Polyphenols as potential enhancers of stem cell therapy against neurodegeneration

The potential of polyphenols for treating chronic-degenerative diseases(particularly neurodegenerative diseases)is attractive.However,the selection of the best polyphenol for each treatment,the mechanisms by which they act,and their efficacy are frequently discussed.In this review,the basics and the advances in the field,as well as suggestions for using natural and synthetic polyphenols alone or in a combinatorial strategy with stem cell assays,are compiled and discussed.Thus,stem cells exhibit several responses when polyphenols are added to their environment,which could provide us with knowledge for advancing the elucidation of the origin of neurodegeneration.But also,polyphenols are being included in the innovative strategies of novel therapies for treating neurodegenerative diseases as well as metabolic diseases related to neurodegeneration.In this regard,flavonoid compounds are suggested as the best natural polyphenols due to their several mechanisms for acting in ameliorative effects;but increasing reports are involving other polyphenols.Even if some facts limiting bioactivity prevent them from conventional use,some natural polyphenols and derivatives hold the promise for being improved compounds,judged by their induced effects.The current results suggest polyphenols as enhancers of stem cell therapy against the targeted diseases.

Diabetes is a disease with a high global burden.Current strategies have failed to limit the advancement and impact of the disease.Successful early diagnosis and treatment will require the development of new agents.In this sense,boroncontaining compounds have been reported as agents with the ability to reduce glycemia and lipidemia.They have also been used for labeling and measuring carbohydrates and other molecules linked to the initial stages of diabetes and its progression.In addition,certain boron compounds bind to molecules related to diabetes development and their biological activity in the regulation of elevated glycemia.Finally,it should be noted that some boron compounds appear to exert beneficial effects on diabetes complications such as accelerating wound healing while ameliorating pain in diabetic patients.

Global longitudinal strain is superior to ejection fraction for detecting myocardial dysfunction in end-stage renal disease with hyperparathyroidism

BACKGROUND The estimation of left ventricular ejection fraction(LVEF)by 2D echocardiography(2D-ECHO)is the most used tool to assess LV systolic function(LVSF).Global longitudinal strain(GLS)has recently been suggested as a superior method for several evaluations.This study explored the association and prevalence of LV systolic dysfunction(LVSD)by using these methods in patients with end-stage renal disease(ESRD)and severe hyperparathyroidism(SHPTH);both associated with cardiovascular events(CEs).AIM To evaluate the myocardial function in patients with ESRD and SHPTH by using the GLS and LVEF measured through conventional 2D-ECHO.METHODS In 62 patients with ESRD and SHPTH,asymptomatic,and without a history of CEs,LVSF was evaluated by 2D-ECHO,obtaining the EF,by the Simpson biplane method,and GLS by speckle tracking.RESULTS The total patients with ESRD had a preserved LVEF(>50%)but abnormal GLS(<13.55%).Additionally,multivariate analysis showed an independent association of GLS and serum parathyroid hormone(PTH),LV mass index,and hemoglobin.Also,PTH was independently associated with lateral e'wave and tricuspid regurgitation velocity.CONCLUSION In patients with SHPTH linked to ESRD,the use of GLS by 2D-ECHO is a more sensitive tool than LVEF for detecting LVSD.

Impacts of electrochemical disinfection on the viability and structure of the microbiome in secondary effluent water

