Metastatic breast cancer leads to poor prognoses and worse outcomes in patients due to its invasive behavior and poor response to *** is still unclear what biophysical and biochemical factors drive this more aggressive phenotype in metastatic cancer;however recent studies have suggested that exposure to fluid shear stress in the vasculature could cause *** this study a modular microfluidic platform capable of mimicking the magnitude of fluid shear stress(FSS)found in human vasculature was designed and *** device provides a platform to evaluate the effects of FSS on MCF-7 cell line,an estrogen receptor positive(ER+)breast cancer cell line,during circulation in the *** of the effects of FSS on MCF-7 cells was carried out utilizing two approaches:single cell analysis and bulk *** single cell analysis,cells were trapped in a microarray after exiting the serpentine channel and followed by immunostaining on the device(on-chip).Bulk analysis was performed after cells were collected in a microtube at the outlet of the microfluidic serpentine channel for western blotting(off-chip).It was found that cells exposed to an FSS magnitude of 10 dyn/cm^(2) with a residence time of 60 s enhanced expression of the proliferation marker Ki67 in the MCF-7 cell line at a single cell *** understand possible mechanisms for enhanced Ki67 expression,on-chip and off-chip analyses were performed for pro-growth and survival pathways ERK,AKT,and JAK/*** demonstrated that after shearing the cells phosphorylation of p-AKT,p-mTOR,and p-STAT3 were ***,there was no change in p-ERK1/*** is a mediator of ER rapid signaling,analysis of phosphorylated ERαwas carried out and no significant differences between sheared and non-sheared populations were *** together these results demonstrate that FSS can increase phosphorylation of proteins associated with a more aggressive phenotype in circulating cancer *** findin

Metabolic(dysfunction)-associated fatty liver disease(MAFLD)affects a third of the population and is a leading cause of liver-related *** no effective treatments exist,novel approaches to drug development are ***,outdated terminology and definitions of the disease are hampering efforts to develop new drugs and *** international consensus panel has put forth an influential proposal for the disease to be renamed from nonalcoholic fatty liver disease(NAFLD)to MAFLD,includ-ing a proposal for how the disease should be *** allies with the many stakeholders in MAFLD care―including patients,patients’advocates,clinicians,researchers,nurse and allied health groups,regional societies,and others―we are aware of the negative consequences of the NAFLD term and *** share the sense of urgency for change and will act in new ways to achieve our *** there is much work to be done to overcome clinical inertia and reverse worrisome recent trends,the MAFLD initiative provides a firm foundation to build *** provides a roadmap for moving for-ward toward more efficient care and affordable,sustainable drug and device innovation in MAFLD *** hope it will bring promising new opportunities for a brighter future for MAFLD care and improve care and outcomes for patients of one of the globe’s largest and costliest public health *** this viewpoint,we have revisited this initiative through the perspectives of drug development and regulatory science.

法定有机酸硷盐的含量测定方法操作繁复,消耗时间长,如分析四个样品最快需要三小时半;经灼燒、煮沸、过滤等操作易造成誤差,并取检品量很大(一般需二克)。应用非水滴定法則能避免以上缺点,不仅分析时间减少,操作簡捷,减少接间誤差机会,并且需要检品量小(小于100毫克也能正确测定)。然尚存在困难,例如酒石酸盐等溶解度低的盐难溶于冰醋酸;以甲紫作指示剂时,終点为藍到藍綠

Temporo-spatial dynamics and behavioural patterns of 2012 cholera epidemic in the African mega-city of Conakry,Guinea

Background:Cholera is endemic in Guinea,having suffered consecutive outbreaks from 2004 to 2008 followed by a lull until the 2012 *** we describe the temporal-spatial and behavioural characteristics of cholera cases in Conakry during a three-year period,including the large-scale 2012 ***:We used the national and African Cholera Surveillance Network(Africhol)surveillance data collected from every cholera treatment centre in Conakry city from August 2011 to December *** prevalence of suspect and confirmed cholera cases,the case fatality ratio(CFR),and the factors associated with suspected cholera were described according to three periods:pre-epidemic(A),epidemic 2012(B)and post epidemic(C).Weekly attack rates and temporal-spatial clustering were calculated at municipality level for period *** was confirmed by culture at the cholera national reference ***:A total of 4559 suspect cases were reported:66,4437,and 66 suspect cases in periods A,B and C,*** the 204 suspect cases with culture results available,6%,60%,and 70%were confirmed in periods A,B,and C,*** 0.3%,the CFR was significantly lower in period B than in periods A(7.6%)and C(7.1%).The overall attack rate was 0.28%in period B,ranging from 0.17%to 0.31%across ***,a cluster of cases was identified in two districts in the northern part of *** 14%,rice water stools were less frequent in period A than in period B and C(78%and 84%).Dehydration(31%vs 94%and 89%)and coma(0.4%vs 3.1%and 2.9%)were lower during period B than in periods A and *** treatment of drinking water was less frequent in period A,while there were more reports of recent travel in period ***:The epidemic dynamic and the sociological description of suspect cases before,during,and after the large-scale epidemic revealed that the Vibrio cholerae was already present before the ***,it appeared that infected individuals react

Calculation of the pH of Buffer Solution of 2-[<i>N</i>-Morpholino]ethanesulfonic Acid (MES) from 5°C to 55°C

This paper reports the results for the pH of three buffer solutions free of chloride ion. The remaining six buffer solutions have saline media of the ionic strength I = 0.16 ***-1, matching closely to that of the physiological sample. Conventional paH values for the three buffer solutions without the chloride ion and six buffer solutions with the chloride ion at I = 0.16 ***-1 from 5°C to 55°C have been calculated. The operational pH values for five buffer solutions at 5°C and 55°C have been determined based on the difference in the values of the liquid junction potentials between the blood phosphate standard and the experimental buffer solutions. Five of these buffers are recommended as standards for the physiological pH range 7.5 to 8.5.

本文报道由于碘离子对不稳定的Ni(Ⅱ)-S(芳基硫醇盐)键的亲核作用,使得Fe(CO)4I2和Ni(SR)2(dppe(SR=芳基硫醇盐)之间的反应生成NiI2(dppe)。含碘和二芳基硫醇盐离子桥联的Ni-Ni双核配合物[(dppe)Ni(μ-I)(μ-pdt)Ni(dppe)]I和[(dppe)Ni(μ-I)(μ-edt)Ni(dppe)]I可方便地由[NiI2(dppe)]和[Ni(pdt)(dppe)]或[Ni(edt)(dppe)]在二氯甲烷的溶液中的反应制得;该类反应可认为是由于硫醇盐离子基团中S-供体上的孤对电子对Ni-I键的进攻所致。另一方面,我们观察到[FeCp(CO)2I]和[Ni(pdt)(dppe)]或[Ni(edt)(dppe)]在二氯甲烷中的反应极其缓慢;但当向上述反应体系中加入NH4PF6进行复分解置换后,源于碘离子和Ni(Ⅱ)-S键的作用同样可得到含碘与二芳基硫醇盐离子桥联的Ni-Ni双核配合物[(dppe)Ni(μ-I)(μ-pdt)Ni(dppe)]PF6和[(dppe)Ni(μ-I)(μ-edt)Ni(dppe)]PF6。实验结果说明在本文所讨认的镍(Ⅱ)-硫醇盐离子-膦配合物中,Ni(Ⅱ)-S键的反应活性随桥联的第二金属离子和不同的碘离子基元而改变。
