Loss of efficacy and safety of the switch from infliximab original to infliximab biosimilar(CT-P13)in patients with inflammatory bowel disease

BACKGROUND Infliximab original has changed the natural history of inflammatory bowel diseases(IBD) over the past two decades. However, the recent expiration of its patent has allowed the entry of the first Infliximab biosimilar into the European and Spanish markets. Currently switching drugs data in IBD are limited. AIM To compare the efficacy of infliximab biosimilar, CT-P13, against infliximab original, analyzing the loss of response of both at the 12 mo follow-up in patients with *** An observational study of two cohorts has been conducted. One retrospective cohort that included patients with IBD treated with Infliximab original, and a prospective cohort of patients who were switching from infliximab original to infliximab biosimilar(CT-P13). We had analyzed the overall efficacy and loss of efficacy in patients in remission at the end of one year after treatment with the original drug compared to the results of the year of treatment with the ***98 patients(CD 67, CU 31) were included in both cohorts. The overall efficacy for infliximab original per year of treatment was 71% vs 68.2% for infliximab biosimilar(P = 0.80). The loss of overall efficacy at 12 mo for infliximab original was 6.6% vs 14.5% for infliximab biosimilar(P = 0.806). The loss of efficacy in patients who were in basal remission was 16.3% for infliximab original vs 27.1% for infliximab biosimilar. Adverse events were 9.2% for infliximab original vs 11.2% for infliximab biosimilar. CONCLUSION The overall efficacy and loss of treatment response with infliximab biosimilar(CT-P13) is similar to that observed with infliximab original in patients who were switching at the 12 mo follow-up. There is no difference in the rate of adverse events.

Rapid emergence and a vigorous growth prevent the seed tubers from infections by soil microbes and allow a rapid interception of solar radiation. In this work, the effect of the potassium phosphites (KPhi) applied to seed tubers of two potato cultivars on crop emergence and early growth was studied. Two experiments were performed under greenhouse and field conditions. Emergence of plants, leaf area, dry matter and the number of primary stems were measured in both experiments. Furthermore, mycorrhizal colonization was also measured on roots under field conditions. The application of KPhi reduced the period between planting and emergence, and increased leaf area and dry matter. The ratio between dry matter of aerials and underground organs was not affected by KPhi. Indigenous mycorrhizal colonization increased after KPhi application to seed tubers. These results confirm the benefit of the application of KPhi to seed tubers on early plant growth and suggest that their application in crop production would be advantageous.

The Long Way from Complex Phenotypes to Genes: The Story of Rat Chromosome 4 and Its Behavioral Effects

Quantitative trait loci (QTLs) mapping has been performed during the past decades in an attempt to identify genes, gene products and mechanisms underlying numerous quantitative traits. It’s a strategy based on natural variations in genes and gene products, which facilitates translation from animal models to human clinical conditions. Our team has shown that the inbred rat strains Lewis (LEW) and Spontaneously Hypertensive Rats (SHR) differ with respect to several emotionality- related behaviors, one of which (inner locomotion in the open field) was strongly influenced by a QTL (Anxrr16) on rat chromosome 4. Since then, several other studies not only corroborated the initial description of Anxrr16, but also extrapolated its effects to a broader context (rats from both sexes and regardless of the estrous cycle phase) and suggested that this same region influences other emotionality-related behaviors as well as alcohol intake. Other QTLs affecting neurobiological traits were also found on rat chromosome 4 and several candidate genes have been pointed out as possibly influencing those phenotypes. Altogether, these studies suggest that rat chromosome 4 constitutes an interesting target for the study of the molecular bases of anxiety and other traits related to emotional reactivity.

Identification of IL11RA and MELK amplification in gastric cancer by comprehensive genomic profiling of gastric cancer cell lines

AIM To identify common copy number alterations on gastric cancer cell *** Four gastric cancer cell lines(ACP02, ACP03, AGP01 and PG100) underwent chromosomal comparative genome hybridization and array comparative genome hybridization. We also confirmed the results by fluorescence in situ hybridization analysis using the bacterial artificial chromosome clone and quantitative real time PCR *** The amplification of 9p13.3 was detected in all cell lines by both methodologies. An increase in the copy number of 9p13.3 was also confirmed by fluorescence in situ hybridization analysis. Moreover, the interleukin 11 receptor alpha(IL11RA) and maternal embryonic leucine zipper kinase(MELK) genes, which are present in the 9p13.3 amplicon, revealed gains of the MELK gene in all the cell lines studied. Additionally, a gain in the copy number of IL11 RA and MELK was observed in 19.1%(13/68) and 55.9%(38/68) of primary gastric adenocarcinoma samples, respectively. CONCLUSION The characterization of a small gain region at 9p13.3 in gastric cancer cell lines and primary gastric adenocarcinoma samples has revealed MELK as a candidate target gene that is possibly related to the development of gastric cancer.

