Claudin-11 and occludin are major contributors to Sertoli cell tight junction function, in vitro

The Sertoli cell tight junction （T J） is the key component of the blood-testis barrier, where it sequesters developing germ cells undergoing spermatogenesis within the seminiferous tubules. Hormonally regulated claudin-11 is a critical transmembrane protein involved in barrier function and its murine knockout results in infertility. We aimed to assess quantitatively the significance of the contribution of claudin-11 to TJ function, in vitro, using siRNA-mediated gene silencing. We also conducted an analysis of the contribution of occludin, another intrinsic transmembrane protein of the TJ. Silencing of claudin-11 and/or occludin was conducted using siRNA in an immature rat Sertoli cell culture model. Transepithelial electrical resistance was used to assess quantitatively TJ function throughout the culture. Two days after siRNA treatment, cells were fixed for immunocytochemical localization of junction proteins or lyzed for RT-PCR assessment of mRNA expression. Silencing of claudin-11, occludin, or both resulted in significant decreases in TJ function of 55% （P 〈 0.01）, 51% （P 〈 0.01）, and 62% （P 〈 0.01）, respectively. Data were concomitant with significant decreases in mRNA expression and marked reductions in the localization of targeted proteins to the Sertoli cell TJ. We provide quantitative evidence that claudin-11 contributes significantly （P 〈 0.01） to Sertoli cell TJ function in vitro. Interestingly, occludin, which is hormonally regulated but not implicated in infertility until late adulthood, is also a significant （P 〈 0.01） contributor to barrier function. Our data are consistent with in vivo studies that clearly demonstrate a role for these proteins in maintaining normal TJ barrier structure and function.

Objective: Short historical description of the evolution of C1-C2 fixation methods, anatomical tricks and free hand fixation technique description. Method: five patients were operated on using free hand technique, X-rays were only used after the positioning of the patient in the ventral position with Mayfield and at the end before locking the lateral rods. Results: no complications documented, in all the cases CT scans revealed good screw position. Conclusion: Using these landmarks and with a good preoperative planning, it is easy for experienced surgeons to perform the technique in a free hand manner, that avoids X-ray exposure for the whole team and the patient. This technique is not recommended for common use, especially for surgeons with few experiences in this field.

Shallow landslide susceptibility assessment under future climate and land cover changes: A case study from southwest China

There is no doubt that land cover and climate changes have consequences on landslide activity,but it is still an open issue to assess and quantify their *** County in southwest China was selected as the test area to study rainfall-induced shallow landslide susceptibility under the future changes of land use and land cover(LULC)and *** used a high-resolution meteorological precipitation dataset and frequency distribution model to analyse the present extreme and antecedent rainfall conditions related to landslide *** future climate change factors were obtained from a 4-member multimodel ensemble that was derived from statistically downscaled regional climate *** future LULC maps were simulated by the land change modeller(LCM)integrated into IDRISI Selva software.A total of six scenarios were defined by considering the rainfall(antecedent conditions and extreme events)and LULC changes towards two time periods(mid and late XXI century).A physically-based model was used to assess landslide susceptibility under these different *** results showed that the magnitude of both antecedent effective recharge and event rainfall in the region will evidently increase in the *** the scenario with a return period of 100 years,the antecedent rainfall in summer will increase by up to 63%whereas the event rainfall will increase by up to 54%for the late 21st *** most considerable changes of LULC will be the increase of forest cover and the decrease of farming *** magnitude of this change can reach+22.1%(forest)and–9.2%(farmland)from 2010 until 2100,*** found that the negative impact of climate change on landslide susceptibility is greater than the stabilizing effect of LULC change,leading to an over decrease in stability over the study *** is one of the first studies across Asia to assess and quantify changes of regional landslide susceptibility under scenarios driven by LULC and climate ***

Effective transport width——A methodology to describe the spatial variability of bedload transport

The monitoring of bedload transport processes in rivers is a still challenging task in research and the parameter effective transport width(ETW) allows, for the first time, a process description of the local variability of the bedload transport. In this paper, the distribution of bedload transport over a river crosssection was studied by statistically analyzing long-term monitoring data. Using an innovative integrated measurement system, combining geophones distributed over the river width with bedload traps and basket sampling, allows for continuous, high-resolution, long-term monitoring data of bedload transport characteristics to be generated. Geophone data can be used to record and describe bedload transport processes such as intensity, temporal variability, and local variability. One parameter observed was the ETW, which describes the width where bedload transport is taking place for different discharge classes in gravel bed rivers. With the help of a data series recorded over more than a decade, it is now possible, for the first time, to statistically describe the continuously and directly measured ETW. Hence, it became evident that the effective bedload transport width depends on the discharge rate and local transport capacity, and that bedload transport over the entire river width only occurs at high flows that prevail a few days a year.

Sex-specific effects of inbreeding and early ife conditions on the adult oxidative balance

Inbreeding negatively affects various life-history traits, with inbred individuals typically having lower fit- ness than outbred individuals （=inbreeding depression）. Inbreeding depression is often emphasized under environmental stress, but the underlying mechanisms and potential long-lasting consequences of such inbreeding-environment interactions remain poorly understood. Here, we hypothesize that inbreeding-environment interactions that occur early in life have long-term physiological effects, in partic- ular on the adult oxidative balance. We applied a unique experimental design to manipulate early life conditions of inbred and outbred songbirds （Serinus canaria） that allowed us to separate prenatal and postnatal components of early life conditions and their respective importance in inbreeding-environment interactions. We measured a wide variety of markers of oxidative status in adulthood, resulting in a com- prehensive account for oxidative balance. Using a Bayesian approach with Markov chain Monte Carlo, we found clear sex-specific effects and we also found only in females small yet significant long-term effects of inbreeding-environment interactions on adult oxidative balance. Postnatal components of early life conditions were most persuasively reflected on adult oxidative balance, with inbred females that experienced disadvantageous postnatal conditions upregulating enzymatic antioxidants in adulthood. Our study provides some evidence that adult oxidative balance can reflect inbreeding-environment inter- actions early in life, but given the rather small effects that were limited to females, we conclude that oxida- tive stress miaht have a limited role as mechanism underlvina *** ***.q_
