Trigonelline protects the cardiocyte from hydrogen peroxide induced apoptosis in H9c2 cells

Objective:To elucidate the key parameters associated with hydrogen peroxide induced oxidative stress and investigates the mechanism of trigonelline(TG)for reducing the H_2O_2induced toxicity in H9c2 ***:Cytotoxicity and antioxidant activity of TG was assessed by EZ-CYTOX *** extraction and cDNA synthesized according to the kit manufacture *** was measured by the Flowcytometry,general PCR and ***:It was found that the TG significantly rescued the morphology of the H9c2 *** of cells with TG attenuated H_2O_2 induced cell deaths and improved the antioxidant *** addition,TG regulated the apoptotic gene caspase-3,caspase-9 and anti-apoptotic gene ***-XL during H_2O_2 induced oxidative stress in H9c2 *** results were comparable with quercetin *** evident,flow cytometer results also confirmed the TG significantly reduced the H_2O_2 induced necrosis and apoptosis in H9c2 ***,further increment of TG concentration against H_2O_2 could induce the necrosis and apoptosis along with H_2O_***:It is suggested that less than 125μM of TG could protect the cells from H_2O_2 induced cell damage by down regulating the caspases and up regulating the Bcl-2 and Bcl-XL ***,we suggest the trigonelline could be useful for treatment of oxidative stress mediated cardiovascular diseases in future.

Isolation and identification of alkaloids and anthocyanins from flower and bulb of Lycoris radiata using HPLC and LC-ESI-MS

Three anthocyanins (cyanidin 3-diglucoside, cyanidin 3-sambubioside and cyanidin 3-glucoside) together with eleven known alkaloids (lycoricidine, hipppeastrine,O-Demethyllycoramin, lycoricidinol, galanthine, lycorine, lycorenine, lycoramine, galanthamine, homolycorine and pretazettine) were identified in the flower and bulb of Lycoris radiata using high-performance liquid chromatography coupled with UV detection and electrospray ionization mass spectrometry. Anthocyanins play a major role in protecting plant’s DNA from the UV spectrum of sunlight and also in attracting insects for the purpose of pollination. Thus, knowledge on the contents and types of anthocyanins of L. radiata will help to evaluate the adaptive evolution of flowers and provide useful information for the ornamental breeding.

Quantitative differentiation of phenolic compounds in different varieties of buckwheat cultivars from China, Japan and Korea

The aim of this study was to determine the variation of phenolic compounds in common and tartary buckwheats collected from China, Japan and Korea. Two buckwheat varieties of each country were cultivated using nutrient solution and vermiculite in the greenhouse from February to May, 2012 and harvested at two-week intervals. Phenolics such as chlorogenic acid, C-glycosylflavones (orientin, isoorientin, vitexin, and isovitexin), rutin, and quercetin were quantified using high performance liquid chroma-tography (HPLC). At 17, 29, 44, 58, and 72 days after sowing (DAS), before harvesting each organs, rutin content of cultivars from China and Korea at 17 DAS was the highest (mean 43.63 and 39.95 mg?g–1 DW, respectively) than that of 29 and 44 DAS. However, two varieties from Japan at 44 DAS were documented maximum;whereas, the amount of rutin in “Hokkai T10” harvested at 44 DAS (58.36 mg?g–1 DW) was the highest of all cultivars. Rutin level in the leaves and stems at 72 DAS were comparatively higher than at 58 DAS. The highest of rutin content was found at 72 DAS (73.33 mg?g–1 DW) in “Hokkai T10” leaves followed by “Daegwan No.3-3” (61.13 mg?g–1 DW), “rice tartary” (53.89 mg?g–1 DW). Rutin content of flowers was presented as the highest amount in “Hokkai T10” at 72 DAS (88.3 mg?g–1 DW) was approximately 14-fold higher than that of 58 DAS (6.44 mg?g–1 DW). However, rutin content of flower in “Xiqiao No.2” was 3-fold higher at 72 DAS than at 58 DAS. Rutin and total phenolic compounds content in flowers, leaves, and stems were higher in the order. The content of four C-glycosylflavones in common buckwheat was higher than those in tartary buckwheat, but rutin content which accounted >90% of the total phenolic compounds was higher in tartary buckwheat. The highest amounts of chlorogenic acid and quercetin were measured in the flower of “Xiqiao No.2” (6.85 and 11.69 mg?g–1 DW, respectively). Identification and Determination of Phenolic Compounds in Rapeseed Meals (Brassica napus L.)

