BACKGROUND Toll-like receptor-2(TLR2) is responsible for recognizing Helicobacter pylori(***) and activating the immune response. Polymorphisms in TLR2 may modulate gastric *** To evaluate whether the TLR2 19216 T/C(rs3804099) and TLR2-196 to-174 ins/del(rs111200466) polymorphisms contribute to gastric carcinogenesis in the Brazilian population, and to determine the influence of both polymorphisms and H. pylori infection on TLR2 mRNA *** DNA was extracted from 854 peripheral blood leukocyte or gastric tissue samples[202 gastric cancer(GC), 269 chronic gastritis(CG), and 383 control/healthy(C)]and genotyped by allele-specific PCR or restriction fragment length polymorphism(RFLP)-PCR. Quantitative polymerase chain reaction by Taq Man■ assay was used to quantify TLR2 mRNA levels in fresh gastric tissues(48 GC, 36 CG, and 14 C).RESULTS Regarding the TLR2-196 to -174 polymorphism, the ins/del and del/del genotypes were associated with a higher risk of GC by comparison with the C in all of the analyzed inheritance models(codominant, dominant, recessive, overdominant and log-additive;P < 0.0001). Similarly, an increased risk was observed when comparing the GC and CG groups [codominant(P < 0.0001), dominant(P <0.0001), recessive(P = 0.0260), overdominant(P < 0.0001) and log-additive(P <0.0001)]. In contrast, TLR2 19216 T/C was associated with a protective effect in the GC group compared to the C group [dominant(P = 0.0420) and log-additive(P =0.0300)]. Regarding the association of polymorphisms with H. pylori infection,individuals infected with H. pylori and harboring the TLR2-196 to-174 ins/del polymorphism had an increased risk of gastric carcinogenesis [codominant(P =0.0120), dominant(P = 0.0051), overdominant(P = 0.0240) and log-additive(P =0.0030)], while TLR2 19216 T/C was associated with a protective effect[codominant(P = 0.0039), dominant(P < 0.0001), overdominant(P = 0.0097) and log-additive(P = 0.0021)]. The use of an internal control in a multiplex-PCR assay for sex determination of In Vitro-produced bovine embryos was evaluated in biopsies of groups of 54 fresh and 44 frozen embryos. The internal controls used were the primers BOV 1 and BOV 2, which amplify a product with 626 base pairs (bp) of bovine mitochondrial DNA ND5 gene. The primers BRY.4aF and BRY.4aR were used for bovine Y chromosome sequence amplification. The specificity of multiplex- PCR reactions realized in biopsies corresponding to about 20% of each fresh embryo (10 male and 10 female) by means of confirming the sexing in the remaining embryo content (~80%) presented 100% specificity. Amplicons of the internal control and Y chromosome were both amplified until dilution corresponding to 6.25% of total extracted DNA from a male embryo. Sex determination was possible in 53 (98.1%) fresh embryos and 40 (90.9%) frozen embryos. The products related to the Y chromosome and mitochondrial DNA were simultaneously amplified in 34 (63%) fresh embryos and 27 (61.4%) frozen embryos, showing a male embryo. The female sex, distinguished by internal control amplification only, was detected in 19 (35.2%) and 13 (29.5%) biopsies, respectively, of fresh and frozen embryos. In one (1.8%) and four (9.1%) biopsies of fresh and frozen embryos, respectively, neither product was amplified, most likely due to the absence of embryonic cells or the presence of embryonic cells going through apoptosis. The multiplex-PCR assay developed in this work showed avoided the limitation of a lack of an internal standard, and was also sensitive, specific, and efficient in reaction failure identification. This technique shows great potential for use on a commercial scale in routine sex determination of In Vitro-produced embryos.

Background： Leptin has a strong relation to important traits in animal production, such as carcass composition,feed intake, and reproduction. It is mainly produced by adipose cells and acts predominantly in the *** this study, circulating leptin and its gene expression in muscle were evaluated in two groups of young Nellore bulls with divergent feed efficiency. Individual dry matter intake（DMI） and average daily gain（ADG） of 98 Nellore bulls were evaluated in feedlot for 70 d to determinate the residual feed intake（RFI） and select 20 animals for the high feed efficient（LRFI） and 20 for the low feed efficient（HRFI） groups. Blood samples were collected on d 56 and at slaughter（80 d） to determine circulating plasma leptin. Samples of Longissimus dorsi were taken at slaughter for leptin gene expression ***： DMI and RFI were different between groups and LRFI animals showed less back fat and rump fat thickness,as well as less pelvic and kidney fat weight. Circulating leptin increased over time in all animals. Plasma leptin was greater in LRFI on 56 d and at slaughter（P = 0.0049）. Gene expression of leptin were greater in LRFI animals（P = 0.0022） in accordance with the plasma levels. The animals of the LRFI group were leaner, ate less, and had more circulating leptin and its gene ***： These findings demonstrated that leptin plays its physiological role in young Nellore bulls, probably controlling food intake because feed efficient animals have more leptin and lower residual feed intake.

