Dietary supplementation of sulfur amino acids improves intestinal immunity to Eimeria in broilers treated with anti-interleukin-10 antibody

Oral antibody to interleukin-10(anti-IL-10)enhances the intestinal immune defense against *** sulfur amino acids methionine and cysteine(MtC)play essential roles in inducing and maintaining protective immune responses during intestinal ***,increased dietary MtC may support the anti-IL-10-induced intestinal immunity to ***(n=640)were arranged in a 2×2×2 factorial design with 2 levels of each of the 3 main factors:dietary standardized ileal digestible(SID)MtC levels(0.6%or 0.8%),dietary anti-IL-10 supplementation(with or without),and coccidiosis challenge(control or challenge).Briefly,the broilers were supplied with either 0.6%or 0.8%SID MtC,each with or without anti-IL-10(300 mg/kg),from d 10 to *** d 14,broilers from each diet were gavaged with either PBS or *** resulting Eimeria infection induced fecal oocyst shedding and intestinal *** fed 0.8%SID MtC(main effects,P≤0.05)had decreased feed-to-gain ratio,increased duodenum and cecum luminal anti-Eimeria IgA titers,and decreased fecal oocyst counts,when compared to 0.6%SID *** supplementation of anti-IL-10(main effects,P≤0.05)increased cecum luminal total IgA concentration and decreased cecum ***(P≤0.05)were detected for growth performance and cecum luminal IfN-γ.Briefly,the highest body weight gain and feed intake were reached in PBS-gavaged broilers fed 0.8%SID MtC with no anti-IL-10 and in Eimeria-challenged broilers fed 0.8%SID MtC with *** Eimeria-infected broilers,anti-IL-10 increased intestinal luminal IfN-γ and body weight gain only at 0.8%SID ***,anti-IL-10 increased intestinal luminal IfN-γ levels,decreased cecum lesions and restored growth only when fed with adequate amounts of sulfur amino *** findings underscore the importance of providing sufficient essential nutrients to support the anti-IL-10 induced immunity against coccidiosis.

KAI2-and MAX2-Mediated Responses to Karrikins and Strigolactones Are Largely Independent of HY5 in Arabidopsis Seedlings

Karrikins are butenolide compounds released from burning vegetation that stimulate seed germination and enhance seedling photomorphogenesis. Strigolactones are structurally similar plant hormones that regulate shoot and root development, and promote the germination of parasitic weed seeds. In Arabidopsis, the f-box protein MAX2 is required for responses to karrikins and strigolactones, and the a/~ hydrolase KAI2 is necessary for responses to karrikins. Both MAX2 and KAI2 are essential for normal light-dependent seedling development. The bZIP transcription factor HY5 acts downstream of multiple photoreceptors and promotes photomorphogenesis, but its relationship with MAX2 and KAI2 in terms of seedling development and responses to karrikins and strigolactones is poorly defined. Here, we dem- onstrate that HY5 action is genetically separable from that of MAX2 and KAI2. While by5 mutants have weak hypoco- tyl elongation responses to karrikins and the artificial strigolactone GR24, they have normal transcriptional responses, suggesting that HY5 is not involved in perception or action of karrikins or strigolactones. furthermore, we show that overexpression of KAI2 is sufficient to enhance responses to both karrikins and GR24 in wild-type seedlings, and that KAI2 overexpression partially suppresses the hy5 long hypocotyl phenotype. These results suggest that KAI2 and MAX2 define a regulatory pathway that largely operates independently of HY5 to mediate seedling responses to abiotic signals such as smoke and light.

Genetic analysis of the maximum germination distance of Striga under fusarium oxysporum *** biocontrol in sorghum

Maximum germination distance（MGD） is an important component of Striga resistance in sorghum. The objective of this study was to determine gene action influencing MGD of Striga hermonthica and Striga asiatica among selected sorghum lines treated with a biocontrol agent, fusarium oxysporum f. sp. strigae（fOS） for effective breeding with Striga resistance, and fOS compatibility. Twelve sorghum genotypes were selected based on their Striga resistance, fOS compatibility, and superior agronomic performance. Selected genotypes were crossed using a bi-parental mating design to generate six families for genetic analysis. Agar-gel assays were used to determine low haustorium initiation factor（LHf） using the 12 parental lines, their f_1 progenies, backcross derivatives, and f_2 segregants in two sets. One set had S. hermonthica seed and the other one had S. asiatica seed. Both were treated with and without fOS. Genotypes were evaluated using a split-plot design with three replications and MGD data were recorded followed by generation mean analysis. fOS reduced MGD by 1cm under both S. hermonthica and S. asiatica infestations. Additive, dominance, and epistatic gene actions were involved in the control of MGD of the two Striga species in the evaluated populations. On average, the relative contribution of additive, additive×additive and dominance×dominance genetic effects on the MGD of S. hermonthica and S. asiatica, with fOS, were 20, 33, and 36%; and 21, 32, and 35%, respectively. Breeding methods exploiting these genetic effects may provide enhanced response to selection for Striga resistance and fOS compatibility in integrated Striga management（ISM） programmes.

