Morphological and Genetical Variability among Rhizoctonia solani Isolates Causing Sheath Blight Disease of Rice

Eighteen isolates of Rhizoctonia solani collected from infected rice plants in four different locations of Bangladesh were studied by using morphological characters and molecular markers. Anastomosis study with a reference isolate confirmed that all the isolates belonged to R. solani. Significant variation was observed in sclerotial size, shape and distribution. Un-weighted pair group method with arithmetic mean dendrogram constructed based on the Gower＇s general similarity coefficient showed that these isolates were grouped into four clusters at the 0.68 similarity coefficent according to morphological characters. Cluster I was a major cluster consisting of 13 isolates, while clusters Ⅱ to Ⅳ consisted of 1 or 2 isolates. Analyses by variable number of tandem repeat and amplified fragment length polymorphism markers showed that the isolates were grouped into five and three clusters at a similarity coefficient of 0.64 and 0.69, respectively. Although most of the variability was found between isolates from different regions as expected, significant variation was observed within the isolates collected from similar agro-ecological regions. Our results suggest the presence of different races of R. solani within the same local geographic regions.

The present study was conducted to analyze the fresh and post-thaw semen quality and fertility from native bulls of Red Chittagong Cattle (RCC), BLRI Cattle Breed 1 (BCB1), and Munshiganj Cattle of Bangladesh. One hundred and seventy-two ejaculates were collected by artificial vagina set and semen analysis was performed using Computer Assisted Sperm Analyzer (CASA) at Bangladesh Livestock Research Institute. Commercial extender (AndroMed) was used to dilute the fresh semen. After equilibration (4°C for 4 hr), freezing was done using a programmable bio-freezer. Post-thawed semen was evaluated for sperm motility and kinematics. Cryopreserved semen straws were used for artificial insemination (AI) and determined the bull fertility based on 60 days non-return rate. Motility of the sperm differs significantly (p 0.05) among the bulls but the highest concentration was found in Munshiganj bull (1669.60 ± 192.07 million/ml) followed by RCC (1648.70 ± 91.07 million/ml) and BCB1 bull (1481.60 ± 167.35 million/ml). Moreover, the highest bent tail (5.89 ± 0.75%), coiled tail (1.01% ± 0.22%) and distal mid-piece reflex (2.26% ± 0.28%) were observed in BCB1 followed by Munshiganj and RCC. Amplitude of lateral head displacement (ALH) was recorded higher in post-thaw than in fresh semen. Kinematics parameters of post-thaw semen decreased than fresh semen irrespective of genotypes. More number of doses/ejaculates can be produced from Munshiganj bull (394.34 ± 127.95) followed by RCC (349.01 ± 120.91) and BCB 1 bulls (331 ± 98.99). Fertility rate does not differ among the bulls (p > 0.05) but the highest value was found for RCC (62.06% ± 1.94%) bulls. Therefore, it can be concise that, the quality of Red Chittagong Cattle semen is better than BCB 1 and Munshigang bull.

To know the position, problems and suggestion of harvest mechanization of rice, the study conducted in 21 upzillas under seven regions (Dhaka, Mymensingh, Sylhet, Comilla, Bogra, Rajshahi and Rangpur) based on different agro-climatic condition and cropping intensity and 126 farmers participated in the interview. For collecting data, a structured questionnaire was used and statistical analysis of the collected data was carried out. In study area, the average cultivable area was 225 to 1239 decimal (1 decimal = 40 m2) and 97.60% farmer reaping by sickle and only 0.80% used combine harvester. Cut paddy transported by head (13.49%), shoulders (12.7%) or both of them of 11.11% farmer and 44.44% of farmers used small truck or field threshed. Threshing was mainly done using (49.21%) closed drum thresher and open drum thresher (26.20%), likewise 1 of 126 farmer use head-feed combine harvester. The study area, most farmers (37.3%) used kula for the cleaning of rice, but only 8.74% of farmers used winnower. In considerable amount, 80.95% of farmers had machinery, while the remaining (19.05%) did not have machines. Farmers agreed that harvesting was a labor-intensive method (35.71%) and time consuming (28.57%). Transporting of harvested paddy was dangerous and painful for head, shoulder, hand, waist, backbone, leg, etc. With mechanization and the introduction of machinery, 40.48% of farmers identified the main ceiling as the price of the machine and information/lack of credit system (21.43%). In survey area, most farmers (36.51%) identified the vital advantage of mechanized harvesting as lower labor requirement, while 21.43% of farmers said that a lower loss of rice. All farmers who participated in the interview expressed their need for machinery and mentioned agricultural credit with easy terms and conditions, subsidy for buying machinery and the ensuring of a fair market price for their rice.

