Objective： The study aimed to evaluate the genotypic profiles of C. albicans （Candida albicans） sequentially isolated throughout the course of HIV infections, and to determine its MIC （minimal inhibitory concentrations） to AMB （amphotericin B）, FLC （fluconazole）, KTC （ketoconazole）, and ITC （itraconazole）. Design： samples were collected from the oral cavity of HIV-positive individuals during 4 years, with a sterilized swab. MIC was performed by using the microdilution method AFST/EUCAST. The genetic similarities within and between sequential clones of C. albicans were assessed by DNA fingerprinting using the random amplification ofpolymorphic DNA technique. Results： A total of 142 oral samples were isolated from 59 HIV-infected individuals who attempted up to five visits each, with or without symptoms of oropharyngeal candidiasis. Profile analysis revealed that yeasts isolated over sequential visits from symptomatic or asymptomatic individuals showed 78% or 87% relatedness, respectively. The degree of similarity among C. albicans was higher for isolates from colonization than for those from infection. Genetically identical C. albicans samples also formed connected subelusters in sequential visits. In regard to susceptibility profile, all isolates were susceptible to AMB, FLC, KTC, and ITC and maintained this pattern all along, no differences in MICs of any given antifungal compound were observed for sequential C. albicans isolates. Conclusions： These data suggest that genotype and susceptibility to antifimgal drugs were maintained over time in sequentially isolates of C. albicans colonization and a diverse evolutionary genetic trend in C. albicans sequentially isolated from the oral eandidiasis of HIV infected individuals.

Objective: As isolates of Cryptococcus are frequently kept collection stocks in institutions, sometimes without proper characterization, we sought to determine the genotype profiles, protease and phospholipase activities “in vitro” and the susceptibility testing for azoles and amphotericin B. Methodology: 84 isolates from several regions of Brazil (40 samples from clinical origin and 44 isolates from environmental origin) were maintained at the microorganism’s bank of the Biomedical Science Institute (ICB-USP) of the São Paulo University, in São Paulo, Brazil. This isolates was submitted fungal strains determination, DNA extraction and purification, determination of genotype by URA5 gene RFLP of CGB-positive isolates, protease and phospholipase activity and susceptibility to antifungals. Results: Of six CGB positive isolates tested by RFLP-PCR, only four presented a genomic profile consistent C. gattii species (VGII), while two other were C. neoformans (VNI and VNIII), indicating the existence of canavanine-resistante C. neoformans isolates in the culture collections. The clinical isolates secreted higher levels of phospholipase and environmental isolates but no differences were observed for the protease levels. Almost all isolates were sensible to azoles and amphotericin B. Conclusion: We point out in this research the existence of C. neoformans strains resistant to canavanine and intrinsic characteristic of C. gatti. These results demonstrate the importance to perform a detailed characterization of isolates kept in culture collections.

Rock slope hazard assessment is an important part of risk analysis for open pit *** main parameters that can lead to rock slope failures are the parameters traditionally used in geomechanical classifications,the slope geometrical parameters and external factors like rainfall and *** paper presents a methodology for a hazard assessment system for open pit mine slopes based on 88 cases collated around the world using principal components analysis,discriminant analysis and confidence *** historical cases used in this study included copper,gold,iron,diamond,lead and zinc,platinum and claystone *** variables used in the assessment methodology are uniaxial compressive strength of intact rock;spacing,persistence,opening,roughness,infilling and orientation of the main discontinuity set;weathering of the rock mass;groundwater;blasting method;and height and inclination of the *** principal component analysis was used to quantify the data,the discriminant analysis was used to establish a rule to classify new slopes about its stability *** provide a practical hazard assessment system,confidence ellipses were used to propose a hazard graph and generate the *** discriminant rule developed in this research has a high discrimination capacity with an error rate of 11.36%.

Trauma to the peripheral nervous system often results in loss of motor and sensory functions of the affected area of the body,leading to a series of functional impairments(Allodi et al.,2012).Injuries to peripheral nerves initiate a series of complex events,known as Wallerian degeneration,which allows the injured axons to regenerate and reinnervate their targets(Allodi et al.,2012).Damage to a nerve induces multiple alterations,which include:axonal degeneration,breakdown of myelin sheath,Schwann cells(SC)proliferation and conversion to a repair phenotype which express cytokines and chemokines that allow the infiltration and activation of macrophages(Mietto et al.,2015;Jessen and Mirsky,2016).

Injuries to the spinal cord result in permanent disabilities that limit daily life *** main reasons for these poor outcomes are the limited regenerative capacity of central neurons and the inhibitory milieu that is established upon traumatic *** decades of research,there is still no efficient treatment for spinal cord *** strategies are tested in preclinical studies that focus on ameliorating the functional outcomes after spinal cord *** these,molecular compounds are currently being used for neurological recovery,with promising *** molecules target the axon collapsed growth cone,the inhibitory microenvironment,the survival of neurons and glial cells,and the re-establishment of lost *** this review we focused on molecules that are being used,either in preclinical or clinical studies,to treat spinal cord injuries,such as drugs,growth and neurotrophic factors,enzymes,and *** mechanisms of action of these molecules are discussed,considering traumatic spinal cord injury in rodents and humans.

