Effects of undigested protein-rich ingredients on polarised small intestinal organoid monolayers

Here, we describe the use of monolayers of intestinal epithelial cells derived from intestinal organoids and transcriptomics to investigate the direct effects of dietary protein sources on epithelial function. Mechanically dissociated 3 D organoids of mouse duodenum were used to generate a polarized epithelium containing all cell types found in the tissue of origin. The organoid-derived cell monolayers were exposed to 4%(w/v) of ‘undigested(non-hydrolysed)-soluble' fraction of protein sources used as feed ingredients [soybean meal(SBM) and casein], or alternative protein sources(spray dried plasma protein, and yellow meal worm), or controls for 6 h prior to RNA isolation and transcriptomics. All protein sources altered expression of unique biological processes in the epithelial cells. Exposure of intestinal organoids to SBM downregulated expression of retinol and retinoid metabolic processes as well as cholesterol and lipid biosynthetic pathways, consistent with the reported hypotriglyceridaemic effect of soy protein in vivo. These findings support the use of intestinal organoids as models to evaluate complex interactions between dietary ingredients and the intestinal epithelium and highlights some unique host effects of alternative protein sources in animal feed and potentially human food.

tRNA^(Glu)-derived fragments from embryonic extracellular vesicles modulate bovine embryo hatching

Transfer RNA-derived small RNAs(tsRNAs)have been shown to be involved in early embryo development and repression of endogenous retroelements in embryos and stem ***,it is unknown whether tsRNAs also regulate embryo *** this study,we mined the sequencing data of a previous experiment in which we demonstrated that the microRNA(miRNA)cargo of preimplantation embryonic extracellular vesicles(EVs)influences embryo *** thus profiled the tsRNA cargo of EVs secreted by blastocysts and *** majority of tsRNAs was identified as tRNA halves originating from the 5'ends of *** the 148 differentially expressed tsRNAs,the 19 nt tRNA fragment(tRF)tDR-14:32-Glu-CTC-1 was found to be significantly up-regulated in EVs derived from ***-qPCR assays confirmed its significant up-regulation in non-blastocyst embryos and their conditioned medium compared to the blastocyst group(P<0.05).Inhibition of tDR-14:32-Glu-CTC-1 by supplementing antagomirs to the conditioned medium improved embryo hatching(P<0.05).Transcriptomic analysis of embryos treated with tDR-14:32-Glu-CTC-1 antagomirs further showed differential expression of genes that are associated with embryo hatching and *** summary,tDR-14:32-Glu-CTC-1 is up-regulated in non-blastocyst embryos and their secretions,and inhibition of tDR-14:32-Glu-CTC-1 promotes embryo hatching,while influencing embryo implantation-related genes and *** results indicate that embryonic EVs containing specific tRFs may regulate preimplantation embryo development.

Forces and energetics of intermittent swimming

Experiments are reported on intermittent swimming motions. Water tunnel experiments on a nominally two-dimensional pitching foil show that the mean thrust and power scale linearly with the duty cycle, from a value of 0.2 all the way up to continuous motions, indicating that individual bursts of activity in intermittent motions are independent of each other. This conclusion is corroborated by particle image velocimetry（PIV） flow visualizations, which show that the main vortical structures in the wake do not change with duty cycle. The experimental data also demonstrate that intermittent motions are generally energetically advantageous over continuous motions. When metabolic energy losses are taken into account, this conclusion is maintained for metabolic power fractions less than 1.

This paper reviews the use of fiber-reinforced polymers （FRPs） in architectural and structural bridge design in the Netherlands. The challenges and opportunities of this relatively new material, both for the architect and the engineer, are discussed. An inventory of recent structural solutions in FRP is included, followed by a discussion on architectural FRP applications derived from the architectural practice of the author and of other pioneers.

