Coating independent cytotoxicity of citrate-and PEG-coated silver nanoparticles on a human hepatoma cell line

The antibacterial potential of silver nanoparticles（AgNPs） resulted in their increasing incorporation into consumer,industrial and biomedical ***,human and environmental exposure to AgNPs（either as an engineered product or a contaminant）supports the emergent research on the features conferring them different toxicity *** this study,30 ran AgNPs coated with citrate or poly（ethylene glycol）（PEG） were used to assess the influence of coating on the effects produced on a human hepatoma cell line（HepG2）,namely in terms of viability,apoptosis,apoptotic related genes,cell cycle and cyclins gene *** types of coated AgNPs decreased cell proliferation and viability with a similar toxicity *** the concentrations used（11 and 5 μg/mL corresponding to IC50 and-IC10 levels,respectively） the amount of cells undergoing apoptosis was not significant and the apoptotic related genes BCL2（anti-apoptotic gene）and BAX（pro-apoptotic gene） were both ***,both AgNPs affected HepG2 cell cycle progression at the higher concentration（11 μg/mL） by increasing the percentage of cells in S（synthesis phase） and G2（Gap 2 phase） *** the cell-cycle related genes,the expression of cyclin B1 and cyclin E1 genes were ***,this work has shown that citrate- and PEG-coated AgNPs impact on HepG2 apoptotic gene expression,cell cycle dynamics and cyclin regulation in a similar *** research is needed to determine the properties that confer AgNPs at lower toxicity,since their use has proved helpful in several industrial and biomedical contexts.

In vitro antileishmanial effects of Physalis angulata root extract on Leishmania infantum

Objective： In the present study, we evaluated the effects of the aqueous extract of Physalis angulata root （AEPa） on Leishmania infanturn proliferation, morphology, and the driving mechanism in leishmanicidal activity and modulatory action on macrophages. Methods： I., infantum promastigotes were treated with 50 and 100 μg/mL AEPa for 72 h and then antipro- mastigote assay was performed by counts in a Newbauer chamber, morphological changes were analyzed by transmission electron microscopy and the mechanism of the leishmanicidal activity was detected. In addition, macrophages were infected with L. infantum and were used to evaluate anti-amastigote activity of AEPa and effects of AEPa on cytokine secretion after 72-hour treatment. Results： Treatment with AEPa reduced the numbers ofL infantum promastigotes （50% inhibitory concen- tration （ICso） = 65.9μg/mL; selectivity index （SI）= 22.1） and amastigotes （ICso = 37.9 μg/mL; SI = 38.5） compared with the untreated control. Amphotericin B reduced 100% of the promastigote numbers after 72 h of treatment （IC50 = 0.2μg/mL）. AEPa induced several morphological changes and increased the production of reactive oxygen species and apoptotic death in promastigotes after treating for 72 h. AEPa （100 μg/mL） promoted tumor necrosis factor-α secretion in macrophages infected with L. infantum after 72 h of treatment, but did not induce an increase in this cytokine in noninfected macrophages. In addition, AEPa showed no cytotoxic effect on J774-A1 cells （50% cytotoxic concentration 〉1000μg/mL）. Conclusion： AEPa presented antileishmanial activity against the promastigotes and amastigotes of I.. infantum without macrophage cytotoxicity; these results show that natural products such as P. angulata have leishmanicidal potential and in the future may be an alternative treatment for leishmaniasis.

Calibration and validation of FAO-AquaCrop model to estimate the total biomass and yacon root yield

Freshwater environments are the main ecosystems subjected to various anthropogenic impacts, which have caused the waterborne transmission of diseases caused mainly by bacteria. The main objective of this work was to carry out a systematic analysis through the literature on the main microorganisms found in impacted aquatic environments and to relate the data with the risks to public health. This review was planned, conducted and reported following the PRISMA protocol, which was used the PECO strategy. There performed searches for studies using the electronic databases PubMed, Medline and LILACS in September 2021, using the controlled search strategy: “Bacteria” and “Ecosystem” and “Environment” and “Aquatic” and “Impacted. The studies that met the eligibility criteria were considered eligible, with the participation of two independent reviewers. There found 1058 studies, of which were chosen only 31 to compose the results of this systematic review. Because of the results, there observed that the predominant bacterial genera in freshwater are: Pseudomonas, Aeromonas, and Escherichia, with the Asian continent presenting more varieties of bacterial species detected. These microorganisms are responsible for causing diseases that can lead to death, which shows the importance of constantly monitoring these ecosystems.

