Different patterns of intestinal response to injury after arterial,venous or arteriovenous occlusion in rats

AIM： To investigate the differences in injury patterns caused by arterial, venous or arteriovenous mesenteric occlusion. METHODS： Male Wistar rats were separated equally into four groups. Occlusion was performed by clamping the superior mesenteric artery （A）, the mesenteric vein （V） or both （AV） for 30 min, followed by 60 min of reperfusion. A control group received sham surgery only. Intestinal sections were examined for histological damage and serum tumor necrosis factor-α （TNF-α）, endothelin-1 （ET-1）, P-selectin, antithrombin Ⅲ （AT Ⅲ） and soluble intracellular adhesion molecule-1 （ICAM-1） concentrations were measured. RESULTS： All groups showed significant mucosal injury compared to controls. Furthermore, mucosal injury was significantly more severe in the V and AV groups compared to the A group （3.6 ± 0.55, 3.4 ± 0.55 and 2± 0.71, respectively, P = 0.01）. ICAM-1 was similarly elevated in all groups, with no significant differences between the groups. P-selectin levels were significantly elevated in the V and AV groups but not the A group （1.4 ± 0.5 ng/mL, 2.52 ± 0.9 ng/mL and 0.02± 0.01 ng/mL, respectively, P = 0.01） and ET-1 was significantly elevated in the A and V groups but not the AV group （0.32 ± 0.04 pg/mL, 0.36 ± 0.05 pg/mL and 0.29 ± 0.03 pg/mL, respectively, P = 0.01） compared to sham controls. ATⅢ levels were markedly depleted in the V and AV groups, but not in the A group （29.1 ± 5.2 pg/mL, 31.4 ±21.8 pg/mL and 55.8± 35.6 pg/mL, respectively, P = 0.01）, compared to controls. Serum TNF-α was significantly increased in all groups compared to sham controls （1.32 ± 0.87 ng/mL, 1.79 ± 0.20 ng/mL and 4.4 ± 0.69 ng/mL, for groups A, V and AV, respectively, P = 0.01）, with higher values in the AV group. CONCLUSION： Different patterns of response to ischemia/reperfusion are associated with venous, arterial or arteriovenous occlusion. Venous and arteriovenous occlusion was associated with the most severe alterations

Tailoring the partitioning of proteins using ionic liquids as adjuvants in polymer-polymer aqueous biphasic systems

Aqueous biphasic systems(ABS)are promising for proteins purification;however,when dealing with samples comprising several proteins,the selectivity towards a target protein is difficult to *** this work,the addition of ionic liquids(ILs)as adjuvants(5 wt%and 10 wt%)in ABS composed of polyethylene glycol(PEG)and dextran was investigated to tailor proteins partitioning between the coexisting *** liquid-liquid phase diagrams were determined at 298 K,followed by partition studies of three proteins(bovine serum albumin(BSA),immunoglobulin G(IgG),and cytochrome C(Cyt C)).Partition coefficients of IgG and BSA indicate the preference of both proteins to the dextran-rich phase,whereas Cyt C has no preferential partitioning between the *** addition of chloride-based ILs as adjuvants allows to increase or decrease the partition coefficients,thus tailoring the proteins partitioning between the *** partitioning essentially depends on the IL content in each phase,whereas Cyt C and IgG partitioning is ruled by the ILs chemical structure and established *** docking was carried out to address the ILs effect on the proteins partitioning,supporting experimental observations,while identifying the specific interactions *** partition of each protein in polymer-salt ABS with ILs as adjuvants was determined,demonstrating the higher tailoring ability of polymer-polymer ABS when adding ***,the partition of each protein in presence of the remaining ones was determined,as well as the selectivity of the studied systems to separate each pair of proteins,paving the way for their use in liquidliquid chromatography.

Intestinal barrier: A gentlemen's agreement between microbiota and immunity

Our body is colonized by more than a hundred trillion commensals, represented by viruses, bacteria and fungi. This complex interaction has shown that the microbiome system contributes to the host's adaptation to its environment, providing genes and functionality that give flexibility of diet and modulate the immune system in order not to reject these symbionts. In the intestine,specifically, the microbiota helps developing organ structures, participates of the metabolism of nutrients and induces immunity. Certain components of the microbiota have been shown to trigger inflammatory responses, whereas others, anti-inflammatory *** diversity and the composition of the microbiota,thus, play a key role in the maintenance of intestinal homeostasis and explain partially the link between intestinal microbiota changes and gut-related disorders in humans. Tight junction proteins are key molecules for determination of the paracellular permeability. In the context of intestinal inflammatory diseases, the intestinal barrier is compromised, and decreased expression and differential distribution of tight junction proteins is observed. It is still unclear what is the nature of the luminal or mucosal factors that affect the tight junctionproteins function, but the modulation of the immune cells found in the intestinal lamina propria is hypothesized as having a role in this modulation. In this review,we provide an overview of the current understanding of the interaction of the gut microbiota with the immune system in the development and maintenance of the intestinal barrier.

