Bone marrow-derived mesenchymal stem cells versus adipose-derived mesenchymal stem cells for peripheral nerve regeneration

Studies have confirmed that bone marrow-derived mesenchymal stem cells （MSCs） can be used for treatment of several nervous system diseases. However, isolation of bone marrow-derived MSCs （BMSCs） is an invasive and painful process and the yield is very low. Therefore, there is a need to search for other alterative stem cell sources. Adipose-derived MSCs （ADSCs） have phenotypic and gene expression profiles similar to those of BMSCs. The production of ADSCs is greater than that of BMSCs, and ADSCs proliferate faster than BMSCs. To compare the effects of venous grafts containing BMSCs or ADSCs on sciatic nerve injury, in this study, rats were randomly divided into four groups： sham （only sciatic nerve exposed）, Matrigel （MG; sciatic nerve injury ＋ intravenous transplantation of MG vehicle）, ADSCs （sciatic nerve injury ＋ intravenous MG containing ADSCs）, and BMSCs （sciatic nerve injury ＋ intravenous MG containing BMSCs） groups. Sciatic functional index was calculated to evaluate the function of injured sciatic nerve. Morphologic characteristics of nerves distal to the lesion were observed by toluidine blue staining. Spinal motor neurons labeled with Fluoro-Gold were quantitatively assessed. Compared with sham-operated rats, sciatic functional index was lower, the density of small-diameter fibers was significantly increased, and the number of motor neurons significantly decreased in rats with sciatic nerve injury. Neither ADSCs nor BMSCs significantly improved the sciatic nerve function of rats with sciatic nerve injury,increased fiber density, fiber diameters, axonal diameters, myelin sheath thickness, and G ratios （axonal diameter/fiber diameter ratios） in the sciatic nerve distal to the lesion site. There was no significant difference in the number of spinal motor neurons among ADSCs, BMSCs and MG groups. These results suggest that neither BMSCs nor ADSCs provide satisfactory results for peripheral nerve repair when using MG as the conductor for engraftment.

Nutritional supplements use in high-performance athletes is related with lower nutritional inadequacy from food

Background: The use of nutritional supplements(NS) among athletes is widespread. However, little is known about the relationship between nutritional adequacy and NS usage. The aims of this study were to evaluate the NS usage and to compare the nutritional intake from food and prevalence of micronutrient inadequacy(PMI) between NS users and ***: Portuguese athletes from 13 sports completed an NS usage questionnaire and a semi-quantitative food-frequency questionnaire assessing information over the previous 12 months. The estimated average requirement cut-point method was used to calculate PMI. General linear models were used to compare nutritional intake and NS usage. Chi-squared tests and logistic regression were performed to study, respectively,relationships and associations between PMI and NS ***: From the 244 athletes(66% males, 13–37 years), 64% reported NS usage. After adjustment, NS users showed a higher intake from food(p < 0.05), for at least 1 gender, for energy, and for 7 of the 17 studied nutrients. The highest PMI were seen for vitamins D and E, calcium, folate,and magnesium. After adjustment, NS users, irrespective of gender, reported lower PMI for calcium(OR = 0.28, 95%CI: 0.12–0.65), and female users for magnesium(OR = 0.06, 95%CI: 0.00–0.98).Conclusion: Athletes using NS reported a higher nutritional intake from food, and a lower PMI for several nutrients. Perhaps, those who were taking NS were probably the ones who would least benefit from it.

Myenteric neurons and intestinal mucosa of diabetic rats after ascorbic acid supplementation

AIM: To investigate the effect of ascorbic acid (AA) dietary supplementation on myenteric neurons and epithelial cell proliferation of the jejunum of adult rats with chronic diabetes mellitus. METHODS: Thirty rats at 90 d of age were divided into three groups: Non-diabetic, diabetic and diabetic treated with AA (DA) (1 g/L). After 120 d of treatment with AA the animals were killed. The myenteric neurons were stained for myosin-V and analyzed quantitatively in an area of 11.2 mm2/animal. We further measured the cellular area of 500 neurons per group. We also determined the metaphasic index (MI) of the jejunum mucosa layer of about 2500 cells in the intestinal crypts, as well as the dimensions of 30 villi and 30 crypts/animal. The data area was analyzed using the Olympus BX40 microscope. RESULTS: There was an increase of 14% in the neuronal density (792.6 ± 46.52 vs 680.6 ± 30.27) and 4.4% in the cellular area (303.4 ± 5.19 vs 291.1 ± 6.0) respectively of the diabetic group treated with AA when compared to control diabetic animals. There were no signifi cant differences in MI parameters, villi height or crypt depths among the ***: Supplementation with AA in the diabetic animal promoted moderate neuroprotection. There was no observation of alteration of the cellular proliferation of the jejunum mucosa layer of rats with chronic diabetes mellitus with or without supplementation with AA.

