Intestinal anti-inflammatory activity of Ground Cherry(Physalis angulata L.)standardized CO2 phytopharmaceutical preparation

AIM To investigate the effects of Ground Cherry(Physalis angulata L.)standardized supercritical CO_2 extract in trinitrobenzenesulphonic acid(TNBS)model of rat intestinal *** The animals were divided into groups that received vehicle or *** extract(PACO_2)orally at the doses 25,50 and 100 mg/kg daily by 5 d before TNBS *** effects of PACO_2 were assessed by macroscopic analysis,biochemical determinations of the levels of myeloperoxidase(MPO),alkaline phosphatase(ALP),glutathione and cytokines(such as INF-γ,IL-1β,IL-6,IL-10 and TNF-α),gene expression evaluation(including Hsp70,heparanase,NF-κB,mitogenactivated protein kinases(Mapk)1,3,6 and 9,and the mucins genes Muc 1,2,3 and 4)and histopathological studies using optical,and electronic(transmission and scanning)*** PACO2 extract promoted a significant reduction in MPO and ALP activities,reducing oxidative stress and neutrophil *** effects were accompanied by significant reduction of colonic levels of IFN-γand IL-6 and down-regulation of heparanase,Hsp70,Mapk3,Mapk9,Muc1 and Muc2 genes expression when compared with TNBS-control *** addition,protective effects were also evidenced by reduced neutrophil infiltration,recovery of cell architecture and replacement of mucin by histopathological and ultrastructural *** Physalis angulata supercritical CO2 extract is an intestinal anti-inflammatory product that modulates oxidative stress,immune response and expression of inflammatory mediators,with potentially utility for treating inflammatory bowel disease.

Bile duct tumors are comprised of tumors that originate from both intrahepatic and extrahepatic bile ducts and gallbladder *** are aggressive tumors and chemotherapy is still the main treatment for advanced-stage disease and most of these cases have a poor overall *** are aimed at treatments with better outcomes and less toxicity which makes immunotherapy an area of significant *** Food and Drug Administration approvals of immune checkpoint inhibitors(ICI)for agnostic tumors based on biomarkers such as microsatellite instability-high and tumor mutation burden-high are important steps in the treatment of patients with advanced bile duct *** limited responses with isolated checkpoint inhibitors in later lines of systemic treatment in advanced disease,drug combination strategies have been demonstrating encouraging results to enhance ICI efficacy.

Leukocyte-technetium-99m uptake in Crohn's disease:Does it show subclinical disease?

AIM:To evaluate inflammatory activity in patients with Crohn's disease (CD) using technetium-99m-hexamethylpropyleneamine oxime (99mTc-HMPAO) granulocyte ***: Twenty patients (7 male and 13 female) with CD and five healthy volunteers were selected for 99mTc-HMPAO granulocyte scintigraphy. The Crohn's Disease Activity Index (CdaI), blood tests and C-reactive protein (CRP) of each patient were performed 7 d before the scintigraphic images. The leukocytes were labeled according to the International Society of Radiolabeled Blood Elements (ISORBE) consensus protocol and the scintigraphic images, including single photon emission computed tomography, were obtained 30 min and 2 h after injection of the radiolabeled ***:The labeling yield of the leukocytes with the lipophilic complex 99mTc-HMPAO was 55.0%±10%. Six of the 20 patients (30%) presented congruent results for the three parameters investigated (CdaI, Scintigraphic Index and CRP). On the other hand, 14 patients (70%) did not show congruent results. There was no significant correlation between the indices analyzed according to the Spearman test (P>0.05,n=20).CONCLUSION: The results suggest that 99mTc-HMPAO-labeled leukocyte scintigraphy could be important for determining inflammatory activity in CD even in the absence of clinical symptoms.

