Role of 9-Lipoxygenase and α-Dioxygenase Oxylipin Pathways as Modulators of Local and Systemic Defense

Plant 9-1ipoxygenases （9-LOX） and α-dioxygenases （α-DOX） initiate the synthesis of oxylipins after bacterial infection. Here, the role of these enzymes in plants＇ defense was investigated using individual Arabidopsis thaliana Ioxl and doxl mutants and a double Ioxl doxl mutant. Studies with Pseudomonas syringae pv. tomato （Pst） revealed the enhanced susceptibility of Ioxl to the virulent strain Pst DC3000 and the partial impairment of Ioxl and doxl mutants to activate systemic acquired resistance. Notably, both defects were enhanced in the Ioxl doxl plants as compared with individual mutants. We found that pre-treatment with 9-LOX- and α-DOX-generated oxylipins protected plant tissues against bacterial infection. The strongest effect in this respect was exerted by 9-ketooctadecatrienoic acid （9-KOT）, which is produced from linolenic acid by 9-LOX. Quantification of 9-KOT revealed its accumulation after bacterial infection. The levels were reduced in Ioxl and Ioxl doxl plants but strongly increased in the doxl mutant due to metabolic interaction of the two pathways. Transcriptional analyses indicated that 9-KOT pre-treatment modifies hormone homeostasis during bacterial infection. The nature of the changes detected suggested that 9-KOT interferes with the hormonal changes caused by bacterial effectors. This notion was substantiated by the finding that 9-KOT failed to reduce the growth of PstDC3OOOhrpA, a mutant compromised in effector secretion, and of the avirulent strain Pst DC3000 avrRpml. Further support for the action of the 9-LOX- and α-DOX-oxylipin pathways as modulators of hormone homeostasis was the ob- servation that Ioxl doxl seedlings are hypersensitive to the growth-inhibitory effect of ABA and showed enhanced ac- tivation of ABA-inducible marker genes as compared with wild-type plants.

An Ecological Alternative for the Control of Gastrointestinal Nematodes in Sheep in Northern Veracruz （Mexico）

The objective was to evaluate the effect of aqueous infusion of cracked soursop seeds on two different concentrations as an organic dewormer compared to a commercial one in GNE （gastrointestinal nematodes） egg population in crossbred hair sheep （Blackbelly-Khatadin） of the northen region of Veracruz. The aqueous infusion was prepared adding 93.5 g of cracked soursop seeds in 1,875 mL of boiled water and let stand for 12 h. Crossbred ewes were randomly assigned to receive every 19 days：（1） 10 mL of cracked soursop infusion orally （n = 27）, （2） 15 mL of cracked soursop infusion orally （n = 27）, and （3） 2 mL of Febendazole subcutaneously （n = 26）. Feces （2-5 g） were collected directly from the rectum of each animal on the following days： 0, 19, 38 and 57 days post treatment. Nematode egg population was determined using the technique of McMaster. Statistical analysis was done using ANOVA （analysis of variance）. There was a reduction overtime （P = 0.05） in all treatments in parasite egg population. There were no differences （P = 0.10） in egg population across treatments. In conclusion, aqueous infusion of soursop cracked seeds proved to be an environmentally friendly and effective alternative in the control of gastrointestinal parasites in crossbred hair sheep in the region of northern Veracruz.

The photoreceptor layer of the human retina has several characteristics that are unique. Their energy requirements are the highest in the organism;in proportion, rods and cones require 10-fold the energy consumed by the cerebral cortex, 6-fold more than the cardiac muscle, and 3-fold more than the renal cortex. Astonishingly, the photoreceptor layer has no blood vessels at all. So, where is the energy to this tissue coming from? In this article we’ll describe the hitherto unknown explanation.

The biological properties of therapeutic peptides,such as their pharmacokinetics and pharmacodynamics,are correlated with their structure and aggregation ***,we studied the aggregation properties of a therapeutic peptide(CIGB-814),currently in phase 2 clinical trial,for the treatment of rheumatoid arthritis over a wide range of concentrations(μM-mM).We applied spectroscopic techniques(fluorescence,circular dichro-ism,resonance,and dynamic light scattering),atomic force microscopy,and molecular dynamics simulations to determine the aggregation mechanism of *** found that the hierarchical aggregation of CIGB-814 at micromolar concentrations was initiated by the formation of peptide ***,the peptide oligomers trigger the nucleation and growth of peptide nanostructures(cac=123μM),ultimately leading to the fibrillization of CIGB-814(cac’=508μM).These results pave the way for a deeper understanding of the CIGB-814 therapeutic activity and may give important insights on its pharmacokinetics.

