Apolipoprotein M(apoM) participates in both high-density lipoprotein and cholesterol metabolism. Little is known about how apo M affects lipid composition of the liver and serum. In this study, we systemically investigated the effects of apo M on liver and plasma lipidomes and how apo M participates in lipid cycling, via apo M knockout in mice and the human SMMC-7721 cell line. We used integrated mass spectrometry-based lipidomics approaches to semiquantify more than 600 lipid species from various lipid classes, which include free fatty acids, glycerolipids, phospholipids, sphingolipids, glycosphingolipids, cholesterol, and cholesteryl esters(CEs), in apo M^(-/-)mouse. Hepatic accumulation of neutral lipids, including CEs, triacylglycerols, and diacylglycerols, was observed in apo M^(-/-)mice;while serum lipidomic analyses showed that, in contrast to the liver, the overall levels of CEs and saturated/monounsaturated fatty acids were markedly diminished. Furthermore, the level of Apo B-100 was dramatically increased in the liver, whereas significant reductions in both Apo B-100 and low-density lipoprotein(LDL) cholesterol were observed in the serum of apo M^(-/-)mice, which indicated attenuated hepatic LDL secretion into the circulation. Lipid profiles and proinflammatory cytokine levels indicated that apo M^(-/-)leads to hepatic steatosis and an overall state of metabolic distress. Taken together, these results revealed that apo M knockout leads to hepatic steatosis, impaired lipid secretion, and an overall state of metabolic distress.

Effect Evaluation of CBL Combined with Rain Classroom Teaching Method in Medical Statistics

Objective: To explore the application effect of CBL combined with rain classroom teaching method in medical statistics courses. Methods: The undergraduate students of medical imaging technology in 2019 and 2020 in a university were selected as the research objects. A cluster sampling method was used to select 79 undergraduate students from 2019 in the control group and 75 undergraduate students from 2020 in the experimental group. Traditional teaching method and CBL combined with rain classroom teaching method was used in the control group and experimental group respectively. The final examination scores of the two groups were compared. In experimental group, the correlation between the average score in the rain classroom and the final examination score was tested, and the teaching effect was evaluated. Results: The average score of final examination in experimental group and control group was 79.13 ± 10.32 points and 71.54 ± 14.752 points, respectively, which had a statistically significant difference (Z = 2.586, P = 0.012);the final examination scores of the students in the experimental group were positively correlated with the average scores of the rain classroom (r = 0.372, P = 0.001), and the proportion of satisfaction in the experimental group was 94.7%. Conclusion: The CBL combined with rain classroom teaching method can improve the teaching effectiveness of medical statistics courses.

An iterative approach to enhance the clinical learning experience in Macao nursing education

Objective: The aim of this study was to reduce or eliminate the stressors to enhance nursing students' clinical learning experience through implementation of an iterative approach that developed a personalised response to student ***: A prospective cohort research design was applied to collect data from a sample of undergraduate nursing students across the four study *** iterative approach was employed to improve students' learning experience and the Stressors in Nursing Students Scale-Chinese Version (SINS-CN) was used to measure student *** problems encountered by students were identified,refined and the responding solutions were worked out and implemented among this group of students through their first year to fourth ***: The overall SINS-CN mean score (2.17-2.82) of students was declined to a moderate ***: Having implemented iterative approach to address factors that led to stressful environments encountered by the nursing students,the overall stress score and each sub-dimension score decreased ***,it is recommended that this approach could be adopted by other colleagues in the nursing arena around the world.

FSAP aggravated endothelial dysfunction and neurological deficits in acute ischemic stroke due to large vessel occlusion

Revascularization and angiogenesis,as substrates of sustained collateral circulation,play a crucial role in determining the severity and clinical outcome of acute ischemic stroke(AIS)due to large vessel occlusion(LVO).Developing an adjunct biomarker to help identify and monitor collateral status would aid stroke diagnosis and *** screen the potential biomarkers,proteomic analysis was performed in this study to identify those distinct plasma protein profiles in AIS due to LVO with different collateral ***,we found that levels of Plasma Factor VII Activating Protease(FSAP)significantly increased in those AIS patients with poor collaterals,and were correlated with worse neurological ***,both in vitro and in vivo models of ischemic stroke were used to explore pathological mechanisms of FSAP in endothelial *** demonstrated that the FSAP inhibitor,high-molecular-weight hyaluronan(HMW-HA),enhanced the pro-angiogenic vascular factors,improved the integrity of brain blood barrier,and promoted newly formed cerebral microvessels in the ischemic penumbra,consequently improving neurological *** elucidate the pathways that might contribute to revascularization during LVO,we applied transcriptomic analysis via unbiased RNA sequencing and showed that Wnt signaling was highly involved in FSAP mediated endothelial ***,inhibition of Wnt5a largely reversed the protective effects from HMW-HA treatment,implying that FSAP might aggravate endothelial dysfunction and neurological deficits by regulating Wnt5a ***,FSAP may represent a potential biomarker for collateral status after LVO and a promising therapeutic target to be explored in the treatment of stroke.

