Efficient Numerical Optimization Algorithm Based on New Real-Coded Genetic Algorithm, AREX + JGG, and Application to the Inverse Problem in Systems Biology

In Systems Biology, system identification, which infers regulatory network in genetic system and metabolic pathways using experimentally observed time-course data, is one of the hottest issues. The efficient numerical optimization algorithm to estimate more than 100 real-coded parameters should be developed for this purpose. New real-coded genetic algorithm (RCGA), the combination of AREX (adaptive real-coded ensemble crossover) with JGG (just generation gap), have applied to the inference of genetic interactions involving more than 100 parameters related to the interactions with using experimentally observed time-course data. Compared with conventional RCGA, the combination of UNDX (unimodal normal distribution crossover) with MGG (minimal generation gap), new algorithm has shown the superiority with improving early convergence in the first stage of search and suppressing evolutionary stagnation in the last stage of search.

Ionic Liquid Assisted Extraction of Nitrogen and Sulphur-Containing Air Pollutants from Model Oil and Regeneration of the Spent Ionic Liquid

Removal of air pollutants, such as nitrogen and sulphur containing compounds from a model oil (dodecane) was studied. An ionic liquid (1-ethyl-3-methylimidazolium chloride [C2mim] [Cl]) was used as an extractant. Liquid-liquid extraction by using 1-ethyl-3-methylimidazolium chloride [C2mim] [Cl] was found to be a very promising method for the removal of N- and S-compounds. This was evaluated by using a model oil (dodecane) with indole as a neutral nitrogen compound and pyridine as a basic nitrogen compound. Dibenzothiophene (DBT) was used as a sulphur compound. An extraction capacity of up to 90 wt% was achieved for the model oil containing pyridine, while only 76 wt% of indole in the oil was extracted. The extraction capacity of a model sulphur compound DBT was found to be up to 99 wt%. Regeneration of the spent ionic liquid was carried out with toluene back-extraction. A 1:1 toluene-to-IL wt ratio was performed at room temperature. It was observed that, for the spent ionic liquid containing DBT as a model compound more than 85 wt% (corresponding 3852 mg/kg) could be removed from the oil. After the second regeneration cycle, 86 wt% of the DBT was recovered from the ionic liquid to toluene. In the case of indole as the nitrogen containing species, more than 99 wt%, (corresponding to 2993 mg/kg) of the original indole was transferred from the model oil to the ionic liquid. After the first-regeneration cycle of the spent ionic liquid, 54 wt% of the indole–in-IL was transferred to toluene. Thus, both extractions of nitrogen and sulphur model compounds were successfully carried out from model oil and the back-extraction of these compounds from the ionic liquids to toluene demonstrated the proved the concept of the regeneration point of view.

Amount of Calcium Elution and Eroded Lesion Depth in Bovine Enamel Derived from Single Short Time Immersion in Carbonated Soft Drink <i>in Vitro</i>

Erosion is one of serious oral health problems among Japanese children. Some dentists argue that brushing just after food/drink intake induces enamel abrasion. Objectives of this study were to evaluate amount of calcium elution from bovine enamel due to single and short immersion into carbonated soft drink, to calculate depth of the eroded lesion, and to determine when to brush teeth after carbonated soft drink intake from the view point of preventing enamel abrasion. Four enamel specimens were made from each of eight bovine teeth. The specimens were covered by quick-cure resin except for enamel surfaces. The four specimens from each bovine tooth were classified into three, six, nine, and 12 minutes immersion (IM3, IM6, IM9, and IM12) groups and immersed separately in five mL of carbonated soft drink. After the immersion, the calcium concentration of the original drink and the drink samples were evaluated using atomic absorption spectrophotometry. The dimension of each enamel specimen was calculated using a planimeter. The amount of eluted calcium per unit area of the enamel specimen into each drink sample was obtained. The depth of the demineralized lesion was obtained by dividing the amount of calcium elution per unit area by the concentration of calcium in enamel and the specific gravity of enamel. The lesion depth of the IM3 group was significantly lower than those in the IM6, IM9, and IM12 groups. The mean lesion depth in the IM12 group which showed the deepest lesion depth was 0.21 μm. As conclusions, the erosive lesion depth due to intake of carbonated soft drink was much shallower than remineralized enamel surface of a white spot lesion which can be repaired in plaque fluid in a short time, suggesting such erosion hardly causes tooth wear, hence it was suggested that brushing teeth just after the intake was recommended.

