Metabolic surgery and intestinal gene expression:Digestive tract and diabetes evolution considerations

AIM: To investigate the effects of bariatric surgery on metabolic parameters, incretin hormone secretion, and duodenal and ileal mucosal gene ***: Nine patients with type 2 diabetes mellitus(T2DM), chronic serum hyperglycemia for more than2 years, and a body mass index(BMI) of 30-35 kg/m2 underwent metabolic surgery sleeve gastrectomy with transit bipartition between May 2011 and December2011. Blood samples were collected pre and 3, 6 and12 mo postsurgery. Duodenal and ileal mucosa samples were collected pre- and 3 mo postsurgery. Pre- and postoperative blood samples were collected in the fasting state before ingestion of a standard meal(520kcal) and again 30, 60, 90, and 120 min after the mealto determine hemoglobin A1c(HbA1c) levels and the lipid profile, which consisted of triglyceride and total cholesterol levels. Intestinal gene expression of p53 and transforming growth factor(TGF)-β was analyzed using quantitative reverse-transcription PCR. Gastric inhibitory polypeptide(GIP) and glucagon-like peptide-1(GLP-1)were quantified using the enzyme-linked immunoassay method and analyzed pre- and ***'s t test or repeated measurements analysis of variance with Bonferroni corrections were performed as ***: BMI values decreased by 15.7% within the initial 3 mo after surgery(31.29 ± 0.73 vs 26.398± 0.68, P < 0.05) and then stabilized at 22% at 6mo postoperative, resulting in similar values 12 mo postoperatively(20-25 kg/m2). All of the patients experienced improved T2 DM, with 7 patients(78%)achieving complete remission(HbA1c < 6.5%), and 2patients(22%) achieving improved diabetes(HbA1c< 7.0% with or without the use of oral hypoglycemic agents). At 3 mo postoperatively, fasting plasma glucose had also decreased(59%)(269.55 ±18.24 mg/dL vs 100.77 ± 3.13 mg/dL, P < 0.05)with no further significant changes at 6 or 12 mo postoperatively. Background： Leptin has a strong relation to important traits in animal production, such as carcass composition,feed intake, and reproduction. It is mainly produced by adipose cells and acts predominantly in the *** this study, circulating leptin and its gene expression in muscle were evaluated in two groups of young Nellore bulls with divergent feed efficiency. Individual dry matter intake（DMI） and average daily gain（ADG） of 98 Nellore bulls were evaluated in feedlot for 70 d to determinate the residual feed intake（RFI） and select 20 animals for the high feed efficient（LRFI） and 20 for the low feed efficient（HRFI） groups. Blood samples were collected on d 56 and at slaughter（80 d） to determine circulating plasma leptin. Samples of Longissimus dorsi were taken at slaughter for leptin gene expression ***： DMI and RFI were different between groups and LRFI animals showed less back fat and rump fat thickness,as well as less pelvic and kidney fat weight. Circulating leptin increased over time in all animals. Plasma leptin was greater in LRFI on 56 d and at slaughter（P = 0.0049）. Gene expression of leptin were greater in LRFI animals（P = 0.0022） in accordance with the plasma levels. The animals of the LRFI group were leaner, ate less, and had more circulating leptin and its gene ***： These findings demonstrated that leptin plays its physiological role in young Nellore bulls, probably controlling food intake because feed efficient animals have more leptin and lower residual feed intake.

An Application of Paraconsistent Annotated Logic for Design Software Testing Strategies

Nowadays, application model systems for decision-making based on non-classical logic such as Paraconsistent Logic are used successfully in the treatment of uncertainties. The method presented in this paper is based on the fundamental concepts of Paraconsistent Annotated Logic with annotation of 2 values (PAL2v). In this study, two algorithms based on PAL2v are presented gradually, to extract the effects of the contradiction in signals of information from a database of uncertain knowledge. The Paraconsistent Extractors Algorithms of Contradiction Effect-Para Extrctr is applied to filters of networks of analyses (PANets) of signal information, where uncertain and contradictory signals may be found. Software test case scenarios are subordinated to an application model of Paraconsistent decision-making, which provides an analysis using Paraconsistent Logic in the treatment of uncertainties for design software testing strategies. This quality-quantity criterion to evaluate the software product quality is based on the characteristics of software testability analysis. The Para consistent reasoning application model system presented in this case study, reveals itself to be more efficient than the traditional methods because it has the potential to offer an appropriate treatment to different originally contradicting source information.

