Structural optimization of patient-specific temporomandibular joint replacement implants for additive manufacturing:novel metrics for safety evaluation and biomechanical performance

Total temporomandibular joint(TMJ)replacement is recommended only when there is irreversible damage to the joint and no conservative treatment can provide functional improvements.Several stock and custom-made TMJ implants have been made available;however,retrospective and comparative studies were unable to find significant differences between the two types of solutions.The introduction of additive manufacturing(AM)techniques in medical practice allows for a greater freedom of design and a higher degree of device customisation.The combination of AM with structural optimisation may streamline development and provide the key for fabricating biomechanic ally enhanced TMJ implants.In this study,structural optimis ation techniques were applied to develop and numerically validate a patient-specific TMJ implant.The biomechanical behaviour of each intermediate TMJ design was assessed under four different nominal and maximum biting tasks using finite element analyses.In addition,a new set of metrics were proposed to compare each design regarding biomechanical performance and implant safety.The results suggest that 55-82%of the natural/intact strain patterns can be recovered with the finally selected TMJ implant.This represents an increase of 15%in biomechanical performance for incisor biting,15%for right molar biting,17%for left molar biting and a decrease of 2%for left group biting compared with the initial design.The results also suggest that load transfer at the proximal ramus reduces the implant’s impact on the mandible’s strain patterns.Finally,structural optimisation allows for a volume reduction of up to 44%with a minimum loss of implant safety and biomechanical performance.

Fecal calprotectin and endoscopic scores: The cornerstones in clinical practice for evaluating mucosal healing in inflammatory bowel disease

Managing inflammatory bowel disease(IBD)is becoming increasingly complex and personalized,considering the advent of new advanced therapies with distinct mechanisms of action.Achieving mucosal healing(MH)is a pivotal therapeutic goal in IBD management and can prevent IBD progression and reduce flares,hospitalization,surgery,intestinal damage,and colorectal cancer.Employing proactive disease and therapy assessment is essential to achieve better control of intestinal inflammation,even if subclinical,to alter the natural course of IBD.Periodic monitoring of fecal calprotectin(Fc)levels and interval endoscopic evaluations are cornerstones for evaluating response/remission to advanced therapies targeting IBD,assessing MH,and detecting subclinical recurrence.Here,we comment on the article by Ishida et al Moreover,this editorial aimed to review the role of Fc and endoscopic scores in predicting MH in patients with IBD.Furthermore,we intend to present some evidence on the role of these markers in future targets,such as histological and transmural healing.Additional prospective multicenter studies with a stricter MH criterion,standardized endoscopic and histopathological analyses,and virtual chromoscopy,potentially including artificial intelligence and other biomarkers,are desired.

complexed amino acid minerals vs. bis-glycinate chelated minerals:Impact on the performance of old laying hens

The present study was to evaluate the effect of trace minerals(Zn, Mn, and cu) from complexed amino acid minerals(ZMcAA) and bis-glycinate chelated minerals(ZMcGly) in laying hen diets on performance,internal and external egg quality, yolk mineral deposition, intestinal morphometry, and bone characteristics. From 78 to 98 weeks of age, 400 White LSL-Lite strain laying hens were distributed in a randomized design with 4 treatments with 10 replicates per treatment. Treatments were distributed in a 2 ×2 factorial arrangement using either Zn, Mn, and cu of ZMcAA or ZMcGly source at 2 levels: low(20, 20,and 3.5 mg/kg of Zn, Mn, and cu, respectively) or high(40, 40, and 7 mg/kg of Zn, Mn, and cu,respectively). The analysis of variance was performed, and in cases where differences were observed, the means were compared using Tukey?s test(P ce and level of trace mineral supplementation had a significant impact on the performance of laying hens. Hens fed ZMcAA had higher egg production(P = 0.01), egg weight(P = 0.02), egg mass(P = 0.01), and lower feed conversion ratio(P =0.05) compared to those fed ZMcGly. The ZMcAA supplementation showed higher albumen height(P =0.01), albumen weight(P = 0.01), and eggshell thickness(P cu(P ceived ZMcAA. Tibia weight(P = 0.04)and bone densitometry(P cAA supplementation. In the small intestine, ZMcAA resulted in longer villi(P = 0.02) and shorter crypt depth(P = 0.01) in the duodenum.Jejunum and ileum measurements were influenced by the level and source of trace minerals(P cAA exhibited superior performance, egg quality, deposition of trace minerals in the egg yolk, and bone density compared to hens fed ZMcGly. In this study, older laying hens supplemented with ZMcAA at lower levels demonstrated adequate levels of supplementation.

