Valproic Acid,a Drug with Multiple Molecular Targets Related to Its Potential Neuroprotective Action

Valproic acid (VA) is used worldwide as an antiepileptic drug and a mood stabilizer. Recently, VA was shown to act on cell growth, differentiation and apoptosis, by regulating gene expression at the molecular level, through epigenetic mechanisms. Thus, VA was demonstrated to act on the chromatin remodeling what is a consequence of the drug inhibition of histone deacetylases (HdaCs) activity. Other studies uncovered the potential of VA to interfere with multiple regulatory mechanisms besides HdaCs, as the GSK3 alpha and beta, Akt, ERK and phosphoinositol pathways, tricarboxylic acid cycle, GABA and OXPHOS system. The review focuses on the mechanisms of action of VA, showing that HdaC inhibitors, as VA, can be successfully used in the treatment of neurodegenerative disorders. This molecule, whose biological activities range from interactions with receptors and ion channels to the regulation of many catalytic reactions, has a central role in cellular cascades that regulate gene expression. Thus, inhibitors of HdaCs, by positively affecting both neuronal degeneration and cognitive deficits, appear as promising drugs against various pathological conditions and neurodegenerative diseases. VA is known to present anti-inflammatory and antioxidative properties. And, since inflammation and oxidative stress are common links in neurodegeneration, VA is a drug that, from a clinical point of view, shows a great potential as a candidate for the treatment of neurodegenerative diseases related to excitotoxic events.

Progress in the characterization of genes involved in the control of iron homeostasis in humans and in mice has improved the definition of iron overload and of the cells affected by it. The cell involved in iron overload with the greatest effect on immunity is the macrophage. Intriguing evidence has emerged, however, in the last 12 years indicating that parenchymal iron overload is linked to genes classically associated with the immune system. This review offers an update of the genes and proteins relevant to iron metabolism expressed in cells of the innate immune system, and addresses the question of how this system is affected in clinical situations of iron overload. The relationship between iron and the major cells of adaptive immunity, the T lymphocytes, will also be reviewed. Most studies addressing this last question in humans were performed in the clinical model of Hereditary Hemochromatosis. data will also be reviewed demonstrating howthe disruption of molecules essentially involved in adaptive immune responses result in the spontaneous development of iron overload and how they act as modifiers of iron overload.

What have we learned about the kallikrein-kinin and renin-angiotensin systems in neurological disorders?

The kallikrein-kinin system(KKS) is an intricate endogenous pathway involved in several physiological and pathological cascades in the brain. Due to the pathological effects of kinins in blood vessels and tissues, their formation and degradation are tightly controlled. Their components have been related to several central nervous system diseases such as stroke, Alzheimer's disease, Parkinson's disease, multiple sclerosis, epilepsy and others. Bradykinin and its receptors(B1R and B2R) may have a role in the pathophysiology of certain central nervous system diseases. It has been suggested that kinin B1R is up-regulated in pathological conditions and has a neurodegenerative pattern, while kinin B2R is constitutive and can act as a neuroprotective factor in many neurological conditions. The renin angiotensin system(RAS) is an important blood pressure regulator and controls both sodium and water intake. AngⅡ is a potent vasoconstrictor molecule and angiotensin converting enzyme is the major enzyme responsible for its release. AngⅡ acts mainly on the AT1 receptor, with involvement in several systemic and neurological disorders. Brain RAS has been associated with physiological pathways, but is also associated with brain disorders. This review describes topics relating to the involvement of both systems in several forms of brain dysfunction and indicates components of the KKS and RAS that have been used as targets in several pharmacological approaches.

Effect of lead chloride on spermatogenesis and sperm parameters in mice

Aim: To evaluate the effect of acute lead chloride exposure on testis and sperm parameters in mice. Methods: PbCl2, 74 mg/kg, was daily administered to sexually mature male mice for 3 days and the effects on the testicular histology and ultrastructure as well as the motility and density of spermatozoa in cauda epididymis were observed. An additional group of mice were treated for 1-3 days and were allowed to recover for 32 days to determine the reversibility of lead-induced changes. Results: The testicular weight, seminiferous tubular diameter and sperm counts were significantly decreased following 3 days of PbCl2 treatment, but were unaffected by shorter-term exposures. The changes caused by lead are mostly reversible. Conclusion: Acute lead chloride exposure injures the fertility parameters of male mice and the effects are partially reversible.

AIM: To evaluate changes in neurotransmission induced by a psychoactive beverage ayahuasca in the hippocampus and amygdala of naive rats. METHODS: The level of monoamines, their main metabolites and amino acid neurotransmitters concentrations were quantified using high performance liquid chromatography(HPLC). Four groups of rats were employed: saline-treated and rats receiving 250, 500 and 800 mg/kg of ayahuasca infusion(gavage). Animals were killed 40 min after drug ingestion and the structures stored at-80 ℃ until HPLC assay. The data from all groups were compared using Analysis of variance and Scheffé as post test and P dala of rats that received 500 of ayahuasca. Animals that ingested 800 mg/kg of ayahuasca also showed a reduction of GLY level(0.11 ± 0.01 vs 0.29 ± 0.07, P dala after ayahuasca administration in doses: 250 mg/kg(noradrenaline: 0.16 ± 0.02 vs 0.36 ± 0.06, P data suggest increased release of inhibitory amin