Evaluating the sensitivity of SARS-CoV-2 infection rates on college campuses to wastewater surveillance

As college campuses reopened in fall 2020,we saw a large-scale experiment unfold on the efficacy of various strategies to contain the SARS-CoV-2 *** individual surveillance testing via nasal swabs and/or saliva is among the measures that colleges are pursuing to reduce the spread of the virus on ***,some colleges are testing wastewater on their campuses for signs of infection,which can provide an early warning signal for campuses to locate COVID-positive ***,a representation of wastewater surveillance has not yet been incorporated into epidemiological models for college campuses,nor has the efficacy of wastewater screening been evaluated relative to traditional individual surveillance testing,within the structure of these ***,we implement a new model component for wastewater surveillance within an established epidemiological model for college *** use a hypothetical residential university to evaluate the efficacy of wastewater surveillance for maintaining low infection *** find that wastewater sampling with a 1-day lag to initiate individual screening tests,plus completing the subsequent tests within a 4-day period can keep overall infections within 5%of the infection rates seen with traditional individual surveillance *** results also indicate that wastewater surveillance can effectively reduce the number of false positive cases by identifying subpopulations for surveillance testing where infectious individuals are more likely to be *** a Monte Carlo risk analysis,we find that surveillance testing that relies solely on wastewater sampling can be fragile against scenarios with high viral reproductive numbers and high rates of infection of campus community members by outside *** results point to the practical importance of additional surveillance measures to limit the spread of the virus on campus and the necessity of a proactive response to the initial signs of outbreak.

Correlation between Leukocyte Numbers and Body Size of Rainbow Trout

Immune cells in rainbow trout Oncorhynchus mykiss comprise granulocytes (neutrophils, basophils and eosinophils), macrophages/monocytes and lymphocytes (B- and T-cells). These cellular elements occur early during the ontogenetic development of trout and allow both innate and adaptive responses towards an antigen to be initiated even in fry. The number of leukocytes in individual fish at different developmental stages is likely to influence the capacity of the fish to respond simultaneously to several antigens (pathogens and vaccine components). This parameter may therefore be crucial for both wild and cultured fish and we show that the size of the leukocyte population increase exponentially with body size of rainbow trout. Four groups (5 fish/ group) of naive rainbow trout (Oncorhynchus mykiss) with a mean body weight of 2-4 g (group I), 4-6 g (group II), 25-30 g (group III), and 650-780 g (group IV) were investigated. The number of lymphocytes was generally higher in head kidney compared to blood and spleen but they dominated in all samples (blood, head kidney, and spleen) and their numbers increased exponentially with fish size. Percentages of lymphocytes in relation to neutrophils and macrophages were higher in spleen (98%-99%) compared to blood and head kidney in all groups. Fish fry is therefore equipped to respond specifically against one or a few vaccine antigens but the capacity to raise protective responses against a repertoire of pathogens may be limited until the larger fingerling stage has been reached. The implications for vaccination of early fry are discussed.

Background We were interested in determining how the tumor suppressor gene RBM5 is regulated in lung cancers. Previous studies suggested that the gene expression is related to histological subtype and smoking exposure, since in small cell lung cancers the RBM5 gene is deleted whereas in non-small cell lung carcinomas （NSCLC） RBM5 expression is reduced. Of particular interest was the recent finding that in lung adenocarcinomas, a histological subtype of NSCLC, smoking exposure correlated with mutational activity in the transforming growth factor alpha （TGF-c~） signaling pathway. Lung adenocarcinomas from smokers were associated with activating KRAS mutations, whereas lung adenocarcinomas from never-smokers were associated with activating epidermal growth factor receptor （EGFR） mutations. We hypothesized that inhibition of RBM5 in lung adenocarcinomas is achieved indirectly via these activating mutations. The objective of the research described herein was to determine if EGFR activation and RBM5 expression are negatively correlated. Methods EGFR expression in the lung adenocarcinoma cell line NCI-H1975 was inhibited using small interfering RNA. RBM5 expression was examined by real-time quantitative polymerase chain reaction and Western blotting. Results Reduced EGFR expression did not correlate with any change in RBM5 expression at either the RNA or protein level. Conclusion These results suggest that RBM5 expression is not directly regulated by EGFR in non-smoker related lung adenocarinomas, and that some other mechanism operates to inhibit either the expression or function of this potential tumour suppressor in lung cancers that retain the RBM5 gene.

Cerebral Microbleeds Identified by Susceptibility-Weighted Imaging in Two Cases of Fabry Disease without Neurological Symptoms

Cerebrovascular disease is one of the fatal causes of Fabry disease (FD). Brain magnetic resonance imaging findings typically show lacunar infarcts in young patients with FD, but brain hemorrhages in FD are rarely reported. We report two cases of FD focusing on cerebral microbleeds (CMBs). Susceptibility-weighted imaging (SWI) and T2*-weighted imaging reveal several lobar and deep CMBs in two patients with no medical history of stroke symptoms, hypertension, and anticoagulant/antiplatelet treatment. SWI can detect a greater number of CMBs than T2*-weighted imaging. Thus, SWI is an excellent tool for identifying underlying CMBs in FD.
