Influence of uncertainties: A focus on block volume and shape assessment for rockfall analysis

Block size and shape depend on the state of fracturing of the rock mass and,consequently,on the geometrical features of the discontinuity sets(mainly orientation,spacing,and persistence).The development of non-contact surveying techniques applied to rock mass characterization offers significant advantages in terms of data numerosity,precision,and accuracy,allowing for performing a rigorous statistical analysis of the *** fact is particularly evident when dealing with rockfall phenomena:uncertainties in spacing and orientation data could significantly amplify the uncertainties connected with in situ block size distribution(IBSD),which represents a relation between each possible value of the volume and its probability of not being *** addition to volume,block shape can be considered as a derived parameter that suffers from *** attempts to model the possible trajectories of blocks considering their actual shape have been proposed,aiming to reproduce the effect on *** authors proposed analytical equations for calculating the expected value and variance of volume distributions,based on the geometrically correct equation for block volume in the case of three discontinuity *** quantify and discuss the effect of both volume and shape variability through a synthetic case ***,a fictitious rock mass with three discontinuity sets is assumed as the source of *** IBSDs obtained considering different spacing datasets are quantitatively compared,and the overall uncertainty effect is assessed,proving the correctness of the proposed ***,block shape distributions are obtained and compared,confirming the variability of shapes within the same ***,a comparison between trajectory simulations on the synthetic slope is reported,aiming to highlight the effects of the propagation of uncertainties to block volume and shape *** benefits of an approach that can quantify the uncertainties are discussed

An integrated multiscale approach for characterization of rock masses subjected to tunnel excavation

The design of tunnels must be conducted based on the knowledge of the territory. The longer the structure, the larger the area to be investigated, and the greater the number of surveys and tests to be performed in order to thoroughly examine all the relevant features. Therefore, optimization of the investigation process is strongly required to obtain complete and reliable data for the design of the infrastructure. The fast development of remote sensing technologies and the affordability of their products have contributed to proving their benefits as supports for investigation, encouraging the spreading of automatic or semi-automatic methods for regional scale surveys. Similarly, considering the scale of the rock outcrop, photogrammetric and laser scanner techniques are well-established techniques for representing geometrical features of rock masses, and the benefits of non-contact surveys in terms of safety and time consumption are acknowledged. Unfortunately, in most cases, data obtained at different scales of investigations are only partially integrated or compared, probably due to the missing exchange of knowledge among experts of different fields（e.g. geologists and geotechnical engineers）. The authors,after experiencing such a lack of connection among the results of different surveys concerning tunnels,propose a multiscale approach for the optimization of the investigation process, starting from the regional scale, to obtain the data that can be useful not only for planning more detailed surveys in a preliminary phase, but also for making previsions on the discontinuity sets that are present in the rock masses subjected to excavations. A methodological process is proposed and illustrated by means of a case study. Preliminary results are discussed to highlight the potentiality of this method and its limitations.

Damaging effects of gliadin on three-dimensional cell culture model

AIM： To evaluate the effects of gliadin on the oxidative environment in the “in vivo-like”model of a three-dimensional cell culture system. METHODS： LoVo cell line （intestinal adenocarcinoma） multicellular spheroids were treated with digested gliadin （with albumin used as a control）. Spheroid volumes, cell viability and morphology, lactate dehydrogenase （LDH） release, content of reduced glutathione （GSH） and activity of GSH-related enzymes were examined. The data were statistically analyzed using the Student＇s t-test （P〈0.05）. was considered statistically significant. RESULTS： Gliadin reduced cell viability （from 20% to 60%） and led to morphological alterations characterized by apoptotic findings and cytoskeletal injuries. LDH activity increased. The content of GSH reduced （-20% vscontrols）, and activity of GSH-related enzymes was significantly inhibited. CONCLUSION： Gliadin treatment induces an imbalance in the antioxidative mechanism of cells cultured by the three-dimensional technique. This alteration may explain cell damage directly caused by gliadin and the subsequentmorphological abnormalities.

To the best of our knowledge no multicenter studies have been published using standardized semi-quantitative evaluation of [123I]FP-CIT scan (DAT-SPECT). The aims of this study were: 1) to cross-compare semi-quantitative software-assisted evaluations of DAT-SPECTs performed in three centers with different equipments;2) to assess the accuracy of semi-quantitative evaluations of DAT-SPECT and 3) to identify the threshold with the best accuracy, sensitivity and specificity in a patient population with suspected parkinsonian syndrome. Materials and Methods: Two hundred twenty patients (mean age at the time of SPECT acquisition, 67.4 ± 9.5 yy) acquired in three centers (Ospedale San Luigi Gonzaga;Ospedale San Giovanni Battista Molinette;Ospedale Mauriziano Umberto I) were included. All of them underwent DAT-SPECT from January 2006 to July 2010. All exams were analyzed with the freely available software BASGAN and semi-quantitative data were used to predict disease. In particular, analyses were based on the values from the most deteriorated putamen and caudate, normalized for age and corrected for equipment. ROC analysis was performed and area under the curve (AUC) was estimated. Results: Analysis showed high AUCs (0.898, 0.864, 0.900 and 0.891 for each center and for the multicenter setting, respectively) confirming the very good accuracies reached. The best cut-off were 0.72 and 0.82 for putamen and caudate respectively. These thresholds allowed sensitivities and specificities in each center and in the multicenter setting of 76% and 96%, 91% and 82%, 93% and 90%, 86% and 89% respectively. No significant differences were observed between sensibility and specificity in each center. Conclusion: A unique threshold useful for all centers with high and similar sensitivities and specificities is possible after correction for age and equipments. Searching for the neutrinoless double beta decay(NLDBD)is now regarded as the topmost promising technique to explore the nature of neutrinos after the discovery of neutrino masses in oscillation *** X-III(particle and astrophysical xenon experiment III)will search for the NLDBD of136Xe at the China Jin Ping Underground Laboratory(CJPL).In the first phase of the experiment,a high pressure gas Time Projection Chamber(TPC)will contain 200 kg,90%136Xe enriched gas operated at10 *** pitch micro-pattern gas detector(Microbulk Micromegas)will be used at both ends of the TPC for the charge readout with a cathode in the *** signals can be used to reconstruct the electron tracks of the NLDBD events and provide good energy and spatial *** detector will be immersed in a large water tank to ensure～5 m of water shielding in all *** second phase,a ton-scale experiment,will consist of five TPCs in the same water tank,with improved energy resolution and better control over backgrounds.
