Hepatocellular carcinoma(HCC)is a liver cancer,highly heterogeneous both at the histopathological and molecular *** arises from hepatocytes as the result of the accumulation of numerous genomic alterations in various signaling pathways,including canonical WNT/β-catenin,AKT/mTOR,MAPK pathways as well as signaling associated with telomere maintenance,p53/cell cycle regulation,epigenetic modifiers,and oxidative *** role of WNT/β-catenin signaling in liver homeostasis and regeneration is well established,whereas in development and progression of HCC is extensively ***,we review recent advances in our understanding of how WNT/β-catenin signaling facilitates the HCC development,acquisition of stemness features,metastasis,and resistance to *** outline genetic and epigenetic alterations that lead to activated WNT/β-catenin signaling in *** discuss the pivotal roles of CTNNB1 mutations,aberrantly expressed non-coding RNAs and complexity of crosstalk between WNT/β-catenin signaling and other signaling pathways as challenging or advantageous aspects of therapy development and molecular stratification of HCC patients for treatment.

AIM:To assess the mucosa-associated bacterial microflora and mucus layer in adolescents with inflammatory bowel disease(IBD) .METHODS:Sixty-one adolescents(mean age 15 years,SD ± 4.13) were included in the *** biopsies from inflamed and non-inflamed mucosa of IBD patients and from controls with functional abdominal pain were cultured under aerobic and anaerobic *** number of microbes belonging to the same group was calculated per weight of collected *** mucus thickness in frozen samples was measured under a fluorescent ***:The ratios of different bacterial groups in inflamed and non-inflamed mucosa of IBD patients and controls were specific for particular *** *** predominant in the inflamed mucosa of Crohn's disease(CD) patients(80% of all bacteria) ,and Lactobacillus *** predominant in ulcerative colitis patients(90%) .The differences were statistically significant(P = 0.01-0.001) .Lower number of bifidobacteria was observed in the whole IBD group.A relation was also found between clinical and endoscopic severity and decreased numbers of Lactobacillus and,to a lesser extent,of Streptococcus in biopsies from CD *** mucus layer in the inflamed sites was significantly thinner as compared to controls(P = 0.0033) and to non-inflamed areas in IBD patients(P = 0.031) .CONCLUSION:The significantly thinner mucosa of IBD patients showed a predominance of some aerobes specific for particular diseases,their numbers decreased in relation to higher clinical and endoscopic activity of the disease.

配体构象应变能(ligand conformational strain energy,LCSE)是计算机辅助药物发现中考虑的一个重要参数.通过量子力学(quantum mechanical,QM)方法对自由态配体和蛋白质口袋中的结合态配体的结构进行计算并比较,可以获得配体构象应变能.然而,对配体构象应变能的合理范围以及计算方法仍然存在争议.选择8个柔性较大、特征结构模式多样的配体,这些结构的分辨率在高分辨率(0.86×10^(−10))与中等分辨率(3.10×10^(−10))之间,分别采用3种不同的量子力学方法:密度泛函(density functional theory,DFT)M062XD3、Hartree-Fock和GFN1-xTB,对8个配体进行逐步优化并计算配体能量,分析在结合态配体逐步优化到局域能量最小的自由态配体的过程中配体在结构和能量上的差异.量子力学计算结果显示,配体构象应变能小于6.0 kcal/mol,结合态配体结构中显著的几何参数误差是结合态和自由态配体结构的主要能量差异.因此,在分析配体构象应变能之前,对结合态配体结构进行精确实验测定非常重要.

