Haplotype Diversity at Sub1 Locus and Allelic Distribution Among Rice Varieties of Tide and Flood Prone Areas of South-East Asia

Single nucleotide polymorphisms and restriction digestion-based haplotype variations among 160 flood prone rice varieties were analyzed with enzymes Alu I and Cac8 I to generate polymorphisms at Sub1A and Sub1C loci （conferring submergence tolerance）, respectively. Haplotype associated with phenotype was used to study the haplotype variations at Sub1A and Sub1C loci and to determine their functional influence on submergence tolerance and stem elongation. Three patterns at Sub1A locus, Sub1A0 （null allele）, Sub1A1 （does not cut） and Sub1A2 （one SNP）, and four patterns at Sub1C locus, Sub1C1, Sub1C2, Sub1C3 and Sub1C4, were generated. Both tolerant Sub1A1 and intolerant Sub1A2 had the same length, but the difference was presence of a restriction site in the Sub1A2, but absent at the Sub1A1. Further, two types of polymorphism were detected at the Sub1C, one included major length polymorphisms （165, 170 and 175 bp） and the other was a single restriction site at different position. Eight haplotypes （different combinations of the two loci）, A1C1, A1C2, A1C4, A2C2, A2C4, A0C2, A0C3 and A0C4, were detected among 160 varieties. Haplotype A1C1 was comparatively more related to haplotypes A1C2 and A1C4, having the same Sub1A allele, and these haplotypes were found only in Bangladeshi, Sri Lankan and Indian varieties. Most tolerant varieties in A1C1 haplotype showed slow elongation, having tolerant specific Sub1A1 and Sub1C1 alleles. Further, the varieties Madabaru and Kottamali （A2C2） also showed moderate level of tolerance without Sub1A1 allele. These varieties were different with FR13A and also suspected to carry different novel tolerant genes at other loci. These materials could be used for hybridization with Sub1 varieties for pyramiding additional tolerant specific alleles into a single genotype for improving submergence tolerance in rice.

The current study investigated the comparative oxidative damage in twomaize seedlings induced by saline,drought,and combined stress and the ameliorative role of two different doses(20 and 80μM)of 5-aminolevulinic acid(ALA)against the above-mentioned *** grown 10-day-old maize(Zea mays,*** Hybrid Maize-7(BHM-7)and BARI Hybrid Maize-9(BHM-9))seedlings were exposed to 12 dS/m of saline solution,200 mM mannitol-induced drought stress alone and their combined stress for 7 *** revealed that individual stresses retard the plant growth to some degrees,however,their combined stress has more detrimental effects,which might be correlated with lipid peroxidation(md.)-induced oxidative stress in seedlings,enhanced Na+/K+ratio,and augmented generation of superoxide(O_(2)•^(−))and hydrogen peroxide(H_(2)O_(2)).In contrast,exogenous ALA supplementation at 20μM concentration markedly recovered from chlorosis and growth inhibition,substantially scavenged reactive oxygen species(ROS)and md. by preserving ionhomeostasis and relaxing oxidative stress,also,by boosting catalase(CAT)and glutathione S-transferase(GST),and exclusively via depressing the activity of lipoxygenase(LOX)antioxidant *** the contrary,80μM ALA made things worse,nevertheless,higher activities shown by other antioxidant enzymes,like,superoxide dismutase(SOD),ascorbate peroxidase(APX),peroxidase(POD),and glutathione peroxidase(GPX),which were related to lessen the oxidative damage by highly produced O_(2)•^(−)and H_(2)O_(2) under combined ***-denaturing gel electrophoresis was done for further ***,ALA importantly increased the photosynthetic pigment contents in both genotypes irrespective of ***,GST might have assisted the plants to escape from the herbicidal effect by ***,in the combined stress condition,high ALA concentration may have some positive role to *** findings also showed that BHM-9 performed better than BHM

Effect of Natural Disasters and Their Coping Strategies in the Kuakata Coastal Belt of Patuakhali Bangladesh

Bangladesh has experienced a number of severe natural disasters. Most recently, tropical storms Aila, Sidr and Mohaseen have affected the Kuakata coastal belt. These natural disasters have had substantial negative impacts on natural resources. Development of coping strategies for the protection of natural resources across this belt is significantly important. The present research is focused on the effects of tropical storms on crops, humans, fish, livestock and infrastructure. There were 99 fatalities recorded during the Sidr and Aila natural disasters. These disasters impacted 910.94 ha (Sidr) and 973.69 ha (Aila) of cropland. The total number of affected livestock and fishes such as, 18,200 fish ponds, 1209 cattle, 3324 goats, 6888 chickens, 1716 ducks, 103 buffalo and 144 sheep. After tropical cyclones of Sidr, Aila, Heavy rainstorms and Mohaseen 12,970, 17,703, 10,050 and 2500 households respectively reported some form of damage. Cattle, goat, chicken and buffalo were found more injured than other livestock due to the natural disasters of Sidr. Temperature and wind speed were both found to be statistically significant (p ≤ 0.05) with humidity. Climatic parameters trend has been increased significantly since 1979. Regular monitoring of climatic variables, preparedness activities, and coping strategies would be significantly important for this coastal communities.

