The purpose of this work was to introduce a new concept of coated pellets containing chitosan microspheres loaded with didadosine for oral administration,aiming at reducing the frequency of administration and improving the bioavailability by a suitable release *** microspheres were produced under fiuidized bed,followed by extrusion and spheronization to obtain pellets with a mean diameter of about 1 *** pellets were then coated with Kollidon VA64 and Kollicoat MAE100P in water dispersion to depict a sustained release *** hard gelatine capsules were loaded with these pellets and tested in vitro for their release profile of *** testing confirmed that chitosan microsphere pellets provides appropriate sustained release up to 2 h behavior for didanosine.

A 74-year-old man with terminal chronic kidney disease, under hemodialysis and with residual diuresis, was admitted due to myalgia, arthralgia, fever and pyuria in the previous 10 days. The patient had a recent diagnosis of high-grade non-invasive bladder cancer and was doing weekly BCG intravesical administrations. The symptoms started three days before the fifth administration. He had done cefixime as an outpatient and started piperacillin-tazobactam on hospital admission, but the fever persisted, and there was no bacterial isolation in urine or blood culture. On the tenth and seventeenth day after the last BCG intravesical administration Mycobacterium bovis was still isolated in the urine culture. The diagnosis of BCGitis was made and treatment was started, with a good response. Forty days after the last administration and under treatment, the culture remained positive for Mycobacterium bovis in the urine. We raise the question about the safety of BCG administration in patients with residual diuresis.

Identification of novel specialized metabolites or bioactive compounds represents the main objective in the research field of natural product leads and drug discovery. Mass spectrometry (MS) provides a central tool to expedite and make more efficient the discovery and isolation phases, while minimizing the waste of resources on rediscovery of known compounds. MS contributes acutely to elucidation and identification of numerous species because it allows molecular mass and structural features determination. In particular, identification of the elemental composition of a precursor ion of interest by accurate mass measurement and investigation of dissociative processes undergone by the molecule, represent a worthy methodology to access the structure assignment. The aim of this study was to discover and identify novel antibacterial drugs from microbial source in a jungle of already known compounds. The focus of this paper is on the analytical strategy that permitted the disclosure of a new compound, otherwise confused with other substances. Emphasis is placed on the interpretation of the ESI-MS/MS fragmentation pattern that combined with high resolution mass determination, allowed step by step to properly deduce the exact molecular formula of an unknown component with a molecular weight higher than 1500 Daltons.

AIM: To evaluate and compare detection of lymphatic and blood vessel invasion (lVI and BVI) by hematox-ylin-eosin (HE) and immunohistochemistry (IHC) in gastric cancer specimens, and to correlate with lymph node status. METHODS: IHC using D2-40 (a lymphatic endothelial marker) and CD34 (a pan-endothelial marker) was performed to study lVI and BVI in surgical specimens froma consecutive series of 95 primary gastric cancer cases. The results of the IHC study were compared with the detection by HE using McNemar test and kappa index. The morphologic features of the tumors and the presence of lVI and BVI were related to the presence of lymph node metastasis. A χ2 test was performed to obtain associations between lVI and BVI and other prognostic factors for gastric cancer. RESUlTS: The detection rate of lVI was considerably higher than that of BVI. The IHC study identified eight false-positive cases and 13 false-negative cases for lVI, and 24 false-positive cases and 10 false-negative cases for BVI. The average Kappa value determined was moderate for lVI (k=0.50) and low for BVI (k=0.20). Both lVI and BVI were statistically associated with the presence of lymph node metastasis (HE: P=0.001, P=0.013, and IHC: P=0.001, P=0.019). The mor-phologic features associated with lVI were location of the tumor in the distal third of the stomach (P=0.039), Borrmann's macroscopic type (P=0.001), organ inva-sion (P=0.03) and the depth of tumor invasion (P=0.001). The presence of BVI was related only to the depth of tumor invasion (P=0.003). CONClUSION: The immunohistochemical identification of lymphatic and blood vessels is useful for increasing the accuracy of the diagnosis of vessel invasion and for predicting lymph node metastasis.

Objective:Features of colorectal cancer such as natural history,molecular,chromosomal,and epigenetic alterations have been well ***,there is still a lack of accurate prognostic markers,which is evident by the lower overall survival rates of patients with advanced *** alterations in parkin protein expression have been described in colorectal cancer,the functional significance of this protein remains *** present study aimed to investigate the involvement of parkin expression in colorectal adenocarcinoma development and progression by evaluating the association between its expression,clinicopathological parameters,and expression of known proteins involved in colorectal ***:Tissue microarrays consisting of 73 tumor and 64 normal tissue samples were generated to examine parkin expression and localization by ***:A positive correlation of parkin and APC expression was observed in the superficial,intermediate,and profound regions of all cases(ρ=0.37;P=0.001).Parkin expression was also significantly associated with tumors in men(P=0.049),those of the mucinous subtype(P=0.028),and of advanced stage(III+IV,P=0.041).In addition,increased parkin expression was observed in the invasive front tumor region(P=0.013).More importantly,a positive correlation was found between parkin expression and the overall survival of patients with advanced colorectal cancer(P=0.019).Multivariate analysis showed that parkin expression was independent of any of the clinicopathological parameters evaluated in relation to patient ***:These results suggest that parkin expression status can be used as a potential independent prognostic marker of survival in advanced colorectal cancer.

Serrated adenocarcinoma is a recently described subset of colorectal cancer(CRC),which account for about10%of all CRCs and follows an alternative pathway in which serrated polyps replace the traditional adenoma as the precursor lesion to *** polyps form a heterogeneous group of colorectal lesions that includes hyperplastic polyps(HPs),sessile serrated adenoma(SSA),traditional serrated adenoma(TSA)and mixed *** are the most common serrated polyp followed by SSA and *** distinct histogenesis is believed to have a major influence in prevention strategies,patient prognosis and therapeutic ***,serrated polyps exhibited also a distinct pattern,with KRAS and BRAF having an important contribution to its *** other molecular changes that have been implicated in the serrated pathway include microsatellite instability and the CpG island methylator *** the present review we will address the current knowledge of serrated polyps,clinical pathological features and will update the most recent findings of its molecular *** understanding of their biology and malignancy potential is imperative to implement a surveillance approach in order to prevent colorectal cancer development.

Characterization and Modification of a Clay Mineral Used in Adsorption Tests

Clay minerals are widespread in natural systems and have been widely used for the removal of pollutants. In this study, natural expanded vermiculite was used in adsorption tests to remove ammonium nitrogen from landfill leachate. The modification of vermiculite was carried out using NaOH and HCl, and for both modifications the best concentration was 0.1 mol/l. The results produced by XRD (X-ray diffraction) showed that Al replaced K after modification of the vermiculite using HCl and that Mg and Na replaced K after modification using NaOH. It was observed that the adsorption capacity increased as the percentage in mass of K diminished. The langmuir is the isotherm that presents the best fit of the data, and the values of Rl (the langmuir coefficient) suggest that the adsorption is linear. The thermodynamic parameters indicate that the process is spontaneous and endothermic, that there is a high affinity between the adsorbate and the adsorbent, and that physical adsorption is prevalent.
