Investigation of the Potential Association between Clustered Regularly Interspersed Short Palindromic Repeats (CRISPR) and Antibiotic Resistance Pattern of Bacterial Strains Isolated from Medical Waste and Environmental Water

Enterobacteriaceae are important human pathogens that cause many food and waterborne illness. Rapid emergence of multi-drug resistant bacteria in Bangladesh has become a serious problem. Phage-host interaction is now considered as the major driving force for the conversion of non-pathogenic bacteria to pathogenic ones. Evolution of highly pathogenic and antibiotic resistant bacteria largely depends upon the horizontal gene transfer by means of plasmid, megaplasmid and bacteriophages. Conversely, bacteria may acquire a novel defence mechanism called CRISPR (Clustered regularly interspersed short palindromic repeats) that can restrict horizontal transfer of plasmids and bacteriophages to limit the spread of antibiotic resistance genes among bacterial species. In this study, twenty bacterial strains were isolated from water of different medical waste and Buriganga river. Therefore, CRISPR locus was investigated following various biochemical and molecular analysis of those bacterial isolates. Identification of the bacterial isolates was conducted by Polymerase Chain Reaction (PCR) based assay of 16S rDNA extracted from those isolated strains. Results indicated that most strains were identified as Proteus mirabilis and Citrobacter freundii which mainly cause septicemia, and pneumonia in human. Thereafter, antibiogram of these strains was performed by using 11 different antibiotic discs where bacterial isolates from medical drainage system showed more resistant to antibiotics than the river water. In this study, CRISPR locus was also investigated within the genome of the isolated bacterial stains but unexpectedly, we did not find any CRISPR locus in their genome. In conclusion, we confirm that multi-drug resistant bacterial strains are devoid of CRISPR locus suggesting a possible negative association between CRISPR locus and antibiotic resistance. Impact of Channel Length and Width for Charge Transportation of Graphene Field Effect Transistor

The effect of channel length and width on the large and small-signal parameters of the graphene field effect transistor have been explored using an analytical *** the case of faster saturation as well as extremely high transit frequency,the graphene field effect transistor shows outstanding *** the transfer curve,it is observed that there is a positive shift of Dirac point from the voltage of 0.15 V to 0.35 V because of reducing channel length from 440 nm to 20 nm and this curve depicts that graphene shows ambipolar ***,it is found that because of widening channel the drain current increases and the maximum current is found approximately 2.4 mA and 6 mA for channel width 2μm and 5μm ***,an approximate symmetrical capacitance-voltage(C-V)characteristic of the graphene field effect transistor is obtained and the capacitance reduces when the channel length decreases but the capacitance can be increased by raising the channel *** addition,a high transconductance,that demands high-speed radio frequency(RF)applications,of 6.4 mS at channel length 20 nm and 4.45 mS at channel width 5μm along with a high transit frequency of 3.95 THz have been found that demands high-speed radio frequency applications.

Optical and electrical properties of crystalline indium tin oxide thin film deposited by vacuum evaporation technique

Indium tin oxide(ITO) thin film was deposited on glass substrate by means of vacuum evaporation technique and annealed at 200 ℃, 300℃ and 400 ℃ in air for 1 h. The characterization and properties of the deposited film samples were analyzed by X-ray diffraction(XRD), scanning electron microscopy(SEM), and UV-VIS-NIR spectroscopy techniques. From the XRD patterns, it was found that the deposited thin film was of crystalline at an annealing temperature of 400℃. The crystalline phase was indexed as cubic structure with lattice constant and crystallite size of 0.511 nm and 40 nm, respectively. The SEM images showed that the films exhibited uniform surface morphology with well-defined spherical grains. The optical transmittance of ITO thin film annealed at 400 ℃ was improved from 44% to 84% in the wavelength range from 250 nm to 2 100 nm and an optical band gap was measured as 3.86 e V. Hall effect measurement was used to measure the resistivity and conductivity of the prepared film.

