Huanglongbing (HLB) or citrus greening is currently the most important citrus disease, caused by the bacterium Candidatus Liberibacter asiaticus (CLas). The impossibility of isolating it causes understanding its pathogenic mechanisms to be a complicated task. Recent studies identified 16 proteins with the signal peptide needed to be secreted in the plant and cause the disease. The present study aims to perform a bioinformatic analysis of these proteins with the function prediction approach by gene ontology (GO) and the detection of conserved domains. It was observed that of the 16 proteins analyzed not all are found in different infective strains reported in the literature. The GO analysis allowed us to relate different proteins with the biological process of energy and pathogenic activity, especially CLIBASIA_03315 and CLIBASIA_05115, respectively. The domain analysis allowed the observation of a β-CA domain, tentatively related to the damage caused to the chloroplast and a PAAR domain associated with the T6SS secretory system. Our results provide information on the possible function of potential pathogenicity effectors in CLas.

American trypanosomiasis is a zoonosis of worldwide medical importance and currently there is no effective treatment in chronic patients, hence the importance of the study of protein function of the parasite with the objective of finding new drug targets and to know better the biology of the agent causal (Trypano-soma cruzi). T. cruzi is an RNAi-negative parasite, therefore the silencing genes strategies by RNAi is not possible;for that reason, antibodies may be taken as a tool for studying the parasite proteins function by blocking these molecules with specific antibodies. The aim of this work was to establish a methodology for antibody delivery (antibody transfection) into viable parasites. We used anti-cyclin-A antibody (human origin) in western blot assay with epimastigote of T. cruzi proteins and this recognized a ~55 kDa polypeptide. Several methods for antibody transfection (electroporation, saponin permeabilization and a lipid-based formulation) were tested. The first two methods were unsuccessful. In electroporation was impossible to visualize the antibody inside parasites and with saponin permeabilization, antibodies were successfully introduced, but with loss of parasites viability. The lipid-based formulation method forms noncovalent complexes with antibodies. These complexes are internalized by cells and antibodies are released into the cytoplasm. With this method, a successful antibody delivery was achieved. Anti-cyclin antibodies were visualized in the cytoplasm from fixed transfected parasites (immunofluorescence assays). At 24 h post-transfection, parasites maintained their viability (90%) and were able to arrest the cell cycle in G0/G1-phase of cultured epimastigotes (cell population increased in G0/G1-phase from 50.5% to 66.2% and decreased in S-phase from 47.2% to 26%). Effect of Pre-Analytical Conditions on Salivary Nitrite Levels

The aim of the present paper was to analyze the influence of sampling and storage procedures on nitrite con- centration values in the saliva of healthy persons. The samples were obtained and stored under varied condi- tions, and processed using the Griess method. Results: when the salivary nitrite concentration was measured immediately after collection a significant dependence on the collection time was observed. A mean value of 94 μmol/L (range 3 - 625) was obtained at 8:30 am. This value decreased significantly with time (p < 0.05) reaching a value of 68 at 12:30 noon. Concerning the sample storage, a significant increase in the nitrite concentration was observed after 2 hrs, either at 4?C or at room temperature (p < 0.05). In spite of the high variability between individuals the values for each individual showed a marked constancy independent of the sampling day. According to our results, by controlling pre-analytical parameters, principally sampling and storage procedures, reproducibility is improved.

Occurrence of tetracycline residues and antimicrobial resistance in gram negative bacteria isolates from cattle farms in Spain

The usage of antibiotics in animal husbandry has dramatically increased the concentration of antibiotic residues and has promoted the development and abundance of antibiotic resistance in manure. When it is spread onto agricultural land, both residues of antibiotics and bacteria carrying antibiotic resistance genes may be introduced into the environment. In this research, we isolated resistant gram negative bacteria from manure produced in two dairy and two beef cattle farms, located in Madrid (Spain), to determine their resistance to seventeen representative antibiotics commonly used in veterinary therapy. A total of 63 isolates were used to assess the overall bacterial antimicrobial resistance on cattle manure samples. Predominant species were Escherichia coli and Comamonas testosteroni accounting for 25% and 19.6% of the total, respectively. The most found antimicrobial resistances in gram-negative bacteria were to tetracycline (66.7%), sulphamethoxazole (55.6%), ampicillin (52.4%), cephalothin (46.0%), chloramphenicol (44.4%), nalidixic acid (39.7%) and trimethroprim- sulphamethoxazole (33.3%). The mean of resistance and the percentage of multi-resistant bacteria in beef farms were higher and statistically significant when compared to dairy farms which is opposite from the findings of the previous studies. The presence of three tetracyclines in all manure samples was also examined with stable recoveries (76% - 82%) and high sensitivity (limit of quantification 0.015 - 0.03 μg/kg). The concentrations of tetracyclines detected (<0.015 - 10 mg/kg) were consistent to the theoretical tetracycline levels in manure in Spain according to the ex- cretion rate of these antibiotics and the values re-ported in scientific literature in other European coun- tries.