The complex meniscus tissue plays a critical role in the knee. The high susceptibility to injury has led to an intense pursuit for better tissue engineering regenerative strategies, where scaffolds play a major role. In this study, indirect printed hierarchical multilayered sca ffolds composed by a silk fibroin (SF) upper layer and an 80/20 (w/w) ratio of SF/ionic-doped β-tricalcium phosphate (TCP) bottom layer were developed. Furthermore, a comparative analysis between two types of sca ffolds pro- duced using di fferent SF concentrations, i.e., 8% (w/v) (Hi8) and 16% (w/v) (Hi16) was performed. In terms of architecture and morphology, the produced sca ffolds presented homogeneous porosity in both layers and no di fferences were observed when comparing both sca ffolds. A decrease in terms of mechanical performance of the sca ffolds was observed when SF concentration decreased from 16 to 8% (w/v). Hi16 revealed a static compressive modulus of 0.66 ± 0.05 MPa and dynamical mechanical properties ranging from 2.17 ± 0.25 to 3.19 ± 0.38 MPa. By its turn, Hi8 presented a compressive modulus of 0.27 ± 0.08 MPa and dynamical mechanical properties ranging from 1.03 ± 0.08 MPa to 1.56 ± 0.13 MPa. In vitro bioactivity studies showed formation of apatite crystals onto the surface of Hi8 and Hi16 bottom layers. Human meniscus cells (hMCs) and human primary osteoblasts were cultured separately onto the top layer (SF8 and SF16) and bottom layer (SF8/TCP and SF16/TCP) of the hierarchical sca ffolds Hi8 and Hi16, respectively. Both cell types showed good adhesion and proliferation as denoted by the live/dead staining, Alamar Blue assay and DNA quanti fication analysis. Subcutaneous implantation in mice revealed weak in flammation and sca ffold’s integrity. BACKGROUND The evaluation of periportal fibrosis(PPF)is essential for a prognostic assessment of patients with Schistosomiasis *** WHO Niamey Protocol defines patterns of fibrosis from abdominal ultrasonography,^(1)H-nuclear magnetic resonance(NMR)-based metabonomics has been employed to assess liver fibrosis in some *** To build 1H-NMR-based metabonomics models(MM)to discriminate mild from significant periportal PPF and identify differences in the metabolite *** A prospective cross-sectional study was performed on schistosomiasis patients at a University Hospital in Northeastern *** evaluated 41 serum samples from 10 patients with mild PPF(C Niamey pattern)and 31 patients with significant PPF(D/E/F Niamey patterns).MM were built using partial least squares-discriminant analysis(PLS-DA)and orthogonal projections to latent structures discriminant analysis(OPLS-DA)*** PLS-DA and OPLS-DA resulted in discrimination between mild and significant PPF groups with R2 and Q2 values of 0.80 and 0.38 and 0.72 and 0.42 for each model,*** OPLS-DA model presented accuracy,sensitivity,and specificity values of 92.7%,90.3%,and 100%to discriminate significant *** metabolites identified as responsible by discrimination were:Nacetylglucosamines,alanine,glycolaldehyde,carbohydrates,and *** MMs discriminated mild from significant PPF patterns in patients with Schistosomiasis mansoni through identification of differences in serum metabolites profiles.

The Role of Nanodispersed Nitrides in the Sintering of Silicon Nitride Ceramics

The synthesis of ceramics based on silicon nitride using nanopowders of TiN and Si3N4 as additives was studied. The ceramic compositions were pressurelessly sintered under ni- trogen atmosphere at different temperatures （1550℃, 1650℃ and 1750℃） with a heating rate of 10℃/min and a holding time of 2 h. The nanodispersed nitrides （NDN） were produced by electric-arc plasma synthesis and characterized. The ceramic composites obtained with nanoparticles of 1wt% to 5wt% TiN and 20wt% Si3N4 were characterized by scanning electron microscopy （SEM）, atom force microscopy （AFM） and energy-dispersive spectrometry （EDX）. The effect of the addition of nanodispersed powders on the mechanical properties and microstructure of Si3N4 ceramics was investigated.