NMR relaxometry is one of the techniques that allow observing changes in the molecular mobility that come from materials’ morphology. T1H has been used to monitor food and polymer sciences. However, T2>H, although being a revealing as T1>H, is rarely used to analyze changes in thermoplastic systems it is more sensitive to the mobile region. High Impact Polystyrene nanomaterials were prepared through solution casting and were exposed for different times to UV light in the air. The samples, removed after each exposure interval, were characterized by T2>H, focusing on the changes in the relaxation data. The results for this parameter showed that the changes in the relaxation data come from the competition of chain scission and chain recombination processes, which occurs due to the UV light influence with increased time. The T2>H data indicated that the clay ratio can influence the chain degradation processes, acting to inhibit or accelerate the aging process [1] [2].

The concern of the ethics and the meaning of life in young adults became universal and are present in all contexts of human life. Such universality is due to the development of modern rationality, to establish a relationship between the intrinsic theoretical dimensions (scientific) and practical dimensions (ethical), originated that are present in the own array of knowledge. This descriptive cross-sectional study, aims to identify the values of the ethical dimension and meaning of life defended by the college students of the Universities of Madeira, Portugal and Fortaleza, Brazil. The sample consists of 605 University students (Madeira = 225 and Fortaleza = 380), with an average age of 21 years and a standard deviation of 2. We used the questionnaire on values (adapted from the European Values Survey). Most of the young people referred to have a religion (60% wood and 78.4% stronghold), being the most Catholic, however, present in your frequency disinterest and, yet, in Brazil there is a greater diversity of religions. Supporting the intervention of religious institutions in situations of everyday life is accepted mostly by Brazilians, with the exception of politics (35.5%). On the other hand, the university students of the University of Madeira understand that Religious Institutions should only give their opinion on racial discrimination (76.6%). Believe in God (90.8% and 55.1% Stronghold) and Soul (80.3% and 66.4% Stronghold) are beliefs more referred to by college students. These results are intended to contribute in the field of research on ethical values and meaning of life in young people. This study contributes to future comparative research among university students and to developing strategic programs in education and health, increasing the success and excellence in the formation of a full citizenship, preventing the fanaticisms and the social discrimination that today devastate the collective globally.

Candida parapsilosis is an increasingly important human pathogen. However, little is known about its potential to cause disease. The aims of the present study were to analyse the production of acid proteinases by clinical isolates of C. parapsilosis in the presence of different keratinous substrates from human sources (stratum corneum, nail and hair) and to verify the capability of yeast cells to adhere and grow as biofilm on these substrates. By scanning electron microscopy, it was observed that all C. parapsilosis sensu stricto isolates adhered to the keratinous substrates. For the isolate recovered from onychomycosis, the cell population attached to stratum corneum and hair keratin consisted mainly of blastoconidia. Differently, on nail keratin, pseudohyphae production was observed. Overall, there was a loose association between yeast cells and keratinous substrates. However, on stratum corneum, flocculent extracellular material was seen evolving cells from the onychomycosis isolate by forming a biofilm-like structure. The isolates recovered from onychomycosis and cutaneous lesion produced higher amount of acid proteinases in medium supplemented with nail keratin and stratum corneum keratin, respectively, than that in salt medium (absence of keratin). Furthermore, no differences were observed in the amount of acid proteinases produced by the isolate recovered from tracheal secretion in the media tested (absence and presence of keratin substrates). The information derived from this study will further our understanding of acid proteinase production by C. parapsilosis isolates and provide an insight into pathogenic mechanisms in C. parapsilosis particularly from isolates recovered from superficial mycoses.

Ectonucleotidases are enzymes involved in nucleotides metabolism. The amount of circulating nucleotides may modulate distinct pathophysiological processes in the vasculature, including platelet aggregation and vascular tone. Onion (Allium cepa L.) phenolic compounds modulate enzymatic activity. The present study evaluated the total phenolic content of red onion methanolic extract, its antioxidant capacity, and its ability to interfere in nucleotides hydrolyses in rat serum. Total phenolic content was determined with the Folin-Ciocalteau reagent using gallic acid as a standard, while total flavonoid content was obtained through the aluminum chloride colorimetric method with quercetin as a standard. Antioxidant capacity was evaluated from the ability of the extract to scavenge ABTS·+ and DPPH· radicals. ATP, ADP, AMP, and p-Nph-5'-TMP hydrolyses were colorimetrically determined in response to different onion extract concentrations (0, 125, 250, 500, or 1000 μg/mL). Phenolic content of the extract was 54.35 mg GAE 100 g-1 sample, while flavonoid content was 7.22 mg quercetin g-1 sample. The IC50 value for ABTS·+ was 374.13 ± 7.52 μg/mL, while it was 440.29 ± 15.17 μg/mL for DPPH·. Red onion extract increased ADP and p-Nph-5'-TMP hydrolysis. The results confirmed that red onion contains high content of antioxidant, mainly flavonoids, and high antioxidant capacity. Additionally, biochemical studies suggest that the increased ADP breakdown may be important to regulate vascular processes. As it occurs for other enzymes, the antioxidant capacity of onion extract may neutralize reactive oxygen species (ROS) formation and favor ectonucleotidase activity and the hydrolysis of ADP, a major platelet agonist.