Physiological Maturity of Corn Seeds(Zea mays)by Tetrazolium Salt

The purpose of this study was to evaluate the relationship of tetrazolium salt testing to common indicators(milk line,black layer,dry matter)used to determine corn seed physiological *** seeds used were the hybrids Pioneer 4285 and Dow *** hybrids were harvested 40 days after polinization(daP),with 4-day intervals until 68 *** harvested seeds were evaluated for viability and vigor(germination test,accelerated aging test and cold test).The parameters used to determine the physiological seed’s maturity were the black layer,the milk line,the seed dry matter,seed water content and the evaluation of seed tissues using the tetrazolium salt to evaluate the endosperm *** germination of both materials was greater than 95%and there was no difference between harvest *** seed physiological maturity(PM)was identified at 56 daP for P4285(F2)and between 48 and 56 daP for 2B587(F2).The maximum vigor was detected by accelerated aging tests and cold tests before PM for the two *** activity of endosperm cells is related to the other PM indicators(milk line,black layer,dry matter and water content).The transport of nutrients from plant to the seed ceases at seed physiological *** of tetrazolium salt allows identifying cell death of the basal region and this parameter corresponds to the others evaluated,thus demonstrating that the tetrazolium salt is effective for corn seed physiological maturity characterization.

A Notation to Support Exploring Collaboration in Design Teams

Collaboration in large projects is a major challenge for contemporary architectural practice and this paper presents a notation for describing and analyzing how these collaborations can take *** on an extensive literature review some opportunities were found for the development of a notation that combined three particular aspects:network diagrams,Euler diagrams and a model from architectural *** exploratory case study was conducted based on the collaboration during a complex architectural project,which combined three approaches:document analysis,semi-structured interviews with key stakeholders and a comparison of the documents with results from the *** notation demonstrated to be suitable for two purposes:to improve the understanding of particular design events and as support material for presenting and exploring *** indicate that the notation was suitable,comprehensible and flexible,and it demonstrated good value when used in speculative ways,such as an intermediary design artifact that supports discussion and improvements in the collaboration process,which indicates promising future *** can also be argued that,similar to a sketch,the notation can also support the process of planning and“designing”the interaction between teams in design fields and even in other project-based organizations.

Modeling Assessment of Renewable Energy to Design Business Plans of Power Options Aimed Energy Planning

The objective of this paper is the evaluation of business opportunities attractiveness in RER （renewable-energy resources） within a specific geographic area, and using some concepts of integrated energy-resource planning. The proposed methodology includes the main figures of economic merit evaluated to each resource assessed, the sensitivity analysis on cash flow, and recommendations of investments to each energy option assessed. A study case is applied within the northwest region of Sao Paulo State （Brazil） for those renewable-energy options. Five options of renewable-energy resources were chosen to be evaluated： three ERs （energy resources） from supply side （small hydro, wind farms and sugarcane bagasse） and two ERs from demand side （replacement of incandescent lighting by fluorescent and replacement of electric showers by central heating systems）. The conclusion is that, only two assessed ERs that have received investment grade were sugarcane biomass and fluorescent lamps replacing incandescent lamps. The rest have restriction due different reasons listed.

A Brazilian experience of the self transglutaminase-based test for celiac disease case finding and diet monitoring

AIM： To evaluate the effectiveness of a rapid and easy fingertip whole blood point-of-care test for celiac disease （CD） case finding and diet monitoring. METHODS： Three hundred individuals, 206 females （68.7%） and 94 males （31.3%）, were submitted to a rapid and easy immunoglobulin-A-dass fingertip whole blood point-of-care test in the doctor＇s office in order to make immediate clinical decisions： 13 healthy controls, 6 with CD suspicion, 46 treated celiacs, 84 relatives of the celiac patients, 69 patients with dyspepsia, 64 with irritable bowel syndrome （IBS）, 8 with Crohn＇s disease and 9 with other causes of diarrhea. RESULTS： Upper gastrointestinal endoscopy with duodenal biopsies was performed in patients with CD suspicion and in individuals with positive test outcome： in 83.3% （5/6） of the patients with CD suspicion, in 100% of the patients that admitted gluten-free diet transgressions （6/6）, in 3.8% of first-degree relatives （3/79） and in 2.9% of patients with dyspepsia （2/69）. In all these individuals duodenal biopsies confirmed CD （Marsh＇s histological classification）. The studied test showed good correlation with serologic antibodies, endoscopic and histological ***： The point-of-care test was as reliable as conventional serological tests in detecting CD cases and in CD diet monitoring.