Litterfall dynamics and soil carbon and nitrogen stocks in the Brazilian palm swamp ecosystems

Background: This study aimed to determine the litterfall production, accumulation, decomposition rate and nutrient stocks, and to estimate the soil carbon(C) and nitrogen(N) stocks in three palm swamp ecosystems with different land use intensities in the Southeast of ***: Three palm swamp ecosystems with different land use intensities were evaluated: Agua Doce(AD),conserved area;Capivara(CV), area with small agricultural practices;and Buriti Grosso(BG), area with an intensive land use. Produced and accumulated litterfall from the Cerrado areas surrounding the palm swamps were collected from October 2014 to September 2015, and the Ca, Mg, K, and P concentrations were determined in the dry and rainy seasons. Soil samples were collected in the surrounding Cerrado and within the palm swamps until 100 cm soil depth to determine bulk density and soil C and N contents and ***: Annual litterfall production in the Cerrado surrounding palm swamps was similar in AD(3.58 Mg·ha^-1·year^-1) and CV(3.79 Mg·ha^-1·year^-1), and was lower in BG(2.84 Mg·ha^-1·year^-1), and was more intense during the dry season. Furthermore, litterfall accumulation was higher in CV(7.12 Mg·ha^-1·year^-1) and BG(6.75 Mg·ha^-1·year^-1), culminating in lower decomposition rates. AD showed the highest decomposition rate(0.60) due to its vegetation structure and conservation. The macronutrient contents from the production and accumulated litterfall had a low influence of the land use, decreasing in the following order: Ca > K > Mg > P. Soils in CV and BG palm swamp areas showed higher values of C and N contents, due to the use of land for agricultural crops and cattle raising in an extensive(rangeland) system. Anal canal gastrointestinal stromal tumors:Case report and literature review

Gastrointestinal stromal tumors (GIST) are an uncommon group of tumors of mesenchymal origin. GIST of the anal canal is extremely rare. At present, only 10 cases of c-kit positive anal GIST have been reported in the literature. There is no widely accepted treatment approach for this neoplasia. Literature is sparse on imaging evaluation of anal canal GIST, usually described as a lesion in the intersphincteric space. We describe the case of a 73-year-old man with a mass in the anal canal, and no other symptoms. Endoanal ultrasound and magnetic resonance imaging showed a well circumscribed solid nodule in the intersphincteric space. The patient was treated by local excision. Gross pathological examination showed a 7 cm × 3.5 cm × 3 cm mass, and histological examination showed a proliferation of spindle cells, with prominent nuclear palisading. The mitotic count was of 12 mitoses/50 HPF. The tumor was positive for KIT protein, CD34 and vimentin in the majority of cells, and negative for desmin and S100. A diagnosis of GIST, with high risk aggressive behavior was made. An abdomino-perineal resection was discussed, but refused. The follow-up included clinical evaluation and anal ultrasound. After 5 years the patient is well, with maintained continence and no evidence of local recurrence.

Antifungal activity of plant extracts with potential to control plant pathogens in pineapple

Objective:To evaluate the in vitro antifungal activity of extracts,resins,oils and mother tinctures from plants against the filamentous fungi Fusarium guttiforme(***)and Chalara paradoxa,and to evaluate the control of the pineapple fusariosis in situ using mother ***:The screening of the antifungal potential of 131 extract forms from 63 plant species was performed in vitro by using plate-hole *** control pineapple fusariosis in situ,preventive and post-infection treatments were performed on detached pineapple leaves of ***(susceptible).Results:The quantitative study indicated that among the 49 mother tincture samples analyzed,46% were effective against *** and 29% for the Chalara *** natural plant extracts,mother tincture of Glycyrrhiza glabra(MTGG1),mother tincture of Myroxylon balsamum(MTBT2),mother tincture of Aloe vera(MTAV3),mother tincture of Allium sativum(MTAS4),resin of Protium heptaphyllum(RESAM5) and crude extracts of Rhizophora mangle(CEMV6),exhibited an antifungal activity against *** the preventive treatment against pineapple fusariosis,MTAV3,MTAS4 and MTGG1 were statistically similar to the treatment with tebuconazol *** curative treatments with MTAV3,MTAS4,MTGG1 and MTBT2 presented similar activity to fungicide(P < 0.05).Conclusions:The findings of the present study concluded that mother tinctures can effectively control *** mother tincture extract of Myroxylon balsamum showed antifungal activity and was used here for the first time for inhibition of phytopathogenic *** study paves the way for the development of bioactive natural products with phytosanitary applications,with the added benefits of an environmentally safe and economically viable product.