Electrochemical disinfection(ECD)is a promising disinfection technique for wastewater reclamation;however,the impacts of ECD on the microbiome in secondary effluent wastewater remain unknown.In this study,Propidium monoazide-qPCR(PMA-qPCR)and the plate count method were used to evaluate the inactivation performance,and the PMA-16S rRNA gene sequences of living cells were targeted to study the microbiome.A discrepancy was found between PMA-qPCR and the plate count method in the evaluation of cell count,with increases of 1.5 to 2.2 orders of magnitude in the disinfection rate after 150 s of disinfection.However,the cell count recovered and occasionally exceeded original levels within 3 d after disinfection.Biodiversity was suppressed after ECD,but the microbiome after 150 s disinfection retained a higher level of evenness and stability in the community with a median Shannon index(>3.7).Pathogenic bacteria remained high in relative abundance even after 150 s of 25 V disinfection,but the biofilm-forming population was effectively suppressed by ECD.The co-occurrence network revealed a centralized and fragile network as disinfection persisted,demonstrating the destabilizing effects of ECD on the microbiome.Functional pathways for cell membrane synthesis and organic compound degradation were enriched after ECD.The reaction of the microbiome after ECD was similar to other disinfection techniques in terms of community structure.

Oxidative stress （OS） is a key process in the development of many neurodegenerative diseases, memory disorders, and other pathological processes related to aging. Tibolone （TIB）, a synthetic hormone used as a treatment for menopausal ymptoms, decreases lipoperoxidation levels, prevents memory impairment and learning disability caused by ozone （O3） exposure. However, it is not clear if TIB could prevent the increase in phosphorylation induced by oxidative stress of the microtubule-associated protein Tau. In this study, the effects of TIB at different times of administration on the phosphorylation of Tau, the activation of glycogen synthase kinase-3β （GSK3β）, and the inactivation of Akt and phosphatases PP2A and PTEN induced by O3 exposure were assessed in adult male Wistar rats. Rats were divided into 10 groups： control group （ozone-free air plus vehicle [C]）, control ＋ TIB group （ozone-free air plus TIB 1 mg/kg [C ＋ TIB]）; 7,15, 30, and 60 days of ozone exposure groups [O3] and 7, 15, 30, and 60 days of TIB 1 mg/kg before ozone exposure groups [O3 ＋ TIB]. The effects of O3 exposure and TIB administration were assessed by western blot analysis of total and phosphorylated Tau, GSK3β, Akt, PP2A, and PTEN proteins and oxidative stress marker nitrotyrosine, and superoxide dismutase activity and lipid peroxidation of malondialdehyde by two different spectrophotometric methods （Marklund and TBARS, respectively）. We observed that O3 exposure increases Tau phosphorylation, which is correlated with decreased PP2A and PTEN protein levels, diminished Akt protein levels, and increased GSK3β protein levels in the hippocampus of adult male rats. The effects of O3 exposure were prevented by the long-term treatment （over 15 days） with TIB. Malondialdehyde and nitrotyrosine levels increased from 15 to 60 days of exposure to O3 in comparison to C group, and superoxide dismutase activity decreased. Diagnostic errors are prevalent in critical care practice and are associated with patient harm and costs for providers and the healthcare system.Patient complexity,illness severity,and the urgency in initiating proper treatment all contribute to decision-making errors.Clinician-related factors such as fatigue,cognitive overload,and inexperience further interfere with effective decision-making.Cognitive science has provided insight into the clinical decision-making process that can be used to reduce error.This evidence-based review discusses ten common misconceptions regarding critical care decision-making.By understanding how practitioners make clinical decisions and examining how errors occur,strategies may be developed and implemented to decrease errors in Decision-making and improve patient outcomes.

More than boric acid:Increasing relevance of boron in medicine

Although boron has been a chemical element of interest since the ancient times,only a few boron-containing compounds(BCCs)had been used for medicinal purposes before the 21^(st) century.Among these,only boric acid has been explored in multiple therapeutic applications.Hence,it is common to extrapolate from boric acid to all BCCs,supposing a similar biological effect.However,boric acid is just one of dozens of BCCs in nature and thousands available from chemical synthesis.Nowadays,there is a boom in research on new BCCs as potential tools in the prevention,diagnosis and therapy of human disease.We herein discuss the new role of BCCs in drug development,with emphasis on the compounds for which a mechanism of action has been proposed or demonstrated.Because of data gathered in recent years,BCCs have expanded beyond the well-known fields of antimicrobial and antineoplastic agents,now being explored for their possible use as enzyme inhibitors,regulators of protein expression and modulators of the immune response,as well as in biomaterials.We suggest that translational medicine can accelerate the medicinal applications of BCCs,which is especially important for the human diseases that are generating a high global burden.