Redox degenerative reactions of the biological system inevitably produce reactive oxygen species (ROS) and their derivatives. Oxidative stress is the result of an imbalance in pro-oxidant/antioxidant homeostasis that leads to the generation of toxic reactive oxygen species (ROS), such as hydrogen peroxide, organic hydro peroxides, nitric oxide, superoxide and hydroxyl radicals etc. Information are accumulating steadily, supporting the general importance of oxidative damage of tissue and cellular components as a primary or secondary causative factor in many different human diseases and aging processes. Many of the recent landmarks in scientific research have shown that in human beings, oxidative stress has been implicated in the progression of major health problems by inactivating the metabolic enzymes and damaging important cellular components, oxidizing the nucleic acids, leading to cardiovascular diseases, eye disorders, joint disorders, neurological diseases (Alzheimer’s disease, Parkinson’s disease and amyotrophic lateral sclerosis), atherosclerosis, lung and kidney disorders, liver and pancreatic diseases, cancer, ageing, disease of the reproductive system including the male and female infertility etc. The advent of a growing number of in vitro and in vivo models for evaluating the human disease pathology is aiding scientists in deciphering the detailed mechanisms of the point of intersection of the oxidative stress with other cellular components or events in the growing roadmap leading to different human disorders. The toxic effect of reactive oxygen and nitrogen species in human is balanced by the antioxidant action of non-enzymatic antioxidants, as well as by antioxidant enzymes. Such antioxidant defences are extremely important as they represent the direct removal of free radicals (prooxidants), thus providing maximal protection for biological sites. A Comparative Study of Heavy Metal Concentration in Different Layers of Tannery Vicinity Soil and Near Agricultural Soil

This research was conducted to bring out the appropriate level of heavy metal contamination in soil at the site of Buriganga river bank near the Hazaribagh tannery area and the normal agricultural area to evaluate heavy metal contamination in soil due to untreated tannery effluents. AAS flame method was used to determine the concentration of Chromium (Cr), lead (Pb), Cadmium (Cd), Zinc (Zn) in several depths of the soil. Results showed that, the heavy metal concentration in soil at the site of Buriganga river bank in the tannery vicinity and normal agricultural area significantly varies, such as Cr varies (561.71 - 31.23) mg/kg, Zn varies (158.23 - 73.5), Pb varies (70.58 - 24) and Cd varies (2.25 - 0.71) mg/kg in the river bank as well as in the normal agricultural area Cr varies (27.869 - 24.5416) mg/kg, Zn varies (134.167 - 28), Pb varies (25.76 - 22.49) and Cd varies (1.30 - 0.64) mg/kg. The study indicated that heavy metals concentration was found to be significantly higher in river bank soils than in the normal agricultural area. This metal concentration in soil is responsible for the promotion of toxicity in agricultural products. Therefore, the human health and environment are affected by these areas.

Comparison between Tencel-Flax Blended Slub Yarn and Cotton-Flax Blended Slub Yarn

The blending of different fibers is done for the manufacturing of yarn to ensure the presence of multiple properties in the resultant yarn which is impossible if we use only one type of fiber. Tencel is regenerated cellulosic fiber & flax is natural bast fiber. Tencel shows more versatility than flax. Tencel has a softer feel with a variety of end uses. Tencel is wrinkle-prone and stiff. Tencel shows extreme strength even in wet conditions and at the same time, it is more durable than linen. The most important fact is that the heat dissipation property of flax is 5 times higher than wool & 19 times higher than silk. Linen clothing can lower the body temperature by 3 to 4 degree celsius in hot weather than clothing made of wool and silk. A yarn made of both Tencel & linen (50:50) will contain the properties of both fibers. At the same time, we can make this yarn more aesthetic by incorporating a slub. It was observed that the slub yarn made by blending Tencel and flax fiber shows better properties than the slub yarn of the same count made from the blending of cotton and flax.
