Mutated genes are rarely common even in the same pathological type between cancer patients and as such, it has been very challenging to interpret genome sequencing data and difficult to predict clinical outcomes. PIK3 CA is one of a few genes whose mutations are relatively popular in tumors. For example, more than 46.6% of luminal-A breast cancer samples have PIK3 CA mutated, whereas only 35.5% of all breast cancer samples contain PIK3 CA mutations. To understand the function of PIK3 CA mutations in luminal A breast cancer, we applied our recentlyproposed Cancer Hallmark network Framework to investigate the network motifs in the PIK3CA-mutated luminal A tumors. We found that more than 70% of the PIK3CA-mutated luminal A tumors contain a positive regulatory loop where a master regulator(PDGF-D), a second regulator(FLT1) and an output node(SHC1) work together. Importantly, we found the luminal A breast cancer patients harboring the PIK3 CA mutation and this positive regulatory loop in their tumors have significantly longer survival than those harboring PIK3 CA mutation only in their tumors. These findings suggest that the underlying molecular mechanism of PIK3 CA mutations in luminal A patients can participate in a positive regulatory loop, and furthermore the positive regulatory loop(PDGF-D/FLT1/SHC1) has a predictive power for the survival of the PIK3 CAmutated luminal A patients.

Differential effects of short term winter thermal stress on diapausing tiger swallowtail butterflies （Papilio spp.）

It is generally thought that insects inhabiting lower latitudes are more severely impacted by changes in their thermal environment than are high latitude species. This is attributed to the wider range of temperatures to which high-latitude species are exposed. By contrast, low-latitude species have typically evolved in more thermally stable environments with a narrower range of temperature variation. However, deviation from this pattern can occur and here we report that under variable winter conditions a higher latitude species may be more sensitive to thermal variation than its lower latitude sister species. Using split broods, we examined the survival and adult emergence success of diapausing pupae of Papilio canadensis and P. glaucus, as well as a unique, recombinant hybrid population （＂late-flight＂） to short periods of mid-winter cold and heat stress. Our results indicate that the higher latitude, univoltine populations （P. canadensis and late-flights） exhibit lower pupal survival than the lower latitude, facultative diapauser （P. glaucus） for all mid-winter thermal stress treatments, both high and low. Size differences alone do not appear to account for the observed differences in survival or metabolic costs in these three pheno- types, as late-flight individuals are similar in size to R glaucus. We attribute the observed differences in survival and weight loss to potential metabolic differences and variation in the intensity of diapause, in addition to divergent adaptation to winter precipitation levels （e.g. snow cover） and the influences this may have on microhabitat temperature moderation.

Predicting excavation damage zone depths in brittle rocks

During the construction of an underground excavation, damage occurs in the surrounding rock mass due in large part to stress changes. While the predicted damage extent impacts profile selection and support design, the depth of damage is a critical aspect for the design of permeability sensitive excavations, such as a deep geological repository(DGR) for nuclear waste. Review of literature regarding the depth of excavation damage zones(EDZs) indicates three zones are common and typically related to stress induced damage. Based on past developments related to brittle damage prediction using continuum modelling, the depth of the EDZs has been examined numerically. One method to capture stress induced damage in conventional engineering software is the damage initiation and spalling limit(DISL) approach. The variability of depths predicted using the DISL approach has been evaluated and guidelines are suggested for determining the depth of the EDZs around circular excavations in brittle rock masses. Of the inputs evaluated, it was found that the tensile strength produces the greatest variation in the depth of the EDZs. The results were evaluated statistically to determine the best fit relation between the model inputs and the depth of the EDZs. The best correlation and least variation were found for the outer EDZ and the highly damaged zone(HDZ) showed the greatest variation. Predictive equations for different EDZs have been suggested and the maximum numerical EDZ depths, represented by the 68% prediction interval, agreed well with the empirical evidence. This suggests that the numerical limits can be used for preliminary depth prediction of the EDZs in brittle rock for circular excavations.

The impact of burning on lion Panthera leo habitat choice in an African savanna

Camivores play a central role in ecosystem processes by exerting top-down control, while fire exerts bottom-up con- trol in ecosystems throughout the world, yet, little is known about how fire affects short-term carnivore distributions across the landscape. Through the use of a long-term data set we investigated the distribution of lions, during the daytime, in relation to burned areas in Serengeti national Park, Tanzania. We found that lions avoid burned areas despite the fact that herbivores, their prey, are attracted to burned areas. Prey attraction, however, likely results from the reduction in cover caused by burning, that may thereby decrease lion hunting success. Lions also do not preferentially utilize the edges of burned areas over unburned areas de- spite the possibility that edges would combine the benefit of cover with proximity to abundant prey. Despite the fact that lions avoid burned areas, lion territory size and reproductive success were not affected by the proportion of the territory burned each year. Therefore, burning does not seem to reduce lion fitness perhaps because of the heterogeneity of burned areas across the landscape or because it is possible that when hunting at night lions visit burned areas despite their daytime avoidance of these ar- eas .

Changes in Urinary Bone Resorption marker after Taking a Combination nutrient Formula in Adults with Scoliosis: A Retrospective, Open-Label, Case-Controlled Series

Background: Scoliosis is a common condition in adult patients, and often causes chronic back pain compared to non-scoliosis. It has also been identified that bone mineral density is very often compromised in patients with scoliosis, even occurring in adolescence. At this time, no study has looked at how bone mineral density and scoliosis incidence or severity may be connected with specific regard to treatment. This study presents data on the outcomes of a scoliosis-specific exercise therapy and its ability to correct scoliotic curvatures in adult patients alone and in combination with bone mineral density supplementation. Methods: The charts of 14 total patients were retrospectively selected based upon specific inclusion criteria. Outcome assessments included the radiographic Cobb angle of the primary curvature, as well as laboratory measures of urinary deoxypyridinoline cross links. These results were compared against 12 patients who did not take the bone density supplement during or after their exercise-based treatment. Results: Patients taking the bone density supplement achieved the same level of Cobb angle reduction as compared to the control group. However, they additionally achieved a significant reduction in urinary deoxypyridinoline cross links as compared to the control group at 6 months. Conclusion: Patients taking a multi-ingredient bone density supplement daily for 6 months after completing a scoliosis-specific exercise program reported statistically significant improvements in urinary deoxypyridinoline cross links as compared to controls. It is unknown if or how bone density loss may contribute to the onset or progression of scoliosis. Long-term follow-up of these patients will be ongoing to assess bone mineral density status and Cobb angle changes longitudinally.
