Diversity of Filamentous Fungi of Area from Brazilian Caatinga and High-Level Tannase Production Using Mango (Mangifera indicaL.) and Surinam Cherry (Eugenia unifloraL.) Leaves under SSF

Tannase is a biotechnologically important enzyme that can be produced during fungal fermentation of organic matter. The Caatinga is an exclusive Brazilian ecosystem that has been largely unexplored by science, particularly its filamentous fungal diversity. This study evaluated the diversity of filamentous fungi in the Caatinga soils of Pernambuco, Brazil, and their potential for tannase production by solid-state fermentation (SSF) of mango (Mangifera indica L.) and Surinam cherry (Eugenia uniflora L.) leaves. A total of 4711 isolates were obtained, 2090 during the rainy seasonand 2621 during the dry season. The isolates belonged to 18 genera and 66 species, with Aspergillus and Penicillium having the highest species richness. The dry season had a higher diversity index. Aspergillus was the dominant genus, and A. flavus, A. sclerotiorum, and A. ochraceus the most abundant species. A representative of each species was tested for tannase production using dried mango and Surinam cherry leaves as substrates;the leaves contained 14.28 and 7.0 g/L tannin, respectively. Most fungal species produced tannase, but the highest yields were obtained when mango leaves were used as substrate for Penicillium restrictum (accession URM 6044), Aspergillus flavofurcatus (URM 6142), and A. stromatoides (URM 6609), which produced 104.16, 87.51, and 81.83 U/mL tannase, respectively. These yields exceeded previously published reports. Filamentous fungi from Caatinga soils have great potential for producing tannase by SSF, and low-cost mango leaves make excellent substrate.

Production of Manganese Peroxidase by Trametes villosa on Unexpensive Substrate and Its Application in the Removal of Lignin from Agricultural Wastes

Manganese peroxidase (MnP) is a ligninolytic enzyme that is involved in the removal of lignin from the cell wall of plants. This removal facilitates the access of hydrolytic enzymes to the carbohydrate polymers that are hydrolyzed to simple sugars, which allows the subsequent fermentation to obtain bioproducts, such as ethanol. In this work, response surface methodology (RSM) was employed to optimize the culture conditions on unexpensive substrate for MnP secretion by Trametes villosa. Three independent variables were evaluated (i.e., temperature, moisture content and pH). The crude extract containing MnP was used in the delignification experiment and it caused a reduction in lignin content for all residues tested: 35.05 ± 1.45 (%) for the sugar cane bagasse;63.11 ± 0.06 (%) for the sisal fiber and 39.61 ± 0.39 (%) for the coconut shell, under the reaction conditions tested after 4 hours of fermentation. The preliminary results exhibited the potential application of this enzyme in the removal of lignin from plant residues. However, the conditions should be evaluated and optimized for each residue type.

Analysis of Autonomic, Respiratory and Motor Function of Infants in Pre- and Post-Liver Transplantation

Purpose: Children with liver impairment are likely to develop changes in autonomic nervous function and delay in motor development. The assessment and identification of these dysfunctions may allow an appropriate physiotherapeutic care. Method: Cross sectional study of 18 infants, 11 controls and 7 infants (post-liver transplantation) with an average age of 10 ± 4.5 months, was evaluated in pre- and post-liver transplant. All infants underwent to assessments of motor skills, body composition, chest and abdominal motion, and cardiac autonomic modulation was measured by heart rate variability. Results: Motor delay and malnutrition were found in all infants. The gravity index (PELD)—pediatric end-stage liver disease—showed a negative correlation with the Alberta Infant Motor Scale (r = 0.83, p = 0.01). In addition, reduced parasympathetic modulation was demonstrated by the rMSSD, SD1 and ApEn, pre- and post-transplant. Conclusion: Infants with liver disease, even after transplantation, have delay in motor development, as well as changes in their nutritional and autonomic dysfunction.

