Correlative CD4 and CD8 T-cell immunodominance in humans and mice:Implications for preclinical testing

Antigen-specific T-cell recognition is restricted by Major Histocompatibility Complex(MHC)molecules,and differences between CD4 and CD8 immunogenicity in humans and animal species used in preclinical vaccine testing are yet to be fully *** this study,we addressed this matter by analyzing experimentally identified epitopes based on published data curated in the Immune Epitopes DataBase(IEDB)*** first analyzed SARS-CoV-2 spike(S)and nucleoprotein(N),which are two common targets of the immune response and well studied in both human and mouse *** observed a weak but statistically significant correlation between human and H-2^(b)mouse T-cell responses(CD8 S specific(r=0.206,p=1.37×10^(−13));CD4 S specific(r=0.118,p=2.63×10^(−5))and N specific(r=0.179,p=2.55×10^(−4))).Due to intrinsic differences in MHC molecules across species,we also investigated the association between the immunodominance of common Human Leukocyte Antigen(HLA)alleles for which HLA transgenic mice are available,namely,A*02:01,B*07:02,DRB1*01:01,and DRB1*04:01,and found higher significant correlations for both CD8 and CD4(maximum r=0.702,p=1.36×10^(−31)and r=0.594,p=3.04−122,respectively).Our results further indicated that some regions are commonly immunogenic between humans and mice(either H-2^(b)or HLA transgenic)but that others are human ***,we noted a significant correlation between CD8 and CD4 S-(r=0.258,p=7.33×10^(21))and N-specific(r=0.369,p=2.43×10^(14))responses,suggesting that discrete protein subregions can be simultaneously recognized by T *** findings were confirmed in other viral systems,providing general guidance for the use of murine models to test T-cell immunogenicity of viral antigens destined for human use.

Intraoperative Glycaemia Following Paracetamol with and without Glucose: A Randomized-Controlled Trial

Background: Hyperglycaemia is conversely a risk factor for perioperative complications. We are currently using a generic 3.3 g glucose containing formula of intravenous 1000 mg paracetamol for perioperative analgesia. Our main goal was to compare the trends of glycaemic values after administration of a generic 3.3 g glucose containing formula with a non-glucose containing branded formula of intravenous 1000 mg paracetamol. Methods: A exploratory proof-of-concept randomized clinical trial was conducted with 150 patients scheduled for elective gynaecologic. Patients were randomly assigned into three groups: control group (saline);active-control group: intraoperative administration of a branded non-glucose containing 1000 mg paracetamol formula;experimental group: intraoperative administration of a generic 3.3 g glucose containing 1000 mg paracetamol formula. The primary outcome was mean change from baseline in glaucoma. In case significant differences were found, the following secondary outcomes were explored: the proportion of patients with high glycaemia values (>150 mg/dL) and the proportion of patients with negative glycaemic variation. Results: Mean glycaemia change was higher after generic 3.3 g glucose containing paracetamol formula both in comparison to placebo (16.3 mg/dL [95% CI: 6.1 to 26.6]) and active-control (19.1 mg/dL [8.2 to 30.0] groups. Similar results were found in the intention-to-treat analysis. In only the experimental group, patients had high glycaemic values (11.3%). Conclusions: This study showed that in non-diabetic, under non-cardiac surgery, administration of a generic glucose-containing formula of intravenous 1000 mg paracetamol was associated with poorer glycaemic control. These results raise the question of a possible increased risk among these patients. Further studies using diabetic patients are recommended.

Rock slope hazard assessment is an important part of risk analysis for open pit *** main parameters that can lead to rock slope failures are the parameters traditionally used in geomechanical classifications,the slope geometrical parameters and external factors like rainfall and *** paper presents a methodology for a hazard assessment system for open pit mine slopes based on 88 cases collated around the world using principal components analysis,discriminant analysis and confidence *** historical cases used in this study included copper,gold,iron,diamond,lead and zinc,platinum and claystone *** variables used in the assessment methodology are uniaxial compressive strength of intact rock;spacing,persistence,opening,roughness,infilling and orientation of the main discontinuity set;weathering of the rock mass;groundwater;blasting method;and height and inclination of the *** principal component analysis was used to quantify the data,the discriminant analysis was used to establish a rule to classify new slopes about its stability *** provide a practical hazard assessment system,confidence ellipses were used to propose a hazard graph and generate the *** discriminant rule developed in this research has a high discrimination capacity with an error rate of 11.36%.