Development of New Submergence Tolerant Rice Variety for Bangladesh Using Marker-Assisted Backcrossing

Submergence tolerant high yielding rice variety was developed using BR11 as a recipient parent applying foreground, phenotypic and background selection approaches. Recombinant selection was found essential to minimize linkage drag by BC2F2 generation. Without recombinant selection, the introgression size in the backcross recombinant lines （BRLs） was approximately 15 Mb on the carrier chromosome. The BRLs were found submergence tolerance compared to the check varieties under complete submergence for two weeks at Bangladesh Rice Research Institute, and produced higher yield compared to the isogenic Subl-line under controlled submerged condition. The BRL IR85260-66-654-Gaz2 was released as BRRI dhan52 in 2010, which was the first high yielding submergence tolerant variety in Bangladesh. BRRI dhan52 produced grain yield ranging from 4.2 to 5.2 t/hm2 under different flash flood prone areas of Bangladesh in three consecutive seasons. The study demonstrated the efficiency of recombinant selection and better adaptability of the newly released submergence tolerant high yielding variety in flash flood prone different areas of the country with respect to submergence tolerance and yield potential.

Reclamation of Coastal Soil Salinity towards Sustainable Rice Production and Mitigating Global Warming Potentials in the Changing Climate

Soil salinity has become a major constraint to rice productivity in the coastal region of Bangladesh, which threatened food security. Therefore, field experiment was conducted at salt stressed Shyamnagor Upazilla of Satkhira district to improve the soil salinity status, sustainable rice production and suppression of global warming potentials. Selected soil amendments viz. trichocompost, tea waste compost, azolla compost and phospho-gypsum (PG) were applied in the field plots one week prior to rice transplanting. In addition, proline solution (25 mM) was applied on the transplanted rice plants at active vegetative stage. Gas samples from the paddy field were collected by Closed Chamber technique and analyzed in by Gas Chromatograph. The 25% replacement of chemical fertilizer (i.e., 75% NPKS) with trichocompost, tea waste compost, Azolla compost and Phospho-gypsum amendments increased grain yield by 4.7% - 7.0%, 2.3% - 7.1% 11.9% - 16.6% and 9.5% - 14.2% during dry boro rice cultivation, while grain yield increments of 5.0% - 7.6%, 2.3% - 10.2%, 12.8% - 15.3% and 10.2% - 15.3% were recorded in wet Aman season respectively, compared to chemically fertilized (100% NPKS) field plot. The least GWPs 3575 and 3650 kg CO2 eq./ha were found in PG Cyanobacterial mixture with proline (T10) and tea waste compost with proline (T8) amended rice field, while the maximum GWPs 4725 and 4500 kg CO2 eq./ha were recorded in NPKS fertilized (100%, T2) and NPKS (75%) with Azolla compost (T5) amended plots during dry boro rice cultivation. The overall soil properties improved significantly with the selected soil amendments, while soil electrical conductivity (EC), soil pH and Na+ cation in the amended soil decreased, eventually improved the soil salinity status. In the present deregulated electricity market, power system congestion is the main complication that an independent system operator (ISO) faces on a regular basis. Transmission line congestion trigger serious problems for smooth functioning in restructured power system causing an increase in the cost of transmission hence affecting market efficiency. Thus, it is of utmost importance for the investigation of various techniques in order to relieve congestion in the transmission network. Generation rescheduling is one of the most efficacious techniques to do away with the problem of congestion. For optimizing the congestion cost, this work suggests a hybrid optimization based on two effective algorithms viz Teaching learning-based optimization (TLBO) algorithm and Particle swarm optimization (PSO) algorithm. For binding the constraints, the traditional penalty function technique is incorporated. Modified IEEE 30-bus test system and modified IEEE 57-bus test system are used to inspect the usefulness of the suggested methodology.