Superior mesenteric artery syndrome in a patient with Charcot marie Tooth disease

The extrinsic compression of the third part of the duodenum as it passes through the aorto-mesenteric angle is known as the superior mesenteric artery syndrome(SMAS).This syndrome is a rare mechanical cause of upper intestinal obstruction,with a reported incidence of between 0.2% and 0.78%.Clinical manifestations of SMAS may be chronic or acute;chronic symptoms include intermittent gastric pain,fullness and occasional episodes of postprandial vomiting,while acute symptoms include incoercible vomiting,oral intolerance,mainly epigastric abdominal distension and abdominal *** is recommended only when initial conservative treatment ***,we report what appears to be the third published case of SMAS associated with hereditary motor and sensory neuropathy or Charcot marie Tooth disease.

Many of the various factors,characteristics,and variables involved in the addictive process can determine an individual’s vulnerability to develop drug *** eating,based on pleasure rather than energy needs,modulates the same reward circuits,as do drugs of *** to the last report of the World Health Organization,the worldwide obesity rate has more than doubled since 1980,reaching especially critical levels in children and young people,who are overexposed to high-fat,high-sugar,energy-dense *** the past few decades,there has been an increase in the number of studies focused on how eating disorders can lead to the development of drug addiction and on the comorbidity that exists between the two ***,we review the most recent research on the subject,focusing especially on animal models of binge eating disorders and drug *** complex profile of patients with substance use and binge eating disorders requires an integrated response to dually diagnosed *** patterns should be considered an important variable in the treatment of substance use disorders,and future studies need to focus on specific treatments and interventions in individuals who show a special vulnerability to shift from one addiction to the other.

Mirodenafil is a 5-phosphodiesterase inhibitor that is currently marketed in Korea for the treatment of erectile dysfunction;however, no information in other populations is available. It has been described that Mirodenafil is metabolized by CYP3A4, a metabolic pathway in which interethnic differences have been reported. The purpose of this study was to characterize the oral pharmacokinetics of Mirodenafil in Mexicans. Seventeen male healthy volunteers were enrolled in this study. After an overnight fast, volunteers received an oral 100 mg dose and blood samples were collected at selected times during 24 h. Plasma was stored frozen and analyzed by an HPLC method. Pharmacokinetic parameters obtained were: Cmax 331.129 ± 32.689 ng/mL, tmax 1.574 ± 0.293 h, AUC24h 883.293 ± 104.088 ng·h/mL, AUC￥ 976.477 ± 108.812 ng·h/mL and t1/2 1.807 ± 0.171 h. Parameter values observed in this study are similar to those reported in Koreans. Since efficacy and safety studies of Mirodenafil have been conducted in Koreans, it is expected that dosage regime to employ in Mexicans should be similar to the approved for Korean population.

Native Trees Forage Alternative Protein Feed for Cattle in Tropical ＂Mexico＂

The objective of this study was to find highly nutritious native plants to feed cattle in northern Veracruz （Mexico） as an alternative to avoid deforestation for pasture establishment. Six fodder tree species （Leucaena, Leucaenaleucocephala, Morera, Morus alba, Chacloco, rubiHamelia patents, Guficima, Guasumaulmifolia, Pichoco-bunting, Erythrinaamericana, Cocuite, Gliricidiasepium） were selected for their leave production, rapid growth and high nutritional quality. The plants were evaluated in the bud stage of senescence and flowering, restricting the fraction of mature foliage in the samples located under 2 meters height. Samples were collected from five plants per specie that were randomly selected. The nutritious parameters were evaluated through proximate analysis with the Van Soest technique. This study used a completely randomized design with five replicates. The nutritious composition showed PC （protein content） differences （P 〈 0.05） among species indicating that Leucaenaleucocephala （20%） had the lowest, Gliricidiasepium was （21%） intermediate and Morus alba （23%） had the highest. The FDN （neutral detergent fiber） and FDA （acid detergent） were lowest for Guasumaulmifolia （23.4%） and the other five species exceeded 40%. In conclusion, native foliage of fodder trees in northern, Veracruz are highly nutritious for cattle feeding and could alternative to avoid deforestation for pasture establishment to continue.
