Bioguided Fractionation and Purification of Natural Bioactives Obtained from<i>Alpinia conchigera</i>Water Extract with Melanin Inhibition Activity

Crude extracts of Alpinia conchigera a species from the Malaysian Ginger (Zingiberaceae) family and its fractions obtained from various extraction methods were assayed for melanogenesis inhibition activity and cell viability. The crude extract obtained from the ethanolic extraction and the super critical fluid extraction did not exhibit significant melanin inhibition activity and was shown to be toxic to the melanocyte cells in comparison to the water extract. The crude aqueous extracts displayed melanin inhibition of 96.38 ± 1.60% and cell viability 109.90 ± 8.32% at a concentration of 500 μg/mL. Bioassay guided fractionation was performed on the water extracts to isolate the active compounds. The actives were identified as trans-cinnamaldehyde and chavicol glucopyranoside with both compound showing potent anti-melanogenesis activity. At 4.9 μg/ml, both trans-cinnamaldehyde and chavicol glucopyranoside gave 85% inhibition of melanin formation in vitro with 77% and 97% cell viability respectively. In comparison, kojic acid, a known skin lightening agent showed 90.0% inhibition at 100 μg/mL. The bioactive composition comprising the extract, active fraction, purified compounds or mixture thereof of Alpinia conchigera may be used for cosmetic and pharmaceutical applications, particularly for the purposes of reducing skin pigmentation.

This paper presents a real approach for thermal unit commitment(UC)problem solution in Niamey(Niger).The proposed methodology consists of four conventional thermal generating units and imported power(IMP)from a neighboring country as the existing grid in addition to the inclusion of Photovoltaic(PV)power,Wind Turbine Generators(WTGs),and Battery Energy Storage System(BESS).Minimization of the total daily operating cost is considered as the objective function in two *** the first case,UC with thermal units considering the IMP,PV and BESS is *** the second case,WTGs are introduced beside high penetration of PV and BESS and the IMP is removed in order to get rid of its economical and mostly political ***(Mixed-Integer Linear Programming)is used here as the optimization technique to obtain an optimal unit commitment problem solution with consideration of PV,WTGs and *** effectiveness and robustness of the proposed scheme is verified by numerical simulations using MATLAB environment.

Objective:Pancreatic ductal adenocarcinoma(PDAC)is a deadly malignancy,due in large part to its resistance to conventional therapies,including radiotherapy(RT).Despite RT exerting a modest antitumor response,it has also been shown to promote an immunosuppressive tumor *** studies demonstrated that focal adhesion kinase inhibitors(FAKi)in clinical development inhibit the infiltration of suppressive myeloid cells and T regulatory(T regs)cells,and subsequently enhance effector T cell *** inhibitors in clinical development have not been investigated in combination with RT in preclinical murine models or clinical ***,we investigated the impact of FAK inhibition on RT,its potential as an RT sensitizer and immunomodulator in a murine model of ***:We used a syngeneic orthotopic murine model to study the effect of FAKi on hypofractionated ***:In this study we showed that IN10018,a small molecular FAKi,enhanced antitumor response to *** activity of the combination of FAKi and RT is T cell *** in combination with RT enhanced CD8+T cell infiltration significantly in comparison to the radiation or FAKi treatment alone(P<0.05).FAKi in combination with radiation inhibited the infiltration of granulocytes but enhanced the infiltration of macrophages and T regs in comparison with the radiation or FAKi treatment alone(P<0.01).Conclusions:These results support the clinical development of FAKi as a radiosensitizer for PDAC and combining FAKi with RT to prime the tumor microenvironment of PDAC for immunotherapy.

Smartphone-Imaged HIV-1 Reverse-Transcription Loop-Mediated Isothermal Ampliflcation(RT-LAMP) on a Chip from Whole Blood

Viral load measurements are an essential tool for the long-term clinical care of human immunodeficiency virus （HIV）-positive individuals. The gold standards in viral load instrumentation, however, are still too limited by their size, cost, and sophisticated operation for these measurements to be ubiquitous in remote settings with poor healthcare infrastructure, including parts of the world that are disproportionately affected by HIV infection. The challenge of developing a point-of-care platform capable of making viral load more accessible has been frequently approached but no solution has yet emerged that meets the practical requirements of low cost, portability, and ease-of-use. In this paper, we perform reverse-transcription loop-mediated isothermal amplification （RT-LAMP） on minimally processed HIV-spiked whole blood samples with a microfluidic and silicon microchip platform, and perform fluorescence measurements with a consumer smartphone. Our integrated assay shows amplification from as few as three viruses in a - 60 nL RT- LAMP droplet, corresponding to a whole blood concentration of 670 viruses per μL of whole blood. The technology contains greater power in a digital RT-LAMP approach that could be scaled up for the determination of viral load from a finger prick of blood in the clinical care of HIV-positive individuals. We demonstrate that all aspects of this viral load approach, from a drop of blood to imaging the RT-LAMP reaction, are compatible with lab-on-a-chip components and mobile instrumentation.

Objectives:To determine the association of Human leukocyte antigen B(HLA‑B)alleles with ankylosing spondylitis and the allelic association of HLAB27 with endoplasmic reticulum aminopeptidase 1(ERAP1)single-nucleotide polymorphism(SNP)rs30187 in patients with ankylosing spondylitis(AS).Methods:We studied 48 consecutively enrolled well-defined AS patients and 48 healthy controls recruited from the outpatient clinic of the rheumatology department of Bangabandhu Sheikh Mujib Medical University(BSMMU),Dhaka,*** genotype HLA-B alleles from peripheral blood DNA,we designed sequence-specific primers for polymerase chain ***'s sequencing method was used to detect the ERAP1 rs30187 *** HLA-B allele distributions were compared between cases and *** infer an allelic association between HLA-B27 and ERAP1,allele distributions from both loci were compared using ***-squared test or Fisher's exact test was used to quantify the association,and their strength of association was tested by odds ratio(OR)with 95%confidence interval(CI).Results:The frequency of HLA-B27 was found to be higher in AS patients(60.4%)than in healthy controls(8.3%).HLA-B27 allele had an OR of 16.8(95%CI=5.2–54.4)for the development of ***,4%of AS patients had both HLA-B27 and *** significant association was found between ERAP1 rs30187 SNP and HLA-B27 in patients with AS(OR=1.7,95%CI=0.5–5.6).Conclusions:HLA-B27 was significantly associated with AS in patients in ***,no association between ERAP1 rs30187 SNP and HLA-B27 was observed.