Culture Circle as a Teaching Approach in the Education of Teenager Health Multipliers on Leprosy Awareness

Objective: The study aims to evaluate the use of a teaching method proposed by Paulo Freire, Culture Circles, in the education of teenagers multipliers on leprosy awareness. Methods: It is an action-research study with a qualitative approach developed in a public school in Pernambuco, Brazil. Five Culture Circles were conducted involving the participation of 26 teenagers. The followings were used as data collection tools: observation, field notes, photography and filming. Results: The educational intervention on health addressed the following topics: 1) definition and transmission of leprosy;2) Characteristics and diagnosis of leprosy;3) Treatment of leprosy;4) Aesthetics, prejudice and mental health related to leprosy;and 5) Planning of educational activities for teenagers as health multipliers on leprosy awareness. The educational action on health provided this age group to perceive themselves, act as political subjects in the development of Culture Circles, and act as protagonists in the dissemination of knowledge on leprosy. Conclusions: This study highlights that the application of active methodologies, such as Culture Circles, is able to encourage the engagement of young people in community empowerment and bring together health professionals and the school community in an intersectoral work in order to develop action strategies involving the promotion of health in the context of neglected diseases such as leprosy.

Background: Most anxiety complaints are treated with pharmacological measures involving barbiturates and benzodiazepines, in which they may end up causing tolerance and pharmacological dependence. Integrative approaches such as aromatherapy are used in addition to medications to improve sleep quality and reduce anxiety. Thus, PinetoninTM, a phytocomplex obtained from a blend of essential oils aims to aid in the symptoms of stress and anxiety. Methods: The cytotoxicity of PinetoninTM was evaluated MTT assay using fibroblasts and astrocytes showed reduction in the cell viability only at high concentrations. Evaluation of intracellular calcium and determination of residual glutamate in the supernatant of astrocyte cultures showed agonist action of dihydroxyphenylglycine (DHPG) increasing linearly the concentration of intracellular calcium and the glutamate levels in the supernatants of the cultures. On the other hand, cultures of astrocytes treated with PinetoninTM showed residual glutamate levels in the supernatants reducing proportionally, as well as, intracellular calcium reduction. The determination of salivary cortisol showed a significant reduction in salivary cortisol levels in the group that received PinetoninTM. The evaluation of the electroencephalogram in patients treated with PinetoninTM had a significant increase (P Results: A reduction in dose-dependent cell viability was observed when compared to cultures of PinetoninTM treated fibroblasts with control culture. When PinetoninTM and linole are administered in astrocytic cells, there was a reduction of the intracellular concentration of Ca2+ against a control group treated with DHPG agonist. The evaluation of salivary cortisol demonstrated a reduction when the patient group was treated with PinetoninTM by purchasing the results against the group of patients treated with saline. Reinforcing the relaxed state of that group, alpha waves have been increased and reductions in beta waves. Conclusion: The results

The present work sought to characterize the cyanogenic glycoside dhurrin in five sugarcane varieties and to determine its possible relationship with the formation of ethyl carbamate in cachaca. For each variety, methanol, ethyl acetate and hexane extracts were prepared and submitted to thin layer chromatography. Chromatographic spots were revealed with iodine vapors. The physical and chemical parameters of the spirits produced from the five different varieties of sugar cane and by different production processes were within the legal limits. Although not exceeding the limit for ethyl carbamate, a significant variation in the final concentration of this compound was observed for each sample analyzed. The Rf values for the dhurrin standard and for the sugarcane variety SP 83-2847 were equal, similar to those for SP 80-3280 and CTC 11 and different from the Rf values for the RB 86-7515 and IAC 86-2480 samples.

Effects of Pereskia aculeataMiller on the Biochemical Profiles and Body Composition of Wistar Rats

