Oxidative DNA Damage Is Elevated in Renal Patients Undergoing Haemodialysis

Background: End stage renal disease (ESRD) is associated with an increase in oxidative stress, cardiovascular disease and cancer. The main treatment for ESRD is haemodialysis (HD), which itself induces repetitive bouts of oxidative stress through membrane biocompatibility and endotoxin challenge. The resulting higher levels of reactive oxygen species in turn produce increased levels of oxidative DNA damage leading to genomic instability which may influence the higher risk of cancer reported in HD patients. Our aims were to measure levels of oxidative DNA damage in HD patients and in age and gender matched control volunteers. Methods: Thirty eight patients receiving HD in the Western Health and Social Services Trust (WHSCT) and 8 healthy volunteers were recruited. Volunteers gave informed consent and non-fasting morning blood samples were taken and assessed for DNA disruption using the comet assay modified to identify oxidative specific damage. Results: The HD patients had significantly elevated levels of alkaline DNA damage (19.46% ± 1.37% vs 3.86% ± 1.36% tail DNA, p < 0.05) and oxidative DNA damage formamidepyrimidine DNA glycosilase (5.81% ± 1.08% vs 1.23% ± 0.43% tail DNA, p < 0.01) and endonuclease III (6.04% ± 1.00% vs 1.98% ± 0.70% tail DNA, p < 0.01) compared to controls, respectively. A positive correlation was observed between the duration on dialysis (months) and levels of Endo III specific damage (p = 0.041). Conclusion: The significant increase in oxidative DNA damage and the positive correlation with duration of HD treatment and Endo III damage may contribute to the increased cancer risk observed in this patient group. Studies are required to investigate the best way to reduce this damage.

Effects of Planting Methods and Seed Density on Vegetable Yield and Nutrient Composition of <i>Solanum macrocarpon</i>and <i>Solanum scabrum</i>in Southwest Nigeria

Two field studies were carried out using Solanum macrocarpon and Solanum scabrum to investigate the effects of planting methods and seed density required for optimum biomass yield and nutrient content in southwest Nigeria. Two planting methods (drilling and broadcasting), and two seed densities (4 and 8 table spoon full) were investigated as a 2×2×3 factorial experiment in randomized complete block design and replicated three times. Four seed spoons gave 25.78 kg seeds ha-1 for S. macrocarpon and 20.67 kg·seeds·ha-1 for S. scabrum. Significant highest S. macrocarpon shoots (2.75 kg·m-2) and Ca, K, Fe and Zn contents and S. scabrum (2.95 kg·m-2) and Zn content, were obtained with drilling and four spoons of seeds compared with broadcasting and 8 spoons of seeds. This treatment gave average values for N, P, K, Mg, Ca, Na, Fe, Zn, Mn, Cu, Ca/P and Na/K ratio were 2.79%, 0.03%, 3.37%, 1.64%, 0.02%, 300 mg·kg-1, 100 mg·kg-1, 300 mg·kg-1, 8 mg·kg-1, 1.22 and 0.008 for S. macrocarpon and 2.05%, 0.27%, 5.81%, 3.27%, 2.91%, 0.09%, 280 mg·kg-1, 52.99 mg·kg-1, 359.35 mg·kg-1 and 22.42 mg·kg-1, 11.63 and 0.016 for S. scabrum. It was concluded that planting in drilling made weeding, fertilizer application, irrigation and harvesting more effective rather than by broadcasting, and four spoons of seeds per 9 m-2 produced deep green and broader leaves and balanced nutrient contents than eight spoons of seeds.

Background:The incorporation of legumes,specifically alfalfa(Medicago sativa L.),into bermudagrass(Cynodon spp.)-based pasture systems in the southeastern United States has increased in recent years as an alternative to synthetic nitrogen(N)***:A small plot evaluation was conducted in Shorter,Alabama,and Tifton,Georgia,USA,to evaluate the impact of harvest height(HH)and harvest frequency(HF)on agronomic characteristics of alfalfa+bermudagrass mixtures in southeastern United ***:Results from both locations revealed that the longer the HF and the shorter the HH,the greater the alfalfa retention was in the stand(p<0.01).HH did not impact any of the reported nutritive value parameters,while longer HF resulted in lower total digestible nutrients,lower crude protein,higher acid detergent fiber,and lower 48 h in vitro dry matter digestibility(p<0.01).Both HH and HF impacted forage accumulation at both locations(p<0.01).HH resulted in different trends at each location,while longer frequencies generally increased forage ***:This research confirmed recent findings from comparable evaluations in the southeastern United States,in that increasing HH and decreasing HF improved alfalfa retention while having a negligible effect on nutritive value or forage accumulation.

