Intestinal anti-inflammatory effect of a plant sterol food supplement in experimental murine colitis and cell co-culture models

The potential of a plant sterol food supplement （PS-FS） in addressing intestinal inflammation in vivo and in vitro was evaluated. As in vivo model， C57BL/6J mice were exposed to 1.5% dextran sulfate sodium in three 5-day periods， with 10-day rest intervals in between， daily reciving PS-FS （35 mg PS/kg）. The in vitro approach involved a bi-cameral system with 8-day-differentiated Caco-2 （apical） and rAW264.7 cells （basolateral）. The bioaccessible fraction of PS-FS， obtained after INFOGeST 2.0 simulated gastrointestinal digestion， was added to the apical part （90 min）， followed by stimulation with lipopolysaccharide （1 μg/mL， 24 h）. PS-FS alleviated rectal bleeding and rebalanced pro- (tumor necrosis factor α， interleukin （IL）-6， and IL-8) and anti-inflammatory cytokines （IL-10）. Additionally， PS-FS ameliorated histopathological damage and enhancing occludin expression. A reduction in oxidative stress was evidenced by decreased myeloperoxidase activity and reactive oxygen species production. The anti-inflammatory mechanism included suppressing cyclooxygenase 2 expression， reducing prostaglandin e2production， and inhibiting the phosphorylation and nuclear translocation of the nuclear factor κB p65 subunit. This study reveals the potential of PS-FS as a therapeutic intervention for colitis. The alignment between in vivo and in vitro outcomes substantiates the appropriateness of the co-culture model to evaluate the anti-inflammatory activity of foods.

Olfactory ensheathing glia promote axonal regeneration in the mammalian central nervous system,including retinal ganglion cell axonal growth through the injured optic ***,it is unknown whether olfactory ensheathing glia also have neuroprotective *** ensheathing glia express brain-derived neurotrophic factor,one of the best neuroprotectants for axotomized retinal ganglion ***,we aimed to investigate the neuroprotective capacity of olfactory ensheating glia after optic nerve *** ensheathing glia cells from an established rat immortalized clonal cell line,TeG3,were intravitreally injected in intact and axotomized retinas in syngeneic and allogeneic mode with or without microglial inhibition or immunosuppressive *** and gene expression analyses were *** bulb-derived primary olfactory ensheathing glia and TeG3 express major histocompatibility complex classⅡ*** and syngenically transplanted TeG3 cells survived in the vitreous for up to 21 days,forming an *** axotomized retinas,only the allogeneic TeG3 transplant rescued retinal ganglion cells at 7 days but not at 21 *** these retinas,microglial anatomical activation was higher than after optic nerve crush *** intact retinas,both transplants activated microglial cells and caused retinal ganglion cell death at 21 days,a loss that was higher after allotransplantation,triggered by pyroptosis and partially rescued by microglial inhibition or ***,neuroprotection of axotomized retinal ganglion cells did not improve with these *** different neuroprotective properties,different toxic effects,and different responses to microglial inhibitory treatments of olfactory ensheathing glia in the retina depending on the type of transplant highlight the importance of thorough preclinical studies to explore these variables.

Plume-lithosphere interaction in the Comei Large Igneous Province: evidence from two types of mafic dykes in Gyangze, south Tibet, China

Two suites of mafic dykes,T1193-A and T1194-A,outcrop in Gyangze area,southeast *** are in the area of Comei LIP and have indistinguishable field occurrences with two other dykes in Gyangze,T0902 dyke with 137.7±1.3 Ma zircon age and T0907 dyke with 142±1.4 Ma zircon age reported by Wang YY et al.(2016),indicating coeval formation *** all the four diabase dykes into consideration,two different types,OIB-type and weak enriched-type,can be ***“OIB-type”samples,including T1193-A and T0907 dykes,show OIB-like geochemical features and have initial Sr-Nd isotopic values similar with most mafic products in Comei Large Igneous Provinces(LIP),suggesting that they represent melts directly generated from the Kerguelen mantle ***“weak enriched-type”samples,including T1194-A and T0902 dykes,have rees and trace element patterns showing withinplate affinity but have obvious Nb-Ta-Ti negative *** show uniform lowerεNd(t)values(−6‒−2)and higher 87Sr/86Sr(t)values(0.706‒0.709)independent of their MgO variation,indicating one enriched mantle *** their closely spatial and temporal relationship with the widespread Comei LIP magmatic products in Tethyan Himalaya,these“weak enriched-type”samples are consistent with mixing of melts from mantle plume and the above ancient Tethyan Himalaya subcontinental lithospheric mantle(SCLM)in different *** weak enriched mafic rocks in Comei LIP form one special rock group and most likely suggest large scale hot mantle plume-continental lithosphere *** process may lead to strong modification of the Tethyan Himalaya lithosphere in the early Cretaceous.