Pereskia aculeata Miller commonly known as “ora-pro-nobis” has been used extensively in folclore medicine. Plant belongs to the family Cactacea and it is found in the Brazil (reaching Bahia and Rio Grande do Sul states). The purpose was to evaluate the effects of plant on biochemical and corporal profiles of Wistar rats. The 80 animals were divided randomly into 4 groups by sex (n = 10 females and 10 males per group) as follows: G1 group (water/rat food ad libitum);G2 group (drink A: commercial condensed milk, sugar and water/rat food ad libitum);G3 group (drink B: P. aculeata juice/rat food ad libitum);G4 group (drink C: commercial condensed milk, sugar and P. aculeate juice/rat food ad libitum). After 35 days their biochemical and corporal profiles were analyzed (cholesterol, glycemia, triglycerides, HDL-c, AST, ALT, Lee Index, weight and visceral fat). Intake of the plant caused no changes in the lipid profile of Wistar rats. However, intake in male rats of the same prevented the increase in triglycerides front of a hypercaloric diet. There were significant changes in glycemic index due to the high carbohydrate content of the plant. The plant was effective in reducing the levels of AST and ALT in male rats and ALT only in females. The use of the plant did not cause significant changes in the index of Lee but the Pereskia aculeata Miller may reduce visceral fat gain in female with a regular diet.

Saturation Levels by Bases and Time of Incubation in the development and Growth of Ornamental Sunflower

Floriculture is a promising segment of Brazilian agriculture. The ornamental sunflower (Helianthus annuus L. cv. Folded Sungold), also known as dwarf sunflower, in addition to being used in gardens can be marketed as cut flower or vase. The sunflower represents a great alternative for small producers because it requires little space, has easy handling and short cycle, besides providing greater economic return. In this context, we aimed to evaluate the development of this culture submitted to different levels of saturation per base, combined with the incubation time of the limestone. The experiment was carried out in a greenhouse at the Federal University of Mato Grosso-University Campus of Rondonópolis. A randomized block design with factorial scheme 6 × 2 was used, with six levels of saturation per base: 0, 20%, 40%, 60%, 80%, 100%;and two incubation times: 0 and 30 days, resulting in 12 treatments with four replicates, totaling 48 experimental units. Each plot consisted of a plastic polyethylene vessel with a capacity of 1.2 dm−3. The following variables were analyzed: Height of the floral stem;number of leaves, diameter of chapters;root volume;fresh mass of leaves;fresh mass of floral stems;fresh mass of chapters;fresh root mass and chlorophyll index (SPAD). Data were submitted to analysis of variance by the test F at 5% probability of error. The qualitative factors were submitted to the Tukey test and quantitative factors to the regression test, both with a 5% probability of error. Basal saturation levels of 80% and 100%, in the treatment with incubated soil, provided higher quality of inflorescence for commercialization purposes.

Increased Risk of Acute Myeloid Leukemia in Patients with CYP1A1 Polymorphisms

Acute Myeloid Leukemia (AML) is a group of genetically diverse hematopoietic malignancies arising from cell progenitors developing in the myeloid pathway or from primitive stem cells. Genetic susceptibility of AML may account for an increased risk of AML due to partial metabolism of or biocativation of carcinogens. Chemical compounds are metabolized by a two-tiered phase detoxifying system. Polymorphisms in these pathways may lead to DNA damage and development of AML. We determined the frequencies of carcinogen metabolism gene polymorphisms (CYP1A1, del{GSTM1} and del{GSTT1}) in a case control-study based on polymorphism analysis. Fifty-eight consecutively AML patients (median age 62 years) and 174 sex and age-matched control group were assessed by a PCR-RFLP assay. There were 51 de novo and 7 secondary AML. CYP1A1*2A and CYP1A1*2C polymorphisms were more frequent in CG than AML p 0.001 and in contrast, CYP1A1*3 and CYP1A1*4 were more frequent in AML than CG p 0.001. There were no differences in del{GSTM1} neither del{GSTT1} between AML and CG (p = 0.999 and p = 0.539). Odds ratio for AML in patients harboring CYP1A1*3 was 2.36 (95% CI 1.2 - 4.5), 2.38 for CYP1A1*4 (95% CI 0.8 - 6.8). Adjusted OR was 2.63 for CYP1A1*3 (95% CI 1.4 - 5.1) and 2.66 for CYP1A1*4 (95% CI 0.9 - 7.8). In the multivariate analysis CYP1A1*3 polymorphism was a risk factor for AML with an OR for 3.99 (95%CI 1.9 - 8.6). To the best of our knowledge this is the first study to show that CYP1A1*3 heterozygous genotypes increase the risk of AML. Our data support that inherited absence of this carcinogen detoxification pathway may be an important determinant of AML.
