Detection transposable elements in Botrytis cinerea in latent infection stage from symptomless apples

Objective:To detect Botrytis cinerea(***)latent infections on apples before storage,which is essential for effective control strategies in the fruit postharvest ***:In the present study,a polymerase chain reaction detection method,based on primers designed on *** transposable elements(boty and flipper)and intergenic spacer region as internal control,were utilized to reveal the presence of symptomless infections on apple *** molecular method proved to be highly specific and sensitive in detecting latent *** revealed the presence of the pathogen in 83%of the samples from infected apples with 104 conidia/mL,whereas those infected with 106 conidia/mL detected 94%as compared to the traditional method that revealed the pathogen in 40%and 66%of the samples inoculated with 10^(4)and 10^(6)conidia/mL ***,the method characterized *** as subpopulation transposa-type by the presence of the transposable elements boty and flipper Results:The results obtained from DnA quantification method were compared with enzyme-linked immunosorbent assay and these studies showed good *** our method has important advantages compared with others detection methods for ***,because the proposed methodology allowed distinguishes between its two subpopulations(vacuma and transposa)and this would allow establish possible appropriate control ***:Finally,the method can be an interesting alternative for its possible application in the phytosanitary programs of the fruit industry worldwide.

Objective:To study the effect of temperature on the morphological characteristics of Botrytiscinerea(***)and its correlated with the genetic *** is a plant-pathogenic fungus that produces the disease known as grey mould in a wide variety of agriculturally important hosts in many ***:Six strains from different host collected have been isolated and characterized by several methods as mycelial growth,fungicide resistance,pathogenicity and the effects of the *** was analyzed by PCR and distinguished by the presence or absence of transposable ***:Results showed that clear morphological differences exist between strains at the temperature of 4,12 and 28℃.All strains analyzed molecularly were classified as Group II(transposa-type).Demonstrating a negative correlation between mycelial growth and other characteristics as the fungicide resistance and ***,it is difficult to establish relationships phenotypic and genotypic between strains of ***:The results indicated that the mycelial growth,resistance at fungicide and pathogenicity are independent of the characteristics molecular,however,are dependent of a factor such as temperature.

灰霉病是导致水果采后巨大经济损失的主要病害,其致病菌为葡萄孢菌。葡萄孢菌通常以潜伏侵染的形式存在,一旦病原菌发展蔓延则引起健康果实的迅速腐烂变质。因此,开发快速、灵敏的葡萄孢菌检测和定量分析方法势在必行。本文介绍了一种用于测定苹果(Red Delicious)、鲜食葡萄(Pink Moscatel)和梨(William’s)组织中葡萄孢菌的间接竞争酶联免疫吸附法(ELISA)。该方法的原理是基于水果组织中葡萄孢菌抗原与交联固定在微量滴定板表面的葡萄孢菌纯化抗原对于单克隆抗体结合位点的竞争作用。为了验证该方法的有效性,进行了如选择性、线性分析、精密度、准确性、灵敏性等大量的参数测定。该方法对于葡萄孢菌抗原的检测限为0.97μ***-1,为了保证测定的稳定性,事先将纯化抗原固定长达4个月。应用该方法可以对尚未表现症状的任何水果中的葡萄孢菌进行检测定量分析。通过与一项DnA定量测试结果进行对比,表明ELISA法的测定结果与DnA分析结果具有很好的相关性。本研究结果表明,新开发的测试方法实现了对无发病症状的果实中葡萄孢菌的检测分析,具有成本低廉、操作简便、快捷等优点,可将其应用于世界各国对水果等植物的病害检疫领域。

Analysis of metal profile in soybean after cadmium-induced oxidative damage

Objective:To analyze the effect of cadmium(Cd)on soybean seedlings growth and the relationship with the distribution and concentration of ***:The ions concentrations were determined by *** extraction efficiency and digestion time were ***,oxidative stress parameters were determined and related with metal ***:The accumulated amount of dry matter in roots and leaves was lower in the Cd-treated *** analysis showed that the exposure to Cd affected the accumulated amount of dry matter as well as the content of mineral elements in the analysis *** Cd treated plants,electrical conductivity increased respect to the controls,indicating that ionic permeability became altered.A strong inhibition of the chlorophylls(chl)biosynthesis in the Cd-treated group was also demonstrated by a decrease of chla and chlb *** result was related with the observed significant decrease in the Mg uptake at the roots and leaves ***:The stress caused by Cd exposure,evidenced by significantly high hydrogen peroxide levels in roots and leaves after 24 h and the content of specific macro-microelements is a factor that affects the accumulation of dry matter,electrical conductivity and chlorophylls concentration.