Quantification of displacement and particle crushing around a penetrometer tip

Particle crushing occurs near the tip of a penetrometer and influences the development of the tip *** study particle crushing near a penetrometer tip,a cone penetrometer was monotonically jacked and then load-tested in medium dense and dense silica sand samples prepared in a halfcylindrical calibration chamber with viewing *** the tests,images of the advancing penetrometer and the surrounding soil were taken using digital cameras and analyzed to obtain the displacement and strain fields around the penetrometer using the Digital Image Correlation(DIC)***,soil samples were collected near the tip of the penetrometer using a novel agarimpregnation technique and digitized using an X-ray *** digitized samples were analyzed to reconstruct the three-dimensional models of individual particles,generating the gradation and relative breakage parameters of the sand around the cone penetrometer.

This research aims to assess the sensitive coastal zones by analyzing the shoreline changes such as accretion and erosion in the eastern coastal part of Ramanathapuram District,Tamil Nadu,*** study focused on Mandapam,Pirappanvalsai,Enmanamcondam,Nagachi,Terbogi,Athyuthu,Chitrakkottai,Pathengal,Attangarai,Alagankulam,and Devipattinam *** of these areas are distinguished as highly sensitive ecological *** erosion of an area leads to irreplaceable disturbances to ***,erosion and accretion studies are top priorities when studying the coastal *** addition to GIS technology,which has been widely used in the past 20 years,the End Point Rate(EPR)and Linear Regression Rate(LRR)analyses were conducted to better understand the shoreline *** stability of the coastal region was analyzed by observing the slope variations of the beach during the profile *** the study,we analyzed that the Quaternary deposits of sand are the dominant geological feature of the study *** reefs and salt pans are highly distributed along the coastal *** coast of Mandapam village has been detected as a highly sensitive region,with an average erosion rate of-3.1 m/yr and an average accretion rate of 1.22 m/yr,resulting in high erosion and low *** coast is a steep coast with an average slope angle of 0.24°.This study provides insight into sustainable coastal zone management.

Comprehensive understanding of marine biodiversity is essential for conservation,especially for vulnerable *** this paper,we implemented and illustrated a checklist of molluscs from the Abrolhos Parcel,Brazil,the most important coralline area in South Atlantic *** list features 100 species belonging to 4 classes and 85 genera,of which 15 are new records for Bahia State,northeast *** results highlight the potential for new records in the *** species came from 54 unconsolidated sediment samples collected from the bottom of the reef with a 0.042-m^(2) van Veen *** the listed species,Curveulima sp.,Notarchus *** Guyanella clenchi are new records for the South Atlantic,while Caecum metamorphosicum,Eratoidea janeiroensis,Volvarina serrei,Solemya notialis,Solariorbis shimeri,and Amphithalamus glabrus are endemic to Brazilian waters.A comparison of our results with those obtained in other reef environments indicates that the Abrolhos Parcel has a rich malacofauna and suggests that many more species can still be found with more samplings.

Litter production and leaf nutrient concentration and remobilization in response to climate seasonality in the central Amazon