Bee Pollen in Transylvania （Romania）： Palynological Characterization and ORACFL Values of Lipophilic and Hydrophilic Extracts of Monofloral Pollen Pellets

A melissopalynological study carried out on 35 bee pollen samples from Romania （Transylvania） shows predominant species as Taraxacum officinale, Tilia sp., Ferbascum sp., Zea mays L., Onobrychis viciifolia Scop., Geranium sanguineum L., Filipendula ulmaria L., Cydonia oblonga L., Calluna vulgaris L. and Brassica sp.. This is the first time for the contribution of total phenolic （TP） and total carotenoid （TC） content to the modified oxygen radical absorbance capacity （ORACFL） values in 29 monofloral samples. Hydrophilic ORACFL （H-ORACFL） values ranged between 7.13-10.12 μmol TE/g and 6.94-22.46 μmol TE/g, whereas lipophilic ORACFL （L-ORACFL）values were between 2.19-7.79 μmol TE/g and 1.65-9.96 μmol TE/g, in multifloral and monofloral samples, respectively. It has proved the complex involvement of botanical origins on the antioxidant features with a specific occurrence. Some monofloral samples presented particular high antioxidant potential, such as Salix sp., Taraxacum officinale, Matricaria chamomilla, Cichorium intybus and the poorly studied Cirsium arvense （L.） Scop. and Scilla bifolia L.. New variables, such as soil characteristics, climates together with botanical origins, were introduced in a multivariate analysis of antioxidant data matrix, given a possible important involvement of all of them in affecting, not only the phytochemical composition, but thereafter the antioxidant capacity. All these data could be crucial to a new way of gathering, by beekeepers, depending on the market demand and the purposes for the product that has a potential for further therapeutic bioactivities investigations, and added value in the enrichment of certain products.

Broad Antibacterial Activity of Bothrops jararaca Venom against Bacterial Clinical Isolates

Purpose: To evaluate the antibacterial activity of Bothrops jararaca venom against bacterial clinical isolates. Methods: Antibacterial activity of Bothrops jararaca venom was evaluated through agar diffusion method against the following bacteria: Acinetobacter baumannii, Oxacillinase-producing Acinetobacter baummanii, extended-spectrum β-lactamase-producing (ESBL) Enterobacter aerogenes, Escherichia coli, Escherichia coli ESBL, Klebsiella pneumoniae, Klebsiella pneumoniae ESBL, Proteus mirabilis, Pseudomonas aeruginosa, metallo β-lactamase-producing Pseudomonas aeruginosa, Staphylococcus aureus, oxacillin resistant Staphylococus aureus (ORSA), Staphylococcus epidermidis, and oxacillin resistant Staphylococus epidermidis.?Minimum inhibitory concentration was determined through microdilution plate protocol. Results: The venom presented antibacterial activity against all tested bacteria. More pronounced results were observed to Gram- positive bacteria, especially against ORSA. Conclusion: The present study evidenced the great antibacterial potential of Bothrops jararaca venom showing promising results even with resistant bacterial clinical isolates.

Creationism and Evolution Views of Brazilian Teachers and Teachers-to-Be

This paper aims at assessing the evolutionist and creationist conceptions of Brazilian teachers. The work is developed within the framework of the European project BIOHEAD-CITIZEN （biology, health and environmental education for better citizenship）, which takes into account that scientific knowledge and teachers＇ attitudes and values can influence the teaching practices. The large questionnaire BIOHEAD-CITIZEN, which was constructed to be applied in 19 countries in Europe, Africa and Middle East countries, was applied, in this study, to six groups of Brazilian teachers： primary school teachers, biology teachers and Portuguese language teachers and corresponding teachers-to-be. For this paper the answers to questions about evolution were used as dependent variables. Descriptive and multivariate analyses were carried out. Biology teachers and biology teachers-to-be gave more importance to the natural selection and the evolution process than the other groups of teachers. Comparing to the BIOHEAD-CITIZEN countries, the total Brazilian sample showed a higher percentage of creationist conceptions, particularly the Brazilian biology teachers and teachers-to-be. As discussed herein, this may not be an obstacle for teaching evolution as they accept both creationist and evolutionist concomitantly.

Zea mays L. is one of the biggest cropping systems among the sustainable development agronomy. Pollen from this crop source is unexplored and apiculture can be a good partner adding value to the product and creating new jobs helping to solve some social issues as unemployment. However, food safety is crucial, thus the aim of this study was to explore the flavonoid/phenolic profiles from Z. mays L. pollen as a fingerprint for this plant identification and also to demonstrate how the method of bee pollen samples （honeybee collected pollen） is applied. For this purpose, several sources ofZ. mays L. pollen were analyzed, including corn hybrids and genetic modified samples collected at the breeding fields. For this work, samples were taken at several years from 2000 to 2012 and collected from different countries and locations, such as Portugal, Mexico and Brazil. Results showed, for the first time, that the fingerprint （flavonoid/phenolic profile） for Z. mays L. pollen does not change over the time of sampling neither with the region of harvesting. The high performance liquid chromatography-diode array detector （HPLC/daD） fingerprints of phenolic/flavonoid extract from Z. mays remain unchanged for all samples analyzed from different countries, hybrids and/or genetic modified plants. This is also the first study reporting these phenolic compounds not only in pollen collected directly from hybrid plants, but also in Z. mays bee pollen. The described fingerprinting method is easy, fast and accurate for the characterization of Z. mays L. pollen samples and complete microscopic analysis because it is species-specific.