Using a double-blind,randomized and counterbalanced,cross-over design,we assessed naproxen's effects on gastrointestinal(GI)distress and performance in eleven volunteers(6 male,5 female).Participants completed 4 trials:1)placebo and ambient);2)placebo and heat;3)naproxen and ambient;and 4)naproxen and *** variables were one placebo or 220 mg naproxen pill every 8 h(h)for 24 h and ambient(22.7±1.8℃)or thermal environment(35.7±1.3℃).Participants cycled 80 min at a steady heart rate then 10 min for maximum *** exertion was measured throughout *** distress was assessed pre-,during,post-,3 h post-,and 24 h post-cycling using a GI index for upper,lower,and systemic *** statistically significant differences occurred between conditions at any time for GI symptoms or perceived exertion,distance,or heart rate during maximum effort.A 24 h naproxen dose did not significantly affect performance or cause more frequent or serious GI distress when participants were euhydrated and cycling at moderate intensity in a thermal environment.

Epigastric hernias in children and the use of ultrasound in its diagnosis

Introduction There is a paucity of clinical data on pediatric epigastric hernias despite them accounting for up to 6%of all hernia repairs in *** aimed to provide additional data to supplement those 117 cases of a recent systematic review and to further clarify the role of ultrasound in diagnosing pediatric epigastric *** We retrospectively included all 60 patients treated for epigastric hernias in children in two tertiary pediatric surgical departments within 12 *** were tested via point-biserial correlation *** Epigastric hernias primarily affected preschool children with a median age of 39 *** vast majority of patients(88%)presented with swelling that was occasionally(30%)accompanied by *** defects could be found during clinical examination in 45%of patients with a median size of 5 mm(95%CI 3 to 10).Smaller defects were less likely to be palpable(r=-0.44,95%CI-0.08 to-0.7,p=0.021).Likewise,ultrasound was used more frequently with smaller fascial defect sizes(r=-0.51,95%Cl-0.16 to-0.74,p=0.007).Laparoscopic repair was used in 11 patients(19%)and more often(4/11)in combination with another simultaneous procedure than open repair(11/48).Conclusions Epigastric hernias are primarily a condition of the preschool *** can be beneficial if the diagnosis cannot be made clinically;otherwise,it is abdicable if it does not change the management of the patient's epigastric *** repairs might be beneficial for children with multiple defects or simultaneous procedures.

Prepubertal boys treated for cancer may exhibit impaired fertility in later life. A number of chemotherapeu tic agents have been identified as being gonadotoxic, and certain treatment regimens, such as that used for Hodgkin's disease, are particularly associated with subsequent infertility. Radiotherapy may also cause gonadal damage, most notably following direct testicular irradiation or total body irradiation. Because of the varied nature of the cytotoxic insult, it can be difficult to predict the likelihood of infertility in later life. Currently it is not possible to detect gonadal damage early due to the lack of a sensitive marker of gonadal function in the prepubertal age *** cryopreservation is currently the only method of preserving fertility in patients receiving gonadotoxic therapy. This is only applicable to postpubertal patients and can be problematic in the adolescent age group. At present there is no provision for the prepubertal boy, although there are a number of experimental methods currently being investigated. By harvesting testicular tissue prior to gonadotoxic therapy, restoration of fertility could be achieved following treatment, either by germ cell transplantation or by in vitro maturation of the germ cells harvested. Alternatively, rendering the testes quiescent during cytotoxic treatment may protect the germ cells from subsequent damage. In addition to the many scientific and technical issues to be overcome prior to clinical application of these techniques, a number of ethical and legal issues must also be addressed to ensure a safe and realistic prospect for future fertility in these patients.

Metabolic and Signaling Aspects Underpinning the Regulation of Plant Carbon Nitrogen Interactions

In addition to light and water, CO2 and mineral elements are required for plant growth and development. Among these factors, nitrogen is critical, since it is needed to synthesize amino acids, which are the building elements of protein, nucleotides, chlorophyll, and numerous other metabolites and cellular components. Therefore, nitrogen is required by plants in higher quantities and this investment in nitrogen supports the use of CO2, water, and inorganic nitrogen to produce sugars, organic acids, and amino acids, the basic building blocks of biomass accumulation. This system is maintained by complex metabolic machinery, which is regulated at different levels according to environmental factors such as light, CO2, and nutrient availability. Plants integrate these signals via a signaling network, which involves metabolites as well as nutrient-sensing proteins. Due to its importance, much research effort has been expended to understand how carbon and nitrogen metabolism are integrated and regulated according to the rates of photosynthesis, photorespiration, and respiration. Thus, in this article, we both discuss recent advances in carbon/nitrogen metabolisms as well as sensing and signaling systems in illuminated leaves of C3-plants and provide a perspective of the type of experiments that are now required in order to take our understanding to a higher level.
