ABCB1 and ABCB19 auxin transporters have synergistic effects on early and late Arabidopsis anther development

Arabidopsis abcbl abcb19 double mutants defective in the auxin transporters ABCB1/PGP1 and ABCB19/PGP19 are altered in stamen elongation, anther dehiscence and pollen maturation. To assess the contribution of these transporters to stamen development we performed phenotypic, histological analyses, and in situ hybridizations on abcbl and abcbl9 single mutant flowers. We found that pollen maturation and anther dehiscence are precocious in the abcbl but not in the abcb19 mutant. Accordingly, endothecium ligniflcation is altered only in abcbl anthers. Both abcbl and abcb1 abcb19 stamens also show altered early development, with asynchronous anther Iocules and a multilayer tapetum. DAPI staining showed that the timing of meiosis is asynchronous in abcbl abcb19 anther Iocules, while only a small percentage of pollen grains are non- viable according to Alexander＇s staining. In agreement, TAM （TARDY ASYNCHRONOUS MEIOSIS）, as well as BAM2 （BARELY ANY MERISTEM）involved in tapetal cell development--areoverexpressed in abcbl abcb19 young flower buds. Corre- spondingly, ABCB1 and ABCB19 mRNA localization supports the observed phenotypes of abcbl and abcbl abcb19 mutant anthers. In conclusion, we provide evidence that auxin transport plays a significant role both in early and late stamen development： ABCB1 plays a major role during anther development, while ABCB19 has a synergistic role.

An experimental investigation of the crystallographic,Raman and transport properties of the Ce_(1-x)(Nd_(0.74)Tm_(0.26))_xO_(2-x/2)(0.1≤x≤0.6) doped ceria system was performed with the aim of setting out correlations between structural features and ionic conductivity of the *** chosen composition ensures that the average size of the Nd^(3+) and Tm^(3+) doping ions coincides with the one of Sm^(3+);even so,the studied system presents larger cell parameters and a wider compositional extent of the CeO_(2)-based solid solution than Sm-doped ***,the occurrence of two different activation energies to ionic conduction below and above ~750 K determines the existence of two distinct conduction *** described experimental results agree with the formation below the threshold temperature of1 V_O2 Tm'_(Ce) trimers,which promote the incorporation of Nd'_(Ce) isolated defects into the CeO_(2)-based solid *** the high temperature range the dissociation of trimers induces the appearance of a lower activation energy;the extrapolation of its value at infinite dilution provides a result in good accordance with the expected binding energy of 1 V_ORE'_(Ce) dimers,pointing at their stability even in the high temperature conduction regime.

Hepatitis C virus micro-elimination:Where do we stand?

Hepatitis C virus (HCV) elimination by 2030, using direct-acting antiviraltreatments, has been promoted by the World Health Organization. Thisachievement is not attainable, however, particularly after the 2020 pandemic ofthe coronavirus disease 2019. Consequently, the more realistic objective ofeliminating HCV from population segments for which targeted strategies ofprevention and treatment are easily attained has been promoted in Europe, as avalid alternative. The underlying idea is that micro-elimination will ultimatelylead to macro-elimination. The micro-elimination strategy may target differentspecific populations and at-risk groups. Different settings, including prisons andhospitals, have also been identified as micro-elimination scenarios. In addition,dedicated micro-elimination strategies have been designed that are tailored at thegeographical level according to HCV epidemiology and individual country’sincome. The main elements of a valid and successful micro-elimination project arereliable epidemiological data and active involvement of all the *** involvement represents another essential component for a successfulprogram.

Interaction of major genes predisposing to hepatocellular carcinoma with genes encoding signal transduction pathways influences tumor phenotype and prognosis

Studies on rodents and humans demonstrate an inherited predisposition to hepatocellular carcinoma （HCC）. Analysis of the molecular alterations involved in the acquisition of a phenotype resistant or susceptible to hepatocarcinogenesis showed a deregulation of G1 and S phases in HCC of genetically susceptible F344 rats and a G1-S block in lesions of resistant Brown norway （BN） rats. Unrestrained extracellular signal-regulated kinase （ERK） activity linked to proteasomal degradation of dual-specificity phosphatase 1 （DUSP1）, a specific ERK inhibitor, by the CKS1-SKP2 ubiquitin ligase complex occurs in more aggressive HCC of F344 rats and humans. This mechanism is less active in HCC of BN rats and human HCC with better prognosis. Upregulation of iNos cross-talk with IKK/NF-KB and RAS/ERK pathways occurs in rodent liver lesions at higher levels in the most aggressive models represented by HCC of F344 rats and c-Myc-TGF-α transgenic mice. iNOS, IKK/NF-κB, and RAS/ERK upregulation is highest in human HCC with a poorer prognosis and positively correlates with tumor proliferation, genomic instability and microvascularization, and negatively with apoptosis. Thus, cell cycle regulation and the activity of signal transduction pathways seem to be modulated by HCC modifier genes, and differences in their efficiency influence the susceptibility to hepatocarcinogenesis and probably the prognosis of human HCC.

