Estrogen receptor expression in chronic hepatitis C and hepatocellular carcinoma pathogenesis

AIM To investigate gender-specific liver estrogen receptor(ER) expression in normal subjects and patients with hepatitis C virus(HCV)-related cirrhosis and hepatocellular carcinoma(HCC).METHODS Liver tissues from normal donors and patients diagnosed with HCV-related cirrhosis and HCV-related HCC were obtained from the NIH Liver Tissue and Cell Distribution System. The expression of ER subtypes, ERα and ERβ, were evaluated by Western blotting and real-time RT-PCR. The subcellular distribution of ERα and ERβ was further determined in nuclear and cytoplasmic tissue lysates along with the expression ofinflammatory [activated NF-κB and IκB-kinase(IKK)] and oncogenic(cyclin D1) markers by Western blotting and immunohistochemistry. The expression of ERα and ERβ was correlated with the expression of activated NF-κB, activated IKK and cyclin D1 by Spearman's correlation. RESULTS Both ER subtypes were expressed in normal livers but male livers showed significantly higher expression of ERα than females(P < 0.05). We observed significantly higher m RNA expression of ERα in HCV-related HCC liver tissues as compared to normals(P < 0.05) and ERβ in livers of HCV-related cirrhosis and HCV-related HCC subjects(P < 0.05). At the protein level, there was a significantly higher expression of nuclear ERα in livers of HCV-related HCC patients and nuclear ERβ in HCV-related cirrhosis patients as compared to normals(P < 0.05). Furthermore, we observed a significantly higher expression of phosphorylated NF-κB and cyclin D1 in diseased livers(P < 0.05). There was a positive correlation between the expression of nuclear ER subtypes and nuclear cyclin D1 and a negative correlation between cytoplasmic ER subtypes and cytoplasmic phosphorylated IKK in HCV-related HCC livers. We present here for the first time,the Raman and infrared spectroscopic investigation of amphiboles from the World's deepest borehole,the Kola super-deep borehole,at the depth of 11.66 *** Kola Super-deep borehole(SG-3)(henceforth referred as KSDB)is located in the northwest of the Kola Peninsula in the northern frame of the Pechenga structure,*** was drilled in the north-eastern part of the Baltic Shield(69о5’N,30о44’E)and reached a depth of 12.262 *** has been drilled in the northern limb of the Pechenga geosyncline composed of rhythmically inter-bedded volcanogenic and tuffaceous-sedimentary strata extending to the NW at 300°–310°and dipping to SW at angles of 30°–50°.The SG-3 geological section is represented by two complexes–Proterozoic and *** facies is dominant in the depth region from 6000 m to 12,000m to the *** Raman spectra of the sample reveal abundant presence of plagioclase and *** most distinct Raman peak in this study indicates the tremolite-ferro-actinolite rich enrichment of the borehole samples at this depth corroborating earlier conventional petrographic studies.

Changes in symmetry of anterior chamber following routine cataract surgery in non-glaucomatous eyes

Background:To assess minimum and maximum changes in anterior chamber dimensions following routine cataract surgery in non-glaucomatous ***:Forty-two eyes(42 participants)underwent routine cataract surgery with same surgeon and were assessed preoperatively,1,3,6 and 12 months *** outcome measure:Angle-to-angle diameter(AAD)(at 0-180°,45-225°,90-270°,135-315°),Anterior-chamber-angle(ACA)(at 0°,45°,90°,135°,180°,225°,270° and 315°)and central anterior chamber depth(ACD)at all *** outcome measures:relationship to axial length(AL).Results:The mean AAD and ACA increased post-operatively in all meridians at all visits *** 12 months,there was a maximum change in AAD in horizontal meridian(506.55±468.71μm)and least in vertical meridian(256.31±1082.3μm).The mean percentage increase in ACA postoperatively was least at 90o(5%increase compared to 29–35%elsewhere).Central ACD deepened at all postoperative visits and this did not change over 12 *** was no correlation between AAD,ACA and ACD with AL at any ***:The AAD,ACA and ACD increases following cataract surgery in non-glaucomatous eyes,but at 12 months increase in AAD is least in vertical compared to horizontal ***,ACA was narrower(only 5%increase)superiorly compared to elsewhere(29–35%increase in ACA).This may have implications with regards to surgeries performed in the anterior chamber and corneal endothelial cell loss.

