There is a need to develop interventions to slow or reverse the degeneration of dopamine neurons in Parkinson’s disease after *** that preclinical and clinical studies suggest benefits of dietary n-3 polyunsaturated fatty acids,such as docosahexaenoic acid,and exercise in Parkinson’s disease,we investigated whether both could synergistically interact to induce recovery of the dopaminergic ***,mice received a unilateral stereotactic injection of 6-hydroxydopamine into the striatum to establish an animal model of nigrostriatal *** weeks after lesion,animals were fed a docosahexaenoic acid-enriched or a control diet for the next 8 *** this period,the animals had access to a running wheel,which they could use or *** acid treatment,voluntary exercise,or the combination of both had no effect on(i)distance traveled in the open field test,(ii)the percentage of contraversive rotations in the apomorphine-induction test or(iii)the number of tyrosine-hydroxylase-positive cells in the substantia nigra pars ***,the docosahexaenoic acid diet increased the number of tyrosine-hydroxylase-positive terminals and induced a rise in dopamine concentrations in the lesioned *** to docosahexaenoic acid treatment or exercise alone,the combination of docosahexaenoic acid and exercise(i)improved forelimb balance in the stepping test,(ii)decreased the striatal DOPAC/dopamine ratio and(iii)led to increased dopamine transporter levels in the lesioned *** present results suggest that the combination of exercise and docosahexaenoic acid may act synergistically in the striatum of mice with a unilateral lesion of the dopaminergic system and provide support for clinical trials combining nutrition and physical exercise in the treatment of Parkinson’s disease.

Zeros in Accelerometer Files A Comparison Study of Ten Published Nonwear Algorithms

Despite the increasing use of accelerometers, the handling of zero values in data files remains understudied. Using data from the QUALITY （QUebec Adipose and Lifestyle Investigation in Youth） cohort, output from 10 published nonwear algorithms were compared in 520 children aged 8-10 years who wore an Actigraph accelerometer for 7 days. These algorithms differed by： 1） the minimal duration of consecutive zeros required to be designated as nonwear （10-90 minutes）; 2） the presence and magnitude of an allowance period, and 3） the presence of a specific time frame. Significant differences were obtained between all but two algorithms, including： 1） wear time differences up to 117 minutes per day; 2） differences in counts per minutes as high as 80; and 3） differences in sedentary time up to two hours daily. Estimates of light and moderate-to-vigorous physical activity were less affected. This study suggests that the nonwear algorithm used can have a significant effect on estimates of wear duration, overall physical activity level and sedentary time in children. Until technological developments bring to zero nonwear time, specifying the nonwear algorithm used when publishing results is essential to ensure accurate comparison between studies.

We studied late-entry commercial thinning effects on growth, yield, and regeneration in a 48-year-old jack pine(Pinus banksiana Lamb.) stand. Applied thinning intensities were 27, 32, and 47% of merchantable basal area(BA) excluding skidding trails. After 15 years, mean diameter at breast height of surviving trees in the 47% BA removal increased by 4.9 cm(25%) compared to the unthinned control. The 47% BA removal also increased gross merchantable volume(GMV) tree-1by 46% compared to the control. The 27% BA removal had twice as much GMV ha-1compared to the 47% BA removal after15 years. Moreover, cumulative GMV ha-1was much higher in the 27% BA removal than in the unthinned control. The highest thinning intensity produced larger trees on average, while the lowest thinning intensity maximized volume production per hectare. Maintenance of acceptable growing stock throughout the 15-year period in the 27% BA removal could provide other ecosystem functions such as biodiversity enhancement or wildlife habitat by delaying senescence. Regeneration data showed that a shift in species composition occurred in the understory. After 15 years, the understory was dominated by black spruce(Picea mariana(Mill.) B.S.P.), white birch(Betula papyrifera Marsh.), and trembling aspen(Populus tremuloides Michx.). If regenerating jack pine is an objective after final overstory removal, additional efforts will be needed to re-establish this species.