Liver injury has been reported in coronavirus disease 2019(COVID-19)cases but the impact of pre-existing liver damage and related etiology have not been completely *** research interests include the potential reciprocal influence of COVID-19 and pre-existing liver damage related to hepatitis C virus(HCV)infection,in *** this end,we have evaluated three cohorts of patients admitted at three Italian hospitals during the coronavirus pandemic;these included 332 patients with COVID-19 and 1527 patients with HCV who were from established real-world antiviral treatment study cohorts(sofosbuvir/velpatasvir),with either liver disease(various severities;n=1319)or cirrhosis(n=208).Among the COVID-19 patients,10 had cirrhosis(3%),including 7 of metabolic origin and 3 of viral *** among the COVID-19 patients was 27.1%,with 70%of those with cirrhosis of metabolic etiology having ***,older age,low white blood cell count and lymphocyte count being identified as risk predictors of death[odds ratio(OR)=13.7,95%confidence interval(CI):2.59-83.01,P=0.006;OR=1.05,95%CI:1.03-1.08,P=0.0001;OR=1.09,95%CI:1.36-1.16,P=0.001;OR=0.61,95%CI:0.39-0.93,P=0.023,respectively].In the two cohorts of HCV patients,COVID-19 diagnosis was made in 0.07%of those with liver disease and 1%of those with ***,the prevalence of HCV antibodies among COVID-19-infected patients was comparable to that currently reported for the general population in *** the COVID-19 patients,pre-existing metabolic cirrhosis appears to be associated with higher mortality,while HCV antibodies may be suggestive of“protection”against COVID-19.

Assay of ghrelin concentration in infant formulas and breast milk

AIM: To test if total ghrelin is present in infant formulas. METHODS: Using a radioimmunoassay, we measured total ghrelin concentrations in 19 samples of commercial infant formulas and in 20 samples of human milk. We also determined ghrelin concentration in the serum of infants and lactating mothers. RESULTS: Ghrelin concentrations were significantly higher in artificial milk (2007.1 ± 1725.36 pg/mL) than in human milk (828.17 ± 323.32 pg/mL) (P = 0.005). The mean ghrelin concentration in infant serum (n = 56) was 1115.86 ± 42.89 pg/mL, and was significantly higher (P = 0.023) in formula-fed infants (1247.93 ± 328.07 pg/mL) than in breast-fed infants (1045.7 ± 263.38 pg/mL). The mean serum ghrelin concentration (mean ± SD) in lactating mothers (n = 20) was 1319.18 ± 140.18 pg/mL. CONCLUSION: This study provides evidence that total ghrelin is present in infant formulas. This findingraises diverse questions regarding the uptake, absorption and metabolic effects of this hormone.

The authors focused on the current surgical treatment of resectable gastric cancer, and significance of peri- and post-operative chemo or chemoradiation. Gastric cancer is the 4th most commonly diagnosed cancer and the second leading cause of cancer death worldwide. Surgery remains the only curative therapy, while perioperative and adjuvant chemotherapy, as well as chemoradiation, can improve outcome of resectable gastric cancer with extended lymph node dissection. More than half of radically resected gastric cancer patients relapse locally or with distant metastases, or receive the diagnosis of gastric cancer when tumor is disseminated; therefore, median survival rarely exceeds 12 mo, and 5-years survival is less than 10%. Cisplatin and fluoropyrimidine-based chemotherapy, with addition of trastuzumab in human epidermal growth factor receptor 2 positive patients, is the widely used treatment in stage IV patients fit for chemotherapy. Genetic susceptibility to Candida infection： a new look at an old entity

The identification of the first molecular defect leading to Mendelian Susceptibility to Mycobacterial Disease ;MD）,1 a rare syndrome conferring predisposition to（MSMD）,1 a rare syndrome conferring predisposition to disease caused by weakly virulent mycobacteria （such as Mycobacterium bovis, Bacille Calmette Gu6rin vaccines and environmental mycobacteria）, has led to a paradigm shift in the field of primary immunodeficiencies in the last two decades. The ＂classic＂ patient with multiple immunologic abnormalities, conferring a broad susceptibility to multiple and recurrent infectious diseases caused by both weakly pathogenic and more virulent microorganisms, was the main Primary Immunodeficiency （PID） phenotype identified. Since 1996, mutations causing MSMD have been found in at least 6 genes related to interferon （IFN）-7-mediated immunity.2

Allergic and nonallergic rhinitis in children: The role of nasal cytology

Nasal cytology is a diagnostic tool currently used in rhinology to study either allergic and vasomotor rhinological disorders or infectious and inflammatory rhinitis. Over the past few years nasal cytology has been rarely used in pediatrics, nevertheless its clinical and scientific applications seem to be very promising. The advantages of this technique are different: the ease of performance, the noninvasiveness allowing repetition and the low cost. We evaluated 100 children, from 2 to 15 years old, referred to our outpatient service for allergic children for suspected allergic rhinitis (AR). After skin prick test (SPT) or Radio Allergo Sorbent Test (RAST), 59/100 subjects were classified as affected by AR, while 8 children refused to be tested. According to ARIA guidelines, the 59 children with AR (4 - 15 years old) were divided in 56 with persistent AR and 3 with an intermittent form. Nine out of 59 children with AR had a significant number of neutrophils and eosinophils at the nasal cytology, documenting the presence of “minimal persistent inflammation”. Eleven out of 59 AR patients showed a positive swab for bacteria. Children with nonallergic rhinitis (NAR) were 33/100 (2 - 15 years old). After nasal cytology, 17/33 children were classified as NARES (nonallergic rhinitis with eosinophils), including one X-linked agammaglobulinemia (XLA) child, 1/33 as NARESMA (nonallergic rhinitis with eosinophils and mast cell) and another 1/33 as NARMA (nonallergic rhinitis with mast cell). In conclusion, nasal cytology allowed us to correctly classify children with NAR and to better assess the condition of children with AR.