The determination of accurate orthometric or normal heights remains one of the main challenges for the geodetic community in *** heights are required for geodetic and geodynamic scientific research as well as for extensive engineering *** main objective of this study is to estimate the geoid-to-quasi geoid separation(GQS)in Ethiopia(ETH-GQS).Such separation would be required for the conversion between geoid and quasigeoid models,which is mandatory for the determination of accurate geodetic heights in mountain *** airborne free-air gravity anomalies and the topo-graphic information retrieved from the SRTM3(Shuttle Radar Topography Mission of a spatial resolution 3 arc-second)digital elevation model were used to compute the ETH-GQS model according to the Sjoberg's strict formula for the geoid-to-quasigeoid *** ETH-GQS was then validated using GNSS-levelling data as well as geoid heights determined from different Global Geopotential Models(GGMs),namely the EGM2008,EIGEN-6C4 and *** results reveal that the standard deviation of differences between the geoid heights obtained from the EIGEN-6C4 model and the geometric geoid heights obtained from GNSS-levelling data were improved by~75%(***~24 to~6 cm)when considering GQS values obtained from the ETH-GQS.

Type 2 diabetes mellitus(T2DM)and sleep disorders(SD)have become important and costly health issues world-wide,particularly in *** are common diseases related to brain functional and structural abnormalities involving the hypothalamic-pituitary-adrenal(HPA)*** brains of individuals who suffer from both diseases simultaneously might be different compared to healthy *** review assessed current neuroimaging findings to develop alternative targeted treatments for T2DM and *** articles published between Jan-uary 2002 and September 2021 were searched in PubMed and Web of Science *** treatment methods for T2DM include dietary/weight-loss management,metformin or a combination of two non-insulin drugs,and melatonin for SD,though alternative therapies including electroacupuncture(EA)have been utilized in treating both of these diseases separately because they are convenient,affordable,and *** and al-ternative treatments for T2DM were somehow effective in treating *** studies of these disorders can achieve higher treatment efficacy by targeting brain areas,such as the hypothalamus(HYP),as visualized via diffusion tensor imaging(DTI),and functional magnetic resonance imaging(fMRI).DTI and fMRI can map the human brain and are utilized in many ***,we propose that neuroimaging studies could be used in treatment of SD in T2DM.

Perturbation finite element solution for chemo-elastic boundary value problems under chemical equilibrium

Modeling the elastic behavior of solids in energy conversion and storage devices such as fuel cells and lithium-ion batteries is usually difficult because of the nonlinear characteristics and the coupled chemo-mechanical behavior of these *** this work,a perturbation finite element(FE)formulation is developed to analyze chemo-elastic boundary value problems(BVPs)under chemical *** perturbation method is applied to the FE equations because of the nonlinearity in the chemical potential expression as a function of solute *** compositional expansion coefficient is used as the perturbation *** the perturbation expansion,a system of partial differential equations for the displacement and dimensionless solute concentration functions is obtained and solved in consecutive *** presence of a numerical solution enables modeling 3D chemo-elastic solids,such as battery electrodes or ionic gels,of any geometric shape with defects of different *** proposed method is tested in several numerical examples such as plates with circular or elliptical holes,and *** numerical examples showed how the shape of the defect can change the distribution of solute concentration around the *** in chemo-elastic solids create sharp peaks in solute concentration around crack tips,and the intensity of these peaks increases as the far field solute concentration decreases.

Defects in the endothelial cell barrier accompany diverse malfunctions of the central nervous system such as neurodegenerative diseases,stroke,traumatic brain injury,and systemic diseases such as sepsis,viral and bacterial infections,and *** endothelial sealing leads to leaking blood vessels,followed by vasogenic *** edema as the most common complication caused by stroke and traumatic brain injury is the leading cause of *** microvascular endothelial cells,together with astrocytes,pericytes,microglia,and neurons form a selective barrier,the so-called blood-brain barrier,which regulates the movement of molecules inside and outside of the *** that regulate blood-brain barrier permeability in health and disease are complex and not fully *** newly discovered molecules that are involved in the regulation of cellular processes in brain microvascular endothelial cells have been described in the literature in recent *** of these molecules that are highly expressed in brain microvascular endothelial cells is protocadherin gamma *** this review,we discuss recent evidence that protocadherin gamma C3 is a newly identified key player involved in the regulation of vascular barrier function.
