Identification of QTL and underlying genes for root system architecture associated with nitrate nutrition in hexaploid wheat

The root system architecture(RSA) of a crop has a profound effect on the uptake of nutrients and consequently the potential yield. However, little is known about the genetic basis of RSA and resource adaptive responses in wheat(Triticum aestivum L.). Here, a high-throughput germination paper-based plant phenotyping system was used to identify seedling traits in a wheat doubled haploid mapping population, Savannah×Rialto. Significant genotypic and nitrate-N treatment variation was found across the population for seedling traits with distinct trait grouping for root size-related traits and root distribution-related traits. Quantitative trait locus(QTL) analysis identified a total of 59 seedling trait QTLs. Across two nitrate treatments, 27 root QTLs were specific to the nitrate treatment. Transcriptomic analyses for one of the QTLs on chromosome 2 D, which was found under low nitrate conditions, revealed gene enrichment in N-related biological processes and 28 differentially expressed genes with possible involvement in a root angle response. Together, these findings provide genetic insight into root system architecture and plant adaptive responses to nitrate, as well as targets that could help improve N capture in wheat.

Effect of Stabilized Fish Oil Source on Sperm Quality and Production of Boars

Research findings for supplementing boar stud diets with fish oils are inconsistent. This study was designed to address three possible causes of performance variation of boars to fish oil supplementation: stability of the fatty acid source, level of inclusion and breed of boars tested. Three groups of 87 boars each, from two genetic lines (PIC 337 and PIC 800), were assigned to treatment based on age, mean sperm production (previous 12 weeks), and body condition score. All boars received a corn-soybean meal diet with a commercial fish oil supplement providing 1.83 g/boar/day of docosahexaenoic acid (DHA) as a preconditioning diet. On 10-Aug., 2020, the DHA source was changed to a stabilized starch imbedded source of refined fish oil (Salmate®), providing 1.83 g/b/d for the test diet. Two additional levels providing 2.38 and 2.94 g/b/d of DHA were fed for a 9 week pretreatment period and during the test period. Salmate® fed at 2.38 g/b/d of DHA resulted in a reduction in the number of rejected ejaculates (P 0.045) by 7.5% and 6.4% compared to the lowest and highest inclusion rates, respectively. ＜正＞ Bioreductive drugs such as CB 1954. nilutamide （anti - tumour） and nimesulide （NSAID） cause hepatotoxicity possibly due to activation by endogenous reductase （s）. The aim of this study was to investigate the relationship between bioactivationand hepatotoxicity using CB 1954 as a model compound. Drug administration in mice caused a dramatic increase in both alanine （ALT） and aspartate aminotransferase（AST） and in rats only a slight increase in AST. Histopathological examination of the livers revealed centrilobular hepatocyte injury in mouse but periportal（biliary） damage in rat. Aerobic incubation of CB 1954 with mouse or rat liver S9 resulted in formation of cytotoxic 2 - and 4 - nitroreduction metabolites, which were also seen in vivo. In conclusion, both mice and rats are susceptible to the hepatotoxicity of CB 1954. perhaps via different mechanisms, which may involve endogenous bioactivation. These models may be used to investigate potential host toxicity of other bioreductive drugs, some of which are under development in ACSRC.