Injection of bone marrow mesenchymal stem cells by intravenous or intraperitoneal routes is a viable alternative to spinal cord injury treatment in mice

In spite of advances in surgical care and rehabilitation, the consequences of spinal cord injury （SCI） are still challenging. Several experimental therapeutic strategies have been studied in the SCI field, and recent advances have led to the development of therapies that may act on the inhibitory microenvironment. Assorted lineages of stem cells are considered a good treatment for SCI. This study investigated the effect of systemic transplantation of mesenchymal stem cells （MSCs） in a compressive SCI model. Here we present results of the intraperitoneal route, which has not been used previously for MSC administration after compressive SCI. We used adult female C57BL/6 mice that underwent laminectomy at the T9 level, followed by spinal cord compression for 1 minute with a 30-g vascular clip. The animals were divided into five groups： sham （anesthesia and laminectomy but without compression injury induction）, MSC i.p. （intraperitoneal injection of 8×10^5 MSCs in 500 μL of DMEM at 7 days after SCI）, MSC i.v. （intravenous injection of 8 × 10^5 MSCs in 500μL of DMEM at 7 days after SCI）, DMEM i.p. （intraperitoneal injection of 500μL of DMEM at 7 days after SCI）, DMEM i.v. （intravenous injection of 500 μL of DMEM at 7 days after SCI）. The effects of MSCs transplantation in white matter sparing were analyzed by luxol fast blue staining. The number of preserved fibers was counted in semithin sections stained with toluidine blue and the presence of trophic factors was analyzed by immunohistochemistry. In addition, we analyzed the locomotor performance with Basso Mouse Scale and Global Mobility Test. Our results showed white matter preservation and a larger number of preserved fibers in the MSC groups than in the DMEM groups. Furthermore, the MSC groups had higher levels of trophic factors （brain-derived neurotrophic factor, nerve growth factor, neurotrophin-3 and neurotrophin-4） in the spinal cord and improved locomotor performance. High‐temperature superconductors are a powerful technological option to be applied in the current scenario of energy *** applications include fault current limiters,power electrical cables,and electrical machines,for *** to the non‐linearities of superconductors,it is computationally costly to run real models of superconducting ***,it is of paramount importance to have a reliable and fast formulation to model superconducting *** paper proposes a new hybrid J‐A formulation to simulate superconducting *** new formulation is validated with 5 case studies,some of which are *** J‐A formulation agrees in all cases and has a smaller computation time when compared with the T‐A ***,due to the simple implementation,the proposed formulation allows the possibility of running the J and A formulations in the same order and presents itself as a potential feature to speed up and help the design of the superconducting devices.

The application of High‐Temperature Superconductor(HTS)coils made of coated conductors has been investigated for many years.A possible configuration for such coils is the jointless loop,also known as the ring *** double crossed loop coil(DCLC)has been successfully applied in superconducting magnetic bearings(SMBs).The design of SMBs with DCLCs requires flexible modelling to allow all parts of the device to be *** work proposes the T‐A formulation with a thin‐film approximation for modelling SMB with DCLCs in the finite element analysis framework.A 2D representation of the system is coupled with an external electric circuit to model the continuity of the lines that represent the parts of each jointless *** couple the T‐A formulation and the circuit,an average of the total electric field,with both resistive and inductive components,is applied to the *** total current computed by the circuit is applied to the T‐A *** proposed methodology was validated by comparison with levitation force experimental *** types of tests were simulated:five levitation force tests and three guidance force *** is shown that there is a limit to the behaviour of the levitation force related to the high‐loss *** this limit,the stack of DCLCs behaves as an equivalent *** this limit,a high‐loss state appears as a linear growth of the levitation *** is also shown that this high‐loss state in vertical displacement influences the lateral force.

The municipality of Boa Nova,in northeastern Brazil,is in an ecotone zone between the Caatinga and Atlantic Forest phytogeographic *** transition phytophysiognomy is seasonal forest and known locally as mata de cipó.In these phytophysiognomies there are lajedos,which are rock outcrops colonized by vegetation welladapted to extreme microclimatic variation and vegetation diversity is affected by the vegetation types of the surrounding *** to the singularity of these environments and the relevance of floristic studies for conservation,this work aimed to identify the species richness and compare the similarity of the flora on four rock outcrops in Boa *** flora was surveyed during exploratory walks along lajedos between 2016 and *** total,162 species were identified on the Boa Nova *** flora has a composition and structure similar to semiarid outcrops,as well as endemic species that also occur in surrounding *** the proximity,a similarity index revealed there is floristic dissimilarity between the *** new occurrences were recorded for the region,five species are threatened with extinction(Aosa gilgiana,Ficus cyclophylla,Hippeastrum stigmovittatum,Pleroma caatingae and Trixis pruskii),and 43 species are common in anthropogenic *** reinforces the importance of actions to conserve these areas.

Renin-angiotensin system in the pathogenesis of liver fibrosis

Hepatic fibrosis is considered a common response to many chronic hepatic injuries. It is a multifunctional process that involves several cell types, cytokines, chemokines and growth factors leading to a disruption of homeostatic mechanisms that maintain the liver ecosystem. In spite of many studies regarding the development of fibrosis, the understanding of the pathogenesis remains obscure. The hepatic tissue remodeling process is highly complex, resulting from the balance between collagen degradation and synthesis. Among the many mediators that take part in this process, the components of the Renin angiotensin system （RAS） have progressively assumed an important role. Angiotensin （Ang） II acts as a profibrotic mediator and Ang-（1-7）, the newly recognized RAS component, appears to exert a counter-regulatory role in liver tissue. We briefly review the liver fibrosis process and current aspects of the RAS. This review also aims to discuss some experimental evidence regarding the participation of RAS mediators in the pathogenesis of liver fibrosis, focusing on the putative role of the ACE2-Ang-（1-7）- Mas receptor axis.