在油气井完整性评估技术中,测井数据的三维可视化是必不可少的手段。沿着井的实际三维轨迹进行测井数据或图像的可视化,对于数据的理解、分析与解释会有极大的帮助。本文引入专业的大地测量学坐标系统,开发了井下坐标计算的定位数据融合算法,对测井数据解释中的三维可视化方法依据一般的、具有内在一致性的坐标系统进行了拓展。我们将这种方法应用于套管井测井中的油套管情况的绘制,得到了接近真实井下场景的效果,为数据的直观理解与解释提供了很大的便利。

测井解释软件是以直观、高效的方式分析测井数据的重要工具,现代测并数据解释都是通过计算机软件完成的。常用的测井解释软件一般是基于Windows操作系统,而用于开发解释软件的计算机语言有C++、C#Java等。然而,随着测井仪器数据处理方法越来越复杂,单独使用C++或C#开发核心算法模块变得越来越困难。因此,直接利用科学计算常用的工具MATLAB编写算法,然后编译成DLL模块嵌入到解释软件中是一个可行的、便利的选择。不同的计算机语言在运行效率、开发学习难易程度、第三方资源应用等方面都有其独特性,因此在测并数据解释软件开发中,为每个模块选择合适的开发语言对于降低整个系统开发难度至关重要。格威公司的ViewWellTM套管井解释平台在开发过程中高效的利用了如VisualC++、C#、MATLAB、Fortran、OpenGL等编程语言及系统工具在各方面的长处,通过多源代码快速、完备地实现了格威公司常用套管并仪器的解释算法库,保证了解释软件的正常运行,实现了各类型套管井测并仪器算法库模块输出的曲线在二维、三维环境中的高效显示。

AIM: To evaluate the effect of an extract of Geranium schiedeanum(Gs) as a hepatoprotective agent against ethanol(Et OH)-induced toxicity in rats. METHODS: Male Wistar rats weighing 200-230 g were subjected to a 70% partial hepatectomy(PH); they were then divided into three groups(groups 1-3). During the experiment, animals in group 1 drank only water. The other two groups(2-3) drank an aqueous solution of Et OH(40%, v/v). Additionally, rats in group 3 received a Gs extract daily at a dose of 300 mg/kg body weight intragastically. Subsequently, to identify markers of liver damage in serum, alanine aminotransferase, aspartate aminotransferase, albumin and bilirubin were measured by colorimetric methods. Glucose, triglyceride and cholesterol concentrations were also determined. In addition, oxidative damage was estimated by measuring lipid peroxidation [using thiobarbituric-acid reactive substances(TBARS)] in both plasma and the liver and by measuring the total concentration of antioxidants in serum and the total antioxidant capacity in the liver. In addition, a liver mass gain assessment, total DNA analysis and a morpho-histological analysis of the liver from animals in all three groups were performed and compared. Finally, the number of deaths observed in the three groups was analyzed.RESULTS: Administration of the Geranium shiedeanum extract significantly reduced the unfavorable effect of ethanol on liver regeneration(restitution liver mass: PHEt OH group 60.68% vs PH-Gs-Et OH group 69.22%). This finding was congruent with the reduced levels of hepatic enzymes and the sustained or increased levels of albumin and decreased bilirubin in serum. The extract also modified the metabolic processes that regulate glucose and lipid levels, as observed from the serum measurements. Lower antioxidant levels and the liver damage induced by Et OH administration appeared to be mitigated by the extract, as observed from the TBARs(PH-Et OH group 200.14 mmol/mg vs PH-Gs-Et OH group 54.20 mmol/mg; P < 0.05), total status of antioxi
