Chemical composition, antiparasitic and cytotoxic activities of aqueous extracts of Ziziphus joazeiro mart

Objective: To compare the in vitro antiparasitic activity of aqueous extracts from Ziziphus joazeiro leaves and stem bark against Trypanosoma cruzi, Leishmania braziliensis, and Leishmania infantum, as well as to evaluate its cytotoxicity in mammalian cells, in addition to identifying the chemical composition of the extracts. methods: Ziziphus joazeiro leaf and stem bark aqueous extracts were prepared by cold extraction maceration and subjected to ultra-efficient liquid chromatography coupled to a quadrupole/time of flight system. The susceptibility assays used Trypanosoma cruzi CLB5 strains and promastigote forms of Leishmania braziliensis and Leishmania infantum for antiparasitic activity of the extracts. moreover, mammalian fibroblasts NCTC clone 929 were used for cytotoxicity analysis. Results: Terpenoid compounds, flavonoids and phenolic acid were identified in extracts. The stem bark aqueous extracts presented more significant results in terms of antiparasitic activity compared with the leaf aqueous extracts, especially against Leishmania braziliensis and Leishmania infantum promastigote forms with an IC50 mL. The cytotoxicity evaluation showed moderate toxicity of the stem bark aqueous extracts, which is relevant information for the rational use of this plant part since it is widely used by the population. Conclusions: These preliminary results may contribute to the formulation of new therapeutic agents against this group of neglected diseases, so further investigations are required to delineate the mechanisms of action mainly of the aqueous extract of stem bark of Ziziphus joazeiro.

Anionic liposomes for small interfering ribonucleic acid （siRNA） delivery to primary neuronal cells： Evaluation of alpha-synuclein knockdown efficacy

Alpha-synuclein （a-syn） deposition in Lewy bodies （LB） is one of the main neuropathological hallmarks of Parkinson＇s disease （PD）. LB accumulation is considered a causative factor of PD, which suggests that strategies aimed at reducing a-syn levels could be relevant for its treatment. In the present study, we developed novel nanocarriers suitable for systemic delivery of small interfering ribonucleic acid （siRNA） that were specifically designed to reduce neuronal α-syn by RNA interference. Anionic liposomes loaded with an siRNA-protamine complex for α-syn gene silencing and decorated with a rabies virus glycoprotein （RVG）-derived peptide as a targeting agent were prepared. The nanoparticles were characterized for their ability to load, protect, and deliver the functional siRNA to mouse primary hippocampal and cortical neurons as well as their efficiency to induce gene silencing in these cells. moreover, the nanocarriers were evaluated for their stability in serum. The RVG-decorated liposomes displayed suitable characteristics for future in vivo applications and successfully induced α-syn gene silencing in primary neurons without altering cell viability. Collectively, our results indicate that RVG-decorated liposomes may be an ideal tool for further studies aimed at achieving efficient in vivo α-syn gene silencing in mouse models of PD.

Dear Editor,Coronavirus disease 2019(COVID-19)is a viral infection caused by severe acute respiratory syndrome coronavirus 2(SARS-Co V-2).From march 2020,several studies indicate that many subjects affected by mild-to-moderate COVID-19 presented olfactory/gustatory dysfunction(OD/GD)that appeared strongly correlated between them but not with the other symptoms suggestive of upper airway infection(Lechien et al.2020a,b;Hopkins et al.2020;Paderno et al.2020).

The magnetic and structural properties of FINEmET alloy with a composition of ***1Nb1Si13.5B9 were investigated after primary and secondary crystallization of amorphous ribbon sample. The crystallization behavior and the nanocrystal formation of the samples were performed by differential thermal analysis （DTA） which in turn was supported by X-ray diffraction （XRD） study. Temperature dependence of initial permeability of amorphous and devitrified toroid shaped samples has been measured. Enhancement of Curie temperature of the amorphous alloy has been observed due to the irreversible structural relaxation. With the appearance of nanocrystalline phase the Curie temperature of the residual amorphous phase gradually decrease with the increase of annealing temperature. Their temperature dependence reflects the characteristic annealing temperature evolution of the basic magnetic parameters in these nanocrystalline systems. Saturation magnetization, ms, increases with annealing temperature Ta for the samples and finally decreases during annealing at a temperature much higher than peak crystallization temperature.

Kinetic scale magnetic holes in the terrestrial magnetosheath:A review

magnetic holes at the ion-to-electron kinetic scale(KSmHs)are one of the extremely small intermittent structures generated in turbulent magnetized *** recent years,the explorations of KSmHs have made substantial strides,driven by the ultra-high-precision observational data gathered from the magnetospheric multiscale(mmS)*** review paper summarizes the up-to-date characteristics of the KSmHs observed in Earth’s turbulent magnetosheath,as well as their potential impacts on space *** review starts by introducing the fundamental properties of the KSmHs,including observational features,particle behaviors,scales,geometries,and distributions in terrestrial *** have discovered that KSmHs display a quasi-circular electron vortex-like structure attributed to electron diamagnetic *** electrons exhibit noticeable non-gyrotropy and undergo *** occurrence rate of KSmH in the Earth’s magnetosheath is significantly greater than in the solar wind and magnetotail,suggesting the turbulent magnetosheath is a primary source ***,KSmHs have also been generated in turbulence simulations and successfully reproduced by the kinetic equilibrium ***,KSmHs have demonstrated their ability to accelerate electrons by a novel non-adiabatic electron acceleration mechanism,serve as an additional avenue for energy dissipation during magnetic reconnection,and generate diverse wave phenomena,including whistler waves,electrostatic solitary waves,and electron cyclotron waves in space *** results highlight the magnetic hole’s impact such as wave-particle interaction,energy cascade/dissipation,and particle acceleration/heating in space *** end this paper by summarizing these discoveries,discussing the generation mechanism,similar structures,and observations in the Earth’s magnetotail and solar wind,and presenting a future extension perspective in this active field.