Long-read sequencing with nanopore technology has been widely utilized for the identification of the full-length sequence of RNA molecules."It is especially suitable for the identification of circular RNAs(circRNAs),which are regulatory molecules that are regarded as promising non-invasive biomarkers for various human malignancies,including breast cancer.2 The impact of circRNAs derived from the protein arginine methyltransferase 1(PRMT1)gene has not been clarified yet,even though the effect of PRMT1-mediated methylation on breast cancer cell characteristics has been well established.

Arroyo Verde epithermal deposit,in the North Patagonia region of Argentina,is located within the Lower chon Aike volcanism and is developed between 192.6±2.5 Ma(mineralization host rock)and 189.5±2.6 Ma(overlying lava).Marifil volcanic complex is the host rock for these veins,veinlets and breccias.This particular small deposit has developed low to intermediate sulphidation characteristics,with gangue mineral textures associated with several ore minerals.Electron probe microanalysis determines electrum,silver minerals such as acanthite,and tetrahedrite,related Ag-cu minerals like jalpaite,mckinstryite,stromeyerite,and base metals like chalcopyrite,sphalerite,galena and pyrite.Fluid inclusions in quartz of two out of five events indicate that these Low Jurassic veins were formed due to subtle boiling and dilution mechanisms and by low-salinity(3.4 wt.%to 6.7 wt.%Nacl Eq)magmatic-hydrothermal fluid at 225 to 310℃.The fluid inclusion measurements that indicate boiling(Event 3)were plotted,and based on this data,we interpret that the Arroyo Verde deposit has been eroded between 550 and 700 m.This particular character has not been previously recorded for other sectors of the eastern north Patagonian region.

Shear-wave elastography to predict hepatocellular carcinoma after hepatitis c virus eradication:A systematic review and meta-analysis

BAcKGROUND Direct-acting antiviral agents(DAAs)are highly effective treatment for chronic hepatitis c(cHc)with a significant rate of sustained virologic response(SVR).The achievement of SVR is crucial to prevent additional liver damage and slow down fibrosis progression.The assessment of fibrosis degree can be performed with transient elastography,magnetic resonance elastography or shear-wave elastography(SWE).Liver elastography could function as a predictor for hepato-cellular carcinoma(Hcc)in cHc patients treated with DAAs.AIM To explore the predictive value of SWE for Hcc development after complete clearance of hepatitis c virus(HcV).METHODS A comprehensive literature search of clinical studies was performed to identify the ability of SWE to predict Hcc occurrence after HcV clearance.In accordance with the study protocol,a qualitative and quantitative analysis of the evidence was planned.RESULTS At baseline and after 12 wk of follow-up,a trend was shown towards greater liver stiffness(LS)in those who go on to develop Hcc compared to those who do not[baseline LS standardized mean difference(SMD):1.15,95%confidence interval(95%cI):020-2.50;LS SMD after 12 wk:0.83,95%cI:0.33-1.98].The absence of a statistically significant difference between the mean LS in those who developed Hcc or not may be related to the inability to correct for confounding factors and the absence of raw source data.There was a statist-ically significant LS SMD at 24 wk of follow-up between patients who developed Hcc vs not(0.64;95%cI:0.04-1.24).cONcLUSION SWE could be a promising tool for prediction of Hcc occurrence in patients treated with DAAs.Further studies with larger cohorts and standardized timing of elastographic evaluation are needed to confirm these data.