This research investigated the water qualities and distribution of heavy metals concentration in rain, river, ground and supply waters, providing a baseline in Dhaka Urban, Bangladesh, against which future changes can be assessed. The water sample was collected in the Dhaka central zone, from a site located in the center of the Bengal Delta plain between 23䓨'N - 23䓶'N latitude and 90䓔'E - 90䓟'E longitude which is affected by environmental pollution for high traffic, industrialization and because of its rapid urbanization. The water samples were analyzed for heavy metals by Inductively Coupled Plasma Mass-Spectrometry (ICP-MS), major ions by Ion-Chromatography (IC), water tracer (δ18O and δD) by Mass-Spectrometry (MS), following the standard techniques. Elevated EC and chloride (Cl-) are the markers of wastewater, providing an indication of impacts in the Buriganga River water. The relative concentrations of major ions were presented in the form of Piper and Stiff Diagram. The color maps were constructed to present ions distribution over the areas. The stream reaches directly in Buriganga River from leather industries which may reflect the naturally low dissolved oxygen (DO) 0.3 - 3.5 (mg/L), where aquatic life can’t survive. Uranium contents U238 (0.1 - 2.7 μg/L) are not exceeding the WHO guideline level of 30 μg·L-1. Significant portion of Cs133 (0.05 - 0.32 μg/L), Se (0.03 - 0.12 μg/L), B (50 - 2986 μg/L) and Cr (0.55 - 18.59 μg/L), could be increased in Buriganga River areas by the effect of local sources like leather industry. Zn (10 - 1466 μg/L) in the Dhaka Rain and Pb (4 - 6 μg/L) in Dhaka Water occur mainly in industrial and the high traffic areas, respectively. Plant associated bacteria are promising alternatives to chemical fertilizers for plant growth and yield improvement in an eco-friendly *** this study,rice associated bacteria were isolated and assessed for mineral phosphate solubilization and indole-3-acetic acid(IAA) production activity in *** promising strains,which were tentatively identified as phylotaxon Pseudochrobactrum sp.(BRRh-1),Burkholderia sp.(BRRh-2),Burkholderia sp.(BRRh-3),Burkholderia sp.(BRRh-4),Pseudomonas aeruginosa(BRRh-5 and BRRh-6) based on their 16 S rRNA gene phylogeny,exhibited significant phosphate solubilizing activity in National Botanical Research Institute phosphate growth medium,and BRRh-4 displayed the highest phosphate solubilizing activity,followed by *** p H of the culture broth declined,resulting in increase of growth rate of bacteria at p H 7,which might be due to organic acid secretion by the *** presence of L-tryptophan,five isolates synthesized IAA and the maximum IAA was produced by BRRh-2,followed by *** of two most efficient phosphate solubilizing isolates BRRh-4 and BRRh-5 by root dipping(colonization) of seedling and spraying at the flowering stage significantly enhanced the growth and grain yield of rice variety BRRI ***,application of both strains with 50% of recommended nitrogen,phosphorus and potassium fertilizers produced equivalent or higher grain yield of rice compared to the control grown with full recommended fertilizer doses,which suggests that these strains may have the potential to be used as bioinoculants for sustainable rice production.

Image-Based Vehicle Speed Estimation

Vehicle speed is an important parameter that finds tremendous application in traffic control identifying over speed vehicles with a view to reducing accidents. Many methods, such as using RADAR and LIDAR sensors have been proposed. However, these are expensive, and their accuracy is not quite satisfactory. In this paper, a video-based vehicle speed determination method is presented. The method shows satisfactory performance on standard data sets and gives that error rate of velocity estimation is within 10%.

Detection of liquid mixture of different volume ratio is very important in industrial purposes. The paper reports a sensing mechanism of binary liquid mixture for different volume fraction, based on the measurement of refractive index of the mixture. Here, a highly sensitive liquid filled core Photonic Crystal Fiber structure has been proposed to detect liquid mixture solution. Numerical investigation of the proposed structure is carried out by employing full vectorial Finite Element Method (FEM).