Identification of SNP and InDel variations in the promoter and 5′ untranslated regions of γ-tocopherol methyl transferase(ZmVTE4) affecting higher accumulation of α-tocopherol in maize kernel

Deficiency of vitamin-E or tocopherol causes neurological and cardiovascular disorders in humans. Though maize kernel is rich in total tocopherol, the level of α-tocopherol possessing the highest vitamin-E activity is low. Mutant allele of ZmVTE4 with deletion of 7 bp and 118 bp (0/0: most favorable haplotype) in 5′UTR and promoter region, respectively significantly enhances α-tocopherol in maize kernel than wild type haplotype (7/118).α- tocopherol estimation in 15 diverse inbreds revealed that mean α-/γ-tocopherol and α-/ total-tocopherol was much higher in genotypes with favorable haplotype (1.51 and 0.41) than unfavorable class (0.19 and 0.13), respectively. However even within favorable class,α- tocopherol ranged from 4.76 to 30.07 μg g 1. Sequence analysis of part of 5′UTR and promoter of ZmVTE4 among the genotypes with favorable haplotype revealed 14 SNPs (SNP1 to SNP14) and eight InDels (InDel1 to InDel8). SNP7 at 606 bp (G to A), and InDels viz., InDel1 (27 bp), InDel4 (27 bp) and InDel8 (14 bp) differentiated low and high α-tocopherol accumulating inbreds with favorable haplotype. Hence the newly identified SNP and InDels in addition to the already reported InDels can be useful in selection of favorable genotypes with higher α-tocopherol in maize.

A Review on Therapeutic Intervention of Yoga and Ayurveda in Post-Partum Depression

Introduction: Postpartum depression often starts two weeks to a month after delivery and can linger for months or even a year. According to the World Health Organization, depression affects roughly 10% of pregnant women worldwide and 13% of women in the immediate postpartum period. In India, the Prevalence of Postpartum Depression (PPD) varies from 15% to 20%. Yoga, which originated in ancient India, is recognized as an alternative medicine practice that incorporates mind-body practices. Methodology: This is a review (narrative) study, in which literature has been evaluated using electronic search in databases of PubMed, Google Scholar, Science Direct, ResearchGate, etc. Searching in the databases was made using keywords of postpartum depression, yoga in postpartum depression. There were many articles on yoga in depression but those articles were included in the study that had done research on postpartum depression, which were of research and review article type. Result: This review study includes 4 interventional studies that showed a highly significant (p < 0.001) decrease in depression after the performance of yoga on postpartum women. Conclusion: This study showed the result that different limbs of yoga (i.e., Dhyana, Asanas, and Pranayama, etc.) could considerably reduce depression symptoms in postnatal women. This would also help in reducing the use of antidepressant medications. Therefore, it is recommended that yoga training should be employed in postnatal treatment.

Ultrasonographic assessment of paediatric ocular emergencies： A tertiary eye hospital based observation

BACKGROUND: The purpose of this study is to assess the utility of ocular ultrasound B scan in the emergency at the fi rst point of care for detecting posterior segment and orbital pathologies in cases of paediatric ocular ***: A prospective observational study involving 122 paediatric patients presenting to eye emergency over a period of ninety days were assessed with ultrasonography for the posterior segment as well as orbital pathology whenever indicated. The ocular ultrasound was performed gently over closed ***: Posttraumatic globe injuries were the most common indication for posterior segment evaluation, which constituted 80(65.57%) eyes. Among these 52 patients had an anechoic posterior segment and 28 patients had variable findings such as vitreous haemorrhage(8.19%), retinal detachment(6.55%), choroidal detachment(4.91%), posteriorly dislocated clear lens(0.81%) and retained intraocular foreign body(5.73%). Non-traumatic cases constituted around 42(34.42%) eyes, which included corneal ulcer(7.37%), retinoblastoma(6.55%), endophthalmitis(4.91%), extraocular muscle cysticercosis(4.91%), orbital cellulitis(4.09%), periocular haemorrhage(2.45%), proptosis(1.63%), paediatric cataract(1.63%) and cryptophthalmos(0.81%). No adverse events of performing the ultrasound was ***: First point ultrasonography in paediatric ocular emergencies is a cheap, portable and an effective tool in the assertion of signifi cant posterior segment and orbital diseases.

Objective: To investigate new scolicidal agent from natural resources to cope with the side effects associated with synthetic drugs in Echinococcosis. Methods: The scolicidal potential of methanolic fruit powder extract (10 and 20 mg/mL) of Mallotus philippinensis ( M. philippinensis ) was investigated. Viability of protoscoleces was confirmed by trypan blue exclusion method, where mortality was observed at concentration of 10 and 20 mg/mL in 60 min treatment against Echinococcus granulosus ( E. granulosus ), under in-vitro conditions with reference to the known standard drug Praziquantel . Results: At concentration 10 and 20 mg/mL, the mortality rate was observed 97% and 99% respectively for 60 min treatment; while up to 93% mortality was observed with 20 mg/mL for only 10 min treatment. The concentration above 20 mg/mL for above 2 h showed 100% mortality, irrespective of further incubation. Conclusions: As compared with the standard anti-parasitic drug Praziquantel our extract has significant scolicidal activity with almost no associated side effects.