carbon sequestration in a bamboo plantation:a case study in a Mediterranean area

In the Mediterranean region,despite bamboo being an alien species that can seriously alter plant and ani-mal biocoenosis,the area occupied by bamboo plantations continues to increase,especially for the purpose to seques-ter carbon(c).However,the c dynamics in the soil-plant system when bamboo is grown outside its native area are poorly understood.Here we investigated the c mitigation potential of the fast-growing Moso bamboo(Phyllostachys edulis)introduced in Italy for climate-change mitigation.We analyzed aboveground(AGB)and belowground(as root/shoot ratio)biomass,litter and soil organic c(SOc)at O-15-and 15-30-cm depths in a 4-year-old bamboo plantation in comparison with the former annual cropland on which the bamboo was established.To have an idea of the maximum c stored at an ecosystem level,a natural forest adjacent the two sites was also considered.In the plantation,c accumulation as AGB was stimulated,with 14.8±3.1 Mg c ha^(-1) stored in 3 years;because thinning was done to remove culms from the first year,the mean sequestration rate was 4.9 Mg c ha^(-1) a^(-1).The sequestration rates were high but comparable to other fast-growing tree species in Italy(e.g.,Pinus nigra).SOc was significantly higher in the bamboo plantation than in the cropland only at the 0-15 cm depth,but SOc stock did not differ.Possibly 4 years were not enough time for a clear increase in SOc,or the high nutrient uptake by bamboos might have depleted the soil nutrients,thus inhibiting the soil organic matter formation by bacteria.In comparison,the natural forest had significantly higher c levels in all the pools.For c dynamics at an ecosystem level,the bamboo plantation on the former annual cropland led to substantial c removal from the atmosphere(about 12 Mg c ha^(-1) a^(-1)).However,despite the promising c sequestration rates by bamboo,its introduction should be carefully considered due to potential ecological problems caused by this species in overexploited environments such as the Mediterranean area.

A short climatology of black and brown carbon and their sources at a suburban site impacted by smoke in Brazil

Emissions from biomass burning challenge efforts to curb air pollution in cities downwind of fire-prone regions,as they contribute large amounts of brown carbon(Br c)and black carbon(Bc)particles.We investigated the patterns of Br c and Bc concentrations using Aethalometer data(atλ=370 and 880 nm,respectively)spanning four years at a site impacted by the outflow of smoke.The data required to be post processed for the shadowing effect since,without correction,concentrations would be between 29%and 35%underestimated.The Br c concentrations were consistently higher than the Bc concentrations,indicating the prevalence of aerosols from biomass burning.The results were supported by the Angstrom coefficient(A_(370/880)),with values predominantly larger than 1(mean±standard deviation:1.25±0.31).A_(370/880)values below 1 were more prevalent during the wet season,which suggests a contribution from fossil fuel combustion.We observed sharp Br c and Bc seasonal signals,with mean minimum concentrations of 0.40μg/m^(3)and 0.36μg/m^(3),respectively,in the wet season,and mean maximum concentrations of 2.05μg/m^(3)and 1.53μg/m^(3)in the dry season.The largest concentrations were observed when northerly air masses moved over regions with a high density of fire spots.Local burning of residential solid waste and industrial combustion caused extreme Br c and Bc concentrations under favourable wind directions.Although neither pollutant is included in any ambient air quality standards,our results suggest that transboundary smoke may hamper efforts to meet the World Health Organization guidelines for fine particles.

The brain is,after the adipose tissue,the organ with the greatest amount of lipids and diversity in their composition in the human body.In neurons,lipids are involved in signaling pathways controlling autophagy,a lysosome-dependent catabolic process essential for the maintenance of neuronal homeostasis and the function of the primary cilium,a cellular antenna that acts as a communication hub that transfers extracellular signals into intracellular responses required for neurogenesis and brain development.A crosstalk between primary cilia and autophagy has been established;however,its role in the control of neuronal activity and homeostasis is barely known.In this review,we briefly discuss the current knowledge regarding the role of autophagy and the primary cilium in neurons.Then we review the recent literature about specific lipid subclasses in the regulation of autophagy,in the control of primary cilium structure and its dependent cellular signaling in physiological and pathological conditions,specifically focusing on neurons,an area of research that could have major implications in neurodevelopment,energy homeostasis,and neurodegeneration.