Changes in the Composition and Abundance of Phytoplankton in a Coastal Lagoon of Baja California,Mexico,during 2011

A time series with weekly sampling was conducted from February 20, to December 16, 2011 at a station in the interior of the San Quintín Bay to estimate seasonal changes in the composition and abundance of phytoplankton. Water temperature was recorded and the upwelling index was calculated for the period. Phytoplankton abundance was estimated and phytoplankton were identified by using an inverted microscope and the CHEMTAX program. There were 16 positive phytoplankton anomalies during the year, but only three were considered to be blooms. The blooms were dominated by diatoms and were recorded in the winter, spring and summer. Different genera composed 80% of the total phytoplankton abundance of each of the blooms. The first bloom consisted of diatoms of the genera Pseudo-nitzschia sp. (15.7%), Skeletonema sp. (14.6%), Eucampia sp. (7%), and Navicula sp. (7%);a haptophyte of the genus Imantonia sp. (13.1%);and dinoflagellates of the genus Prorocentrum sp. (4.6%). The second bloom consisted mainly of diatoms of the genera Guinardia sp. (30.6%), Pseudo-nitzschia sp. (21.5%), Skeletonema sp. (14.5%), Chaetoceros sp. (8.4%) and Eucampia sp. (5.3%). The third bloom consisted of Chaetoceros sp. (46.3%), Pseudo-nitzschia sp. (22.6%), Skeletonema sp. (7.29%), and Imantonia sp. (6%). Dinoflagellates were observed in the winter, summer and autumn, but they contributed less biomass than diatoms. Prorocentrum sp., Gymnodinium sp., and Ceratium sp. were among the dinoflagellates that were observed. The differences in abundance and composition of phytoplankton groups in the blooms in San Quintín Bay during 2011 were due primarily to seasonal changes in the physical and chemical factors of the seawater and to upwelling events.

Decrease in the Hospital Stay of Neonates with Suspected Nosocomial Sepsis with the Use of a Molecular Biology Technique

Introduction: Diagnostic alternatives that allow microbial identification in blood, such as molecular biology;have shown utility in neonatal sepsis. Objective: To evaluate the impact of a Light Cycler SeptiFast test (LCSF) on days of hospital stay and the use of newborn (NB) antibiotics with suspected sepsis. Methods: We included NB with LCSF test and blood culture (cases) and NB with only blood culture (controls). We evaluated the days of stay, days of antimicrobial use and number of deaths. Statistic analysis: Descriptive statistics was used. The association between the use of LCSF and variables such as mortality, days of stay, use of antimicrobials, and use of mechanical ventilation was analyzed by odds ratio with 95% CI. Results: We included 20 subjects in the case group and 40 in the control group. 8 (40%) of the cases were positive vs 6 (15%) of the controls in the control group. In the controls, the average days of stay were 65 + 15 vs 35 + 10 days of the cases (p Conclusions: The LCSF test influences the days of hospital stay, but does not impact mortality or the use of antimicrobials.

Changes in the Abundance and Composition of Phytoplankton in a Coastal Lagoon during Neap-Spring Tide Conditions

The objective of this work was to estimate the changes in abundance and composition of phytoplankton in a coastal lagoon in Baja California, México during neap-spring tide conditions. Sampling was conducted from the 7th to the 16th of October 2004. Surface water was collected at 18 stations distributed across the bay during day time at high tide. Also, a time series was collected at a fixed station;surface water was collected every two hours from 8:00 to 18:00. High temperatures, low salinities and low nutrient concentrations at the oceanic end indicated weak or non upwelling conditions during this period. The phytoplankton community was characterized using an inverted microscope and the chemical taxonomy program CHEMTAX, based on pigment concentration estimated by high performance liquid chromatography (HPLC). The phytoplankton concentration was two times lower during this period than during periods of upwelling in the same year. Cryptophytes and diatoms were the most abundant groups estimated by CHEMTAX. Statistical analyses of the effect of tidal conditions on phytoplankton composition indicate that Zone A is strongly affected by tides, and that tidal effects are lessened at the inner zones. Differences in phytoplankton abundance between zones and between tidal conditions indicate that phytoplankton distribution is patchy in the lagoon.

Systematic Conservation Assessment for Most of the Colombian Territory as a Strategy for Effective Biodiversity Conservation

Colombian ecosystems maintain key ecological processes that support thousands of species, including human beings. With the expansion of the country’s population, and the implementation of a government’s development plan based on an economy centred on extraction patterns, the conservation of these ecosystems is at serious risk. It is a priority to implement effective strategies that ensure the protection of the country’s biological diversity as well as the mitigation and prevention of threats and to contribute to its proper use. Colombia’s development strategies as well as its peoples’ wellbeing depend on the suitable condition of its natural assets. The identification of surrogates of conservation, the formulation of conservation goals, the prioritization of key areas and the formulation of conservation strategies based on the preservation, restoration and sustainable use of the territory and its biodiversity are proposed for 60% of the emerged land (~ 700,000 Km2). This research aims at giving proper guidelines to manage the territory and finding common points between development and biodiversity conservation, as well as to use this input for the development and implementation of a National Decision-making Support System (DSS) that will potentially have an impact on Colombia’s environmental policies and territorial planning schemes.