Yield Response of Direct Seeded <i>Aus</i>Rice Varieties under Rainfed Condition

Rainfed Aus rice is a popular conventional rice cultivation technique that through appropriate variety selection can accelerate the benefits of the farmer. On this context, an experiment was carried out at the research field of Bangabandhu Sheikh Mujibur Rahman Agricultural University, Gazipur1706 from April to August in 2014 to evaluate yield performance of some Aus rice varieties under rainfed condition. The experiment was laid out in a split plot design with three replications. The experiment consisted of two sets of treatments: water regime as irrigated (irrigation when necessary) and rainfed (dependent on rainfall) in the main plot and 15 varieties into sub-plots. It was revealed that yield and yield contributing characters were significantly influenced by water regime, variety and their interaction. The traits like number of effective tillers and non-effective tillers·hill-1, number of filled grains and unfilled grains panicle-1, panicle length, test weight, grain yield and harvest index were higher in irrigated crops compared to rainfed. Considering the performance of yield, the highest grain yield recorded in BRRI dhan27 (irrigated-2.47 t·ha-1 and rainfed-2.26 t·ha-1) followed by BRRI dhan55 (irrigated-1.95 t·ha-1 and rainfed-1.88 t·ha-1), BRRI dhan48 and BRRI dhan43 under both irrigated and rainfed treatment. BRRI dhan27 ranked the top under both irrigated and rainfed conditions, however the relative yield was maximum in BRRI dhan55 (reduction was only 3.59%) followed by Nerica10. Hence, considering the yield stability and absolute yield performance, BRRI dhan27 may be considered favorably cultivable for Aus season in Bangladesh followed by BRRI dhan55, BRRI dhan48 and BRRI dhan43.

Groundwater Level Effect on Redox Potential, on Cadmium Uptake and Yield of Soybean

In this greenhouse experiment, we investigated the effects of two constant groundwater levels: 10 cm groundwater level (GW-10) and 40 cm groundwater level (GW-40) and one change groundwater level, which was 40-10-40 cm (GW-40-10-40) on Cadmium (Cd) uptake and seed yield of Soybean plant in Cd contaminated soils (1.57 mg·kg-1). The experimental soil layer was made with gravel layer (14 cm), non-polluted soil (15 cm) and polluted soil (25 cm). The redox potential of every soil layer was measured from sowing to harvesting. The soil layer (10 – 40 cm) of GW-10 was always in reduction condition and that of GW-40 was always in oxidation condition. First 50 days of GW 40-10-40 were in oxidation and next 50 days in reduction and final 20 days again returned in oxidation condition. Soybean seed Cd concentration was significantly highest in GW-40-10-40 (1.16 ± 0.13 mg·kg-1) and lowest in GW-40 (0.81 ± 0.12 mg·kg-1). Cd concentration of stem was found significantly higher in GW-40 (1.7 ± 0.2 mg·kg-1) than GW-10 (0.91 ± 0.08 mg·kg-1) and GW-40-10-40 (1.28 ± 0.13 mg·kg-1). There was no significant difference in root Cd concentration among these 3 treatments. Main stem height of soybean plant and 100 seed weight of GW-40 were significantly higher than those of GW-10. The result revealed that, soil redox condition is an important factor for Cd uptake in soybean plant and seed yield of soybean. This study will help to manage the farming process more appropriately with the aim of minimizing uptake of Cd and other toxic metals in grain crops.

Seed priming has proved to be an effective pre-germination seed invigoration technique for different crops to improve seed and seedling performance under different abiotic *** Bangladesh,winter rice is very often exposed to cold waves just after sowing in the nursery bed resulting in poor seed germination and seedling emergence,yellowish and thin seedlings production,and a very low survival *** priming may mitigate the cold stress during seed germination and seedling emergence and helps in the quality seedling production of winter *** evaluate the efficacy of different seed priming techniques in increasing seedling emergence,growth,vigor and survivability of winter rice cultivars under cold stress,a pot experiment was conducted at the Department of Agronomy,Bangladesh Agricultural University during December 2018 to January *** experiment comprised two factors,(A)Winter rice variety namely,(i)BRRI dhan29 and (ii)BRRI dhan36;(B)Seed priming agent namely (i)Control(no priming),(ii)20000 ppm NaCl,(iii)30000 ppm NaCl,(iv)20000 ppm KCl,(v)30000 ppm KCl,(vi)20000 ppm CaCl2,(vii)30000 ppm CaCl2,(viii)50 ppm CuSO4,(ix)75 ppm CuSO_(4),(x)10000 ppm ZnSO4,(xi)15000 ppm ZnSO4,(xii)2 ppm Na2MoO4,(xiii)3 ppm Na2MoO4,(xiv)100 ppm PEG(Polyethylene glycol 4000)and (xv)150 ppm *** were sown on two different dates viz.,1st December and 1st January so that seedlings are exposed to cold stress at different *** experiment was laid out in a completely randomized design(CRD)with three *** indicated that(in most of the cases)seed priming has a positive impact on seedling emergence rate(%),root length,shoot length,root shoot ratio,root dry weight,shoot dry weight,seedling dry weight and survival rate(%).Among the priming agents,KCl and CaCl2 performed best;while priming with NaCl and PEG showed no advantages over no priming for both the sowing *** general,BRRI dhan36 performed better than BRRI dhan29 in terms of seedling growth because