Characteristics of low altitude ionospheric electric field over Hainan Island,China

A sounding rocket experiment undertaken by the Chinese Meridian Project from a low latitude station on Hainan Island(19.5°N,109.1°E),China,measured the DC electric field during 05:45-05:52 LT on April 5,*** data observed using a set of electric field double probes,as part of the rocket's scientific payload,revealed the special profile of how the vectors of the DC electric field vary with altitude between 130 and 190 *** the experiment,the vertical electric field was downward,and the maximum vertical electric field was nearly 5.1 mV/m near the altitude of 176 *** zonal electric field was eastward and slightly less than 0.6 mV/*** plasma drift velocity was estimated from the E×B motion,and the zonal drift velocity was eastward and of the order of 100 m/*** zonal wind velocity was also estimated using the drift velocity near the maximum density height in the F1-region,and it was found to be nearly 120 m/*** work constituted the first in situ measurement of the DC electric field conducted within the Fl-region(between 130 and 190 km) in the East Asian Sector.

PuMYB21/PuMYB54 coordinate to activate PuPLDβ1 transcription during peel browning of cold-stored“Nanguo”pears

Refrigeration is commonly used to extend the storage life of“Nanguo”pears,but fruit in long-term refrigeration is prone to peel browning,which is related to membrane lipid *** determine the mechanism of membrane lipid degradation,we identified two R2R3-MYB transcription factors(TFs),PuMYB21 and PuMYB54,from“Nanguo”pears,which were notably expressed in response to cold stress and during the peel-browning *** results from yeast one-hybrid,electrophoretic mobility shift,and transient expression assays indicated that both PuMYB21 and PuMYB54 directly bind to the promoter of PuPLDβ1(a key enzyme catalyzing the hydrolysis of membrane phospholipids)and activate its expression,which probably enhances the degradation of membrane phospholipids and eventually results in peel ***,the overexpression of PuMYB21 and PuMYB54 can greatly activate the transcription of endogenous PuPLDβ1 in both“Nanguo”pear fruits and calli,and their silencing can inhibit its ***,yeast two-hybrid,bimolecular fluorescence complementation,and pull-down assays verified that PuMYB21 interacts with PuMYB54 to enhance the expression of PuPLDβ*** summary,we demonstrate that PuMYB21 and PuMYB54 may have roles in membrane lipid metabolism by directly binding to the downstream structural gene PuPLDβ1 during the low temperature-induced peel browning of“Nanguo”pears.

Plexiform angiomyxoid myofibroblastic tumor treated by endoscopic submucosal dissection:A case report and review of the literature

BACKGROUND Plexiform angiomyxoid myofibroblastic tumor(PAMT)is a rare mesenchymal tumor characterized by multiple nodular plexiform growth patterns and an immunophenotype with *** pathological characteristics,immunohistochemistry,diagnostic criteria,differential diagnosis,and gene-level changes of PAMT have been reported in many *** present,the main treatment for PAMT in the reported cases is surgery;only eight cases were treated via endoscopy(excluding 1 thoracoscopic resection),and the lesions were all smaller than 5 *** are no reports on the prognosis and follow-up of young patients with lesion sizes reaching 5 cm who undergo endoscopic submucosal dissection(ESD).Herein,we present the first case of a young patient with a lesion size reaching 5 cm who was diagnosed with PAMT via endoscopic submucosal *** SUMMARY A 15-year-old young man with upper abdominal pain for 2 years presented to the Gastroenterology Department of our *** gastroscopy showed a semicircular bulge approximately 5 cm in size in the lesser curvature near the cardia of the fundus;the surface was eroded,and shallow ulcers had *** pathological manifestations of the biopsy were spindle cell proliferative lesions with interstitial mucinous changes,and the surface mucosa showed chronic inflammatory changes with active lesions;immunohistochemistry showed smooth muscle actin(SMA)(+),CD117(-),CD34(-),DOG-1(-),S-100(-),and Ki67(LI:displayed typical manifestations,such as fat spindle-shaped fibroblasts and myofibroblast-like cells showing irregular nodular *** staining of the tumor cells revealed the following:CD34(partially+),SMA(weakly+),CD117(-),DOG-1(-),S-100(-),SDHB(+),PCK(-),and Ki67(labelling index:2%).There was no recurrence or metastasis during the 3-mo follow-up after the operation,and