This study aimed in the identification and quantification of phenolic compounds in different four varieties of rapeseeds (Brassica napus L.) using LC-MS and HPLC. LC-MS analysis guided to identify 12 phenolic compounds including sinapine, sinapine(4-O-8')guaiacyl, feruloyl choline(4-O-8') guaiacyl, kaempferol-3-O-sinapoyl-sophoroside-7-O-glucoside, kaempferol-3-O-β-sophoroside, cyclic spermidine-alkaloid, feruloyl choline(5-8')guaiacyl, kaempferol-sinapoyl-trihexoside,1,2-di- O-sinapoyl-gentiobiose, 1,2-di-O-sinapoyl-glucose, feruloyl choline(4-O-8')guaiacyl-di-sinapoyl, and sinapine(4-O-8')guaiacyl-di-sinapoyl, respectively. Total phenolic compounds in rapeseed meals were ranged from 38.50 (2012 “Sunmang”) to 63.95 mg/g dry weight (DW) (2014 Jeju local cultivar). Main phenolic compositions were cyclic spermidine-alkaloid, kaempferol, feruloyl choline, and sinapine. Sinapine was predominant compound among all the samples ranged from 29.74 to 52.24 mg/g DW (mean 79% of total phenolic contents). This study provided the information for the variation of phenolic compounds between the varieties of rapeseeds and the cultivation periods. Furthermore, this information can be developed for bio-energy industry as a reducer of the cost of the bio-energy products.

Life cycle cost analysis of new FRP based solar parabolic trough collector hot water generation system

Parabolic trough collectors (PTCs) are employed for a variety of applications including steam generation and hot water generation. This paper deals with the experimental results and an economic analysis of a new fibre reinforced plastic (FRP) based solar PTC with an embedded electronic controlled tracking system designed and developed for hot water generation in a restaurant in Madurai, India. The new collector performance has been tested according to ASHRAE Standard 93 (1986). The performance of a new PTC hot water generation system with a well mixed hot water storage tank is investigated by a series of extensive tests over ten months period. The average maximum storage tank water temperature observed was 74.91 °C, when no energy is withdrawn from the tank to the load during the collection period. The total cost of the new economic FRP based solar PTC for hot water generation with an embedded electronic controlled tracking system is Rs. 25000 (US$ 573) only. In the present work, life cycle savings (LCS) method is employed for a detailed economic analysis of the PTC system. A computer program is used as a tool for the economic analysis. The present worth of life cycle solar savings is evaluated for the new solar PTC hot water generation system that replaces an existing electric water heating system in the restaurant and attains a value of Rs. 23171.66 after 15 years, which is a significant saving. The LCS method and the MATLAB computer simulation program presented in this paper can be used to estimate the LCS of other renewable energy systems.

Heat Transfer Analysis of an Elevated Linear Absorber with Trapezoidal Cavity in the Linear Fresnel Reflector Solar Concentrator System

This paper describes various aspects of the design methodology and heat transfer calculations for an elevated linear absorber. The absorber is a part of the linear Fresnel reflector solar concentrator system, in which hot fluid is generated. The design of the absorber is an inverted trapezoidal air cavity with a glass cover enclosing a multi tube absorber. In a trapezoidal cavity absorber, a set of linear multi tube absorber with plate(named as "plane surface") and without plate(named as "tube surface") underneath are considered. An analytical simulation is done for different gaps between the tubes and for different depths of the cavity. A better design of the absorber is found out to maximize the heat transfer rate supplied to the absorber tube fluid. Also, the experimentally obtained overall heat loss coefficients are compared with the analytical values for the considered arrangements of absorber set up and results are discussed in details.

Characterization and phylogenetic analysis of novel polyene type antimicrobial metabolite producing actinomycetes from marine sediments:Bay of Bengal India

Objective:To isolate and indentify the promising antimicrobial metabolite producing Streptomyces strains from marine sediment samples from Andraprudesh coast of ***:Antagonistic aetinomycetes were isolated by starch casein agar medium and modified nutrient agar medium with 1%glucose used as a base for primary *** antimicrobial metabolite producing strains were selected and identified by using biochemical and 16S rDNA *** inhibitory concentrations of the organic extracts were done by using broth micro dilution ***:Among the 210 actinomyeetes,64.3%exhibited activity against Gram positive bacteria,48.5%showed activity towards Cram negative bacteria,38.8%exhibited both Cram positive and negative bacteria and 80.85%isolates revealed significant antifungal ***,five isolates AP-5,AP-18,AP-41 and AP-70 showed significant antimicrobial *** analysis of cell wall hydrolysates showed the presence of LL-diaminopimelic acid and glycine in all the *** analysis indicated that the isolates shared 98.5%-99.8%sequence identity to the 16S rDNA gene sequences of the Streptomyces *** antimicrobial substances were extracted using hexane and ethyl acetate from spent medium in which strains were cultivated at 30X for five *** antimicrobial activity was assessed using broth micro dilution *** of the culture extracts from these five strains showed a typical polyenelike *** lowest minimum inhibitory concentrations of ethyl acetate extracts against Escherichia coli and Cumularia lunula were 67.5 and 125.0μg/mL,***:It can be concluded that hexane and ethyl acetate soluble extracellular products of novel isolates are effective against pathogenic bacteria and fungi.