Botryosphaeria, Neofusicoccum, Neoscytalidium and Pseudofusicoccum species associated with mango in Brazil

Members of the Botryosphaeriaceae are well known fungi associated with dieback,canker and fruit rot on various hosts worldwide,including *** aim of this study was identify a large collection of Botryosphaeriaceae species associated with dieback and stem-end rot of mango in the semi-arid region of Northeastern Brazil,and compare the species in relation to mycelial growth,pathogenicity and virulence.A total of 115 isolates were sampled and based on morphology and DNA sequence data(ITS and EF1-α)seven taxa were identified,namely,Botryosphaeria dothidea,***,Fusicoccum fabicercianum,Neofusicoccum parvum,*** ***,Neoscytalidium dimidiatum and Pseudofusicoccum *** and *** were the most commonly isolated species,which represented 37%and 33%of all isolates *** is reported for the first time in association with mango diseases *** were significant differences among the species obtained in this study in relation to optimum temperature for mycelial growth and mycelial growth *** species were pathogenic on mango *** were significant differences in virulence among the species,with *** and *** being the most virulent species,while *** was the least virulent.

The emerging contaminants(ECs)are organic compounds including *** products are consumed in large quantities,favoring their continuous entrance to the environment and affecting the health of living *** is a non-steroidal anti-inflammatory drug with analgesic properties of prolonged *** is the commonest postoperative analgesic,and it is highly consumed without *** recent years,the capability of microalgae to remove pharmaceuticals in water has been tested as a promising *** this work,the removal efficiency of diclofenac(16µg/mL)by the microalga Nannochloropsis oculata CCAP 186/7 was *** major nutrient concentrations in Bold’s Basal Medium were modified(nitrogen and carbon:50 and 100%of their original concentration)to know their effect in the removal of *** degradation by light was also *** maximum removal capacity of diclofenac in the medium was 68.8%.The percentage of diclofenac adsorbed on the cell wall was between 6 and 12.7%.Around 23.6%of diclofenac was degraded by light after 18 *** did not show growth inhibition due to the presence of *** results obtained point to the promissory application of *** as a bioremediation system for diclofenac removal.

Genome-wide association studies(GWASs) have been instrumental in understanding complex phenotypic traits. However, they have rarely been used to understand lineage-specific pathways and functions that contribute to the trait. In this study, by integrating lineage-specific enhancers from mesenchymal and myeloid compartments with bone mineral density loci, we were able to segregate osteoblast-and osteoclast(OC)-specific functions. Specifically, in OCs, a PU.1-dependent transcription factor(TF)network was revealed. Deletion of PU.1 in OCs in mice resulted in severe osteopetrosis. Functional genomic analysis indicated PU.1 and MITF orchestrated a TF network essential for OC differentiation. Several of these TFs were regulated by cooperative binding of PU.1 with BRD4 to form superenhancers. Further, PU.1 is essential for conformational changes in the superenhancer region of Nfatc1. In summary, our study demonstrates that combining GWASs with genome-wide binding studies and model organisms could decipher lineage-specific pathways contributing to complex disease states.

Validation of a FFQ （Food Frequency Questionnaire） for Older People

Nutrition data for older adults is an area that has few studies mainly in Brazil. Due to the importance to know the behavior of this age group, the aim of this study was to develop and validate a frequency food questionnaire （FFQ） for older people in Brazil. It was considered a sample of 73 subjects from the city of Avar6, Sao Paulo, Brazil, and it was applied three 24-hour recalls and a FFQ developed for adults. Data from the three 24-hour recalls were transformed in nutrient intake as well as the data from FFQ. Statistical analysis was performed in order to get correlations between data from the mean of three 24-hour recalls and FFQ for some nutrients. It was found high correlations among them, concluding that the FFQ is valid to get nutrient intakes for older people.
