2%to 43.6%.Conferring to the consequences,the association analysis methodology may operative system for quantitative,qualitative,and biochemical traits related to gene position mapping and support breeders in improving novel approaches for advancing the grass pea quality.

Upgrade from aerated static pile to agitated bed systems promotes lignocellulose degradation in large-scale composting through enhanced microbial functional diversity

Composting presents a viable management solution for lignocellulose-rich municipal solid ***,our understanding about the microbial metabolic mechanisms involved in the biodegradation of lignocellulose,particularly in industrial-scale composting plants,remains *** study employed metaproteomics to compare the impact of upgrading from aerated static pile(ASP)to agitated bed(AB)systems on physicochemical parameters,lignocellulose biodegradation,and microbial metabolic pathways during largescale biowaste composting process,marking the first investigation of its *** degradation rates of lignocellulose including cellulose,hemicellulose,and lignin were significantly higher in AB(8.21%-32.54%,10.21%-39.41%,and 6.21%-26.78%)than those(5.72%-23.15%,7.01%-33.26%,and 4.79%-19.76%)in ASP at three thermal stages,*** AB system in comparison to ASP increased the carbohydrate-active enzymes(CAZymes)abundance and production of the three essential enzymes required for lignocellulose decomposition involving a mixture of bacteria and fungi(i.e.,Actinobacteria,Bacilli,Sordariomycetes and Eurotiomycetes).Conversely,ASP primarily produced exoglucanase andβ-glucosidase via fungi(i.e.,Ascomycota).Moreover,AB effectively mitigated microbial stress caused by acetic acid accumulation by regulating the key enzymes involved in acetate conversion,including acetyl-coenzyme A synthetase and acetate ***,the AB upgraded from ASP facilitated the lignocellulose degradation and fostered more diverse functional microbial communities in large-scale *** findings offer a valuable scientific basis to guide the engineering feasibility and environmental sustainability for large-scale industrial composting plants for treating lignocellulose-rich *** findings have important implications for establishing green sustainable development models(e.g.,a circular economy based onmaterial recovery)and for achieving sustainable development goals.

Multi-strain probiotics provide the most dependable approaches to improve health,immune response,and disease resistance in farmed *** the present study,we examined the effects of multi-species probiotics on the survival,growth,immune response,and disease resistance of rohu(Labeo rohita)*** hatched larvae from the day of first feeding(average weight of 0.003 g)were reared with multi-species probiotics having a combination of Bacillus subtilis(109 colony forming units(cfu)/mL)and Lactobacillus spp.(Lactobacillus plantarum,Lactobacillus buchneri-1011 cfu/mL)in water containing doses of 0(control-C),0.5 mL/L(treatment 1-T1),and 1.0 mL/L(treatment 2-T2)in triplicates for 90 *** the experiment,a challenge test was performed to assess the fish's resistance to pathogenic Aeromonas *** higher survival was recorded in larvae of treated groups(87%in T2 and 79%in T1)compared to the control(62%).Significantly higher growth performance(weight gain and specific growth rate—SGR)was shown by the probiotic-treated larval groups compared to the *** supplementation resulted in significantly higher counts of total viable colony(TVC)and lactic acid bacteria(LAB)in the *** immunological parameters(mucosal fold fattening,goblet cell abundance,expansion of lamina propria and enterocytes)of the gut were significantly better in probiotic-treated *** liver of treated fish showed irregular shape nuclei turning into regular shape and reducing spaces between the hepatic ***-treated fish had the highest post-challenge survival rate(90%)against *** *** erythrocytes of challenged fish treated with probiotics had significantly lower frequencies of various nuclear and cellular *** findings suggest that multi-species probiotic supplements could improve the survival,growth,health status,and immune response of rohu in the early stages of its development.

Host associated probiotics(HAPs)provide health benefits to the host when administered as dietary ***,a short-term probiotics application strategy has yet to be optimized.A 90-days study was conducted to evaluate the response of Malaysian mahseer,Tor tambroides post larvae fed with basal diet enriched HAPs for 30-days,and its response following another 60-days feeding with only basal *** experimental diets(Enterococcus faecalis strain 2674(T1),Aeromonas *** A8-29(T2)and *** strain FC11682(T3))were prepared by spray-coating each HAPs on a basal diet at 1×108 CFU/g *** in growth performances,whole-body proximate and fatty acid composition,muscle morphometry,and gut morphology were *** showed that after 30 days,T3 fish produced highest *** HAP treatment groups showed better muscle distribution profile,improved fatty acid composition,and higher villus length,width and area,than control *** 90 days,the growth of T3 fish was still the *** distribution profile and villus growth were higher in HAP treatments,although only total n-6 PUFA,total MUFA,linoleic acids,and linolenic acids in HAP treated fish remained high after probiotics *** difference in whole-body proximate composition was observed in both 30 and 90 *** findings demonstrated that short-term application of HAPs at an early stage could be used to boost *** growth,with *** strain FC11682 showing the best efficacy.

Background: The robustness is a measurement of an analytical chemical method and its ability to contain unaffected by little with deliberate variation of analytical chemical method parameters. The analytical chemical method variation parameters are based on pH variability of buffer solution of mobile phase, organic ratio composition changes, stationary phase (column) manufacture, brand name and lot number variation;flow rate variation and temperature variation of chromatographic system. The analytical chemical method for assay of Atropine Sulfate conducted for robustness evaluation. The typical variation considered for mobile phase organic ratio change, change of pH, change of temperature, change of flow rate, change of column etc. Purpose: The aim of this study is to develop a cost effective, short run time and robust analytical chemical method for the assay quantification of Atropine in Pharmaceutical Ophthalmic Solution. This will help to make analytical decisions quickly for research and development scientists as well as will help with quality control product release for patient consumption. This analytical method will help to meet the market demand through quick quality control test of Atropine Ophthalmic Solution and it is very easy for maintaining (GDP) good documentation practices within the shortest period of time. Method: HPLC method has been selected for developing superior method to Compendial method. Both the compendial HPLC method and developed HPLC method was run into the same HPLC system to prove the superiority of developed method. Sensitivity, precision, reproducibility, accuracy parameters were considered for superiority of method. Mobile phase ratio change, pH of buffer solution, change of stationary phase temperature, change of flow rate and change of column were taken into consideration for robustness study of the developed method.