Assessment of the knowledge, attitude and practice of rural women of northeast Nigeria on risk factors associated with cancer of the Cervix

Cervical cancer is the leading cancer-related cause of death among women in Nigeria. An estimated 70,700 new cases occur each year, representing one quarter of all female cancers in sub Saharan Africa. The magnitude of the problem has been under recognised and under prioritised compared with the competing health priorities of infectious diseases such as HIV/ AIDS, tuberculosis and malaria. Studies in the United States and Nigeria have indicated that the disease has the highest incidence among the lowest socio-economic groups especially residing in rural areas. The peak age for the disease has been shown to be within 35-45 years age group. Knowledge of the risk factors of the disease is deemed important in its early detection and prevention. The objective of this study was to assess the knowledge, attitude and practice of rural women with cancer of the cervix. A cross sectional descriptive study was conducted among 1600 rural women aged 15-55 years (randomly selected from 28 villages) who were interviewed using a structured questionnaire between April and june, 2010. The majority (82.2%) were married before the age of 20 years and 19.3% before 15 years, 40% in polygamous union, 22.6% have had 2 or more sexual partners, 71.3% were primi and grand multiparous, 7.5% have had previous treatment for STIs and 10.1% were on various types of contraceptive. 454 (28.4%) have heard of Ca cervix, 358 (22.4%) knew the location of the cervix. 2.3% had Pap smear test of which 72.6% were within 2 years. The majority (89.9%) will avail themselves for screening.

Background:The public health response to COVID-19 has shifted to reducing deaths and hospitalizations to prevent overwhelming health *** amount of SARS-CoV-2 RNA fragments in wastewater are known to correlate with clinical data including cases and hospital admissions for *** developed and tested a predictive model for incident COVID-19 hospital admissions in New York State using wastewater ***:Using county-level COVID-19 hospital admissions and wastewater surveillance covering 13.8 million people across 56 counties,we fit a generalized linear mixed model predicting new hospital admissions from wastewater concentrations of SARS-CoV-2 RNA from April 29,2020 to june 30,*** included covariates such as COVID-19 vaccine coverage in the county,comorbidities,demographic variables,and holiday ***:Wastewater concentrations of SARS-CoV-2 RNA correlated with new hospital admissions per 100,000 up to ten days prior to *** that included wastewater had higher predictive power than models that included clinical cases only,increasing the accuracy of the model by 15%.Predicted hospital admissions correlated highly with observed admissions(r¼0.77)with an average difference of 0.013 hospitalizations per 100,000(95%CI¼[0.002,0.025])Interpretation:Using wastewater to predict future hospital admissions from COVID-19 is accurate and effective with superior results to using case data *** lead time of ten days could alert the public to take precautions and improve resource allocation for seasonal surges.

Heart disease prognosis(HDP)is a difficult undertaking that requires knowledge and expertise to predict early *** failure is on the rise as a result of today’s *** healthcare business generates a vast volume of patient records,which are challenging to manage *** it comes to data mining and machine learning,having a huge volume of data is crucial for getting meaningful *** methods for predictingHDhave been used by researchers over the last few decades,but the fundamental concern remains the uncertainty factor in the output data,aswell as the need to decrease the error rate and enhance the accuracy of HDP assessment ***,in order to discover the optimal HDP solution,this study compares multiple classification algorithms utilizing two separate heart disease datasets from the Kaggle repository and the University of California,Irvine(UCI)machine learning *** a comparative analysis,Mean Absolute Error(MAE),Relative Absolute Error(RAE),precision,recall,fmeasure,and accuracy are used to evaluate Linear Regression(LR),Decision Tree(j48),Naive Bayes(NB),Artificial Neural Network(ANN),Simple Cart(SC),Bagging,Decision Stump(DS),AdaBoost,Rep Tree(REPT),and Support Vector Machine(SVM).Overall,the SVM classifier surpasses other classifiers in terms of increasing accuracy and decreasing error rate,with RAE of 33.2631 andMAEof 0.165,the precision of 0.841,recall of 0.835,f-measure of 0.833,and accuracy of 83.49 percent for the dataset gathered from *** SC improves accuracy and reduces the error rate for the Kaggle dataset,which is 3.30%for RAE,0.016 percent for MAE,0.984%for precision,0.984 percent for recall,0.984 percent for f-measure,and 98.44%for accuracy.

Osteoporosis and People with Down Syndrome: A Preliminary Descriptive Examination of the Intellectual Disability Supplement to the Irish Longitudinal Study on Ageing Wave 1 Results

Background: People with Down syndrome (DS) are a high “at risk” group to develop osteoporosis. Increased morbidity associated with older age, higher prevalence of Alzheimer dementia, hypotonia, hypogonadism, and thyroid disease, are all features of DS and predispose this cohort to musculoskeletal complications. Methods: The Intellectual Disability Supplement to The Irish Longitudinal Study on Ageing (IDS-TILDA) is a representative study of the aging of adults with intellectual disability in Ireland. The sample (N = 753) included 147 people with DS. Data was gathered on participants’ health status, behavioural health, health screenings and activities of daily living. The prevalence of osteoporosis and related risk factors among people with DS was specifically examined. Findings: Of the 147 participants with DS, 9.4% reported a doctor’s diagnosis of osteoporosis;a much lower figure than prevalence of risk factors would suggest. Predisposing factors identified included higher than general population rates of thyroid disease (37.4%), epilepsy (19.3%), sedentary lifestyle (51.7%) and the majority of the females reporting having experienced menopause (61%). Bone health screening was low at (8%) despite the presence of such high levels of risk factors in this population. Conclusion: Given the risk factor findings and the hidden nature of osteoporosis, underreported incidence among people with Down syndrome seems probable. Further investigations and systematic screening are required.