Molecular evaluation of glutathione S transferase family genes in patients with sporadic colorectal cancer

AIM To evaluate the association between polymorphismsin glutathione S transferases(GSTs) and the risk of sporadic colorectal cancer(SCRC), tumor progression and the survival of *** A case-control study of 970 individuals from the Brazilian population was conducted(232 individuals from the case group with colorectal cancer and 738 individuals from the control group without a history of cancer). PCR multiplex and PCR-RFLP techniques were used to genotype the GST polymorphisms. The tumors were categorized according to the TNM classification: tumor extension(T), affected lymph nodes(N), and presence of metastasis(M). Logistic regression, multiple logistic regression and survival analysis were used to analyze the data. The results are presented in terms of odds ratio(OR) and 95% confidence interval(CI). The level of significance was set at 5%(P ≤ 0.05).RESULTS Age equal to or over 62 years(OR = 8.79; 95%CI: 5.90-13.09, P < 0.01) and female gender(OR = 2.91; 95%CI: 1.74-4.37; P < 0.01) were associated with increased risk of SCRC. Analysis of the polymorphisms revealed an association between the GSTM1 polymorphisms and a risk of SCRC(OR = 1.45; 95%CI: 1.06-2.00; P = 0.02), as well as between GSTT1 and a reduced risk of the disease(OR = 0.65; 95%CI: 0.43-0.98; P = 0.04). An interaction between the presence of the wild-type allele of GSTP1 Ile105 Val polymorphism and tobacco consumption on risk of SCRC(OR = 2.33; 95%CI: 1.34-4.05; P = 0.05) was observed. There was an association between the GSTM1 null genotype and the presence of advanced tumors(OR = 2.33; 95%CI: 1.23-4.41; P = 0.009), as well as increased risk of SCRC in the presence of a combination of GSTT1 non-null/GSTM1 null genotypes(OR = 1.50; 95%CI: 1.03-2.19; P = 0.03) and GSTT1 non-null/GSTM1 null/GSTP1 Val~*(OR = 1.85; 95%CI: 1.01-3.36, P = 0.04). The CaO-P_(2)O_(5)-SiO_(2)-MgO system presents several compounds used as biomaterials such as hydroxyapatite(HA),tricalcium phosphate(TCP)and TCP with magnesium substituting partial calcium(TCMP).Theβ-TCMP phase with whitlockite structure has interesting biological features and mechanical properties,meeting the requirements of a bioactive material for bone *** this work,the production of Mg-doped TCP,β-TCMP,has been investigated by crystallization from a glass composed of 52.75 wt%3CaO·P_(2)O_(5),30 wt%SiO_(2)and 17.25 wt%MgO(i.e.,31.7 mol%CaO,10.6 mol%P_(2)O_(5),26.6 mol%MgO and 31.1 mol%SiO_(2))using heat treatments between 775℃and 1100℃for up to 8 *** devitrification process of the glass has been accompanied by differential scanning calorimetry(DSC),high-resolution X-ray diffraction(HRXRD),relative density and bending strength *** characterization by HRXRD and DSC revealed the occurrence of whitlockite soon after the bulk glass preparation,a transient non-cataloged silicate between 800℃and 1100℃,and the formation of diopside in samples treated at 1100℃as crystalline *** overall crystalline fraction varied from 26%to 70%depending on the heat ***,contraction of the a-axis lattice parameter and expansion of the c-axis lattice parameter of the whitlockite structure have been observed during the heat treatments,which were attributed to theβ-TCMP formation with the partial substitution of Ca^(2+)by Mg^(2+).Relative densities near 99%and 97%for the glass and glass-ceramics respectively indicated a discrete reduction as a function of the devitrification *** strengths of 70 MPa and 120 MPa were determined for the glass and glass-ceramic material crystallized at 975℃for 4 h,respectively.