Zn metal anode suffers from dendrite issues and passive byproducts,which severely plagues the practical application of aqueous Zn metal ***,a polyzwitterionic cross-linked double network hydrogel electrolyte composed of physical crosslinking(hyaluronic acid)and chemical crosslinking(synthetic zwitterionic monomer copolymerized with acrylamide)is introduced to overcome these *** the one hand,highly hydrophilic physical network provides an energy dissipation channel to buffer stress and builds a H_(2)O-poor interface to avoid side *** the other hand,the charged groups(sulfonic and imidazolyl)in chemical crosslinking structure build anion/cation transport channels to boost ions’kinetics migration and regulate the typical solvent structure[Zn(H_(2)O)_(6)]^(2+)to r-SO_(3)^(−)[Zn(H_(2)O)_(4)]^(2+),with uniform electric field distribution and significant resistance to dendrites and parasitic *** on the above functions,the symmetric zinc cell exhibits superior cycle stability for more than 420 h at a high current density of 5 mA·cm^(−2),and Zn||MnO_(2)full cell has a reversible specific capacity of 150 mAh·g^(−1)after 1000 cycles at 2 C with this hydrogel ***,the pouch cell delivers impressive flexibility and cyclability for energy-storage applications.

BACKGrOUND In patients with metastatic colorectal cancer(mCrC),the treatment options are limited and have been proved to be affected by rat sarcoma virus(rAS)mutational *** rAS wild-type(wt)patients,the combination of antiepidermal growth factor receptor(eGFr)monoclonal antibodies with chemotherapy(CT)is more effective than CT *** the other hand,rAS-mutated patients are not eligible for treatment with anti-eGFr *** SUMmarY eleven patients with initially rAS-mutated mCrC were followed from diagnosis to May *** the time of cell-free DNA determination,five patients had undergone one CT line,five patients had undergone two CT lines,and one patient had undergone three CT lines(all in combination with bevacizumab).At the second and third treatment lines[second line(2L),third line(3L)],patients with neo-rAS wt received a combination of CT and *** neo-rAS wt patients treated with anti-eGFr,our findings indicated an increase in progression-free survival for both 2L and 3L(14.5 mo,P=0.119 and 3.9 mo,P=0.882,respectively).regarding 2L overall survival,we registered a slight increase in neo-rAS wt patients treated with anti-eGFr(33.6 mo vs 32.4 mo,P=0.385).At data cut-off,two patients were still alive:A rAS-mutated patient undergoing 3L treatment and a neo-rAS wt patient who received 2L treatment with anti-eGFr(ongoing).CONCLUSION Our case series demonstrated that monitoring rAS mutations in mCrC by liquid biopsy may provide an additional treatment line for neo-rAS wt patients.

Background:One of the pathological hallmarks distinguishing Alzheimer’s disease from other dementias is the accumulation of amyloid beta(Aβ).Higher physical activity is associated with decreased dementia risk,and one potential path could be through Aβlevels *** aimed to explore the relationship between physical activity and Aβin middle-aged and older ***:A systematic search of PubMed,Web of Science,PsycINFO,Cochrane Central register of Controlled Trials,and SPOrTDiscus was performed from inception to April 28,*** were eligible if they included physical activity and Aβdata in adults aged 45 years or ***-level metaanalyses of intervention and observational studies were performed to examine the role of physical activity in modulating Aβ***:In total,37 articles were included(8 randomized controlled trials,3 non-randomized controlled trials,4 prospective longitudinal studies,and 22 cross-sectional studies).The overall effect size of physical activity interventions on changes in blood Aβwas medium(pooled standardized mean difference=-0.69,95%confidence interval(95%CI):-1.41 to 0.03;I^(2)=74.6%).However,these results were not statistically significant,and there were not enough studies to explore the effects of physical activity on cerebrospinal fluid(CSF)and brain Aβ.Data from observational studies were examined based on measurements of Aβin the brain using positron emission tomography scans,CSF,and *** physical activity was positively associated with Aβonly in the CSF(estimate r=0.12;95%CI:0.05-0.18;I^(2)=38.00%).Conclusion:Physical activity might moderately reduce blood Aβin middle-aged and older ***,results were only near statistical significance and might be interpreted with caution given the methodological limitations observed in some of the included *** observational studies,higher levels of physical activity were positively associated with Aβonly in ***,further research is needed t

Control of dust in underground coal mines is critical for mitigating both safety and health *** decades,the National Institute of Occupational Safety and Health(NIOSH)has led research to evaluate the effectiveness of various dust control technologies in coal *** studies have included the evaluation of auxiliary scrubbers to reduce respirable dust downstream of active mining and the use of canopy air curtains(CACs)to reduce respirable dust in key operator *** detailed dust characterization was not a focus of such studies,this is a growing area of *** preserved filter samples from three previous NIOSH studies,the current work aims to explore the effect of two different scrubbers(one wet and one dry)and a roof bolter CAC on respirable dust composition and particle size *** this,the preserved filter samples were analyzed by thermogravimetric analysis and/or scanning electron microscopy with energy dispersive *** indicate that dust composition was not appreciably affected by either scrubber or the ***,the wet scrubber and CAC appeared to decrease the overall particle size *** an effect of the dry scrubber was not consistently observed,but this is probably related to the particular sampling location downstream of the scrubber which allowed for significant mixing of the scrubber exhaust and other return *** from the insights gained with respect to the three specific dust control case studies revisited here,this work demonstrates the value of preserved dust samples for follow-up investigation more broadly.
