Transcriptomic responses to drought stress in the Patagonian southern beech Nothofagus alpina

Background Deciphering the genetic architecture of drought tolerance could allow the candidate genes identification responding to water stress. In the Andean Patagonian forest, the genus Nothofagus represents an ecologically relevant species to be included in different genomic studies. These studies are scarce in South American ecosystems however represent an important source of genomic data in order to interpret future climate-change environment scenarios of these emblematic forests. Here, we achieved the assemblage of the transcriptome of N. alpina while searching for key genes of activated or suppressed metabolic pathways in response to drought *** De novo transcriptome assembly resulted in 104,030 transcripts. Following confirmation of drought conditions, based on reduction of leaf water potential and stomatal conductance, a differential gene expression analysis resulted in 2720 significantly expressed genes(1601 up-regulated and 1119 down-regulated). Enrichment analysis(over-representation analysis and gene set enrichment analysis) resulted in more than one hundred stress-responsive term ontologies(i.e. biological processes) and pathways. Terms such as response to abscisic acid and pathways such as plant hormone signal transduction or starch and sucrose metabolism were over-represented. Protein–protein interaction assessment resulted in networks with significantly expressed top common hub gene clusters(e.g. planttype cell wall biogenesis among down-regulated or ABA-signalling among up-regulated). Black soils, characterized by their thick, dark horizons enriched with organic matter, epitomize highly fertile soils. However, their fertility precipitates intense land use, engendering challenges such as soil erosion, nutrient depletion, pollution, compaction, salinization, and acidification. Notably, these soils are significant contributors to global greenhouse gas emissions, primarily due to substantial losses in soil organic carbon. Despite these challenges, black soils are pivotal for global food production. This paper delineates the implementation of digital soil mapping for the global cartography of black soils and human interference on these soils. Predominantly distributed in Eastern Europe, Central and Eastern Asia, and North and South America, black soils cover an approximate area of 725 Mha, with the Russian Federation,Kazakhstan, and China collectively have over half of this area. Agriculturally,these soils underpin significant proportions of global crop yields, producing 66% of sunflower, 30% of wheat, and 26% of potato outputs. The organic carbon content in the upper 30 cm of these soils is estimated at 56 *** management of black soils is imperative for ensuring food security and addressing climate change on a global scale.

Two intestinal microbiota-derived metabolites, deoxycholic acid and butyrate, synergize to enhance host defense peptide synthesis and alleviate necrotic enteritis

Modelling of the variation of granular base materials resilient modulus with material characteristics and humidity conditions

This study aims to quantify the susceptibility of granular materials used in pavements to changes in moisture content and propose a correlation model to incorporate this susceptibility into seasonal *** fines content and the percentage of fractured coarse aggregates were identified as direct indicators of the resilient modulus susceptibility to changes in water *** results showed that the percentage of fractured coarse aggregates particles(FR)has a more significant impact on the resilient modulus(Er)of crushed granular materials used in pavement construction than the combined indicator of the fines content and sample volumetrics(nf).Crushed granular materials with a higher percentage of fractured coarse aggregates are relatively insensitive to changes in the degree of saturation,but become more sensitive as the fine fraction porosity *** adjusted model was proposed based on the existing formulation,but considers a complex parameter to describe and adjust the sensitivity of base granular materials to variations in moisture content with respect to fabrication charac-teristics,fines content and volumetric *** model shows that the variation of Er values is below10%for fully crushed granular ***,it reaches approximately±12%for materials with 75%of crushed coarse aggregates andþ40%and-25%for materials with FR=50%.This model could help select good ag-gregates characteristics and adjust grain-size distribution for environments where significant moisture content variations can occur in the pavement system,such as in the Province of Quebec(Canada).As it is based on pa-rameters that can be easily determined or estimated,it also represents a valuable tool for detailed design and analysis that can consider material characteristics.

Implementation of a New Solution for the Preservation of Anatomical Specimens Made of Non-Toxic Substances

The preservation of anatomical pieces in Veterinary Anatomy is essential since it is not possible to dissect all domestic species. Most techniques use reagents with high levels of toxicity such as formaldehyde. The objective of this work was to develop a new preservation technique that uses reagents with zero toxicity and that allows obtaining preserved pieces suitable for anatomical studies. The alcohol propylene glycol technique was developed, the method of which uses a fixation step with alcohol, sodium chloride, commercial vinegar and subsequently the impregnation of the preservation solution made from propylene glycol and commercial vinegar, which are non-toxic. As a result of this work, adequately preserved sheep hearts were obtained that preserved their morphology with slight changes in size and weight, which did not affect their external and internal anatomical structure. Its coloration was not substantially affected, remaining a little lighter. The pieces obtained showed flexibility which allowed dissections to be carried out. The time to develop the technique was 20 days. A comparative study was carried out with the phenolated glycerin technique that uses toxic reagents (formaldehyde and phenol) and the pieces obtained with the alcohol propylene glycol technique were of better quality, observing that the pieces with phenolated glycerin tend to darken and are more rigid. And the time to develop the technique is 24 days. In conclusion, a preservation technique for anatomical pieces was developed that allowed the preservation of the organs under study, which allow their use for anatomical studies, and which have been preserved without changes until the time of this publication (8 months) and there are pieces preserved with this technique for 2 years.

Carbon stock estimation in halophytic wooded savannas of Uruguay:An ecosystem approach

Savannas constitute a mixture of trees and shrub patches with a more continuous herbaceous *** contribution of this biome to the soil organic carbon(SOC)and above-ground biomass(AGB)carbon(C)stock globally is ***,they are frequently subjected to land use changes,promoting increases in CO_(2) *** Uruguay,subtropical wooded savannas cover around 100,000 ha,of which approximately 28%is circumscribed to sodic soils(i.e.,subtropical halophytic wooded savannas).Nevertheless,there is little background about the contribution of each ecosystem component to the C stock as well as site-specific allometric *** study was conducted in 5 ha of subtropical halophytic wooded savannas of the national protected area Esteros y Algarrobales del Rio *** work aimed to estimate the contribution of the main ecosystem components(e.g.,soil,trees,shrubs,and herbaceous plants)to the C ***-specific allometric equations for the most frequent tree species and shrub genus were fitted based on basal diameter(BD)and total height(H).The fitted equations accounted for between 77%and 98%of the aerial biomass variance of Netuma affinis and Vachellia *** shrubs(Baccharis sp.),the adjusted equation accounted for 86%of total aerial biomass.C stock for the entire system was 116.71±11.07 Mg·ha^(-1),of which 90.7%was allocated in the soil,8.3%in the trees,0.8%in the herbaceous plants,and 0.2%in the *** results highlight the importance of subtropical halophytic wooded savannas as C sinks and their relevance in the mitigation of global warming under a climate change scenario.