Experimental and Modeling Study of Forest Fire Effect on Soil Thermal Conductivity

An understanding of soil thermal conductivity after a wildfire or controlled burn is important to land management and post-fire recovery efforts. Although soil thermal conductivity has been well studied for non-fire heated soils, comprehensive data that evaluate the long-term effect of extreme heating from a fire on the soil thermal conductivity are limited. The purpose of this study was to evaluate the long-term impact of fire on the effective thermal conductivity of soils by directly comparing fire-heated and no-fire control soils through a series of laboratory studies. The thermal conductivity was measured for ten soil samples from two sites within the Manitou Experimental Forest, Colorado, USA, for a range of water contents from saturation to the residual degree of saturation. The thermal conductivity measured was compared with independent estimates made using three empirical models from literature, including the Campbell et al. （1994）, CSt~ and Konrad （2005）, and Massman et al. （2008） models. Results demonstrate that for the test soils studied, the thermal conductivity of the fire-heated soils was slightly lower than that of the control soils for all observed water contents. Modeling results show that the Campbell et al. （1994） model gave the best agreement over the full range of water contents when proper fitting parameters were employed. Further studies are needed to evaluate the significance of including the influence of fire burn on the thermal properties of soils in modeling studies.

This article describes the FAIRification process(which involves making data Findable, Accessible, Interoperable and Reusable—or FAIR—for both machines and humans) for data related to the impact of COVID-19 on migrants, refugees and asylum seekers in Tunisia, Libya and Niger, according to the scheme adopted by GO FAIR. This process was divided into three phases: pre-FAIRification, FAIRification and postFAIRification. Each phase consisted of seven steps. In the first phase, 118 in-depth interviews and 565 press articles and research reports were collected by students and researchers at the University of Sousse in Tunisia and researchers in Niger. These interviews, articles and reports constitute the dataset for this research. In the second phase, the data were sorted and converted into a machine actionable format and published on a FAIR Data Point hosted at the University of Sousse. In the third phase, an assessment of the implementation of the FAIR Guidelines was undertaken. Certain barriers and challenges were faced in this process and solutions were found. For FAIR data curation, certain changes need to be made to the technical process. People need to be convinced to make these changes and that the implementation of FAIR will generate a long-term return on investment. Although the implementation of FAIR Guidelines is not straightforward, making our resources FAIR is essential to achieving better science together.

Photoionization Modelling of H II Region for Oxygen Ions

We present phoionization model calculations of ionization fraction of hydrogen, helium and oxy-gen at low density of photoionized nebulae using the photoionization code CLOUDY. The ionization parameter is between -2.0 ≤ log(U) ≤ 1.0, and we study them by changing the abundances of oxy-gen from -2.5 ≤ log(O/H) ≤ -5.0. This was tested first by Campbell (1988) [1]. But, this was differ-ent in highest abundances of oxygen, filling factor and carbon abundances. We change the abun-dances of carbon by 0.5 dex of solar value, to avoid formation CO in the process. We include the charge transfer interaction between oxygen ions and hydrogen atom. Based on this process, we have calculated the ionization profiles of O0, O+, O2+ O3+ and O4+ in the presence and absence of charge exchange between hydrogen and oxygen. In this work, we assumed the effective rate of co-efficient for the charge transfer is the order of ≈10-9 cm-3-s-1.

Model of Hydrogen Deficient Nebulae in H II Regions at High Temperature

In current photoionization models, determination of the ionization structure of the nebulae is a crucial part of the calculations for non-uniform hydrogen density. We have been tried to compute temperature fluctuations of electron and electron density by changing hydrogen density by scale factor of 10 at high temperature of blackbody. We have studied the ionization structure of hydrogen and helium by means of cloudy C13.01 codes. We find a reasonable close agreement of the computed value. Moreover, the ionization structure of oxygen at ionization stages of, O I - O IV, in gaseous nebulae are studied by changing non uniform hydrogen density with power law. Our simulation also shows that how the change of this density affect temperature fluctuations, electron density and ionization structures of oxygen, helium and hydrogen with and without grains. The result also shows the impact of the forbidden lines of [O III] and [O II] to cool down the temperature of electron.