The persistent challenges in attracting and retaining a diverse healthcare workforce, with a specific focus on nurses, have become increasingly pronounced in recent years. These hurdles have been exacerbated by a growing difficulty in retaining young nurses, thereby exacerbating labor shortages driven by demographic shifts and the retirement of experienced nursing professionals. While most research efforts have concentrated on the broader issue of nurse retention, our study is centered on a specific demographic—young nurses. Our research endeavors to shed light on the unique challenges faced by young nurses through a qualitative survey involving nursing students who are simultaneously employed. We seek to discern the multifaceted obstacles they encounter in both their academic environment and the healthcare organizations where they work. While certain challenges are linked to course organization, examinations, and the time required for studying, our respondents overwhelmingly emphasize the pivotal role of the work environment in facilitating the harmonization of work, family, and educational commitments. This reconciliation is achieved through measures such as flexible working arrangements and the efficient organization of nursing duties. The primary objective of our research is to provide insights into how these diverse challenges can be effectively addressed and how a range of measures can significantly contribute to the attraction and retention of nursing students, as well as the long-term retention of nurses within the healthcare system. Our recommendations are intended to be of practical use to a wide array of stakeholders, including academic institutions, particularly colleges and universities offering nursing programs, as well as hospitals, clinics, and other healthcare institutions that hire nurses. Farnesoid X receptor （FXR） is a ligand-activated nuclear receptor that plays a central role in regulating genes involved in bile acid homeostasis, and fat and glucose metabolism. Here, we demonstrate a post-translational interplay between FXR phosphoryl- ation, SUMOylation, and ubiquitination that directs the receptor into an activation-degradation pathway in hepatocytes. We iden- tify a non-canonical SUMOylation motif termed pSuM that conjugates SUM02 at Lys-325 of FXR under the direct control of casein kinase 2 （CK2）, which provides the required negative charge for Ubc9 and PIAS1 to perform SUMOylation, by phosphorylating Ser-327. Lys-325 SUMOylation is indispensable to the promotion of efficient ligand activation and transcriptional coactivation of FXR. Constitutive pSuM activation using a phospho-mimic Ser-327 mutant or catalytic CK2 expression strongly induces SUM02 conjugation, which directs FXR ubiquitination and proteasome-dependent degradation. We also determine that such SUMOylation-dependent ubiquitination of FXR is mediated by the E3 ubiquitin ligase RNF4, which is required to achieve maximal induction of FXR and optimal up- or downregulation of responsive genes involved in bile acid homeostasis and liver regeneration. Our findings identify a highly regulated atypical SUMO conjugation motif that serves to coordinate FXR transcriptional compe- tence, thereby expanding the intricate dynamics of the SUMOylation process used by incoming signals to govern metabolic gene regulation.

AIM To investigate the use of droplet digital polymerase chain reaction(dd PCR) for detecting host m RNA markers in stools as a non-invasive test for colorectal cancer *** dd PCR and quantitative PCR were compared side by side for their performance in the detection of ITGA6 and ITGA6 A transcripts in stool samples obtained from patients with various types of colorectal lesions(advanced adenomas and stage Ⅱ-Ⅳ colorectal cancers) and control(patients displaying no pathological findings) using duplex Taq Man reactions for both methods. ITGA6 and ITGA6 A were chosen for this proof-of-concept study based on their relative medium and low abundance in stool samples, respectively, as established in a previous *** We found that the dd PCR and q PCR methods per-formed equally well in this Taq Man duplex assay for the detection of ITGA6 and ITGA6 A transcripts in stools of patients with colorectal lesions. For ITGA6, receiver operating characteristic(ROC) curve analysis showed comparable areas under the curve of 0.91(P evels in control patients, was found to be significantly elevated(over 40 times) in stage Ⅱ and Ⅲ colorectal cancers(P evealed a strong correlation of the copy numbers obtained by dd PCR and q PCR for both ITGA6 and ITGA6 *** We found that ITGA6 and ITGA6 A detection in stools of patients with colorectal cancers with dd PCR is comparable to that of q PCR using Taq Man assays.