Background: Primary effusion lymphoma (PEL) is a lymphoid proliferation related to Kaposi sarcoma herpesvirus 8/human herpesvirus 8 (KSHV/HHV8) that affects mainly human immunodeficiency virus (HIV) infected individuals but can also occur in other immunodeficiency settings. It is characterized by lymphomatous effusions in different serous body cavities without the presence of a detectable tumor mass. The diagnosis is challenging and the clinical outcomes are poor. Aim: The aim of this paper is to report a rare case of PEL in a man who have sex with women (MSW) with HIV-1/2 infection, history of visceral Kaposi sarcoma (KS) and the development of a seronegative arthritis previous to the lymphoproliferative disease diagnosis. PEL presented with ascites, was treated with high-dose chemotherapy and autologous stem cell transplantation, with a good clinical outcome. Case Presentation: We describe a case of a 48-year-old HIV-1/2-infected patient from a high HHV8 seroprevalent country, hospitalized following a three-month history of increased abdominal volume and general constitutional symptoms. Laboratory data revealed normocytic normochromic anemia and a high level of lactate dehydrogenase. A diagnostic paracentesis was performed with cytology compatible with high-grade B-cell lymphoma. Peritoneal fluid cytology showed large lymphoid cells expressing leucocyte-common antigen CD45 without expression of the CD20 antigen (B-lymphocytes) and positivity for HHV8 by immunocytochemical staining, compatible with the diagnosis of PEL.

Background: In patients with Parkinson’s disease (PD), real-time quaking-induced conversion (RT-QuIC) detection of pathological α-synuclein (α-syn) in olfactory mucosa (OM) is not as accurate as in other α-synucleinopathies. It is unknown whether these variable results might be related to a different distribution of pathological α-syn in OM. Thus, we investigated whether nasal swab (NS) performed in areas with a different coverage by olfactory neuroepithelium, such as agger nasi (AN) and middle turbinate (MT), might affect the detection of pathological α-syn. Methods: NS was performed in 66 patients with PD and 29 non-PD between September 2018 and April 2021. In 43 patients, cerebrospinal fluid (CSF) was also obtained and all samples were analyzed by RT-QuIC for α-syn. Results: In the first round, 72 OM samples were collected by NS, from AN (NSAN) or from MT (NSMT), and 35 resulted positive for α-syn RT-QuIC, including 27/32 (84%) from AN, 5/11 (45%) from MT, and 3/29 (10%) belonging to the non-PD patients. Furthermore, 23 additional PD patients underwent NS at both AN and MT, and RT-QuIC revealed α-syn positive in 18/23 (78%) NSAN samples and in 10/23 (44%) NSMT samples. Immunocytochemistry of NS prepara-tions showed a higher representation of olfactory neural cells in NSAN compared to NSMT. We also observed α-syn and phospho-α-syn deposits in NS from PD patients but not in controls. Finally, RT-QuIC was positive in 22/24 CSF samples from PD patients (92%) and in 1/19 non-PD. Conclusion: In PD patients, RT-QuIC sensitivity is significantly increased (from 45% to 84%) when NS is performed at AN, indicating that α-syn aggregates are preferentially detected in olfactory areas with higher concentration of olfactory neurons. Although RT-QuIC analysis of CSF showed a higher diagnostic accuracy compared to NS, due to the non-invasiveness, NS might be considered as an ancillary procedure for PD diagnosis.

Developing new imaging methods needs to establish some proofs of concept before implementing them on real-time scenarios. Nowadays, the high computational power reached by multi-core CPUs and GPUs have driven the development of software-based beamformers. Taking this into account, a library for the fast generation of ultrasound images is presented. It is based on Synthetic Aperture Imaging Techniques (SAFT) and it is fast because of the use of parallel computing techniques. Any kind of transducers as well as SAFT techniques can be defined although it includes some pre-built SAFT methods like 2R-SAFT and TFM. Furthermore, 2D and 3D imaging (slice- based or full volume computation) is supported along with the ability to generate both rectangular and angular images. For interpolation, linear and polynomial schemes can be chosen. The versatility of the library is ensured by interfacing it to Matlab, Python and any programming language over different operating systems. On a standard PC equipped with a single NVIDIA Quadro 4000 (256 cores), the library is able to calculate 262,144 pixels in ≈105 ms using a linear transducer with 64 elements, and 2,097,152 voxels in ≈ 5 seconds using a matrix transducer with 121 elements when TFM is applied.

Much of the genetic predisposition to polyposis,and particularly to serrated polyposis(SP),remains *** germline pathogenic variants in RNF43,a tumor suppressor that exerts negative feedback in the Wnt/β-catenin signaling pathway,have been causally linked to some SP cases(<2%),a disease associated with increased risk of colorectal cancer(CRC).^(1) Most known hereditary CRC and polyposis genes affect DNA repair,BMP/TGF-β,or Wnt signaling,being the latter associated with adenomatous and serrated polyposis phenotypes.2 Based on this observation,we evaluated the presence and role of germline variants in those pathways in unsolved polyposis patients.