Background： Beef cattle breeding programs in Brazil have placed greater emphasis on the genomic study of reproductive traits of males and females due to their economic importance. In this study, genome-wide associations were assessed for scrotal circumference at 210 d of age, scrotal circumference at 420 d of age, age at first calving, and age at second calving, in Canchim beef cattle. Data quality control was conducted resulting in 672,778 SNPs and 392 ***： Associated SNPs were observed for scrotal circumference at 420 d of age（435 SNPs）, followed by scrotal circumference at 210 d of age（12 SNPs）, age at first calving（six SNPs）, and age at second calving（four SNPs）. We investigated whether significant SNPs were within genic or surrounding regions. Biological processes of genes were associated with immune system, multicellular organismal process, response to stimulus, apoptotic process, cellular component organization or biogenesis, biological adhesion, and ***： Few associations were observed for scrotal circumference at 210 d of age, age at first calving, and age at second calving, reinforcing their polygenic inheritance and the complexity of understanding the genetic architecture of reproductive traits. Finding many associations for scrotal circumference at 420 d of age in various regions of the Canchim genome also reveals the difficulty of targeting specific candidate genes that could act on fertility; nonetheless,the high linkage disequilibrium between loci herein estimated could aid to overcome this issue. Therefore, al relevant information about genomic regions influencing reproductive traits may contribute to target candidate genes for further investigation of causal mutations and aid in future genomic studies in Canchim cattle to improve the breeding program.

The aim of this study was to characterize tinctures and microcapsules loaded with an ethanol extract of red propolis through chemical, physicochemical and microbiological assays in order to establish quality control tools for nutraceutical preparations of red propolis. The markers(isoflavonoids, chalcones, pterocarpans,flavones, phenolic acids, terpenes and guttiferones) present in the tinctures A and B were identified and confirmed using LC/ESI/FTMS/Orbitrap. Four compositions(A, B, C and D) were prepared to contain B tincture of the red propolis with some pharmaceutical excipients and submitted to two drying processes, i. ***-drying and freeze-drying to obtain microcapsules loaded with the red propolis extract. The tinctures and microcapsules of the red propolis were submitted to the total flavonoid content and antioxidant activity *** antibacterial activity and minimum inhibitory concentration(MIC) were tested using Staphylococcus aureus ATCC 25293 and Pseudomonas aeruginosa ATCC 27853 strains. The tinctures and microcapsules presented high flavonoid quantities from 20.50 to 40.79 mg/100 mg of the microcapsules. The antioxidant activity and IC50 were determined for the tinctures A and B(IC50: 6.95 μg/m L and 7.48 μg/m L), the spray-dried microcapsules(IC50: 8.89–15.63 μg/m L) and the freeze-dried microcapsules(IC50: 11.83–23.36 μg/m L). The tinctures and microcapsules were proved to be bioactive against gram-positive and gram-negative bacteria with inhibition halos superior to 10 mm at concentration of 200 μg/m L and MIC values of 135.87–271.74 μg/m L using gram-positive strain and 271.74–543.48 μg/m L using gram-negative strain. The tinctures and microcapsules of the red propolis have a potential application for nutraceutical products.

Helicobacter pylori and EBV in gastric carcinomas:Methylation status and microsatellite instability

AIM:To verify the methylation status of CDH1, DAPK, COX2, hMLH1 and CDKN2A genes and to evaluate their association with Helicobacter pylori (H. pylori)-cagA+ and Epstein Barr virus (EBV) infections in gastric ***: Methylation-specific PCR (MSP) assay was performed in 89 primary gastric carcinomas (intestinal and diffuse types). Microsatellite instability (MSI) analysis was performed using the BAT26 primer set and PCR products were analyzed with the ABI PRISM 3100 Genetic Analyzer using Genescan 3.7 software (Applied Biosystems). Detection of H. pylori and genotyping were performed by PCR, using specifi c primers for ureaseC and cagA genes. The presence of EBV was assessed by in situ hybridization. Statistical analyses were performed using the χ2 or Fisher's exact ***: The most frequent hypermethylated gene was COX-2 (63.5%) followed by DAPK (55.7%), CDH1 (51%), CDKN2A (36%) and hMLH1 (30.3%). Intestinal and diffuse adenocarcinomas showed different methylation profiles and there was an association between methylation of E-CDH1 and H. pylori-cagA+ in the intestinal adenocarcinoma type. MSI was correlated with hMLH1 methylation. There was an inverse correlation between DAPK hypermethylation and ***: We found a strong association between CDH1 methylation and H. pylori-cagA+ in intestinal-type gastric cancer, association of MSI and better prognosis and an heterogeneous COX-2 overexpression.