Litterfall is the largest source of nutrients to for-est soils of tropical ***,variability in lit-terfall production,nutrient remobilization,and changes in leaf nutrient concentration with climate seasonality remain largely unknown for the central *** study meas-ured litterfall production,leaf nutrient remobilization,and leaf area index on a forest plateau in the central *** was measured at monthly intervals during 2014,while nitrogen,phosphorus,potassium,calcium and mag-nesium concentrations of leaf litter and canopy leaves were measured in the dry and rainy seasons,and remobilization rates *** area index was also recorded in the dry and rainy *** litterfall varied from 33.2(in the rainy season)to 87.6 g m^(-2) in the dry season,while leaf area index increased slightly in the rainy *** seasonality had no effect on concentrations of nitrogen,calcium,and magnesium,whereas phosphorous and potassium responded to rainfall seasonality *** phosphorous increased,potassium decreased during the dry *** seasons,nitrogen,potassium,and phosphorous decreased in leaf litter;calcium increased in leaf litter,while magnesium remained unaffected with leaf ***,the five nutrients had similar remobilization rates over the *** absence of climate seasonality on nutrient remobilization suggests that the current length of the dry season does not alter nutrient remobilization rates but this may change as dry periods become more prolonged in the future due to climate change.

To supervise or to self‑supervise:a machine learning based comparison on credit supervision

This study investigates the need for credit supervision as conducted by on-site banking *** builds on a real bank on-site credit examination to compare the performance of a hypothetical self-supervision approach,in which banks themselves assess their loan portfolios without external intervention,with the on-site banking supervision approach of the Central Bank of *** experiment develops two machine learning classification models:the first model is based on good and bad ratings informed by banks,and the second model is based on past on-site credit portfolio examinations conducted by banking *** findings show that the overall performance of the on-site supervision approach is consistently higher than the performance of the self-supervision approach,justifying the need for on-site credit portfolio examination as conducted by the Central Bank.

Aspects of forest restoration and hydrology:the hydrological function of litter

Although forests play important roles in the hydrological cycle,there is little information that relates the water retention capacity of litter in areas under passive restoration,especially in Cerrado *** study relates litter levels to water holding capacity and effective water retention among forest fragments under different passive restoration stages:46,11,and 8 years to better understand litter hydrological functions in the *** retention capacity and effective water retention capacity of litters(unstructured materials,branches and leaves)in the field were monitored on a monthly *** litter accumulation at 46 years was significantly higher than that of the other succession *** litter mass was significantly higher than that of leaves and *** 46-year stage had the highest water holding capacity in the leaf fraction,followed by unstructured material and *** the water holding capacity was lower in the oldest resto-ration,this site showed the highest efficiency under field *** process was quickly reestablished,as the 11-year restoration showed results closer to that for the 46-year stage in comparison to the area at 8 ***,passive restoration plays a key role in soil water mainte-nance due to the influence of litter in Cerrado *** and the imminent need of restoring degraded sites,highlight the need for further studies focused on bet-ter understanding of the process of forest restoration and its temporal effect on soil water recovery dynamics.

Although the frequency of metabolic risk factors for cirrhosis and hepatocellular carcinoma(HCC)is increasing,chronic hepatitis B(CHB)and chronic hepatitis C(CHC)remain the most relevant risk factors for advanced liver disease *** addition to liver damage,hepatitis B virus(HBV)and hepatitis C virus(HCV)infections are associated with a myriad of extrahepatic manifestations including mixed cryoglobulinaemia,lymphoproliferative disorders,renal disease,insulin resistance,type 2 diabetes,sicca syndrome,rheumatoid arthritislike polyarthritis,and autoantibody ***,the list has grown to include *** of muscle mass or muscle function is a critical feature of malnutrition in cirrhotic patients and has been found in approximately 23.0%-60.0%of patients with advanced liver ***,among published studies,there is significant heterogeneity in the aetiologies of hepatic diseases and measurement methods used to determine *** particular,the interaction between sarcopenia,CHB and CHC has not been completely clarified in a real-world *** can result from a complex and multifaceted virus-host-environment interplay in individuals chronically infected with HBV or ***,in the present review,we provide an overview of the concept,prevalence,clinical relevance,and potential mechanisms of sarcopenia in patients with chronic viral hepatitis,with an emphasis on clinical outcomes,which have been associated with skeletal muscle loss in these patients.A comprehensive overview of sarcopenia in individuals chronically infected with HBV or HCV,independent of the stage of the liver disease,will reinforce the necessity of an integrated medical/nutritional/physical education approach in the daily clinical care of patients with CHB and CHC.
