Malate dehydrogenase （MDH） is an enzyme widely distributed among living organisms and is a key protein in the central oxidative pathway. It catalyzes the interconversion between malate and oxaloacetate using NAD＋ or NADP＊ as a cofactor. Surprisingly, this enzyme has been extensively studied in eukaryotes but there are few reports about this enzyme in prokaryotes. It is necessary to review the relevant information to gain a better understanding of the function of this enzyme. Our review of the data generated from studies in bacteria shows much diversity in their molecular properties, including weight, oligomeric states, cofactor and substrate binding affinities, as well as differences in the direction of the enzymatic reaction. Furthermore, due to the importance of its function, the transcription and activity of this enzyme are rigorously regulated. Crystal structures of MDH from different bacterial sources led to the identification of the regions involved in substrate and cofactor binding and the residues important for the dimer-dimer interface. This structural information allows one to make direct modifications to improve the enzyme catalysis by increasing its activity, cofactor binding capacity, substrate specificity, and thermostability. A comparative analysis of the phylogenetic reconstruction of MDH reveals interesting facts about its evolutionary history, dividing this superfamily of proteins into two principle clades and establishing relationships between MDHs from different cellular compartments from archaea, bacteria, and eukaryotes.

Predictors of bowel damage in the long-term progression of Crohn’s disease

BACKGROUND Crohn’s disease(CD)is a chronic inflammatory bowel disorder that progresses to bowel damage(BD)over *** image-based index,the Lémann index(LI),has been developed to measure cumulative *** To characterize the long-term progression of BD in CD based on changes in the LI and to determine risk factors for long-term *** This was a single-center longitudinal cohort *** who had participated in prospective studies on the accuracy of magnetic resonance imaging using endoscopy as a gold standard and who had a follow-up of at least 5 years were reevaluated after 5-12 *** Seventy-two patients were *** increased in 38 patients(52.8%),remained unchanged in 9 patients(12.5%),and decreased in 25 patients(34.7%).The small bowel score and surgery subscale significantly increased(P=0.002 and P=0.001,respectively),whereas the fistulizing subscale significantly decreased(P=0.001).Baseline parameters associated with BD progression were ileal location(P=0.026),CD phenotype[stricturing,fistulizing,or both(P=0.007,P=0.006,and P=0.035,respectively)],disease duration>10 years(P=0.019),and baseline LI stricturing score(P=0.049).No correlation was observed between BD progression and baseline clinical activity,biological markers,or severity of endoscopic *** BD,as assessed by the LI,progressed in half of the patients with CD over a period of 5-12 *** main determinants of BD progression were ileal location,stricturing/fistulizing phenotype,and disease duration.

The article deals the structure of the CaO -Al2O3 - TiO2 -SiO2 system as a basis for obtaining advanced ce- ramics with a complex of high-operational characteris- tics. The most techno- logical system area ,for the production of heat resistant advanced ceramics has been determined. The batches for producing heat and chemical resistant dense advanced ceramics made of natural, technical and technogenic raw materials have been developed.

Tumor biopsies may help to reliably distinguish hepatocellular carcinoma(HCC) from other tumors, mostly cholangiocarcinoma as well as to identify the patient populations who most benefit from target-driven HCC treatments, in order to improve the success rate of experimental therapies. Clarifying tumor biology may also lead to identify biomarkers with prognostic role and/or enabling to predict response or resistance to therapies. Recently, clinical trials have more efficiently included biomarker endpoints and increasingly collected tumor tissue from enrolled patients. Due to their frail status and sometimes fast-progressing disease, the performance status of patients with HCC progressing on first-line therapy can deteriorate quickly, preventing their enrollment in clinical trials. However, the challenge of identifying the proper patient at the proper time can be overcome by periodic inter-department meetings involving the key specialists taking care of HCC patients, and solid networks between research centers and referring institutions. An early planned biopsy would also facilitate timely inclusion of patients in biology-driven clinical trials. Ultimately, institution of multidisciplinary teams can optimize treatment choice, biopsy timing, and quick enrollment of patients in clinical trials, before their performance status deteriorates.

Sleep Disorders in Menopause: Review of the Literature and Occurrence through Menopausal Stages

We here review principal literature data about sleep disorders in menopause and we compared it with data obtained from a systematical screening of a sample of 161 menopausal women through questionnaires and clinical evaluation. Our data reveal high occurrence of sleep disorders in this group of women if compared to literature data about general population, in the absence of statistically significant differences among the three menopausal stages: we find only a trend toward a higher risk of OSAS (Obstructive Sleep Apnea Syndrome) in PM versus early PM and MT. However, none of women in our survey underwent PSG (polysomnography), essential to confirm the diagnosis of OSAS and none was diagnosed with RLS (Restless Legs Syndrome) (prior to our survey) by the Gynecologist or General Pratictioner: these data reveal the issue of too low attention towards sleep disorders in this contest. Generic “sleep complaints” are clearly associated with depressed mood and worse-quality life along the menopausal process. In our sample, a non-specific definition of insomnia shows a peculiar relationship with a mood disorder: it is only variable not associated with higher BDI and KI scores, in fact. This might suggest that the conditions linked to depression of these women could be more specific sleep disturbances, such as RLS and OSAS.

Influence of Filtration Processes on Aqueous Nanostructures by NIR Spectroscopy

Filtration processes are worldwide used for sterilizing solutions and substrates. Filtration seems to induce the formation of aqueous nanostructures. The aim of this work was to verify the influence of filtration processes on water structure detected by spectral variations in NIR region. Samples of ultrapure water （MilliQ-Millipore, Vimodrone, Milan, Italy） before and after iterated filtrations were analyzed. NIR spectra were collected in transmission mode in the whole NIR range, by using NIRFIex N500 spectrometer at constant temperature （40 ± 1 ℃）. NIR data were processed using Unscrambler software v. 9.2 in evaluating qualitative differences between filtered and not filtered samples. The information related to possible solvent physical stresses were highlighted in the range 6500-7500 cm^-1. The shifts observed were ascribable to a different distribution of the number of water molecules involved in hydrogen bonds in filtered and not filtered water samples, at constant temperature. NIR spectroscopy, commonly used to study relationship between spectral changes and hydrogen bonds in water at increasing temperature values, was applied to evaluate effects of filtration processes on water structure. The obtained results are in agreement with literature data and allowed the improvement of the knowledge about pure water characteristics when some mechanical perturbations are applied.