A Resource Allocation Scheme with Delay Optimization Considering mmWave Wireless Networks

This paper presents a resource allocation scheme for wireless networks, aiming at optimizing the users’ data delay. It is proposed to provide optimal delay al-location by solving an optimization problem using idle state prediction and considering 5G characteristics such as mmWave propagation. The perfor-mance of the resource allocation algorithm is verified and compared with oth-ers from the literature using computational simulations in terms of Quality of Service (QoS) parameters such as throughput, delay, fairness index, loss rate and computational complexity. In these simulations, it is also considered the mmWave propagation and carrier aggregation technology for wireless next generation systems, in order to verify the system performance in a high data rate scenario.

PROD and BROD Modulation in Nile Tilapia after Exposure to 17 β-estradiol

The activities of 7-ethoxyresorufin-O-deetylase （EROD）, 7-benzyloxyresorufin-O-debenzylase （BROD）, 7-pentoxyresorufin-O-depentilase （PROD）, and glutathione S-transferase （GST） were measured in Nile tilapias exposed for 7 days of 5 and 15μg/L 17 β-estradiol. EROD and GST activities were unchanged. PROD activity increased in animals exposed to the higher dose of the hormone, while BROD was increased after 7 exposure days to both doses of the compound. These results indicate the usefulness of these enzymes as biomarkers for 17 β-estradiol exposure.

Background:Neglected Tropical Diseases are a set of communicable diseases that affect the population so low socioeconomic status,particularly 1.4 billion people who are living below the poverty *** study has investigated the magnitude and mortality time trends for these diseases in the state of Sergipe,Northeast Region of ***:We conducted an ecological study of time series,based on secondary data derived from the Mortality Information System of the Ministry of *** mortality rates(crude,age-standardized rates and proportional ratio)were calculated from the deaths due to Neglected Tropical Diseases in the state of Sergipe,from 1980 to *** time trends were obtained using the Joinpoint regression ***:Three hundred six thousand and eight hundred seventy-two deaths were certified in the state and Neglected Tropical Diseases were mentioned as the underlying cause in 1,203 certificates(0.39%).Mean number of deaths was 35.38 per year,and crude and age-standardized mortality rates were,respectively:2.16 per 100000 inhabitants(95%CI:1.45-2.87)and 2.87 per 100000 inhabitants(95%CI:1.93-3.82);the proportional mortality ratio was 0.41%(95%CI:0.27-0.54).In that period,Schistosomiasis caused 654 deaths(54.36%),followed by Chagas disease,with 211(17.54%),and by Leishmaniases,with 142(11.80%)*** other diseases totalized 196 deaths(16.30%).There were increasing mortality trends for Neglected Tropical Diseases,Schistosomiasis and Chagas disease in the last 15 years,according to the age-standardized rates,and stability of the mortality trends for ***:The Neglected Tropical Diseases show increasing trends and are a real public health problem in the state of Sergipe,since they are responsible for significant mortality *** following diseases call attention for showing greater number of deaths in the period of study:Schistosomiasis,Chagas disease and *** finally suggest that public managers take appropri

Physiological Effect of Kinetin on the Photosynthetic Apparatus and Antioxidant Enzymes Activities During Production of Anthurium

The results observed in the literature raise the hypothesis according to which cytokinin plays important roles in photosynthetic metabolisms and antioxidant enzymes. Thus, the study aimed to evaluate the effect of foliar application of the isolated cytokinin kinetin at the production cycle, seeking to analyze its effects on enzyme activity and photosynthetic parameters. The plants treated with CK presented reduction of leaf CO_2 assimilation rate(Pn) and stomatal conductance(Gs), while that transpiration rate(Tr) was unaffected. The internal CO_2 concentrations decreased with the increase in cytokinin levels, but were maintained under CK 50 mg·L^(-1). The plants treated with CK 75 mg·L^(-1) was verified higher carboxylation efficiency(Pn/Ci), which was associated to values of CO_2 assimilation and transpiration unaltered. Apparent electron transport rate showed variations in the concentration of 25 mg·L^(-1). Considering the study of enzyme activity, on the other hand, it cannot be stated that kinetin has an effective action in delaying oxidative damage. It presents mixed results, since an efficiency in the application of cytokinin was not observed, presenting induction levels of ascorbate peroxidase activity. Thus, further research is needed to determine more precisely the effects of kinetin on gas exchange and antioxidant enzymes in anthurium plants.
