Prognostic and predictive biomarkers in metastatic colorectal cancer anti-EGFR therapy

AIM: To reviewing genetic and epigenetic make-up of metastatic colorectal cancers (mcRcs) addicted to epidermal growth factor receptor (EGFR) ***: The present study summarizes the potential value of prognostic and predictive biomarkers in selecting mcRc patients treated with anti-EGFR therapy. A meta-analysis was performed using a systematic search of PubMed, Medline and Web of Science to identify eligible papers until March 21st, 2016 using these following terms: ‘‘colorectal cancer’’, “predictive biomarkers’’, “anti-EGFR therapy”, “KRAS”, “NRAS’’, “PIK3cA”, “TP53”, “PTEN”, ‘‘EGFR”, “MET”, “HER2”, “epiregulin”, “amphiregulin”, “prognostic biomarkers”, “BRAF”, “miRNA” and “antibody-dependent cell-mediated cytotoxicity (ADcc) activity”. Two investigators independently evaluated and extracted data from each identified studies based on selected criteria of inclusion and ***: The introduction of agents targeting EGFR such as cetuximab and panitumumab increased overall survival of mcRcs. Nevertheless, it has firstly became evident that response rates to cetuximab regimens in unselected patient populations were typically lower than 30%. clinical data confirmed the predictive value of RAS mutations for resistance to cetuximab and panitumumab leading to the license of these monoclonal antibodies exclusively for the management of patients with RAS-wild type colorectal cancers. So far the identification of predictive biomarkers have generated interesting, though preliminary and, at times, conflicting data on the importance of tumour mRNA levels of EGFR ligands, of activating mutations in other genes such as NRAS and PIK3cA. When decisions are based on empirical observations,a trade-off arises between flexibility of the decision and ability to generalize to new *** this paper,we focus on decisions that are obtained by the empirical minimization of the conditional Value-at-Risk(cVa R)and argue that in cVa R the trade-off between flexibility and generalization can be understood on the ground of theoretical results under very general assumptions on the system that generates the *** results have implications on topics related to order and structure selection in various applications where the cVa R risk-measure is used.A study on a portfolio optimization problem with real data demonstrates our results.

Turning of aluminum metal matrix composites： influence of the reinforcement and the cutting condition on the surface layer of the workpiece

Aluminum metal matrix composites （Al-MMcs） are difficult to machine. The reinforcement of aluminum using ceramic particles accelerates tool wear. Moreover, demanded machining accuracies or properties of the surface layer are difficult to achieve. In the present study, the effect of silicon carbide reinforcement particles on the surface layer of the workpiece was investigated using multiple cutting conditions for dry turning. Three differently reinforced Al-MMcs regarding the volume percent-age （17% and 30%） and the particle size （0.6 μm and 3 μm） and their non-reinforced matrix were considered as the workpiece materials. The reinforcement and the cutting condition affect the results of turning. A greater particle volume percent improves the surface roughness and decreases the tensile stress in the surface. The smaller particle size caused a lower tensile stress in the surface. A general effect of the particle size on the workpiece roughness can not be concluded. The most important cutting parameter for the surface layer of the workpiece is the feed. Greater feeds decrease the tensile stress in the surface, but deteriorate the surface quality.

The pressure versus temperature phase diagram of a shoulder-like potential is exactly computed in one system of particles interacting through a multiscale dimension. The N-shoulder potential exhibits N density anomaly regions in the phase diagram if the length scales can be connected by a convex curve The result is analyzed in terms of the convexity of the Gibbs free energy.

We consider a simple model for the diffusion of heavy quarks in a hot bath,modeling the latter by an ensemble of oscillators distributed according to either a thermal distribution or to an out-of-equilibrium distribution with a saturation *** this model it is easy to introduce memory effects by changing the distribution of oscillators:we model them by introducing a Gaussian distribution,dN/dω,which can be deformed continuously from aδ?function,giving a Markov dissipation,to a broad kernel with *** the equation of motion of the heavy quark in the bath,we remark how dissipation comes out naturally as an effect of the back-reaction of the oscillators on the ***,the exact solution of this equation allows to define the thermalization time as the time necessary to remove any memory of the initial *** find that the broadening of the dissipative kernel,while keeping the coupling fixed,lowers the thermalization *** also derive the fluctuation-dissipation theorem for the bath,and use it to estimate the kinematic regime in which momentum diffusion of the heavy quark dominates over *** find that diffusion is more important as long as K0/E is small,where K0 and E denote the initial energy of the heavy quark and the average energy of the bath,respectively.

Intraocular inflammation after Ultrasound cyclo Plasty for the treatment of glaucoma

This is a prospective interventional clinical study evaluating intraocular inflammation developed after Ultrasound cyclo Plasty(UcP) for the treatment of glaucoma. Eighteen eyes of 18 patients were treated with UcP second-generation probes(Eye OP1). After treatment, the mean intraocular pressure(IOP) significantly decreased from 26.8±7.2 to 18.8±6.1 mm Hg at day 1 and to 14.7±3.4 mm Hg at month 6(all Pcell photometry value steeply increased after surgery from 12.1±7.5 to 64.1±53.9 ph/ms(P=0.001) at day 1, and then progressively decreased to respectively 60.6±49.7 at day 7, 43.5±38.5 at day 14 and 28.2±18.3 at month 1(all Pctively 16.7±6.2 and 12.8±10.2, both P>0.05). A significant negative correlation was found between postoperative increase of aqueous flare values and anterior chamber depth(R=-0.568, P=0.014). This timeframe may be considered reasonable for repeating UcP treatment, when required.

Estimation of carbon storage of forest biomass for voluntary carbon markets:preliminary results

Estimating the carbon storage of forests is essential to support climate change mitigation and promote the transition into a low-carbon emission *** achieve this goal,voluntary carbon markets(VcMs)are *** are promoted by a spontaneous demand,not imposed by binding targets,as the regulated *** Italy,only in Veneto and Piedmont Regions(Northern Italy),VcMs through forestry activities were carried *** camonica District(Northern Italy,Lombardy Region)is ready for a local VcM,but carbon storage of its forests was never *** aim of this work was to estimate the total carbon storage(TcS;t c ha^−1)of forest biomass of Valle camonica District,at the stand level,taking into account:(1)aboveground biomass,(2)belowground biomass,(3)deadwood,and(4)*** developed a user-friendly model,based on site-specifi c primary(measured)data,and we applied it to a dataset of 2019 stands extracted from 45 Forest Management *** results showed that,in 2016,the TcS achieved 76.02 t c ha^−*** aboveground biomass was the most relevant carbon pool(48.86 t c ha^−1;64.27%of TcS).From 2017 to 2029,through multifunctional forest management,the TcS could increase of 2.48 t c ha^−1(+3.26%).In the same period,assuming to convert coppices stands to high forests,an additional TcS of 0.78 t c ha^−1(equal to 2.85 t cO 2 ha^−1)in the aboveground biomass could be achieved without increasing forest *** additional carbon could be certifi ed and exchanged on a VcM,contributing to climate change mitigation at a local level.
